Antimicrobial Effects of the Extract of Galla rhois on the Long-term Swine Clinical Trial (양돈 임상 연구에 의한 천연물 오배자 추출물의 항균.항바이러스 효과 규명)

  • We are aim to evaluate antimicrobial effects of the extract of Galla rhois (GR) on the health status and performance of growing and finishing pigs. This study was conducted on the growing and finishing pigs (n = 200) for 130 days in a swine husbandry. The animals were divided with two groups; GR treated group (n = 100) and commercial diet feeding group (n = 100). GR treated animals had provided with commercial diet adding the extract of GR as 0.2%. During the study period, we compared clinical signs, weight increase rate, diet consumption amount, fecal scores, gross findings, necropsy findings, histopathological findings between the treated group and non treated group. After necropsy, bacteria isolation and PCR analysis were conducted with the clinical samples. As the results of this long-term clinical trial, GR showed the antimicrobial effects on respiratory disease and diarrhea. We were identified that GR had the anti-bacterial and anti-viral effects.

Gall formation on different age, habitat, and parasite position in Rhus javanica L. (붉나무의 수령, 서식지 및 기생위치에 따른 오배자 형성)

  • This study was conducted to investigate the ratio and number of the Rhus javanica gall formed by chinese sumac aphid, Scheleechtendalia chinensis, in several different conditions in which R. javanica lives. The density of 11 to 15-year-old R. javanica was more higher and the number of galls was also higher than 1 to 5- or over 16-year-old one at these sites. The formed gall according to four directions such as the East, West, South and North was founded the highest at the East and followed South, North, and West order. The formative rate of R. javanica gall did not differ in the mixture forest, and in the habitat having a heap of a little stone and mosses near a valley was better than in the site of road perimeter. Through an index of the gall diameter and the number of chinese sumac aphid, we evaluated the gall processing and development formed by the aphid. As the density of aphids increase, the size of the gall was also more plumped. The part of phyllotaxy of R. javanica gall in leaf and pinnate of was increased in the middle and upper part of terminal leaflet and leaflet was more occupied than wing. R. javanica which did not form a gall in natural environmental, formed gall by a artificial inoculation of S. chinensis. Some gall disappeared during a gall development from various environmental conditions.

  • These studies were conducted to carry out mechanism and site of gall formation by Schlectendalia chinensis in Rhus javanica. The formation period of gall hy occurrence of fundatrix was middle of June, and fundatrix was settled and parasited on back side of immature leaves and tissues of Rhus javanica was ride to fore face, then formed Schlectendalia chinensis gall. Scl~lectendalia chinensis was suddenly developed in middle of Auguest. As the density of Schlectertdalia chinensb increases, the Schlectendalia chinensir gall was more plumped. The part of phyllotaxy of Schlectendalia chinensis gall in leaf and pinnate of was increased in closer pan of terminal leaflet and leaflet was more occupied then wing.

  • We investigated the in vivo effect of an aquous extract from Rhois Galla (R-G) on glucokinase and hexokinase activities of diabetes mellitus induced by interleukin-$1{\beta}$ ($IL-1{\beta}$). After 1 week of alloxan injection, the levels of serum glucose and insulin secretion were dramatically increased, however, the insulin secretion was decreased with administration of R-G. $IL-1{\beta}$ injection allowed the serum glucose level increased and the level was decreased by R-G administration. Furthermore, we could observe that R-G was effective in recovering the levels of insulin secretion. Enzyme activities of the glucokinase and hexokinase were decreased by $IL-1{\beta}$ treatment In contrast, R-G administration to the mice allowed proportion increasing. Seemingly, when $IL-1{\beta}$ was injected to the mice, enzyme activities of the glucokinase and hexokinase were decreased. But, R-G stimulated induction of enzyme activities of the glucokinase and hexokinase as high as normal group. These results suggested that R-G is highly effective in treatment of diabetes mellitus.

