Impacts of Contaminated Water Outflow from the Lake Sihwa on the Meiobenthic Animals Living in the Coastal Zones of the Kyonggi Bay (시화호 오염수 방류에 따른 중형저서동물의 군집변동)

  • To alleviate the harmful effects from contamination, the Sihwa Lake waters have been regularly fluxed out into the ocean to mix and circulate with the oceanic waters, and then allowed to flow back into the lake. In the present study, eco-environmental impacts by the outflowing contaminated lake waters to the benthic communities of the coastal zones along Kyonggi Bay were examined at three separate areas. A total of 22 groups of meiobenthic animals were commonly found in the subtidal zones of the Bay. All groups of the meiobenthic animals on the 1st and 7th days after the outflow were remarkably reduced in numbers from the selected areas examined, as compared to those observed one day before the outflow. It was also found that the community structure of meiobenthos was changed drastically: At the station near the water gate, for example, where the immediate outflow of the lake water was encountered, the benthic harpacticoids, the group observed to be one of the predominant groups before the outflow, were found to have disappeared completely. For an area that was relatively far away from the water gate, the reduction rate of meiobenthos after the outflow events was slower and more gradual than the nearer sites. An area that is the farthest from the water gate, no reduction in any of the meiobenthos group was observed. The ratio between nematodes/copepods (N/C) was remarkably changed by the outflows at the station near the water gate: One day before the outflow, the ratio was 0.7 while the ratio was 19.0 at the station near the water gate. In contrast, at the stations relatively far from the water gate, the ratio decreased one day after the outflow event, but increasing trends were observed one week later. In the tidal flats, Station 1 exhibited the common observed regional values of the ratio whereas Station 2 showed a very high ratio of 191.3. The results suggested that the contaminated lake water outflows directly effect meiobenthos in a relatively short time period and thus the meiobenthos may be a good indicator animal group for examining the effect of pollution. Replacement of Saline Water through Injecting Fresh Water into a Confined Saline Aquifer at the Nakdong River Delta Area (염수로 충진된 낙동강 델타지역 피압대수층에서 담수주입에 의한 염수치환 연구)

  • We performed injection tests in a deep-seated confined aquifer to assess the potential of artificial recharge as a means of preventing saltwater contamination, thereby securing groundwater resources for the Nakdong Delta area of Busan City, Korea. The study area comprises a confined aquifer, in which a 10-21-m-thick clay layer overlies 31.5-36.5 m of sand and a 2.8-11-m-thick layer of gravel. EC logging of five monitoring wells yielded a value of 7-44 mS/cm, with the transition between saline and fresh water occurring at a depth of 15-38 m. Above 5 m depth, water temperature is 10-15.5℃, whereas between 5 and 50 m depth the temperature is 15.5-17℃. Approximately 950 m3 of fresh water was injected into the OW-5 injection well at a rate of 370 m3/day for 62 hours, after which the fresh water zone was detected by a CTD Diver installed at a depth of 40 m. The persistence of the fresh water zone was determined via EC and temperature logging at 24 hours after injection, and again 21 days after injection. We observed a second fresh water zone in the OW-2 well, where the first injection test was performed more than 20 days before the second injection test. The contact between fresh and saline water in the injection well is represented by a sharp boundary rather than a transitional boundary. We conclude that the injected fresh water occupied a specific space and served to maintain the original water quality throughout the observation period. Moreover, we suggest that artificial recharge via long-term injection could help secure a new alternative water resource in this saline coastal aquifer.

Effect of Trace Metal on Accumulation and Physiological Response of the Polychaete, Perinereis nuntia (미량금속 노출에 따른 갯지렁이(Perinereis nuntia)의 미량금속 축적 및 생리학적인 반응)

  • Metal exposure experiments using polychaete (Perinereis nuntia) as a bio-indicator of trace metals contamination were conducted to evaluate the bioaccumulation and the biomarkers responses such as metallothionein-like protein (MTLPs) and glutathione S-transferase (GST) which was simultaneously exposed to Cadmium (Cd) and Copper (Cu). Cu and Cd concentrations in polychaete were enhanced with increasing exposure time and their concentrations of aqueous medium. Initial accumulation of Cd was higher than that of Cu. Our results showed that the bioaccumulation of Cu and Cd were prohibited, especially at higher Cu levels, suggesting the different cellular uptake mechanisms when Cu and Cd are co-exist. Net accumulation rate of Cu was declined with exposure time but it did not show any significant change for Cd. Although the highest MTLPs concentration was observed at 6 hr of exposure time, it did not show any significant change related to exposure times and metals concentrations. An increase of GST activity tended to increase as a function of exposure time and metals concentrations. And GST activities in P. nuntia have similar tendency with bioconcentration factors in high concentration of Cu (treatment group IV) at post 24 h of exposure. Our results provide new information of the bioaccumulation and biomarker responses to understand the effects of co-existing contaminants (Cu and Cd) using polychaete. Further studies are required to elucidate the bioaccumulation and biomarkers responses for various contaminants.

