Numerical Taxonomy of Cirsium japonicum Complex(Asteraceae) in Korea (한국산 엉겅퀴군(국화과) 식물의 수리분류학적 연구)

  • Patterns of morphological variation in Cirsium japonicum complex were examined using numerical analysis, and the delimitation of taxa was evaluated. Principal components analysis of taxa using 24 morphological characters and 11 leaf characters revealed the presence of two major groups; C. japonicum var. japonicum and C. japonicum var. spinosissimum. Circium japonicum f. nakaianum could be included in the category of C. japonicum var. japonicum. In conclusion, Korean C. japonicum complex was composed of two varieties and one form, C. Japonicum var. japonicum, C. japonicum var. spinosissimum, and C. japonicum var. spinosissimum f. alba.

Taxonomic Identities of Cirsium setidens, C. chanroenicum and C. toraiense (고려엉겅퀴, 정영엉겅퀴 및 동래엉겅퀴의 분류학적 실체 검토)

  • Numerical analysis using morphological characters was done in order to clarify taxonomic circumscriptions and identities of Korean endemic, Cirsium setidens (Dunn) Nakai, Cirsium chanroenicum Nakai, and Cirsium toraiense Nakai ex Kitam.. Principal components analysis of C. setidens and C. chanroenicum using 29 morphological characters and 12 leaves characters revealed one group which could not separate. Identity of C. toraiense could not be confirmed by the destruction of habitat, similarity of morphological diagnostic characters between many specimens identified C. toraiense including type specimen and C. japonicum var. ussuriense. In considering the variational patterns of morphological characters, it is strongly suggested that identities of C. setidens f. alba, C. setidens var. niveo-araneum, and C setidens var. pinnatifolium should be recognized, but C. chanroenicum var. lanceolata should be included to circumscription of C. setidens.

Phylogenetic and Chemical Analyses of Cirsium pendulum and Cirsium setidens Inhabiting Korea (국내에 자생하는 큰엉겅퀴와 고려엉겅퀴의 분자유전학적 및 화학적 분석)

  • Cirsium pendulum plants were collected from Hongcheon, Pyeongchang, Wonju, Yangyang in Kangwondo, Gapyeong in Gyeongkido, and Choongju in Choongcheongbukdo. Cirsium setidens plants were collected from Taebaek in Kangwondo and Bonghwa in Kyeongsangbukdo. Genomic DNA was prepared from those plants and used for the amplification of 18S rDNA, ITS1, 5.8S rDNA, ITS2, and part of 28S rDNA. The PCR products were sequenced, and the sequence was deposited in the GenBank. The comparison of those sequences has revealed that the rDNA sequences are identical for all six C. pendulum plants, but that the ITS1 and ITS2 sequences contain variable nucleotides. The two C. setidens plants had different nucleotides in 18S rDNA, ITS1, and ITS2. The comparison of the DNA sequences of C. pendulum and C. setidens collected in this study with C. pendulum of Hokkaido in Japan and C. japonicum of Anhui in China indicated that the plants of those three species are clearly divided into three distinct groups. The silymarin content of the collected plants was analyzed and turned out to be quite high. Therefore, it has been found that both C. pendulum and C. setidens plants are producing large amounts of silymarin, which has been reported to have various medicinal effects.

Cirsium japonicum var. maackii Protects Against Osteoarthritic Development by Cartilage Degradation (엉겅퀴의 연골 분해에 의한 골관절염 진행의 억제 효과)

  • 골관절염은 연골 파괴 및 연골 형성의 손상을 초래하는 산화 스트레스 세포사와 관련된 염증성 질환이다. 최근에, 엉겅퀴(Cirsium japonicum var. maackii)는 다양한 염증성 질환에 대하여 보호하는 역할을 하는 것으로 보고되었습니다. 그러나, 연골 퇴행 및 골관절염 진행에 대한 엉겅퀴의 역할은 아직 알려져 있지 않았다. 따라서, 본 연구는 골관절염 및 연골 분해 생쥐 모델에서 엉겅퀴의 보호 효과를 조사하였다. 먼저, 엉겅퀴의 활성 성분 함량을 측정하기 위해 총 폴리페놀 함량과 총 플라보노이드 함량 분석을 수행하였다. 그 결과, 건조된 엉겅퀴 지상부의 물 추출물에서 총 폴리페놀 함량은 $149.2{\pm}24.1mg\;GAE/g$ 및 총 플라보노이드 함량은 $27.9{\pm}2.0mg\;NE/g$을 함유하는 것으로 밝혀졌다. 또한, HPLC 분석으로부터 엉겅퀴 지상부 물 추출물의 주요 화합물은 플라보노이드 계열인 cirsimarin과 cirsimaritin으로 확인되었다. 또한, 엉겅퀴 추출물은 조직 병리학적 분석에 의해 입증된 내측 반월판의 불안정화에 의해 유도된 골관절염 마우스 모델에서 연골파괴 억제효과를 나타냈다. 결론적으로, 본 연구결과는 엉겅퀴 추출물이 골관절염과 연골 파괴를 개선 또는 예방할 수 있는 새로운 식 의약 소재로의 개발 가능성을 제시한다.

