Morphogenetic Identification of Eel's Larva (Leptocephalus) Collected by Set net in Namhae, Korea (남해 정치망에서 채집한 엽상자어(Leptocephalus)의 형태 및 유전학적 특성)

  • The present study was tried to identify whether the eel's larva was close to a conger (Conger myriaster), a pipe conger (Muraenesox cinereus) or four species of Anguilla. Experimental fishes were collected by set net in the gulf of enggang, Namhae, Korea from May to June. Their morphological characteristics were compared with adult fishes of a conger, a pipe conger and four species of Anguilla. For genetic classification, DNA was isolated and amplified by using 12S rRNA and 16S rRNA primer set. The PCR products were direct sequencing in both directions. The nucleotide sequences were analyzed using softwares. As results of morphological measurement on eel's larva, the percentages of head length and preanal length against total length were similar with a conger. Based on the nucleotide sequences, the phylogenetic tree also revealed a close relationship to a conger. Therefore, eel's larva, caught in Namhae from May to June, was identified into a conger's larva.

  • Three specimens of leptocephalus larvae (33.4~35.2 mm in SL) were collected from the southern coast of Jeju Island, Korea, by an aquatic lamp on August 14, 2004. These larvae were identified as Elops hawaiensis because of their having a forked caudal fin, a smaller anal fin than dorsal fin, and anal-fin position posterior to dorsal fin.

  • The seven leptocephali (14.6~68.0 mm in total length) of the family Ophichthidae, from the East Sea of Korea, were identified using 12S rRNA 861-bp sequences and their morphological development were described. Our seven leptocephali were identified to Echelus uropterus based on genetic distance (d=0.000) being well consistent with that of adult E. uropterus. Our seven leptocephali of E. uropterus are characterized by morphological combinations: caudal fin present; the melanophores present in series of nine gut swellings; total myomeres 154~158; body depth in total length less than 10%; eight black horizontal bands present from anus to caudal fin base. Our morphological and molecular results suggest that E. uropterus may be an intermediate group between subfamily Ophichthinae and Myrophinae.

  • Four leptocephali (TL 109.8~129.7 mm) of the family Ophichthidae, collected from Sangju in the South Sea of Korea, were identified using morphological and molecular methods. Our four leptocephali were similar to Ophichthus sp. based on morphological characters: the melanophores present in series of eight gut swellings; total myomeres 144~151; body depth in total length less than 10%; eight or nine black horizontal bands present from anus to caudal fin base. On the other hand, our four leptocephali were identified to Pisodonophis sp. based on 826 base pairs of 12S rRNA gene nucleotide sequences. Our results showed that morphology-based identification did not agree with molecular identification, indicating difficulty in differentiating morphologically between Ophichthus and Pisodonophis leptocephali. We, herein, firstly describe morphological characteristics of Pisodonophis sp. leptocephali from Korea.

Molecular Identification and Morphological Description of Gymnothorax minor (Muraenidae, Anguilliformes) Leptocephali off Jeju Island (제주 근해에서 채집된 나망곰치, Gymnothorax minor (뱀장어목, 곰치과) 엽상자어의 분자동정 및 형태기재)

  • Two leptocephali (36.2~69.2 mm in total length) of the family Muraenidae were collected off Jeju Island, Korea. They were identified using 742 base pairs of mitochondrial DNA 12S rRNA sequences, and their morphological description were presented. The two leptocephali were identified to Gymnothorax minor based on genetic distance (d=0.000), being well consistent with that of G. minor adult collected off Tongyeong. Gymnothorax minor leptocephali are characterized by morphological combinations: total myomeres 137~139, predorsal myomeres 32, preanal myomeres 82~86, body depth in total length more than 10%.

  • One leptocephalus (TL 59.9 mm) from the family Ophichthidae collected from Jeju Island, Korea, was identified using morphological and molecular methods. Our ophichthid leptocephalus was identified as belonging to the genus Muraenichthys based on morphological characters: 157 myomeres; the origin of the dorsal fin located a little in front of the anus; a distinct melanophore present on the opercle; and six gut swellings present. An analysis of 886 base pairs of the 12S rRNA mtDNA sequences showed that our leptocephalus must be Muraenichthys gymnopterus, because its sequences were concordant with those of an adult M. gymnopterus (d=0.001) and next to those of Muraenichthys sp. leptocephalus (d=0.034). Here, we are the first to describe the morphological characteristics of the M. gymnopterus leptocephalus.