Monitoring of the residues of some pesticides in mulberry leaves and their safety evaluation (뽕잎중 잔류농약의 monitoring 및 안전성 평가)

  • In order to evaluate the safety of mulberry leaves for sericultural purpose to the contamination by pesticide residues, the analyses of dichlorvos, acephate, phenthoate, pyrazophos, deltamethrin, and cypermethrin were done for 24 samples collected in spring and fall over two consecutive years (1996 and 1997), each, from 8 provinces across the country, by selecting three representative farmhouses in each province. In spring samples of 1996, the residual amounts of dichlorvos, acephate, and phenthoate were $0.018{\sim}0.032$, $0.013{\sim}0.072$, and 0.051 ppm, respectively, whereas, pyrazophos was not detected, and the detection frequencies were 29, 58, and 4%, respectively. In fall samples, on the other hand, those of the above pesticides were $0.012{\sim}0.048$, $0.020{\sim}0.156$, and $0.018{\sim}0.050$ ppm, respectively, and pyrazophos was not detected either and the detection frequencies were 42, 17, and 13%, respectively. While in spring samples of 1997, the residual amounts of dichlorvos, acephate, and cypermethrin were $0.014{\sim}0.064$, $0.033{\sim}0.061$, and $0.019{\sim}0.068$ ppm, respectively, and deltamethrin was not detected. The detection frequencies were 75, 13, and 8%, respectively. In fall samples, on the other hand, those of the above pesticides were $0.013{\sim}0.062$, 0.015, and $0.009{\sim}0.013$ ppm, respectively, and deltamethrin was not detected either. The detection frequencies were 88, 4, and 17%, respectively. Almost all of the samples turned out to be non-toxic to silkworms, except that one sample (No. 10, spring 1997) contaminated by a high concentration of cypermethrin (0.068 ppm) was judged to do harm to silkworms. In the GC analyses, the selection of columns, the change in column temperature, and the use of MSD made the separation and identification of the vague compounds possible.

Biological Hazard Analysis of Leaf Vegetables and Fruits According to Types of Cultivation and Distribution Systems (엽채류 및 과채류의 재배유형 및 유통경로별 생물학적 위해요소 조사)

  • As the consumption of environmentally friendly agricultural products increases, food safety is at the forefront of public health concerns. We analyzed the biological hazards of 26 species of leaf vegetables and 4 species of fruit according to types of cultivation (conventional or organic filming) and distribution system (giant retailers or organic food stores) using various culture media, automatic bacterial identification systems, and microscopy, Total bacterial count of unwashed agricultural product ranged from $5.2{\times}10^{3}\;to\;1.5{\times}10^{5}\;CFU/mL$ (from 0.1 g of agricultural products), and the average count dropped 25-fold (range, 8-60-fold) after water washing. Microbial levels of washed organic agricultural products were $6.0{\times}10^{2}-1.9{\times}10^{4}\;CFU/mL$, and were not significantly different f개m the microbial loads on conventionally farmed produce. There was no significant difference in bacterial count from agricultural produce purchased from giant retailers or organic food stores. Total microbial count of Chinese cabbage, welsh onion, red chicory and kale were comparatively high, and Enterobacter cloacae was isolated most frequently. Parasites were detected in agricultural products purchased from conventional farm products in the stores of giant retailers, and in organic food stores, and parasite prevalence was especially high in Chinese cabbages and welsh onion. The study indicated that cultivation methods and distribution systems did not cause significant differences in biological contamination levels of agricultural produce. Some vegetables and fruits were highly contaminated effective sanitizing methods to reduce these biological hazards are needed.