Genetic Relationship of Some Cirsium Plants of Korea (국내에 자생하는 일부 Cirsium 속 식물들의 분자유전학적 유연관계 분석)

  • Cirsium chanroenicum, Cirsium nipponicum, and Cirsium schantarense plants were collected from Changwon, Ulleungdo, and Dooryoon Mountain, respectively. Cirsium japonicum plants were also collected from various locations in Korea. Genomic DNA was prepared from the collected plants and used for amplification of the 18S rDNA, ITS1, 5.8S rDNA, ITS2, and part of 28S rDNA. The ITS1 and ITS2 sequences of the PCR products and from other Cirsium plants reported previously were aligned and compared. Cirsium chanroenicum, Cirsium nipponicum, and Cirsium setidens formed distinct branches on the neighbor-joining tree. Cirsium japonicum and Cirsium pendulum appeared to be close to one another, but Cirsium pendulum plants were clearly clustered in an independent clade. Cirsium shantarense was clustered with the other Cirsium japonicum plants. The most important characteristic that distinguished these two species was the direction of the flowers. All Cirsium japonicum flowers point upward, but Cirsium shantarense flowers point downward. Other than this feature, these two species are almost indistinguishable morphologically. Cirsium chanroenicum is indistinguishable morphologically from Cirsium setidens, but it still formed a distinct group on the neighbor-joining tree based on ITS sequences, suggesting that this species is worth considering as an independent species. Silymarin production of the collected plants was analyzed and appeared to be quite high, indicating that the ability to synthesize silymarin is common to all Cirsium plants analyzed so far.

Development of Functional Tea Product Using Cirsium japonicum (플라보노이드 함유 엉겅퀴를 이용한 기능성 다류 개발)

  • The antidepressant effects of Cirsium japonicum was revealed in previous studies using open field test and forced swimming test. The contents of total flavonoids and apigenin of Cirsium japonicum were analyzed and sensory evaluation of Cirsium japonicum tea was performed in order to develop Cirsium japonicum tea as an antidepressant. Among the different plant parts of Cirsium japonicum, leaves had the highest level of total flavonoids and apigenin contents and were followed by flowers and stems. Drying method affected total flavonoids and apigenin content of Cirsium japonicum, but regular pattern was not revealed. In sensory evaluation, overall acceptance of Cirsium japonicum flower-leaf mix tea was higher than those of Cirsium japonicum leaf tea. Also purchase intention of Cirsium japonicum flower-leaf mix tea was higher than those of Cirsium japonicum leaf tea.

Effects of Cirsium japonicum Powder on Plasma Glucose and Lipid Level in Streptozotocin Induced Diabetic Rats (엉겅퀴 섭취가 Streptozotocin 유발 당뇨 흰쥐의 혈당과 지질수준에 미치는 영향)

  • This investigation was conducted to assess the influence of Cirsium japonicum consumption on the plasma glucose and lipid profiles in streptozotocin (STZ)-induced diabetic rats. Diabetes mellitus was induced in male Sprague-Dawley rats weighing 230 g by injection of STZ into the tail vein at 45 mg/kg body weight. The rats were randomly assigned into four groups: a normal and STZ-control fed an AIN-93 diet group, and diabetic groups whose diets were supplemented with 10% Cirsium japonicum powder containing leaf or root for four weeks. To observe the effects of Cirsium japonicum in the animal model, the levels of glucose, cholesterol, HDL-cholesterol, triglyceride and free fatty acid in the plasma and the levels of glycogen in the tissue were determined after four weeks. Treatment of STZ-induced diabetic rats with leaf consumption significantly lowered the diet intake when compared with STZ-control rats. The relative weights of the liver and kidney differed significantly between the normal and diabetic groups. The weights of the kidneys in the leaf group were significantly lower than those in the STZ-control group. The level of hematocrit was also significantly lower in diabetic rats whose diets were supplemented with leaves when compared to those of STZ-control rats. The plasma glucose level was found to be significantly lower in the leaf group than the STZ-control group. The effect was significant after 2 weeks. The HDL-cholesterol levels increased in all of the diabetic experimental groups when compared to the STZ-control group. These results suggest that supplementation with Circicum japonicum leaves induced considerable hypoglycemic effect in STZ-induced diabetic rats and that these leaves may be useful for the management of diabetes mellitus.

