Phytoremediation of Soils Contaminated with Heavy Metal by Long-Term Cultivation (장기재배 시험에 의한 중금속 오염토양의 식물정화)

  • In order to select proper plants for phytoremediation at heavy metal contaminated areas, eight species of non-edible plants were cultivated at the heavy metal contaminated soils near a metal smelter. The content of the absorbed heavy metals (Cd, Cu, Pb and As) at different part of the plants were analyzed. Plants included five tree species (Populus nigra x P. maximowiczii, Euonymus japonica, Acer palmantum, Celtis sinensis, Buxus microphylla), two flower species (Rhododendron lateritium, Calendula officinalis), and lawn (Zoysia japonica). Biomass yield of tree species was higher than those of flower or lawn species. Heavy metals were highly accumulated in roots compared to those In leaves and stems. The concentrations of Cd, Cu, Pb, and As in Buxus microphylla were greater than those in other plant species. Total absorbed Cd and Pb contents, from high to low by each plant in experimental plots were in the order of Populus nigra x P. maximowiczii, Celtis sinensis and Acer palmantum. They were Celtis sinensis, Populus nigra x P. maximowiczii and Buxus microphylla for Cu, and Buxus microphylla, Acer palmantum and Populus nigra x P. maimowiczii for As. It was estimated that among eight plant species used in the experiment Populus nigra x P. maximowiczii, Buxus microphylla, Acer palmantum, and Celtis sinensis were the most effective species for phytoremediation in the heavy metals polluted areas considering biomass yield and heavy metal uptake.

Toxicity, Uptake, and Transformations of Atrazine by Hybrid Poplar Tree

  • 국내에서 자생하는 포플러중 현사시, 양황철, 이태리포플러를 대상으로 제초제인 atrazine의 제거특성을 조사하였다 Atrazine이 포플러에 미치는 독성을 평가하고, 식물체에 의한 atrazine 제거효율 및 제거속도를 조사하였다. 또한 식물체의 체내로의 흡수에 의해 atrazine이 제거되므로 식물체로 흡수된 후의 생물학적 변환 과정에 대해서도 조사하였다. 현사시, 양황철, 이태리포플러 모두 실험이 진행된 전범위의 농도에서 황백화현상, 낙엽증가, 생체량 감소와 같은 식물독성을 나타냈으며, 현사시인 경우 양황청 및 이태리 포플러에 비해 상대적으로 민감한 반응을 나타내었다. Atrazine 독성여부 평가의 하나로 초기 atrazine 농도가 증가할수록 현사시, 양황철, 이태리포플러의 증산량은 상대적으로 감소함을 관찰하였다. 포플러에 의한 atrazine 생물학적 변환실험에서는 atrazine이 중간생성물질인 HA, DEA, I)IA를 거쳐 ammeline을 포함한 HDAP로 전환되는 것을 확인하였다. 이 결과의 중요성은 atrazine 부산물은 상대적으로 독성이 적고, 미생물에 의해 어렵지 않게 완전 무기화할 수 있기 때문이다. 즉, 식물체내에서도 HBAP 이후 과정을 통한 분해가 이루어질 수 있는 가능성이 있다고 추측할 수 있다.

Study on the Crystals in the Populus spp.(I) - Crystals in the Woods and Barks of Populus maximowiczii and Populuss nigra × maximowiczii (포풀라류의 결정에 관한 연구(I) - 황철나무와 양황철나무의 목부 및 수피종의 결정)

  • Crystals in the woods and barks of Populus maximowiczii and Populus nigra ${\times}$ maximowiczii were examined by light microscopy, scanning electron microscopy and soft X-ray. Populus examined had the wetwood and the heartwood of Populus showed higher moisture content than in adjacent sapwood. The crystals can be detected clearly by the soft X-ray due to their higher densities than the wood. Crystals in xylem wood of Populus were found mainly at heartwood. However, they were concentrated in specific areas such as in color-stained area or along the specific annual rings. Crystals showed the highest occurrence around the pith. Within a growth ring the amount of crystals in earlywood was more abundant than in latewood. Crystals were observed mainly at vessels and fibers although they occurred at all kinds of cells in heartwood. Crystals in the bark were scattered evenly both in inner bark and in outer bark. Populus nigra${\times}$maximowiczii showed higher frequency of leaf knots than Populus maximowiczii.

