Immunoregulatory Mechanism of Polysaccharides from Natural Plant Resources (신기술 신소재/해외기술 - 천연소재 유래 다당체의 면역조절 기작)

  • 분자단계에서 천연소재 유래 다당체는 면역 세포 표면(예, 대식세포)에 있는 한가지 또는 여러가지 수용체(${\beta}$-glucan 수용체, mannose 수용체, complement 수용체 3, Toll-like 수용체, scavenger 수용체)와 면역 반응을 시작하기 위하여 결합할 수 있고, 그 후에 신호전달을 위하여 cytokine과 second messenger($Ca^{2+}$, cAMP, cGMP, NO 등등)들을 생성한다. 그 결과로 선천면역과 적응면역을 유도하여 다양한 질환에 영향을 미친다. 추가적으로 다당체들의 면역조절은 신경내분비 체계의 조절과도 연관되어 있다. 최근 수많은 천연소재유래 다당체의 생리활성에 관한 연구가 진행되고 있다. 다양한 연구를 통해 정확한 약효, 낮은 독성과 부작용을 가진 일단의 다당체가 발견되어 현재 임상에서 암 치료에도 사용되고 있다. 이러한 사실들은 면역증진 다당체들이 항 종양치료를 위한 새로운 치료제로서 큰 시장을 개척할 수 있다는 가능성을 보여주고 있다. 이러한 사실에도 불구하고 현재 다당체에 대한 연구는 다음과 같은 취약점을 내포하고 있다. 첫째 다당체의 생리활성에 대한 정해진 비교기준이 없다. 특별히 임상 환경에서는 이러한 연구들이 중국, 일본, 한국과 일부 다른 동양국가들에 국한되어 있어 유럽이나 미국에서의 연구 자료가 결여되어있다. 둘째 현재 연구에 사용되고 있는 대다수의 천연소재 유래 다당체는 순수한 다당체가 아닌 천연 그대로의 것을 사용하고 있어 소재로부터 유래하는 다른 활성성분에 의해 쉽게 영향을 받을 수 있어 순수 다당체가 면역체계에 어떠한 역할을 수행하는지 진단하는데 어려움이 있다. 셋째 면역조절 경로에서 다당체의 정확한 신호전달 경로가 아직 정확하게 밝혀지지 않았고, 또한 동물과 임상실험에서 다당체가 작용하는 수용체 및 신호전달 경로에 대한 상반되는 결과가 발표되고 있기도 하다. 따라서 다당체의 면역조절활성에 대한 연구에 있어 다음과 같은 점들에 주목해야 할 필요가 있다. 첫째 다당체의 구조와 활성간의 상관관계를 밝혀야 할 것이다. 특히 다당체의 세부 구조 및 활성 부위를 밝혀낸다면 분자수준의 면역조절 기작을 명확히 하는데 도움을 줄 수 있을 것이다. 이러한 정보를 확보할 수 있다면 다당체의 구조를 변형시켜 활성을 증진시킬 수 있을 것이며, 또한 활성이 증가된 다당체를 합성하는 일도 가능해 질 것으로 예상된다. 마지막으로 ${\beta}$-glucan과 lentinan과 같은 정제된 다당체의 연구를 지속적으로 수행하여 효과적인 동물실험 및 임상실험 protocol을 확보하고 연구결과들에 대한 database를 구축해야 할 필요가 있다. 이러한 노력들이 성공적으로 수행된다면 천연소재유래 다당체들이 가까운 장래에 암 치료를 포함한 다양한 질병에 적용 가능한 새로운 면역조절제로 사용될 수 있을 것이다.

Study on Eco-friendly Control Effect of Natural Plant Extract Mixtures on Mulberry Popcorn Disease and Mulberry Sucker (천연 식물추출물 복합제를 이용한 오디균핵병 및 뽕나무이에 대한 친환경 방제기술 개발)

  • BACKGROUND: Two Field tests were conducted at Buan and Yangpyung in 2012 and 2013 to confirm controlling effect of mulberry popcorn disease and mulberry Sucker pests which are becoming serious in mass cultivation area of eco-friendly farming mulberry. METHODS AND RESULTS: As the treatments, 4 Natural products and 3 microbials were applied and 4 mulching materials were used. On the prevent effect of mulberry popcorn disease(Sclerotinia shiraiana) by using mulching materials, nonwoven fabric mulching showed worse effect than non-mulched treatment plot. Moreover, rice straw mulching showed significantly worse effect compare to nonwoven fabric and herb mulching treatments. Natural plant extracts and microbials showed 40~65% control value in 2013, which is little bit worse than 2012 results. On the control effect of mulberry Sucker(Amomoneura mori), organic products which combined with neem, sophora and derris showed excellent result as similar control level of Thiophanate-methyl. It means the chemical products can be replaced with organic product. CONCLUSION(S): According to the 2 years studied results, integrate eco-friendly farming measures are recommended for control of mulberry popcorn disease, because any single method is seemed not sufficient enough. However, natural plant extracts mixture is recommended as a product to control of mulberry Sucker.

