Immune Response of the Japanese Eel(Anguilla japonica) to Vibrio anguillarum (Vibrio균에 대한 뱀장어 (Anguilla japonica)의 면역반응)

  • Some eels Anguilla japonica, ranging from 16 to 23 g in their weight(average: 20 g), were sampled at the private eel farming company equipped with water recycling system, located at Kimhae city, Kyungnam Province, Korea. Three kinds of vaccine were prepared with Vibrio anguillarum (EPM-8406) isolated at National Fisheries University in Korea for the immune response experiment against eels; those vaccines were made by inactivating the strain with $0.3\%$ formalin for 24 hrs at $25^{\circ}C$, heating for 3 mins or for 15 mins at $121^{\circ}C$, respectively. The various optimal vaccination conditions for the control of vibriosis in the fish were investigated based on the cultivation temperature, vaccination concentration and booster effect. The maximum titer rapidly increased with higher temperature up to $23^{\circ}C$, but there were little differences between $23^{\circ}C\;and\;28^{\circ}C$. The formalin-killed vaccine showed good efficacy at the injection concentration of above $10^8$ cells per fish and little effect at the below $10^7$ cells. The booster effect on the vaccination showed good efficacy above twice-injections with little difference between the numbers of injection.

  • Field survey on the variational characteristics of water quality and chlorophyll a concentraion were carried out at the 25 stations for four seasons in Kogum-sudo(Straits) southern coast of Korean Peninsula from Feburuary to October in 1993. We made an analysis on biological factor as chlorophyll a concentraion as well as physicio-chemical factors such as water temperature salinity sigma-t transparency dissolved oxygen(DO) chemical oxygen demand(COD) nutrients (ammonia, nitrite, nitrate, phosphate, and silicate) N/P ratio and suspended solid(SS). The waters in the Kogum-sudo were not stratified due to the tidal mixing. And the high productivity in photic layer were supported by high nutrients concentration from bottom waters. The high concen-trations of suspended solid in straits had a bad influence upon marine biology of nature and cultivations. In Kogum-sudo had a sufficient nutrients for primary productivity during a year. Especially phosphate and inorganic nitrogen were high the other side silicate was very low. The source of nutrients supply depend on rather mineralization of organic matters and input of seawater from outside than input of freshwaters from lands. Phytoplankton biomass as measured by chlorphyll a concentratiion was very high all the year round and it was controlled by the combination o f several environmental factors especially of phosphate in summer and dissolved nitrogen in other seasons.

Isolation of Ammonia Oxdizing Bacteria and their Characteristics (암모니아 산화세균의 분리와 그 특성)

  • This study was carried out far isolation and characterization of ammonia oxidizing bacteria (AOB) from aquacultural place and sludges of waste water collected in Pusan. One autotrophic AOB, Nitrosomonas sp. and 8 heterotrophic AOB (2 strains of Bacillus sp., 2 strains of Acinetobacter sp., Xanthomonas sp., Alcaligenes sp., Pseudomonas sp., Sphingobacterium sp.) were isolated. and identified. Variation of total nmmonia nitrogen (TAN) and $NO_2-N$ in mineral salt media containing 10mg/ $\ell$ of NHCl for 15 days in differents 9 strains was measured in order to examine the ablitity of ammonia oxidation. TAN was started to reduce after 4 days incubation and ca. 2 mg/$\ell$ of TAN was decreased after 15 days incubation by Nitrosomonas sp., At that time, $NO_2-N$ was produced to 0.023$\~$0.036 mg/$\ell$. Heterotrophic AOB showed the low ability of ammonia oxidation, 0.02$\~$0,04 mg/$\ell$ of TAN was decreased and $NO_2-N$ was produced to 0.01$\~$0.51 mg/$\ell$ after 15 days. When each strain of 8 heterotrophs was incubated in mimeral salt media containing 10 mg/$\ell$ $NH_4Cl$ and 50 mg/$\ell$ glucose, and 50 mg/$\ell$ $NH_4Cl$ and 5 g/$\ell$ glucose, the diminution of TAN was 1.12$\~$3.38 mg/$\ell$ and 1$\~$20 mg/$\ell$, respectively.