Preliminary Diagnosis of Fishing Ground Environment for Establishing the Management System in Fisheries Resources Protection Area (수산자원보호구역 관리체제 구축을 위한 어장환경 예비진단)

  • For preliminary diagnosis on current fishing ground environment and basic information for establishment of effective and rational management policy in fisheries resources protection area, water and sediment quality and changes of total area in the 10 marine protection areas designated for fisheries resources management in Korea were assessed. Results showed that environmental quality in these areas has been degraded by pollution sources, coastal utilization and development stress, etc. The pattern and degree of contamination differed by protection areas, suggesting that it is necessary for optimum environmental management plan considering the regional characteristics. The total designated area of protection areas in 2003 changed by $-22.9{\sim}2.4%$, on average -6.4%, compared with the first year of designation; Wando-Doam Bay showd the highest increase rate (2.4%), and Hansan Bay has the highest decrease rate (-22.9%) Decrease rate of land and sea in total area showd 6.1% and 6.6%. An integrated management of environmental data in protection areas is required for systematic assessment. Therefore, the suitable environmental and information management is needed specifically considering the environment characteristics such as development and utilization conditions of land and sea area Furthermore, bemuse urbanization and industrialization threats the junctions of the protection areas, authorized ministry (MIFAFF) should develope and establish monitoring and management procedures based on the related laws.

Geochemical and Isotopic Study of the Onjeongri Granite in the Northern Gyeongsang Basin, Korea : Comparison with Cretaceous to Tertiary Granitic Rocks in the Other Part of the Gyeongsang Basin and the Inner Zone of Southwest Japan (경상분지 북부에 분포하는 온정리 화강암에 대한 암석화학적, 동위원소 지구화학적 연구 : 경상분지 다른 지역과 서남 일본 내대에 분포하는 백악기-제 3기 화강암류와의 비교 고찰)

  • We analyzed geochemical and radiogenic isotope data to investigate the genesis and source characteristics of the Onjeongri granite in the northern part of the Gyeongsang Basin. Field observation and K-Ar ages confirm late Cretaceous intrusion (ca. 87 Ma) of the Onjeongri granite. The hornblende geobarometery gives less than 2 kbar for the emplacement pressure of the Onjeongri granite. Geochemical and isotopic compositions suggest that the Onjeongri granite was formed in a relatively immature arc system. $SiO_2$ contents show a negative linear relationship with initial $^{87}Sr/^{86}Sr$ ratios, and an apparent positive correlation with $^{207}Pb/^{204}Pb$ ratios, suggesting an incomplete mixing or assimilation. However, the isotopic data known for any exposed rocks of the study area do not fit as an endmember, implying that the contaminant might reside in the lower crust. A review of published isotopic ages, geochemical, and Sr and Nd isotopic data for the Cretaceous to Tertiary granites in the Gyeongsang Basin indicates the followings. 1) Granitic magmatism in the Gyeongsang Basin were episodic. 2) Granitic rocks in the basin were derived from young (< 0.9 Ga) lower crust, and their isotopic signatures reflect heterogeneous source region. Geochemical and isotopic signatures of granitic rocks in the basin are difficult to explain by upper crustal contamination. 3) Granites in the Gyeongsang Basin have closely related to those in the San in Belt of the Inner Zone of Southwest Japan in terms of age, petrography, and isotopic and geochemical composition. 4) Sr-Nd isotopic signatures of the Onjeongri granite are relatively primitive compared with granitic rocks in the other parts of the Gyeongsang Basin and in the Inner Zone of Southwest Japan.

Changes in Various Quality Characteristics of Short-ripened Salami During Storage at Chilled or Room Temperatures (단기숙성 살라미의 냉장 및 실온저장 중 품질 특성 변화)

