Gene Expression Levels of Growth Hormone, Prolactin and Their Receptors of Olive Flounder Paralichthys olivaceus by Salinity Changes (염분농도에 따른 넙치 (Paralichthys olivaceus)의 성장호르몬, 프로락틴 및 수용체 유전자의 발현변화)

  • To investigate the effects of environmental salinity on the expression of the genes for growth hormone (GH) and prolactin (PRL) in the pituitary, and their receptors (GHR, PRLR) In the kidney, intestine, and gills in teleosts, we acclimated juvenile olive flounders (Paralichthys olivaceus) to different salinities (5, 15, 25, or 32 psu) for 3 days and examined their mRNA levels using the reverse transcription-polymerase chain reaction (RT-PCR). In the fish adapted to low salinity, the PRL mRNA levels in the pituitary were elevated dramatically, whereas the GH mRNA levels did not differ significantly. PRLR mRNA increased significantly in fish exposed to low salinity, whereas GHR mRNA levels did not differ. These results suggest that PRL is an important hormone for flounders that are acclimated to brackish water and it may control ion homeostasis with PRLR in the osmoregulatory organs.

Mortality of the Horned Turban Shell, Batillus cornutus Caused by Vibrio spp (소라(Batillus cornutus)의 비브리오균 감염에 의한 폐사)

  • Mass mortality of the horned turban shell, Batillus cornutus was investigated using histological and bacteriological methods. Some pathogenic bacteria were isolated from mortal or inactive individuals. The pathogenic agents causing mortality of the horned turban shells were as Vibrio alginolyticus and V. anguillarum. Laboratory experiment indicated that optimal growth temperature of two bacteria was 25 to 30$^{\circ}C$ and 3% of NaCl. Histological examination of the horned turban shells showed that gill necrosis is one of the major symptom of infected individuals. It was believed that sudden increase of those two bacterial agents due to environmental change cause mortality of horned turban shells.

  • Acid phosphatase in the radish young plant showed optimal activity at pH 5.5. The activity of acid phosphatase was maintained longer during the ABA (0.5 mM) treatment than those in control, whereas that was similar to the treatment of NaCl (0.5 mM). But during the cold (4$^{\circ}C$) treatment, the activity of acid phosphatase was decreased dramatically compared to the control, which was maintained almost on a constant level and increased gradually during 6 days. It showed that acid phosphatase was in relation to the change of biochemical reaction, which plants were coped with cold, NaCl and ABA stress.

  • Forecasting salinity in an estuary reservoir is essential to promise irrigation water for the reclaimed land. The objective of the research was to assess salinity balance and its temporal and spatial variations in the Iwon estuary reservoir which has been issued by its high contents of salinity in spite of desalination process for four years. Seepage flows through the see dikes which could be one of possible reason of high salinity level of the reservoir was calculated based on the salinity balance in the reservoir, and used as input data for salinity modeling. A three-dimensional hydrodynamic model, Environmental Fluid Dynamics Code (EFDC), was used to simulate salinity level in the reservoir. The model was calibrated and validated based on weekly or biweekly observed salinity data from 2006 to 2010 in four different locations in the reservoir. The values of $R^2$, RMSE and RMAE between simulated and observed salinity were calculated as 0.70, 2.16 dS/m, and 1.72 dS/m for calibration period, and 0.89, 1.15 dS/m, and 0.89 dS/m for validation period, respectively, showing that simulation results was generally consistent with the observation data.

Lipid and Fatty Acid Composition in Nannochloropsis oculata Cultured in Varying Salinities (염분농도에 따른 해양미세조류(Nannochloropsis oculata)의 지질 및 지방산의 변화)

  • The quality and quantity of food organisms in fish seed production are important. The marine microalgae Nannochloropsis oculata are used as initial food organisms in the field. We investigated the effects of salinity (0, 10, 20, 30, 40 and 50 psu) on the lipid and fatty acid composition of N. oculata. Cultivation of N. oculata at varying salinities showed the highest growth rate at 20 psu. Total lipid content ranged from 17.26 to 18.63% at salinities from 0 to 50 psu). The nonpolar lipid content increased markedly at 30 psu and was highest at 15.55%. The polar lipid content was lowest at 30 psu, by 84.45%. It was also found that the omega-3 and EPA contents were inversely proportional to salt concentration. For the polar and nonpolar lipid compositions, there was no significant effect of salinity. Omega-3 polyunsaturated fatty acid content especially the content of EPA in the seawater larvae is the essential fatty acid in this food organism. It is thus advantageous to culture N. oculata at 20 psu.