Effect of Preservative Solutions on Postharvest Vase Life of Hydrangea macrophylla 'Verena Green' (보존용액이 절화 수국 '발레나 그린'의 관상기간 연장에 미치는 영향)

  • 수국 '베레나 그린'의 줄기하단의 엽을 제거하고 5엽 상태에서 수돗물과 0.1% 크리잘 RVB에 각각 담가 4℃에서 24시간 물올림을 하고 물올림을 하고, 줄기를 60~70cm 길이로 끝을 대각선으로 절단하여 물올림용액을 채운 물 대롱을 끼운 다음 꽃을 보호하기 위한 비닐 슬리브를 씌워 유통용 종이상자에 담아 5±1℃ 냉장차로 국립원예특작과학원으로 운반하였으며, 절화수명 및 관련 특성분석을 위해 평가실로 옮기기 전까지 유통용 종이상자에 담긴 상태로 저온저장고(5±1℃)에 보관하였다. 보존용액에 따른 절화특성 조사를 위해 절화를 평가실에 전시하기 전에 절화의 줄기길이를 40cm로 맞춰 절단한 뒤에 보존용액을 800 mL 채운 플라스틱 화병에 꽃아 절화 화관끼리 서로 닿지 않도록 배치하였으며 22±2℃ 실내에서 절화수명을 조사하였다. 보존용액으로 4% 차아염소산나트륨 용액, 1% sucrose + 250 mg/L 8-hydroxquinoline + 100 mg/L citric acid로 이루어진 용액(HQ) 그리고 시중에서 수국 절화 보존제로 판매되는 크리잘 프로 II, III와 크리잘 clear 0.5% 용액을 사용하였다. 수돗물에 물올림하였을 때 보존용액인 0.5% 크리잘 프로 II에서는 33.8일, 0.5% 크리잘 프로 III에서는 27.7~33.5일, 크리잘 클리어는 33.7일, 차아염소산나트륨과 수돗물은 각각 26.2, 28.8일이었고, HQ 용액은 49.0일로 크리잘보다 15일, 수돗물보다 20일 절화수명이 연장되었다. 수국 '베레나 그린'의 보존용액으로 1% sucrose + 250 mg/L 8-hydroxquinoline + 100 mg/L citric acid를 사용하는 것이 관상기간을 연장하는 데 효과적이었다.

First Morphological Description of Ophichthus asakusae (Ophichthidae: Anguilliformes) Leptocephali Collected from the Waters South of Jeju Island (제주도 남방해역에서 채집된 제주바다뱀(Ophichthus asakusae) 엽상자어(Leptocephalus)의 첫 형태기재)

  • Two leptocephali [total length (TL) 68.2-80.5 mm] from the family Ophichthidae, collected from the waters south of Jeju Island, Korea, were identified using morphology. The two ophichthid leptocephali were identified as belonging to the genus Ophichthus based on morphological characters: 131-132 myomeres; nine gut swellings; dorsal fin origin above the middle of the body; five postanal pigments present from the anus to the caudal margin. Ultimately, both leptocephali were identified as Ophichthus asakusae, as the total number of myomeres was consistent with those of adult O. asakusae. Here, we first describe the morphological characteristics of the O. asakusae leptocephalus, and suggest diagnostic characters for leptocephali belonging to the genus Ophichthus.

  • A single leptocephalus specimen of Albula glossodonta belonging to the family Albulidae under the order Albuliformes, was collected from the southern sea of Korea in May 21, 2019. This species is distinguished from A. vulpes in the number of melanophores at the base of the caudal fin. In addition, our specimen was readily distinguished from A. argentea in having no melanophores on the operculum, at the base of the dorsal fin and in front of the pectoral fin. Also it was well distinguished from A. gilberti in having no melanophores on the operculum, in front of the pectoral fin and on the lateral body. A molecular analysis based on 467 bp sequences of the mitochondrial DNA 16S rRNA shows that the leptocephalus is well matched to A. glossodonta adult (K2P distance=0.002). We report the first occurrence of A. glossodonta leptocephalus in the Korean waters, and propose it's new Korean name, "Nam-bang-yeoeul-myeol".
