Cryo-grinding Characteristics of Frozen Fish Meat at Different Temperature (동결온도에 따른 어육의 동결마쇄특성)

  • Cryo-grinding process comprising hammer mill-shattering and colloid mill-grinding without refrigerant was applied to sardine, pollack and squid muscle frozen at $-20^{\circ}C,\;-40^{\circ}C\;and\;-80^{\circ}C$, respectively and its characteristics were investigated. Particle size distribution of shattered product was shown larger in the order of squid, sardine and pollack and particle size of shattered product frozen at $-80^{\circ}C$ was shown smaller than those at $-40^{\circ}C$. Image of shattered product depended on freezing temperatures and fish species, suggesting particle size distribution of rheological properties can be dependent on fish species or freezing temperature. Yield of cryo- grinded product was in the range of $52.5\~62.5\%$ and Ca content of sardine or pollack product was $6\~8$ times higher than its fillet. Emulsion capacity of cryogrinded product was not decreased during processing. Therefore, this method is thought to be applicable to fish precessing, and preparation of fish paste or powaer.

  • We selected horse mackerel Trachurus japonicus, mackerel Scomber japonicus and arrow squid Todarodes pacificus, to be used in the experiment. These fishes migrate to the Cheju coast and were caught by set-net. We studied on the attracting effect and the leading effect on the fish school using incandescent lights in an outdoor water tank at the Marine Research Institute Cheju National University. The results obtained are summarized as follows; 1. The horse mackerels, mackerels and arrow squids were actively attracted to the incandescent light. The optimum illumination and the reactions of each fish school were different though. 2. We turned the first attracting lamp on and turned it off after ten minutes. Then we turned the second attracting lamp on and turned it off after another ten minutes. The reaction of each fish school was a little bit different, but it worked to lead them to their destination. 3. We turned the three attracting lamps on at the same tune. After ten minutes, We turned the first attracting lamp off. After another ten minutes we turned the second attracting lamp off. The reaction of each fish school was a bit different, but it worked to lead them to their destination. 4. One incandescent light was moving. The reaction of each fish school was a little bit different, but it worked to lead them to their destination.

Immuno-Electron Microscopic Studies on the Distribution of Dopamine and $Calbindin-D_{28K}$ in the Optic lobes of Cephalopods (Todarodes pacificus and Octopus minor) inhabiting the Korean waters (한국 연근해산 두족류 (Todarodes pacificus and Octopus minor) 시엽내 Dopamine 및 $Calbindin-D_{28K}$의 분포에 관한 면역전자현미경적 연구)

  • In this study, we carried out immunostaining and immunogold labeling with rabbit anti-dopamine (TH) and rabbit anti-calbindin-$D_{28K}$ to examine the characteristics and functions of the neurons that secrete neurotransmitters in optic lobes of Todarodes pacificus and Octopus minor inhabiting the Korean waters. The obtained results are as follow. In the immunostaining with anti-dopamine, only a few of the large amacrine cells in an the upper part of an outer granule cell layer and the cells forming the islands of medulla showed positive reaction in Todarodes pacificus, while $2{\sim}3$ cells in the upper and middle parts of an outer granule cell layer and more than 5 cells in the islands of medulla reacted positively in Octopus minor. For the case of anti-calbindin case, $2{\sim}3$ small amacrine cells in the upper portion of the outer granule cell layer and $1{\sim}2$ cells which are located in the lower part of an inner granule cell layer showed positive reaction in Todarodes pacificus, while, in Octopus minor, 4 cells in the outer granule cell layer reacted positively, no immunoreactive cell being found in the inner granule cell layer. As a result of performing the immunogold labeling, relative large number ($17{\sim}26$) of gold particles were labeled per $0.5{\mu}m^2$ of the cytoplasm of the cells which showed the immunoreactivity to the anti-dopamine and anti-calbindin in Todarodes pacificus, however, small number (10) of gold particles were labeled in Octopus minor, which reach only half of the number in the Todarodes pacificus.

