Production of Alkaline Carboxymethyl Cellulase and Xylanase by Batch and Fed-batch Cultures of Alkalophilic Cephalosporium sp. RYM-202 (호알카리성 Cephalosporium sp. RYM-202의 회분 및 유가배양에 의한 Alkaline Carboxymethyl Cellulase와 Xylanase의 생산)

  • Production of alkaline carboxymethyl cellulase (CMCase) and xylanase by batch and fed-batch cultures of alkalophilic Cephalosporium sp. RYM-202 was investigated. Of carbon sources tested, wheat bran gave the highest production of those enzymes. The high levels of CMCase on carboxymethyl cellulose and xylanase on birchwood xylan suggest that the biosynthesis of CMCase and xylanase in Cephalosporium sp. RYM-202 is regulated separately at the level of enzyme induction. The temperature and pH for maximal production of those enzymes was $20^{\circ}C$ and 9.0, respectively. High concentration of wheat bran in batch fermentation resulted in the lower and delayed production of the enzymes by catabolite repression. In fed-batch fermentation with controlled feeding of 5% final wheat bran concentration, the highest activities of CMCase and xylanase were 0.39 and 9.2 units/ml, respectively, and 1.22 and 1.36 times higher respectively than those in batch fermentation on 5% wheat bran.

  • In this study, we observed differences in the fruit body genesis in with seasonal changes. The mushrooms generated different spines even when under same environmental conditions in equivalent growth chambers during different seasons. The growth chamber conditions were set at $18^{\circ}$, 1000 ppm CO and 95% humidity for uniform primordium formation. Following this, the humidity was reduced to 85% during growth and development. The characteristics of the fruit body were measured: yield (weight), length (size), hardness, and chromaticity. Each measure was relatively compared owing standard data for. We investigated the environmental factors to assess the cause of variation in fruit body. Change in temperature and humidity were especially focused on, apart from climate that can affect mushroom growth. It was observed that temperature and humidity both influence fruit body growth. Not the absolute value of the elevation, there was a change of climate which the temperature is lowered and lowered the humidity can be assumed that the changes in the environmental conditions inside the growth chamber the mushroom growth.

Analysis of the Cause of the Twig Blight on Mulberry Based on the Weather Conditions (뽕나무 재배기간 중 기상환경과 눈마름병 발생관계 분석)

  • The twig blight of mulberry caused by Fusarium lateritium occurs mainly in Buan area, which is the main area of mulberry. The initial symptom on twigs showed dark edge blight and dark orange tiny particles on the spot, and it was withered and died. In the case of mulberry twig blight, the difference in the degree of occurrence between the years is intense, and an average of 2.7% occurred in Buan in 2013, but in 2014, it was difficult to search for the generated fields. In order to find the cause of the difference in the incidence of mulberry twig blight, we investigated and analyzed the weather conditions during cultivation and as a result, the occurrence of the disease increased under the condition that the temperature of the spring was lower and the number of days passing below the winter freezing point increased. In addition, it was analyzed that the occurrence of the next year increases when the rainfall is high and the number of sunshine is low after removing the harvested branches. Therefore, in the event that weather conditions causing twig blight diseases have elapsed, it is necessary to prevent the disease by spraying the registered applied fungicide prophylactically.

Characterization of ${\alpha}$-amylase Producing Hybrid Constructed between Saccharomycopsis and Saccharomyces (Saccharomycopsis속과 Saccharomyces속의 잡종형성 균주에서 생산하는 ${\alpha}$-amylase의 특성)

