Effect of Preharvest Sprayable 1-Methylcyclopropene (1-MCP) Treatment on Fruit Quality Attributes in Cold Stored 'Gamhong' Apples (수확전 수체살포용 1-MCP 처리가 '감홍' 사과의 저온저장 중 과실품질에 미치는 영향)

  • This work was carried out to evaluate the effect of preharvest sprayable 1-methylcyclopropene (1-MCP: $Harvista^{TM}$) treatment on the fruit quality attributes in cold stored 'Gamhong' (Malus domestica Borkh) apples. Preharvest sprayable 1-MCP treatment did not affect fruit qualities at harvest but titratable acidity (TA) was higher in fruit treated 10 days before harvest (DBH) than in control. During cold storage, fruit firmness and TA were higher in 1-MCP treated fruit than in control fruit as storage duration progressed, while SSC was not affected. Internal ethylene concentration (IEC) levels were not different among treatments at harvest. While the IEC level in control fruit reached to $58.3{\mu}L{\cdot}L^{-1}$ after 6 month cold storage, the IEC levels in 1-MCP treated fruit ranged from 18.2 to $25.0{\mu}L{\cdot}L^{-1}$. Therefore, this result indicates that preharvest sprayable 1-MCP treatment should be another promising approach to retain fruit quality attributes in long-term cold stored 'Gamhong' apples.

  • 비닐 알코올의 함량이 62(EVOH-62) 및 71(EVOH-71) mole%인 에틸렌 비닐알코올 공중합체와 나일론(나일론-6, 나일론-6/12 및 나일론-12)과의 블렌드를 이축압출기에서 제 조하였으며 이들블렌드의 형택학 및 유변학적 성질을 SEM, TEM 및 RDS로 측정하였다. EVOH-61/나일론-6 및 EVOH-7/나일론-6 블렌드는 나일론의 조성이 많은 영역에서는 균 일상을 EVOH 조성이 많은 영역에서는 미세한 상분리(약 0.2$\mu$)를 나타내었다. 한편 EVOH-71/나일론-6/12 및 EVOH-71/나일론-12 블렌드는 상분리가 뚜렸하였으며 EVOH조 성이 많은 영역에서는 피브릴화가 나타났다. 낮은 진동주파수 영역에서 점도 대 조성곡선은 전반적으로 양의 편차를 보였으나 Cole-Cole plot은 나일론-6과의 블렌드가 나일론-6/12나 나일론-12와의 블렌드에 배해 유변학적인 측면에서보다 상용성이 우수함을보였다.

  • 기술적으로는 다짐장비의 개선으로 성토다짐의 시공속도가 매우 빨라지고 있으나 국내에서 적용하고 있는 현장다짐 밀도 및 수분함량 측정 방법(KS F 2311 KS F 2306)$^{(1)}$ 은 신속한 측정을 어렵게 하고 있다. 본 연구는 외국에서 개발 현재 활발히 적용되고있는 방사성동위원소를 이용한 함수량측정기개발의 시작단계로 기본설계 결정을 목적으로 한다. 함수량 측정 RI계기의 원리를 먼저 살펴본 후 실험실 내에서 자연건조된 성토용 흙으로 다짐을 하여 공시체를 제작한 뒤, 실험실용 함수량측정 RI계기를 공시체위에 놓고 폴리에틸렌과 중성자 검출기의 개수를 변화시켜가며 일정시간동안 측정개수를 측정한 값을 분석한 결과 목표측정 시간을 1분으로 하였을 때 신뢰측정개수인 10,000개이상을 계측하면서도 경제적으로 최소인 중성자검출기의 개수는 2개, 폴리에틸렌의 두께는 7cm로 결정되었다.

IAA Synthesis and Polyamine Metabolism in Higher Plants: Effects of IAA on the Activity of Diamine Oxidase in Seedlings of Soybean (고등식물에서 IAA합성과 Polyamine 대사에 관한 연구: 대두의 유식물에서 IAA가 Diamine Oxidase의 활성에 미치는 영향)

  • Vie investigated the effect of indole acetic acid (IAA) on diamine oxidase (DAO; EC 1.4.3.6) in the seedling of soybean (Glycine max L. forma Paldalkong). DAO activity was not detected in the resting soybean seeds. During germination it appeared in the elongating zone on day 3, increased up to day 4 and decreased thereafter. Endogenous IAA content has been shown the same pattern as DAO activity. However, cadaverine content was reduced on day 4. To investigate the effect of IAA on DAO activity, the segments of hypocotyl on day 3 were soaked in various concentration of IAA. The activity increased at low concentrations ($10^{-7}-10^{-6}M$) of 1M as compared to that of control but not at high concentrations ($10^{-5}-10^{-4}M$) of IAA. Differing from DAO activity, cadaverine content increased as concentrations of IAA increased. On the other hand, ethylene was induced by IAA at high concentrations. To study the effect of ethylene on DAO activity, we cotreated IAA and 2,S-Norbonadiene (NDE). In this case, DAO activity was not affected.fected.