Recovery of High Unsaturated Fatty Acid from Squid Processing Wastes using Supercritical Carbon Dioxide Extraction Method (초임계 이산화탄소 추출법을 이용한 오징어 가공 부산물로부터 고도불포화 지방산 회수)

  • A squid viscera oil contains a high content of EPA, DHA, and other valuable polyunsaturated fatty acids. The extractions of squid viscera oil by supercritical carbon dioxide both with/without $3\%$ (v/v) ethanol were performed in a semicontinuous flow extractor at 8.3 to 13.8 MPa and 25 to $50^{\circ}C$. When ethanol was added to $SC-CO_2$, the extraction ratio of lipid increased. The extracts contained high content of unsaturated oils like DHA and EPA. The highest extraction yield of lipid from squid viscera oil extracted by supercritical carbon dioxide was obtained at 12.4 MPa and $40^{\circ}C$ with/without entrainer. The main fatty acids of squid viscera oil extracted by supercritical carbon dioxide were myristric acid (14:0), palmitic acid (16:0), palmitoleic acid (16:1), oleic acid (18:1), arachidic acid (20:0), eicosapentaenoic acid (20:5), and docosahaxaenoic acid (22:6).

Composition and Catch Variation of Fishes by a Set Net in the Coastal Waters off Gwideuk, Jeju Island (제주도 귀덕 연안 정치망 어획물의 조성 및 변동)

  • Fishes by a set net in the coastal waters off Gwideuk, Jeju Island were studied to determine species composition and catch variation, 2004. The fishing period conducted by a set net in Gwideuk fishing ground were from April to December. A total of 45,473.2 kg including 21 species was caught during the survey period. Most of them consisted of fishes of 17 species and a few were cephalopods of 4 species. Dominant species were Trachurus japonicus and Siganus fuscescens, 44.3% in the total. And sub-dominant species were Sepioteuthis lessoniana, Seriola dumerili, Seriola quinqueradiata, Loligo bleekeri, Seriola lalandi accounting for 54.4%. Although the number of species had a tendency to decreased from July to December, the catch by a set net was higher in summer and autumn than in spring. Such catch variations were closely related to recruitment of dominant species and also the seasonal variation of fishing ground temperature.

Effect of High Pressure Processing on the Shelf Life of Seasoned Squid (초고압 가공이 조미오징어의 저장성에 미치는 영향)

  • This study was designed to evaluate the potential of using high pressure processing (HPP) for extending shelf life of seasoned squid during refrigerated storage. The vacuum-packed seasoned squid samples were subjected to 400 MPa for 20 min using a custom-made high pressure processor. Microbial counts, dimethylamine (DMA), trimethylamine (TMA), total biogenic amine, autolytic activity were determined on days 0, 7, 14, and 21 of refrigerated storage. The numbers of indigenous bacteria were effectively reduced by 2.77 log CFU/g after HPP treatment. The amounts of DMA and TMA produced in the control samples increased up to 15.99 and 42.82 mg/g after 7 days of refrigerated storage when compared to 5.27 and 10.21 mg/g the HPP-treated samples, respectively. The autolytic activity of the HPP-treated sample (4.32 nkat/g) significantly lower than that of the control (7.13 nkat/g) after 7 days of refrigerated storage. Therefore, HPP can be applied as a potential squid processing method microbiological safety and shelf life.

Water Properties of Electrolytic Machine by Stainless Diaphragm and Effects of Electrolytic Ice Water Storage For Keeping Freshness of Squid, Todarodes pacificus (스테인레스 극판을 이용한 전해수장치의 수질특성과 오징어 선도유지를 위한 전해수 빙장 효과)

  • This study was undertaken to prolonging the freshness in Squid(Todarodes pacificus) on the effects of ice storage methods(tap water ice, electrolytic water ice) using stainless diaphragm electrolytic instrument and also test an efficiency of instrument. Basically, stainless diaphragm electrolytic instrument studied for changes of pH on difference water flow and ampere. The lower water flower and higher ampere made low pH on acid part of electrolytic instrument. Squid samples were stored in tap water ice, acid part of electrolytic water and base part of electrolytic water and used in studying the changes of VBN and skin color through storage. Acid water had strong sterilization effects and VBN was lower levels of acid water than the others. Base water had not sterilization effects. Tap water ice storage was more sterilizing effects than base water ice storage but less effects than acid water ice storage. Lightness of Squid skin showed getting decrease at storage of ice water of all storage methods but acid water ice storage showed more retarding than the other two storage. From these results, it could be suggested that acid electric water ice storage is effective in extendance the shelf-life of squid at chilled storage.