  • This study has been performed to deveope a yeast strain having high ${\alpha}$-amylase production ability using nuclear transfer method. Hybrids formed between the strains of Saccharomyces fiburigera KCTC 7393 and Saccharomyces cerevisiae KCTC 7049 (tyr-, ura-)were obtained by nuclear transfer technique. Nuclei isolated from the wild type S. fiburigera strain were transfered into auxotrophic mutants S. cerevisiae and selected the hybrids showing an increased starch degrading capability were selected (MN-16). This transformant grew best and produced maximal ${\alpha}$-amylase activity on the medium containing 2% (V/V) soluble starch. ${\alpha}$-Amylase from MN-16 was purified electrophoretically homogenety and its properties were investigated. The enzyme was purified about 10.6 fold with an overall yield 9.7% from the culture medium by ammonium sulfate fractionation. DEAE-Sephacel column chromatography, and Sephacryl S-200 column chromatography. The purified enzyme showed a single band on SDS-polyacrylamide gel electrophoresis. The molecular weight of the ${\alpha}$-amylase was estimated to be 53,000 daltons by SDS-PAGE and by gel permeation chromatography on Sephacryl S-200. The purified enzyme showed the maximum activity at pH 5.5 and 40${\circ}C$. The km value for soluble starch was 2.5㎎/㎖. The enzyme activity increased in the presence of $Ca^{2+}, Co^{2+}, EDTA, Mg^{2+}, Mn^{2+}, Zn^{2+}$, but inhibited by $Cu^{2+}, Fe^{2+}$, and $Ni^{2+}$

Partial Purification and Some Properties of Carboxymethyl Cellulases from Alkalophilic Cephalosporium sp. RYM-202 (호알칼리성 Cephalosporium sp. RYM-202가 생산하는 carboxymethyl cellulase의 부분정제 및 특성)

  • An alkalophilic Cephalosporium sp. RYM-202 capable of producing cellulase components was isolated from soil. This organism grew best at an initial pH 9.0 and produced cellulase maximal at an initial pH 9.5-10.0. Three carboxymethyl cellulases(CMCases), P-I-I, P-I-II and P-II-I, were partially purified by DEAE-Sephadex A-50 ion exchange column followed by Sephadex G-150 gel filtration. The optimum pH values for activity were 7.5 for P-I-I, 8.0-9.5 for P-I-II and 7.5-10.0 for P-II-I. All CMCases were stable between pH 4.5 and 12.0. Temperature optima for activity ranged between 40 and $60^{\circ}C$ and more than 50% of the maximum activity was observed at $20^{\circ}C$ for both of P-I-I and P-II-I. The activity of CMCases was significantly stable in the presence of various laundry components, such as, surfactants, chelating agents and alkaline proteinases.

  • Mulberry fruit is common to keep a frozen storage because of the very high water content. Because freezing-storing cost is generated before the processing or sales, farmers of mulberry fruit production are weighted in financial burden. In addition, mulberry fruit has difficulty in expanding consumption by made in the simple processed products like the mulberry jam, mulberry juice, mulberry enzyme, alcohol, etc. Recent, frozen fruits including frozen blueberries, frozen strawberries, frozen mango, frozen rich, frozen mulberry fruit has been started to sell in the large discount stores and convenience stores. However, there is restricted sales in only special place that have the frozen distribution facilities. So, for the income improvement and consumption promotion of mulberry farmers facing these difficult situations, we developed the processing method of semi-dried mulberry fruit using the vacuum freezing dryer. The frozen mulberry fruit which was stored below $-40^{\circ}C$ is to dry in a vacuum freeze dryer, at $30{\sim}35^{\circ}C$, 23 ~ 24 hours. Semi-dried mulberry fruit seperated respectively. It had soft texture, and maintained shape, size and flavor. In particular, semi-dried mulberry fruit can be stored at room temperature.