Antimicrobial Activity of Rhus javanica Extracts Against Animal Husbandry Disease-Related Bacteria (가축질병 균주에 대한 오배자 추출물의 항균활성)

  • Antimicrobial activity of Rhus javanica (RJ) extract against animal husbandry disease-related bacteria was determined by a paper disc method. The RJ extracts showed a significant antimicrobial activity against Gram positive (+) bacteria and especially the activity was most potent against L. monocytogenes and S. epidermidis. Minimum inhibitory concentrations (MIC) of the MeOH and EtOH extracts of RJ were in the range of 0.8 ∼ 16 mg/mL and 0.8 ∼ 10 mg/mL, respectively. Furthermore, among five solvent fractions (n-hexane, CHC1$_3$, EtOAc, n-BuOH and $H_2O$ frs.) from MeOH extract of RJ, the EtOAc fr. exhibited the most significant antimicrobial activity The antimicrobial activities of RJ extracts against most microbial strains were unstable by either heat treatment or acid treatment. The inhibitory effect of RJ extracts on microbial cell growth was further examined by the addition of 0, 100, 300, and 500 ppm of RJ extracts into growth medium. The growth of gram positive (+) bacteria, S. aureus, S. epidermidis and L. monocytogenes was inhibited for 72 hours when at least 300 ppm of RJ extracts added, but the growth of gram negative (-) bacteria was only inhibited when at least 500 ppm of RJ extracts were added. Taken together, tile antimicrobial activities of RJ extracts were more effective against gram positive (+) bacteria compared to those against gram negative (-) bacteria.

  • This study investigated the antioxidant and anti-inflammatory effects of Galla Rhois (Rhus verniciflua) on LPS (lipopolysaccharide)-stimulated human osteoblastic cells (MG-63). The aim of this study was to evaluate the LPS induced nitric oxide (NO) production and antioxidant radical in human osteoblast-like MG-63 cells. Therefore, the present work indicate that Galla Rhois extracts may be an ideal candidate for further research into their use for dental caries prevention component as well as, natural plant-based products. This suggested that 80% methanol and hexane extracts of Galla Rhois were inhibited NO generation and 1,1-diphenyl-2-pirylhydrazyl (DPPH) radical. Therefore, 80% methanol and hexane extracts of Galla Rhois may be utilized as a good source of protection against inflammation and oxidative stress. This study is intended to pursue wellness convergence in quality of life in the excavations in natural ingredients to create a variety of oral products with fewer side effects, cheap oral products for the dental treatment.

Antiviral activity of methanol extract from Rhus chinensis gall (오배자 추출물의 항바이러스 활성)

  • Trafficking process of viral glycoprotein to cell surface results in the syncytium formation when baby hamster kidney (BHK) cells was infected by Newcastle disease virus (NDV). Rhus chinensis gall, well-known as a medicinal plant, inhibited not only syncytium formation, but also trafficking of glycoprotein, hemagglutinin-neuramidase (HN) to the cell-surface. Modification of viral glycoprotein is processed within the endoplasmic reticulum and golgi body during trafficking into surface. R. chinensis gall extracts showed the strong inhibitory activities ($IC_{50}$ $12.5{\mu}g/mL$) against ${\alpha}-glucosidase$, when compared with the ${\beta}-glucosidase$. And this inhibitory activities is increased by the samples in a dose-depedent pattern. These data showed that the extracts of R. chinensis gall inhibited the cell-surface expression of NDV-hemagglutinin-neuramidase glycoprotein without significantly affecting HN glycoprotein synthesis in NDV-infected BHK cells.

Antimicrobial Synergistic Effects of Gallnut Extract and Natural Product Mixture against Human Skin Pathogens (피부 병원성균에 대한 오배자 천연 복합물의 시너지 항균 효과)