  • This study investigated the microbial contamination levels of raw meats used for short-ripened salami and changes in the microbial and physico-chemical properties of the product during storage at 10 and $25^{\circ}C$ for up to 120 days. The microbial counts of raw meats ranged between 2 and 4 Log CFU/g. Frozen-thawed sow meat showed higher total aerobe and Enterobacteriaceae counts than fresh chilled pork and pork back fat. Staphylococcus aureus was found in all raw materials except fresh chilled pork samples, and Clostridium perfringens was detected in a sample stored for 21 days at $25^{\circ}C$. The counts of total aerobes, lactic acid bacteria and Staphylococcus spp. decreased more rapidly at $25^{\circ}C$ than at $10^{\circ}C$ when the storage time was extended. The growth of Enterobacteriaceae, Pseudomonas spp., Clostridium spp., yeast, and mold were restricted to levels below 2 Log CFU/g during storage. The contents of salt, water, crude protein, crude fat, and ash of salami samples were 3.4, 33.4, 30.8, 32.7, and 4.3%, respectively, which were not affected by storage time or temperature. The pH value of the salami was initially 4.79 and increased to 5.02 and 5.26 after 120 days of storage at 10 and $25^{\circ}C$, respectively, whereas the water activity values decreased from an initial value of 0.91 to 0.90 and 0.88 after 120 days at 10 and $25^{\circ}C$, respectively. The TBA and VBN values increased slowly during storage. The redness value of the salami samples stored at $25^{\circ}C$ decreased more significantly than the samples stored at $10^{\circ}C$. With increased storage time, the values for the rheological characteristics of the salami in terms of hardness, brittleness, elasticity, cohesiveness, gumminess, and adhesiveness tended to decrease more remarkably at $25^{\circ}C$ than at $10^{\circ}C$. Based on sensory evaluation scores, it appears that short-ripened salami is no longer acceptable after 90 days at $10^{\circ}C$ and 30 days at $25^{\circ}C$.

THE EFFECTS OF DRYING AGENTS AND BONDING AGENTS ON THE SHEAR BOND STRENGTH OF SEALANTS TO ENAMEL (치면건조제와 접착제의 사용에 따른 치면열구전색재의 전단결합강도에 관한 연구)

  • The application of sealants is a highly technique-sensitive procedure, requiring an extremely dry field prior to placement. Moisture contamination of the etched enamel surface before sealant placement is cited as the main reason for sealant failure. The purpose of this study was to evaluate the effects of different methods of sealant application on the shear bond strength of sealants to enamel. In groups 1, 2, 3, 4 Teethmate(unfilled sealant) was used, while Ultraseal XTplus(filled sealant) was used in groups 5, 6, 7, 8. Groups 1 and 5(control) were acid etched for 15 seconds using 35% phosphoric acid, washed and then dried. In groups 2, 6 drying agents were applied, and in groups 3, 7 bonding agents were applied and light cured. In groups 4 and 8 both drying agent and bonding agent were applied. Then sealant was cured to the specimen using molds 3mm in diameter and 2mm in height. Thermocycling was performed and shear bond strength was finally measured. The following results were obtained : 1. Groups using filled sealant(groups 5, 6, 7, 8) showed higher shear bond strengths compared to groups using unfilled sealant(groups 1, 2, 3, 4). 2. Among groups using unfilled sealant(groups 1, 2, 3, 4), groups 2, 3, 4 showed significantly higher shear bond strength compared to group 1(p<0.05). There were no significant differences among groups 2, 3 and 4. 3. There were no significant differences(p>0.05) among groups using filled sealant(groups 5, 6, 7, 8). 4. When modes of fracture were examined, cohesive failure was observed in groups 2, 3 and 4.

A Survey of Staphylococcus aureus Contamination and Antibiotic Susceptibility in Retail Meat (국내유통 축산물에서의 Staphylococcus aureus 오염도 및 항생제 감수성 조사)

  • Staphylococcus aureus is a pathogene of major concern in livestock products. This study was conducted to test imported and domestic meat sold by retail stores for the presence S. aureus. In addition, the antibiotic susceptibility of any S. aureus found was also evaluated. The overall isolation rate of S. aureus was 20.2% (13.9% in pork and 33.8% in beef) in retail meats. The percentage of imported meats found to contain S. aureus (33.3% in pork and 40.4% in beef) was higher than that of domestic meat (13.0% in pork and 14.7% in beef). In addition, the detection rate of S. aureus was higher in raw material meat than in ready to cook packaged meat. When the antibiotic susceptibility of S. aureus isolated from the meat products was evaluated, ampicillin was found to be the highest (76.5%), followed by penicillin (75.3%), tetracycline (27.1%) and erythromycin (21.2%). Penicillin and tetracycline resistant were detected in 55.6% and 13.3% of the beef isolates, respectively, and 97.5% and 42.5% of the pork isolates, respectively. The tetracycline and erythromycin resistant plasmids of the isolated strain were transferred into S. aureus DPRMM2429 by the filter mating method and the frequencies of transfer was found to be $1.1{\times}10^{-5}{\sim}1.9{\times}10^{-9}$ and $1.2{\times}10^{-5}{\sim}4.0{\times}10^{-8}$ respectively.