Enhanced tolerance through increasing polyamine contents in transgenic tobacco plants with antisense expression of ACC oxidase gene (ACC oxidase 발현 억제 식물체에서 폴리아민 생합성 증가에 의한 스트레스 저항성 증강)

  • Antisense construct of cDNA for senescencerelated ACC oxidase (CAO) cDNA isolated from carnation flowers were introduced into tobacco by Agrobacteriummediated transformation. The decreasing expression of NtACO and the reduction of ethylene production were observed in these transgenic lines. In contrast, the SAMDC transcripts and spermidine content were increased. The findings that higher content of spermidine in the ethylene suppressed transgenic plants compared with wild-type should be directly resulted in the enhancement of SAMDC activity followed by the increased accumulation of SAMDC transcript. To investigate the pathogenic response in these transgenic plants, wild-type and transgenic plants were inoculated with Phytophthora parasitica pv. nicotianae. Transgenic plants suppressing ethylene production showed the increased resistance against fungal pathogen, comparing with wild-type plant. PR-protein genes expression in CAO-AS-2 and CAOAS-4 were also higher at the normal growth condition and pathogenic response than in wild-type plants. The results of higher spermidine content and SAMDC activity in transgenic plants, CAO-AS-2 and CAO-AS-4, support the possibility that an increase in spermidine content might induce the higher transcripts of PR-protein genes. This results agreed with the phenomena that spermidine promoted the expression of PR1a and a SAMDC inhibitor, MGBG, decreased the expression of PR1a in leaf discs. These results suggest that the resistance against fungal pathogen in transgenic tobacco impaired in ethylene production might be caused by increasing in polyamine, especially spermidine, biosynthesis.

Modified Atmosphere Packaging of ‘Tsugaru’Apple(Malus domestica Borkh) for Distribution (‘쓰가루’사과의 유통용 신선도유지 MA 포장 효과)

  • Modified atmosphere packaging(MAP) technology was applied to ‘Tsugaru’apple (Malus domestica Borkh) in order to extend the shelf-life of apples during distribution. ‘Tsugaru’apples were packed with the PE film of 40 $\mu\textrm{m}$ thickness(40LD), the PE film modified by the addition of 5% (w/w) zeolite (40CK), and the PE film bags containing the ethylene absorbent (40LP). Quality indices of ‘Tsugaru’apples during storage at 10$^{\circ}C$ were measured in terms of weight loss, soluble solids content, pH, titratable acidity, flesh firmness, peel color and sensory properties. Oxygen, carbon dioxide and ethylene concentration in the film bags changed rapidly at the early stage of storage. Weight loss of the control increased up to 3.0% after 60 days storage while those of the packed apples remained less than 0.6%. No significant differences in soluble solids content and titratable acidity could be found in all the treatments, but significant differences in pH between the control and the packed apples. Higher firmness was kept in 40LD and 40LD than other treatments during storage. Color of the peel changed rapidly in control but slowly in 40LD and 40LP. ‘Tsugaru’apples Packed with Packaging films showed a good visual and sensory quality. Results suggest that packaging treatment with LDPE of 40 $\mu\textrm{m}$ thickness and ethylene absorbent can be used for extending the shelf-life of ‘Tsugaru’apples during distribution.

A Comparison of the Storability in MA Storage and the Quality of Paprika Fruit among Cultivars (MA저장 중 파프리카 과실의 품종별 품질 및 저장성 비교)

  • This study was carried out to compare the storability in MA storage and quality of paprika fruits among 9 cultivars grown in 2011 and 12 cultivars in 2012. 'Mosanto 7044' was the most proper cultivar in red-type paprika has grown in 2011 for MA storage, because it showed relatively higher fruit weight, firmness, soluble solid content, and the lowest ethylene production rate, as well as the second longest shelf life. 'Stayer' was suitable cultivar in yellow-type paprika cultivated in 2011, because it showed higher firmness, soluble solid content and lower ethylene production rate than other cultivars. 'Nagano' was the most proper cultivar in red-type paprika has grown in 2012 for MA storage. Because it showed relatively higher fruit weight, firmness, flesh thickness and the lowest ethylene production rate and decay ratio, although the shelf life of 'Nagano' was the third longest (28 days). 'Freestar' was the excellent cultivar in yellow-type paprika cultivated in 2012, because it showed higher firmness, vitamin C content and lower respiration rate than other cultivars and the longest shelf life of 26 days. The shelf life had a high correlationship with ethylene production rate (r = -0.5504), and soluble solid (r = 0.6112), as comparing with the shelf life and the fruit characteristics of 21 cultivars paprika has grown in 2011 and 2012.