Factors Effecting Agrobacterium Mediated Transformation and Regeneration of Populus nigra × P. maximowiczii (Agrobacterium tumefaciens에 의한 양황철나무의 형질전환(形質轉換) 요인(要因))

  • We have demonstrated expression of bacterial genes transferred into cells of Populus nigra ${\times}$ P. maximowiczii by A. tumefaciens strain 6044 (pGA 472). We determined the optimum concentration of kanamycin sulfate for effective selection of punctured leaf transformed using Agrobacterium binary vector pGA 472 containing a neomycine phosphotransferase gene (NPT-II) which confers kanamycin resistance. The combination of cefotaxime (200mg/l) and carbenicillin (300mg/l) showed good performance of discarding Agrobacterium from inoculated punctured leaf. A relatively low concentration (10mg/l) of kanamycin sulfate inhibited callus and shoots induction from punctured leaf. Number of shoots regenerated from co-cultured punctured leaf was 3.0 on MS basal medium supplemented with 10 mg/l kanamycin sulfate, while that of not co-cultured punctured leaf was none. The regeneration rate was 10% from the punctured leaf co-cultured on MS medium with 10 mg/l kanamycin. Regenerated shoots are developing from micropropagation for Southern blot analysis and inheritance of the kanamycin resistance trait (NPT-II).

Studies on the Phytoextraction of Cadmium and Lead Contaminated Soils by Plants Cultivation (토양중 카드뮴과 납의 Phytoextraction을 위한 식물재배 연구)

  • In order to select more proper plants for phytoextraction at the heavy metal polluted areas, 11 species of non-edible plants were cultivated at the cadmium(Cd) and Lead(Pb) treated soils and analyzed the content of the absorbed Cd and Pb in each part of plants. Plants include three fibers(Linum usitatissimum, Cannabis sativa, Gossypium spp.), three flowers(Calendula officinalis, Rhododendron lateritium, Portulaca grandiflora), and five trees(Pinus thunbergii, Magnolia kobus, $Populus\;nigra\;{\times}\;P.$ maximowiczii, Euonymus japonica, Fraxinus rhynchophylla). Yield of tree species were higher than that of fiber and flower species. Cd and Pb were highly accumulated in root rather than leaves and stems. The Cd content of plants was in the order Portulaca grandiflora > Calendula officinalis > Gossypium spp. > Linum usitatissimum, Pb was Cannabis sativa > Linum usitatissimum > Fraxinus rhynchophylla. Total absorbed Cd by each plant was in the order $Populus\;nigra\;{\times}\;P.$ maximowiczii > Euonymus japonica > Rhododendron lateritium, but Pb was $Populus\;nigra\;{\times}\;P.$ maximowiczii > Rhododendron lateritium > Euonymus japonica. Total absorbed Cd and Pb contents in plants were negatively correlated with the residual Cd and Pb in the treated soils. It was estimated that $Populus\;nigra\;{\times}\;P.$ maximowiczii, Euonymus japonica, Fraxinus rhynchophylla, and Rhododendron lateritium were the most effective species for phytoextraction in the polluted area considering yield and heavy metal uptake.

  • Above ground biomass production of one-year-old hybrid poplars was investigated to select the best clones which are suitable for short rotation forestry. The results were as follows: 1. Average dry weight of Populus alba X P. glandulosa Fl clones was 2.86 for stem,0.41 for branches 0.70 for leaves and 3.97 t/ha for total. and the ratios of green weight to dry weight were 0.47 for stem,0.36 for branches,0.31 for leaves and 0.42 for total. Dry weight of Populus nigra X P. maximowiczii Fl clones was 1.48 for stem, 0.54 for branches.0.71 for loaves and 2.73t/ha for total, and the ratios of green weight to dry weight were 0.42 for stem. 0.37 for branches, 0.28 for leaves and 0.36 for total. 2. Biomass production (green and dry weight)of Populus alba X P. glandulosa F1 clones can be predicted by diameter measurement only, but biomass production of Populus nigra X P. maximowiczii F1 clones can not be predicted by any of investigated characteristics. 3. Dry weight distribution of pepulus alba X P. glandulosa Fl clones were 73$\%$ for stem, 10$\%$ for branches and 17$\%$ for leaves, and dry weight distribution of P. Populus nigra X P. maximowiczii F1 clones were 54$\%$ for stem, 20$\%$ for branches and 26$\%$ for leaves.