  • Rotating Biological Contactor (RBC) was tested for the removal of total ammonia nitrogen (TAN) by using simulated aquaculture system. RBC performance was evaluated by controlling revolution rate of disk and hydraulic residence tile (HRT). The optimum revolution rate of disk was 4 rpm, As HRT of RBC was increased, TAN removal efficiency of RBC and TAN concentration of rearing water were increased. HRT for maintaining lowest TAN concentration of rearing water was 9.5 minutes and at that condition TAN concentration of rearing tank was $1.03 g/m^3$

Relationship between the Catches of Tuna and Oceanographic Conditions in the South-East Pacific (남동 태평양의 참치 어획양과 해양환경과의 관계)

  • The relationship between the catches of tuna and hydrographic conditions in South-East Pacific region (latitude $5^{\circ}N-12^{\circ}S$, longitude $135^{\circ}W-115^{\circ}W$) was investigated by using the catch data of tuna and Digital Bathythermograph (DBT) data from December 9, 1980 to April 2, 1981. The results are as follows : The study area were located in South Eguatorial Current regions including equatorial upwelling regions in $5^{\circ}N\;to\;12^{\circ}S$. The horizontal mean temperature at the depth of 10m on the first quarter months in the study area was about $25^{\circ}C$C and the salinity of those fishing areas ranged from 34.8 to $35.0\%_{\circ}$. Yellowfin tuna and bigeye tuna were mainly caught in SW vertical temperature profile type, which the depth of thermocline ranged from loom to 300m, and temperature difference of thermocline was about $12^{\circ}C$. The deeper the depth of thermocline, the more the catches of tuna. While albacore tuna was caught well in SS vertical temperature profile type which the temperature of thermocline ranged from $9^{\circ}\;to\;26^{\circ}C$ and its gradient was very smooth. The depth of 1 ml/l surface of dissoved oxygen content ranged from loom to 200m in the South-East Pacific between longitude $140^{\circ}W-100^{\circ}W$, but it was shallower than 100 m near the North-South American continent. The catches of bigeye tuna were larger than those of yellowfin tuna in South Equatorial Current region. As approaching to the South and North American continent, the catches of yellowfin tuna and bigeye tuna decreased because the thermocline becomes shallower and steeper and the depth of the 1 m1/1 surface of dissolved oxygen content became shallower.

The Effect of Variations in the Tsushima Warm Currents on the Egg and Larval Transport of Anchovy in the Southern Sea of Korea (한국 남해의 대마난류 변동이 멸치 난$\cdot$자어의 연안역 수송에 미치는 영향)

  • The relationship between the transport of eggs and larvae of Anchovy (Engraulis japonica) and the oceanic condition in the southern sea of Korea was examined on August and November 1996. In summer (August), when the Tsushima Warm Current is strong near to the coast, the warm waters such as warm streamers from the Tsushima Warm Current intrude into the coastal area, and cyclonic circulations are formed. The warm water intrusions also generate wakes around Komun Island, Sori Island and Koje Island. In the coastal area where the warm water intrusions occur, the nutrients, dissolved oxygen, suspended solid and chlorophyll are concentrated in probably relation to the upwelling concerned with this warm streamer and/or the wakes. Anchovy eggs and larvae are transported to the coastal area by the cyclonic circulations. The hatching and growth of anchovy larvae are increased because of high primary production in the cyclonic circulations. However, as the amount of Copepods which are a main food for anchovy larvae decrease in the coastal area, anchovy larvae seem to move to the Isushima Warm Water area for seeking a prey. In autumn (November), the Tsushima Warm Current is far away from the coast. In this season the warm water intrusions almost disappear, and the small scaled frontal eddies are formed between the coastal water and the Tsushima Warm Water. As the surface water moves towards offshore, few anchovy eggs and larvae were sampled in the survey area. Chemical and biological substances are concentrated in the leftdown sides of the small scaled frontal eddies because of eddy formation.

Effect of Daily and Alternate Day Feeding Regimens on Growth and Food Utilization by Juvenile Flounder Paralichtys olivaceus (일일 및 격일 습사료 공급방법이 육성 넙치의 성장과 영양소 이용효율에 미치는 영향)

  • Two groups of juvenile flounders (90 g/each) were fed on moist pellet feed thrice a day for a period of 8 weeks; the first group, reared in 4 circular (6m $\times$ 0.8 m) tanks (1200 flounders/tank), was fed daily (ED) and the second one on alternate days (EO). There was no significant difference in growth and feed conversion ratio (FCR) between these groups. There was also no difference in chemical composition, except in lipid, which was significantly higher (4.1%) in the ED group than the EO group (3.4%). Food intake and gain in the whole body nitrogen (N) were higher in the ED group but there was no significant difference between these groups in N retention; they excreted almost equal amount (114 vs 112 g) of N. Although phosphorus intake was significantly different between these groups, the flounders retained equal amount (0.5 g) of P in their body and excreted also equal amount (27.6 vs 26.0 g/kg weight gain) of p. Energy intake (2528 kJ) of the ED group was higher than that (2116 kJ) of the EO group. However, there was no significant difference in their energy retention efficiency. Clearly, alternate day feeding regimen led to more efficient utilization of food by the juvenile flounder.