  • The suitable temperature and salinity for the long-distance transportation for the rockfish were investigated by assessing survival rate, metabolism, histological change. All experimental groups showed survival rates of 100%. Daily Oxygen consumption rhythm was decreased during nights and increased during days. Average oxygen consumption was significantly decreased as temperature and salinity were decreased. Ammonia excretion was significantly increased as temperature and salinity were decreased. Histological changes were observed in the skin and gill of the rockfish exposed to 10 psu under all the temperature conditions, with larger changes at $4^{\circ}C$. Further, nucleus deformation and uniformity in the cytoplasm were also observed.

A Study on the Behaviors of Several Layers in a Solar Pond (태양연못안의 각 층의 거동에 관한 연구)

  • The behaviors of layers developed in a solar pond were studied by experimental and analytical methods. An experimental solar pond heated from below was constructed and operated at the net heat fluxes of 110 and 160W/m$^{2}$ and at the initial salt concentration gradients of 18.2, 27.3 and 36.4%/m. The thicknesses, growth rates, temperature and salt concentration in the top and the bottom mixed layers, the diffusive layer and the upper and the lower interfacial boundary layers were measured. The shadowgraph technique was used in order to observe all layer formation and an electroconductivity-temperature probe consisting of four electrodes was fabricated and used in measuring the salt concentration. Based on the experimental results, a model for the solar pond was developed and the governing equation and the assumptions were established. The governing equations were solved by the numerical method. The calculated results obtained from the analysis were compared with the experimental results.

  • Fatigue strength of offshore structures or ship structures is significantly decreased due to corrosive environment condition such as sea water and/or coal, crude oil of cargoes, compared to that of on shore structures. In corrosive environment, fatigue strength of structures also depends on characteristics of weld material heat affected zone(HAZ). In this research work, rotary bending fatigue tests of parent material and HAZ of TMCP steel were performed in order to investigate the initiation and propagation of cracks both in air and in NaCl solution. Comparison of fatigue strength In relation with the salinity of NaCl were carried out as well. According to the test results weld material or HAZ of TMCP steel showed higher fatigue strength than that of the parent material. The fatigue strength of TMCP steel decreases drastically in NaCl solution compared to that of in air environment. In particular, more reduced fatigue strength is observed in 1% NaCl solution than in 3% NaCl solution.