  • This study was attempted to investigate natural materials with antimicrobial activity and to apply as natural preservatives in cosmetics. The disc diffusion method was used to search for nine species of natural antibacterial material for three species of skin pathogenic bacteria (Staphylococcus aureus, Escherichia coli, Pseudomonas aeruginosa) and Candida albicans. As a result of measuring the size of inhibition zone, Rhus Semialata gall (Gallnut) extract, Oak vinegar, and ε-polylysine were showed strongest antibacterial activities (> 10 mm). The Minimum Bactericidal Concentration (MBC) of gallnut and oak vinegar ranged from 10 to 20 mg/mL and from 20 to 40 mg/mL against five human skin pathogens. The MBC of ε-polylysine ranged from 0.5 to 2 mg/mL in fungus. The synergic effect of gallnut extract/oak vinegar mixture and gallnut extract/ε-polylysine mixture were evaluated by checkerboard test. Compared to when used alone, the MBC of gallnut extract/oak vinegar mixture were at 4 times lower concentration against E. coli, C. albicans, and A. brasiliensis. Also Furthermore, the MBC of gallnut extract/ε-polylysine mixture were at 4 times lower concentration against C. albicans and A. brasiliensis. It was confirmed that the combination of gallnut extract with oak vinegar or ε-polylysine resulted in synergistic antibacterial effect against three human skin pathogens. Thus, it is expected that gallnut extract and natural product mixture can not only demonstrate antibacterial synergies, but also be applied in cosmetics as a natural preservative system with a wide antibacterial spectrum.

The Anti-bacterial Activity of Eco-friendly Farming Material based on Chinese Nut-gall Extraction on Acidovorax citrulli (오배자 추출물 유래 친환경제제의 세균성과실썩음병균에 대한 항세균활성)

  • Bacterial fruit blotch (BFB) is one of most important diseases in Cucurbitaceae due to infection of Acidovorax citrulli, causing huge economic losses damage worldwide. This seedborn disease spread rapidly at period of high temperature and humidity. The eco-friendly farming is getting popular. So far there was no effective agent to control BFB in eco-friendly farming. Therefore, effect of the material based on chinese nut-gall extract with antibacterial activity against BFB to was tested against A. citrulli. Different hosts showed various symptoms of BFB. Liquid formulation among exhibited most effective anti-bacterial activity on A. citrulli. Pot experiment in greenhouse showed the potential as an control agent of BFB in cucurbits. The treatment of material based on chinese nut-gall extract showed the positive effect on survival of the watermelon seedling and on the length of the cucumber seedling treated with A. citrulli. We cautiously conclude that the material based on chinese nut-gall extract used in this study may be good agents against major diseases of cucurbits in the future even though it is require to be tested with more study on field test.

Antimicrobial Activity of Medicinal Herbs against Staphylococcus aureus and Salmonella gallinarum. (Staphylococcus aureus와 Salmonella gallinarum에 대한 한약재의 항균활성)

  • The extracts from approximately 40 different Korean traditional medicines were prepared to investigate the antimicrobial activities against poultry disease-related bacteria. Among tested, the extracts of Schizandra chinensis (SC), Melia azedarach (MA), Caesalpinia sappan (CS) and Rhus javanica (RJ) exhibited significant antimicrobial activities against Salmonella gallinarum, whereas the extracts of Elsholtzia ciliata (EC), Myristica fragrans (MF), Alpinia katsumadai (AK), Poncirus trifoliata (PT), Prunella vulgaris (PV), CS and RJ exhibited antimicrobial activities against Staphylococcus aureus. Minimum inhibitory concentrations (MIC) of MA, CS and RJ extracts against S. gallinarum were 1.2 mg/ml, whereas MIC of RJ extract for S. aureus was 0.6 ㎎/ml, which was the lowest among tested. The antimicrobial activities of SC and RJ extracts against S. gallinarum were reduced, but those of AK and CS extracts against S. aureus were not affected by heating treatment. The antimicrobial activities of SC extract against S. gallinarum and those of EC, PT and RJ extracts against S. aureus were stable by acid treatment but unstable by alkaline treatment. those of CS extract was not effected by either acid or alkaline treatment. The growth of all bacteria was significantly inhibited within 24 hours by the addition of at least 100 ppm and 300 ppm of RJ and CS extracts, respectively, compared with the control group. In conclusion, these findings suggest that RJ and CS extracts may play important roles for antimicrobial activities against poultry disease-related bacteria.