Effects of Diet Supplemented with Squid Intestine on Growth and Body Composition of the Catfish (Parasilurus asotus) (오징어내장 첨가사료가 메기 (Parasilurus asotus)의 성장 및 육조성에 미치는 영향)

  • In order to find out the effect of supplemented diets with squid intestine on the catfish, growth and body compositions of catfish (Parasilurus asotus) were investigated for 112 days with the four experimental diets of different lipid levels. The higher lipid level of diets with squid intestine supplement was fed, the higher body weight was gained (p<0.05). HSI (Hepatosomatic Index) was the lowest in the diets with $10\%$ squid intestine. There were no significant differences in muscle md skin compositions, however the increasing lipid levels in the diets well reflected in the fatty acid pattern of skin and muscle. The percentage of n-3 polyunsaturated fatty acids such as C20 : 5 and C22 : 6 in tissues was increased according to increase of supplemented squid intestine. Saturated and n-6 fatty acid contents did not show the significant difference.

Distribution of Common Squid Todarodes pacificus Larvae in the Southwestern Part of the East Sea in Summer and Autumn, 2015 (2015년 하계 및 추계 동해 남서해역에서 출현하는 살오징어 (Todarodes pacificus) 유생의 분포 양상)

  • To determine the horizontal and temporal distribution of common squid larvae, Todarodes pacificus (hereafter T. pacificus), we conducted surveys using an IKMT net (mesh size: $500{\mu}m$) with a Fisheries Research Vessel (FRV, TAMGU 21) in the southwestern part of the East Sea in summer (August and September) and autumn (November) 2015. A total of 228 larvae, ranging in mantle length (ML) from 1.4 mm to 21.9 mm, were collected at 35 stations over the research period. The monthly average mantle length of T. pacificus larvae did not differ significantly in August, September and November. (p > 0.05). Catch densities at positive stations ranged between 0.1 and $7.9inds./1,000m^3$ over the research period. Incidence rates of T. pacificus larvae were similar over three months, in the study area (62.9 % - 68.6 %). The 4 - 5 mm mantle length range had the highest frequency in size-frequency distributions for T. pacificus larvae. The larval survival temperature ($15-24^{\circ}C$) at positive stations for catch densities was located below a 20 m depth in August whereas it was located at the surface of the water in September and November. The survival temperature for larvae existed from the bottom to the surface of the water where larvae were sampled larvae in shallow sea areas. However, the larval survival temperature occurred in a shallower location than the upper layer of the thermocline in deep sea areas at a depth below 100 m.

Effects of glycine on microbial safety of low-salted squid and myungran jeotgal (글리신을 활용한 저염 오징어 및 명란 젓갈의 미생물 안전성 확보)

  • Seven antimicrobial agents known to be effective in inhibiting the growth of lactic acid bacteria were applied to ensure the microbial safety of low-salted squid and myungran jeotgal with 4-6% salinity. These agents reduced the salt content by 50% compared with the conventional Jeotgal. Lactic acid bacteria such as Lactobacillus sp., Streptococcus sp., and Pediococcus sp. were commonly found to account for 80% of microbial organisms, and yeast and fungi were observed in squid and myungran jeotgal, respectively. The total bacterial counts in squid and myungran jeotgal showed 94.20 and 90.87% reduction after the addition of 0.5% (w/w) glycine. The microbial counts in squid and myungran jeotgal decreased $10^1-10^2CFU/g$ when compared with the control after 21 days at $10^{\circ}C$. Glycine was found to be an effective commercial antimicrobial agent that can be used to control bacterial count in low-salted Jeotgal without affecting sensory qualities such as overall taste and flavor.