마지막 최대빙하기 이후 북극해 스발바드군도 Van Mijenfjorden의 고환경 변화

  • Van Mijenfjorden은 스발바드 군도의 Spitsbergen 서부에 위치하는 두 번째로 큰 피오르드로 온난하고 염분도가 높은 북대서양의 표층수가 북극해로 유입되는 Gateway에 위치하기 때문에 전지구적이며 지역적인 기후변화의 영향을 받는 지역이다. 1999년 IMAGES 프로그램의 일환으로 프랑스 탐사선인 'R/V Marion Dufresne'을 이용하여 북극해의 스발바드 군도에 위치하는 Van Mijenfjorden (77$^{\circ}$ 46.87'N and 15$^{\circ}$ 17.81'E)에서 약 18m의 빙ㆍ해양 퇴적물 코아(MD99-2305)를 시추하여 마지막 최대 빙하기 이후의 고환경변화를 연구하였다. AMS 14C 연대 측정에 의하면 diamicton 층인 하부 2m를 제외한 16m의 퇴적층은 지난 12cal. ka BP경에 피오르드에 존재하던 조수빙하(tidewater glacier)가 해빙되기 시작한 이후에 퇴적되었다. 특히 유기지화학 자료와 부유성과 저서성 유공충의 산출양상 그리고 저서성 유공충인 C. reniforme의 산소ㆍ탄소 안정동위원소 비에 의하면 코아 MD99-2305에는 Van Mijenfjorden에서 홀로세 동안에 일어난 퇴적환경변화를 잘 기록하고 있다. 특히 홀로세 동안에 피오르드내의 퇴적환경 변화는 조수빙하의 확장과 후퇴와 밀접한 관계가 있으며, 유기물의 탄소동위원소(13Corg) 비는 -24에서 -22$\textperthousand$ 값의 변화를 보인다. 이는 Van Mijenfjorden 퇴적물에 공급된 유기물은 육상과 해양기원이 혼합되어 퇴적되었음을 지시한다. 지난 12 cal. ka BP 이후 13Corg 값이 뚜렷하게 변하는 것은 Van Mijenfiorden에서 조수빙하의 변동과 표층수에서의 생산력의 변화를 강하게 반영한다. 강하게 반영한다.53%가 이공계 출신, 100대 기업 CEO의 38.4%가 이공계 출신, 대입수능시험에서 자연계 지원비율이 감소 -40.1%('99)\$\longrightarrow$34.7%('00)\$\longrightarrow$29.4%('01)\$\longrightarrow$26.9%('02)\$\longrightarrow$30.3%('03)\$\longrightarrow$31.5%\$\longrightarrow$'04)필요성, 효과적인 운전자 교육 정책 등이 그것이다. 자동차 긴급 피난 차선은 유용한 시설로 여러곳에서 그 기능이 발휘되고 있으므로 많은 고속도로 관계자들은 설계, 시공 및 유지 관리 측면에서 유의해야 할 것이다.미세조직을 광학현미경으로 압출방향에 평행한 방향과 수직방향으로 관찰하였고, 열간 압출재 이방성을 검토하기 위하여 X선 회절분석을 실실하여 결정방위를 확인하였다. 전기 비저항 및 Seebeck 계수 측정을 위하여 각각 2$\times$2$\times$10$mm^3$ 그리고 5$\times$5$\times$10TEX>$mm^3$ 크기의 시편을 준비하였다.준비하였다.전류를 구성하는 주요 입자의 에너지 영역(75~l13keV)에서 가장 높은(0.80) 상관계수를 기록했다. 넷째, 회복기 중에 일어나는 입자들의 유입은 자기폭풍의 지속시간을 연장시키는 경향을 보이며 큰 자기폭풍일수록 현저했다. 주상에서 관측된 이러한 특성은 서브스톰 확장기 활동이 자기폭풍의 발달과 밀접한 관계가 있음을 시사한다.se that were all low in two aspects, named "the Nonsignificant group". Condition of Processing and Change of Chemical Composition on the Salted-dried Sardine during the Storage (정어리 염건품의 제조 조건과 저장중의 성분변화)

  • An investigation on optimum condition for processing of salted-dried sardine and quality change of the product during the storage was carried out. The fillets of sardine were salted in 5, 10 and $15\%$ NaCl solution for 12, 24 and 36 hours at $9-10^{\circ}C$, respectively and air-blast dried at $24-25^{\circ}C$ with a velocity of 3 m/sec and then stored at room temperature for 30 days. The results were as follows : The optimum condition for the product was salting in $15\%$ NaCl solution for 36 hours as VBN and histamine contents were 27.5 mg/100g and 42.5 mg/100g, respectively, and also showed a good result in sensory evaluation. Histamine, VBN and TMA-N content of the dried fillets immediately after salting in $15\%$ NaCl solution for 36 hours increased to 47.8 mg, 29.5 mg and 5.4mg per 100g of the sample, respectively and increased slowly during the storage. The peroxide value and acid value of the lipid extracted from the product increased from 15.9 meq/kg to 114.0 meq/kg, and 12.6 to 30.5 during the storage. In the fatty acid composition, $C_{22:6}\;and\;C_{20:5}$ acids were decreased during the storage period.