Antioxidant Activity and Protective Effects of Cirsium japonicum against Damaged Mouse Liver Cell (BNL CL.2) (엉겅퀴의 항산화 활성 및 손상된 흰쥐 간세포(BNL CL.2)에 대한 간 보호 효과)

  • We analyzed the content of total phenolic and silymarin compounds of Cirsium japonicum (CJ), and its antioxidant activities and Liver protective effects were compared with those of Silybum marianum (SM). The total phenolic content in the aerial part ($97.22{\pm}5.51mg/g$) of CJ is higher than that in the underground part ($85.32{\pm}3.06mg/g$). The total silymarin content of CJ was 55.56% of SM, with the underground part ($0.47{\pm}0.03mg/g$) having higher content than the aerial part ($0.18{\pm}0.02mg/g$). The antioxidant activity of CJ was generally slightly lower than that of milk thistle, and the underground part of CJ generally had higher activity compared to the aerial part. When CJ extracts were processed at 1 mg/ml, DPPH activities were $83.76{\pm}0.60$ and $88.28{\pm}0.17%$, and FRAP activities were $77.63{\pm}0.70$ and $82.83{\pm}0.39%$ for extracts from aerial part and underground part, respectively. ABTS activities were $68.60{\pm}1.24$ and $63.41{\pm}0.57%$ for underground and aerial part respectively when extracts were processed at 0.1 mg/ml. The Liver protective effects of CJ were higher in the extracts from underground part compared to the aerial part, Liver cells were damaged by treating them with t-BHP, $H_2O_2$ and Ethanol, and then they were treated with 0.2 mg/ml CJ extracts. The survival rates of the damaged liver cells were $49.58{\pm}0.34$, $76.87{\pm}1.10$ and $71.73{\pm}0.58%$ respectively, which were higher than the cells not treated with extracts.

Cirsium japonicum Extracts Show Antioxidant Activity and PC12 Cell Protection against Oxidative Stress (좁은잎 엉겅퀴 추출물의 산화방지 활성 및 산화적 스트레스에 대한 PC12 세포 보호효과)

  • The phenolic compounds, antioxidant activity and neuronal cell protective effect of Cirsium japonicum extract were evaluated in this study. High performance liquid chromatography mass analysis showed that C. japonicum was composed of chlorogenic acid, linarin, and pectolinarin. C. japonicum extract showed its antioxidant activity with half-maximal inhibitory concentrations of 567 and $130{\mu}g/mL$ by DPPH and ABTS radical scavenging activity, respectively. The total antioxidant capacities of C. japonicum via DPPH, ABTS, and FRAP assays were 11.32, 100.15, and $12.76{\mu}g/mL$ trolox equivalents, respectively. In addition, the neuroprotective effect of C. japonicum extract was investigated by measuring cell viability via MTT, LDH and DCF-DA assay using $H_2O_2-damaged$ PC12 cells. C. japonicum extract showed neuronal cell protective effects in a dose-dependent manner. These results indicated that C. japonicum extract has potent antioxidant and neuronal protective effects. Therefore, C. japonicum can be regarded as an effective and safe functional food resource as natural antioxidants, and may decrease the risk of neurodegenerative disorders.

The Inhibition of UVA-induced Matrix Metalloproteinase-1 in Human Dermal Fibroblasts and the Improvement of Skin Elasticity by Cirsium setidens Extract (고려엉겅퀴 추출물의 사람 섬유아세포에 있어서 자외선으로 유도된 MMP-1발현 저해와 피부 탄력 개선 효과)

  • In this study, we measured the anti-oxidative activity of Cirsium setidens and investigated its effect on UVA-induced MMP-1 expression in human dermal fibroblats. And then we examined possible improvement in skin elasticity by topical treatment with fomular including Cirsium setidens extract. The ethanol extract of C. setidens showed free anion radical scavenging effect(87.47 % at 1 mg/mL) and superoxide anion radical scavenging effect(61.71 % at 1 mg/mL) in the xanthine/xanthine oxidase system, respectively. At the concentration of 100 ${\mu}g/mL$, C. setidens extract showed 95.54% inhibition on lipid peroxidation of linoleic acid. UVA-induced MMP-1 expression in human dermal fibroblasts was reduced to 54.69 % by treatment with 100 ${\mu}g/mL$ of C. setidens extract. A human clinical study, in which oil-in-water emulsion with C. setidens extract was topically applied, showed significant increase in skin elasticity. These results suggest that the C. setidens extract can be effective anti-aging ingredient for cosmetics applications.