On the Efficiency of Soybean Meat as a Protein Source Substitute in Fish Feed for Common Carp (잉어 사료 단백질 대체원으로서의 대두박분의 효율)

  • An experiment on the efficiency of soybean meal substituted for fish meal in the diet of common carp was carried out at the Fish Culture Experiment Station of the National Fisheries University of Pusan from June 27 to August 25, 1983. The carp used for the experiment were 50 fish each lot, each averaging 26 g in initial weight (total weight 1300 g). The experimental diets contained $10\%,\;20\%,\;30\%$, and $40\%$ soybean meal ($44\%$-protein) at iso-protein content basis in place of white fish meal in the control diet which contained $35\%$ protein. The growth rate in the control division was highest with progressive decrease in performance of the fish with increasing rates of soybean meal substituted in the test diets. Nevertheless, the $10\%\;and\;20\%$ soybean meal test diet groups showed acceptable conversions and growth rates, i. e. feed coefficients being 1.28 and l.30 and daily growth rates, $1.708\%\;and\;1.694\%$ compared to $1.22\%\;and\;1.758%$ for the control group, respectively. Therefore, soybean meal could be substituted for fish meal in carp diet by $20\%$ without significant differences of performance with some saving in feed cost.

A Study on the Management of the Indian Ocean Tuna Fishing Grounds (인도양 다랭이어업의 어장관리에 관한 연구)

  • Tuna and tuna fishing extend over most of the tropical waters of the world oceans, especially, this study of the Indian Ocean and the fish and the fishing vessels are highly mobile. No country can deal in isolation with the problems of its tuna fishery and of any tuna fishery in its waters, without collaborating with other countries. This cooperation is often best established through some formal international mechanism. The essential requirements are for information-on the resources, the fishery, the trade-and to identify where management actions are needed to consore the resources or to maintain the economic or social function of the fishery. These will also usually require some form of international mechanism. With the changes in the Law of the Sea, the siuuation in respect of management of tuna-considered among the highly migratory species, requirng special treatment-has also changed. At present there are both uncertainty regarding tuna management and political arguments as to how such management can be best implemented. Bearing this in mind, together with the fact that only few tuna stocks have shown clear evidence of serious depletion through over-exploitation, present emphasis will be placed on information requirements. This study included compiling the data necessary to review the state of stoks in the Indian Ocean tuna fishing grounds. Information on the resources-where the fish are, their quantity, their movements, etc. is important, whether in planning development of new fisheries, or considering the need for collaboration with other countries (in ascertaining to what extent their catches may affect catches of the natoinal fleet) or for conservation measures. A major source of information is the statistics of for conservation measures. A major source of information is the statistics of catch and fishing effort from existing commercial vessels. These need to be assembled for all fisheries on the same stock on an oceanwide basis. The statistical data also need to be analysed and intepreted, and combined with biological data(e.g., on growth, mortality and migration rates) to provide information that is intelligible and useful for administrators and other decision-makers. This must also be undertaken on a resource-wide or ocean-wide basis. Finally, because of the world-wide similarities in the methods of datching, processing and marketing fish, there is a particular tut not exclusive need in those countries just beginning to develop their tuna fisheries-to have ready access to a syntheis of information on tuna, tuna biology and tuna fisheries and tuna fisheries economics and management on the Indian Ocean.

Organic Matter Sources in a Reservoir (Lake Soyang); Primary Production of Phytoplankton and DOC, and External Loading (식물플랑크톤의 세포외배출유기물을 고려한 소양호의 1차생산과 유기물 부하)

  • The autochthonous and allochthonous organic carbon loading were measured in Lake Soyang, to estimate the amount of carbon loading into the lake and the contribution of their sources to tile lake's carbon loading. Autochthonous carbon loading was estimated from phytoplankton primary production with the extracellular organic carbon (EOC). Allochthonous loading was determined by measuring dissolved organic carbon (DOC) and particulate organic carbon (POC) concentration in the main inflowing Soyang River. Both autochthonous and allochthonous organic carbon loading were high during the svmmer, from July to September, and accounted for 43.2% and 71.7% of the annual loading, respectively. Primary productivity was elevated up to $1,000\;mgC\;m^{-2}\;d^{-1}$ during summer and lowest in winter. EOC production from phytoplankton was also large in summer, resulting in a high DOC concentration in the lake water. Primary production of phytoplankton and allochthonous organic matter loading from the watershed contributed to 53.6% and 46.4% of total loading, respectively. The EOC production accounted far $4.4{\sim}21.2%$ of POC primary production, implying that EOC production of phytolankton must be considered in estimation of primary production.