Phytoremediation Study of Disel Contaminated Soil by Indigenous Poplar Tree (국내 자생 포플러나무에 의한 디젤오염토양 정화특성 연구)

  • In this study, uptake and toxicity of disel (TPH) by poplar specie, $P.\;nigra{\times}P.\;maximowiczii$ were assessed in laboratory soil column experiments. Poplar cuttings were grown for 2 months and exposed to various concentration (0, 200, 500, 1000, 2000 mg/kg) of disel for a period of 60 days. For disel removal experiments, disel was effectively removed in the range of lower concentration. but, the removal rate of disel was rapidly decreased as increasing initial disel concentrations. For the this reason, toxicity effetcs were evaluated by measuring in poplar cutting mass variation and monitoring transpiration. Exposure on higher disel concentration resulted in decrease of biomass and transpiration accompanied by chlorosis and abscission, indicating toxic effect of disel on the poplar tree. And also, we have observed that both removal efficiency of disel and the microbial activity were higher at the bottom of the soil column. It was suggested that the plant formed the root zone at contaminated soil, stimulated microbial activity by plant root exudates, and played an important role in enhanced biodegradation of disel.

  • To study the wood quality factors of Populus nigra $\times$ maxmowiczii known a rapidly growing species, the variations of green moisture contents, bulk density, wood fibre dimensions, microfibril angles, and number of leaf knot in stem wood were investigated. The heartwood contained a higher moisture content than the corresponding sapwood. Bulk density in radial patterns variations decrease outward from the pith, then increase toward the bark. The wood-fiber length and diameters had somewhat smaller values than on Populus alba $\times$ glandulosa or Populus euramericana. The microfibril angles decreased rapidly toward the outside, and their mean values were about 16 degree. The grain angles run nearly parallel to the cell axies. Number of leaf knot showed a fluctural change above ground level to a point near the base of the crown and then increased rapidly to the top of tree and average number of leaf knot varied exclusively from tree to tree.

  • In order ot obtain the information on wood quality of the Populus nigra $\times$ maximowiczii the distributions of crystalliferous substancess, ty loses and tension woods were examined through light microscopy or scanning electron microscopy. 1. The crystalliferous subtance, presumably calcium oxalate. partially occurred in vessel elements of heartwood, and sporadically in ray parenchyma, but not in the wood fibers The types of crystalliferous subtances were mainly crystal sand, $2-3{\mu}m$ in diameter. 2. The tyloses arranged in a ladder-like series were sporadically formed in the vessel elements of heartwood. 3. Although the woods were cut from the straightly grown stem without any evident disorientation with respect to gravitational axis, single or group of gelatinous fibers were irregularly distributed among normal wood fibers.

In Vitro Selection for Salty Tolerance of Populus nigra ${\times}$ P. maximowiczii (포푸라 내염성(耐塩性) 개체(個體)의 기내선발(器內選拔))

  • The concentration of 50 mM Nacl inhibited the shoot growth of P. nigra ${\times}$ maximowiczii in viro culture. The punctured leaves have produced so many individuals on MS basal medium with cytokinin. Especially MS basal medium with BAP 0.8mg/l showed the best shoot performance in which the average number of shoots were 127.6. For selection of NaCl tolerance shoot of poplar, punctured leaves were inoculated on MS basal medium with BAP 0.8mg/l and various concentrations of NaCl(from 10 mM to 100 mM graded by 10 mM). On the medium with over 50mM of NaCl, 13.7 to 15.7 shoots were obtained. Especially on the medium with go mM and 100mM. 10.7 and 8.3 shoots, respectively. The shoots derived from control medium (non-NaCl) were depressed growth, while the selected shoots from MS with NaCl showed good growth performance on MS basal medium with 50 mM of NaCl. From this results, we suggested that the possibility of in vitro selection to tolerance for inorganic salts in forest tree species.