Artificial Sea Ice Increasing to Mitigate Global Warming (지구 온난화 경감을 위한 인공해빙증가)

  • This study suggests a method of alleviating global warming by the increase of the Earth surface albedo through Artificial Sea ice Increasing (ASI) over the Available Freezing Areas (AFA). The method is developed based on the fact that the large sea surface area in or near the Arctic and the Antarctic has no ice even though both water and air temperatures are below zero and the artificial sea ice generation is thus available. The mean energy of $0.85Wm^{-2}$, which was suspected of adding to the earth by the global warming effect was calculated to offset at once when the sea ice area about $4.09{\times}10^6km^2$ was additionally increased. In addition, three techniques for producing ice plates on the sea surface (using ships, installation apparatus, and floating matter such as Green Cell Foam) for ASI were proposed. According to the result of simple analysis using the energy balance model, when ASI was maximally operated only for 3 months (September, October, and November) over AFA, it is expected that the annual mean temperature of earth surface would be decreased about $0.11^{\circ}C$ in the following year. On the other hand, in case of generating the artificial sea ice in all four seasons, a risk of triggering snowball earth was detected.

A Fundamental Experiment on Preventing Frost Damage at Early Age of Mortar in Low Temperature using Reduction Slag (환원슬래그를 사용한 모르타르의 저온에서의 초기동해 방지에 관한 기초적 실험)

  • In this research, it used cement powder and reduction slag, which generates high hydration heat in hydration reaction without heat cure below $-5^{\circ}C$ degree. Purpose of final research is preventing freezing and thawing by making the compressive strength 5MPa in 3days below zero temperature due to own heat of concrete. and it is the result of physical characteristic and thermal property evaluation of reduction slag. Because reduction slag generates high hydration heat, compressive strength development is excellent. By generating highly hydration heat by $C_{12}A_7$ and $C_3A$ in reduction slag, compressive strength is developed in low temperature. In case of displacing only reduction slag without $SO_3$, it is indicated that quick-setting occurs by shortage of $SO_3$. For preventing quick-setting, gypsum is used essentially. According to this research result, in case of using reduction slag and gypsum as a ternary system, compressive strength developed 5MPa in 3 days below zero temperature. It is identified to prevent early frost damage of concrete below zero temperature.

Effects of Low-Level Laser Irradiation on the ALP Activity and Calcified Module Formation of Rat Osteoblastic Cell (저수준레이저(GaAs 반도체)조사가 골모세포의 알칼리성 인산분해효소의 활성과 석회화결절의 형성에 미치는 영향)