Effect of 1-methylcyclopropene and ethylene-absorbent treatments on quality changes of Prunus mume fruit during storage (1-MCP 처리 및 에틸렌 흡착제가 '백가하' 매실 저장에 미치는 영향)

  • This study was conducted to investigate the effect of 1-methylcyclopropene (1-MCP) and ethylene-absorbent on the qualities of Prunus mume fruit. Prunus mume fruits were stored without film packaging (Cont), packed in LDPE film (Cont-P), and packed with ethylene-absorbent (Cont-PE). Fruits were treated with 1-MCP (1 ppm) for 24 hr at $1^{\circ}C$. After treatment, fruits were packed in LDPE film (MCP-P) and with ethylene-absorbent (MCP-PE) and then stored at $1^{\circ}C$ for 8 weeks. Total soluble solids increased during storage but decreased after 6 weeks while total acidity decreased during storage. Cont was almost completely decayed after 8 weeks of storage while Cont-P, Cont-PE, MCP-PE, and MCP-P were 46, 69, 83, and 5% decayed, respectively. L value decreased but a value increased during storage in all samples. Firmness of peel and flesh of samples decreased gradually for 8 weeks. Respiration rate did not show any significant difference among samples. Ethylene production of Cont showed $0.05{\mu}L/kg/h$ but immediately after 1-MCP treatment, it showed $0.02{\mu}L/kg/h$. Oxalic and malic acids decreased while citric acid increased during storage; fructose and glucose substantially decreased after 8 weeks whereas sorbitol and sucrose increased upto 4 weeks and then decreased thereafter. Based on these results, packing the fruits treated with 1-MCP could extend the freshness of Prunus mume fruit.

  • A functionalization of mesoporous materials with organosilane was carried out via a post-synthesis grafting method and $(n-BuCp)_2ZrCl_2$/methylaluminoxane (MAO) as subsequently immobilized on the functionalized mesoporous materials for ethylene polymerization. Organosilanes having amine, cyano or imidazoline group such as $N$-[(3-trimethoxysilyl)propyl]ethylenediamine (2NS), 4-(triethoxysilyl)butyronitrile (1NCy), 1-(3-triethoxysilylpropyl)-2-imidazoline (2NIm) were used for the surface functionalization of mesoporous materials. In the SBA-15/2NS/$(n-BuCp)_2ZrCl_2$ catalyst preparation, the amount of MAO in feed increased with an decrease in the Zr content of the supported catalyst, and Al content in the supported catalyst increased. The ethylene homopolymerization activity of SBA-15/2NS/$(n-BuCp)_2ZrCl_2$ dramatically increased as the amount of MAO in feed increased. Furthermore, when the immobilization time was 6 hrs, SBA-15/2NS/$(n-BuCp)_2ZrCl_2$ showed the highest activity. The activities of supported 2NS-, 1NCy-, 2NIm-functionalized catalysts decreased in the following order, SBA-15/2NS/ > SBA-15/2NIm/ > SBA-15/1NCy/$(n-BuCp)_2ZrCl_2$. 2NS and 2NIm which have two amine groups per silane molecule were shown to interact with $(n-BuCp)_2ZrCl_2$ strongly compared to 1NCy which has one amine group. Thus, the activities increased with an increase in the nitrogen and the Zr content of the supported catalysts.

Effect of Preharvest Foliar Application of Calcium Chloride on Shelf-life of Red Sweet Pepper 'Ace' (CaCl2 엽면처리가 홍단고추의 저장성에 미치는 영향)

  • This study was conducted to evaluate the effect of calcium chloride treatment (0.1%, 0.3%, and 0.5%) in red sweet pepper 'Ace' on the improvement of shelf-life and the physiological characteristics. $C_2H_4$ production and respiration rates of fruits treated with $CaCl_2$ before harvest decreased during storage of red sweet pepper at $7^{\circ}C$. Ca contents in the leaves and in the fruits showed also a greater increase in treatment of $CaCl_2$ than that in control. No difference was found in total sugar, whereas sucrose content was rapidly reduced after 20 days at $7^{\circ}C$. Ascorbic acid contents were increased by the $CaCl_2$ treatment. As Ca concentration is getting higher, the fruit decay rate was significantly reduced. From this result, we can positively conclude that foliar application with the concentration of 0.3% $CaCl_2$ and 0.5% $CaCl_2$ before harvest is effective in improving storage quality in red sweet pepper.