  • 저수준레에저요법에 대해서는 지난 10여년간 의학계 및 치과계에서 임상적으로 사용하여 좋은 결과가 있다고 많은 보고가 발표되고 있다. 특히 치근의 골결손에 관한 연구에서는 전기요법, 초음파요법, 전자장요법 등 뿐만아니라 저수준레이저를 사용하여 골절부내 Callus형성이 촉진되었음을 보고하고 새로운 치료법의 하나가 될 수 있음을 제안한 바도 있다. 본 연구에서는 칼륨비소를 다이오드로 사용한 저수준레이저조사가 골결손의 치유에 어떠한 영향을 미치는지 확인하고자 골모세포의 알칼리성 인산분해효소의 활성화와 석회화결절의 형성을 평가함으로 골모세포의 기능을 조사코저하였다. 실험은 첫째, 9개군으로 나누어 레이저 조사기간에 따른 알칼리성 인산분해효소의 활성화를 조사하였고, 둘째, 이를 근거로 9일간 계속 매일 1회 1.3 J/cm2의 레이저를 조사한 후 펄스의 종류별 차이를 비교하였으며, 세째,레이저펄스별 석회차 결절의 형성 정도를 광학현미경으로 관찰하여 비교분석하였다. 결과, 7일 계속 레이저를 조사한 경우 다른 군에 비해 서서히 ALP의 활성이 증가하였으나 유의한 차이는 없었으며. 따라서 9일동안 레이저를 계속 조사한 경우에는 전체 에너지량이 5.895 J/cm2 인 펄스13과 15가 뚜렷하게 유의한 증가를 보여주었다. 그러나 석회화결절의 형성은 전체 에너지량이 2.546 J/cm2 인 펄스11에서 가장 많았다. 결론적으로 골형성이나 알칼리성 인산분해효소의 활성을 촉진하는 데에는 적절한 레이저 조사조건이 필요하나, 알칼리성 인산분해효소의 활성을 촉진한 펄스와 석회화결절의 형성을 촉진하는 펄스가 서로 다르게 나타난 것은 골형성을 촉진하는 여러요인 들이 저수준레이저에 자극받았을 가능성이 높음을 보여준다 이러한 결과들로 보아 저수준레이저는 골모세포의 기능을 자극하여 골결손의 치유를 개선하는 데 도움될 것이라 사료된다.다. 각 백서의 양측 창상중 하나는 1,3,5,7일 마다 각 실험의 방법에 따라 레이저를 조사하고 실험동물의 다른 창상은 대조군으로서 사용하였다. 모든 창상의 면적은 실험 1,3,5,7 일째에 일정한 거리에서 사진촬영하여 면적계를 이용, 측정한 후 통계적인 의의를 조사하였다. 본 연구의 결과는 저수준레이저는 특정 조건하에서 S. aureus의 증식을 촉진하였다. 그러나 S. aureus에 감염된 창상을 저수준레이저로 조사시 치유가 촉진되었다. 중앙 조사법고 주변조사법에 의한 창상치유효과는 통계적인 의의가 보이지 않았다. 따라서 결론적으로 S. aureus 에 감염된 창상에 직접 또는 간접적이든 pulse의 종류에 관계없이 조사하는 경우 치유효과가 나타나는 것은 정사주위 조직의 LLLI 자극효과가 염증의 확산을 억제한다고 말할수 있다.4/1$0^{\circ}C$에서는 Shoa-Nan-Tsan과 Lenkwang이 가장 높았으며 백앙벼는 3 온도 조건 모두에서 활성이 낮았다. 발아소요일수와 amylase 활성과는 유의적인 정의 상관관계를 보였다., 다다조, 미국의 건답직파재배 품종 등이었으며 우리 나라 육성종들은 모두 지중에서 신장이 멈추어 제1본엽이 지중에서 추출하였으며, Scm파종심에서 불완전엽이 지면을 뚫고 나오는 품종은 Chinsura Boro뿐이었고 Nato, Labelle, Weld Pally, Italliconaverneco 등도 지면 가까이 까지 신장하였다. 6. 50% 출아일수는 제2절간장을 제외 한 모든 유아 형질의 신장도와 유의한 부의 상관을 보였는데 가장 높은 상관을 보인 것은 중배축장+제1절간장+불완전옆장이었으며, 다음이 불완전엽장이 었다. 7. 출아율은 중배축장+제1절간장+불완전엽장, 중배축장+초엽 장과 모든 파종심에서 높은 정의 상관을 보여 제1본엽의 추출 위치가 높을수록

Study of Hydrotrope Action and Liquid Crystal Behavior of Concentrated Liquid Detergents (농축세정제의 액정 상거동과 하이드로트로프 작용에 대한 연구)

  • Concentrated liquid detergents have 2~3 times higher surfactant contents [35~45% (w/w)] compared to those of normal type detergents. In general, a single surfactant forms a lyotropic liquid crystal (LC) phase when the concentration is in the region of 30~60% (w/w). Whereas the concentrated liquid detergent at about 40% (w/w) concentration in a mixed surfactant system shows an opaque appearance of gel or LC. In order to meet consumer needs and preference for product appearance, we applied hydrotropes and various surfactants systems in concentrated liquid detergents to obtain an opaque gel-phase and also a clear transparent phase at even below zero $^{\circ}C$ temperature. The more effective hydrotropes for making concentrated liquid detergents are 1,6-hexanediol, adipic acid and dipropylene glycol (DPG) which have two hydrophilic groups in both terminated positions. In order to prepare an excellent concentrated liquid detergent, good hydrotropes alongside secondary type surfactants like LAS and SAS were used. The formation of LC phase of concentrated liquid detergents at about 40% (w/w) concentration could be prevented by the use of both hydrotropes and secondary type surfactants. The result indicate that concentrated detergents having excellent low temperature stability and controlled viscosity can be prepared.