Effect of Fruit Extract of Prunus mume on the Scavenging Activity of Reactive Oxygen Species and Melanin Production in B16F1 Cells (매실추출물이 활성산소종 소거효과와 B16F1 세포에서 멜라닌 생성에 미치는 영향)

  • Prunus mume has been traditionally used as a medicinal food in Korea, Japan, and China. In particular, this fruit has been reported to have beneficial biological effects on gastritis and gastric ulcers. However, its action in relation to skin whitening has remained unclear. Accordingly, the effects of fruit extract of P. mume related to antioxidation and skin whitening were examined in this study. First, using the MTT assay, it was observed that fruit extract of P. mume below 0.1% has no cytotoxicity in B16-F1 cells as a result of cell viability. Second, the direct scavenging effects and the reducing power of the fruit extract of P. mume were evaluated in vitro on DPPH radicals, hydrogen peroxide, and superoxide. It exhibited high reducing power and scavenging activity on the aforementioned reactive oxygen species. Furthermore, we found that its protective effect against genomic DNA damage related to oxidative stress was increased in a dose-dependent manner. In addition, the fruit extract of P. mume had an inhibitory effect on melanin production induced by L-dopa. In addition, it reduced the expression level of NRF-2, SOD-1, and SOD-2 related to antioxidation in western blot analysis. These results suggest that fruit extract of P. mume could exert a whitening effect through inhibition of melanin production by its antioxidant effect.

PRELIMINARY STUDY FOR ADHD TREATMENT GUIDELINE (ADHD 최적치료 지침을 위한 예비연구)

  • Objectives:In order to treatment guideline of ADHD, present clinical practise of child psychiatrists and their opinion of optimal intervention were evaluated. Methods:Structured questionnaire items about diagnostic workup, drug choice of 5 different situations according to different co-morbid disorders, and non - pharmacological treatment were applied to 32 child psychiatrists working at university and general hospital. we compared the data with Texas Algorithm Project guideline. Results:(1) Intelligence Test, Sentence Completion Test, sustained attention test, and Conner's questionnaire were the basic routine test that must be performed. (2) Main trend of medication in this study was not different from TAP guideline. (3) In case of co-morbid tic disorder, first recommending drug is still psychostimulant in the TAP guideline. But in this study initial psychostimulant prescription was not main trend. (4) In case of MPH non-response co-morbid disruptive behavior disorder, MPH medication combined with other drug were more common than switching to other drug as suggested the TAP guidelines. (5) In non-pharmacological treatment, most child psychiatrists reported the importance of parent management. Conclusion:There were some difference in medication trend in this study compared with TAP guideline. Further study and conference are needed for experts consensus in Korea.

Effects of Acai Berry Ethanolic Extracts on Production of Nitric Oxide and Activity of Angiotensin Converting Enzyme Related to Blood Circulation (혈액순환과 관련하여 nitric oxide 생성과 angiotensin converting enzyme 활성에 미치는 Acai berry 에탄올 추출물의 영향)

  • The aim of this study is to develop a supplementary therapeutic agent capable of promoting vascular circulation. The effects of Acai berry ethanolic extracts (ABEE) on activity of angiotensin converting enzyme from rabbit lung, production of nitro oxide in both murine macrophage cells and vascular endothelial cells as well as antioxidant effects were investigated in this study. First of all, it was observed the direct effects of ABEE on reducing power and antioxidant effect lipid peroxidation. In addition, ABEE showed a protective effect on DNA oxidation induced by hydroxyl radical. Furthermore, ABEE at 0.01% exerted approximately 50% inhibition on activity of angiotensin converting enzyme. ABEE increased the production of nitric oxide in endothelial cells, but decreased the induction of nitric oxide stimulated by lipopolysaccharide in microphage. The expression levels of superoxide dismutase (SOD)-2 and -3 were enhanced by ABEE treatment, however, the expression level of SOD-1 remained constant. Moreover, the expression level of nitric oxide synthases-1 (NOS-1), a constitutive enzyme, was increased by ABEE, but that of NOS-2, a inducible enzyme, was constant. It was also found that the level of Nrf-2, a transcription factor of SOD, was increased by ABEE. Therefore, these results demonstrate that ABEE could promote blood circulation via above actions, suggesting that may be helpful for health of blood vessel.

  • Chloral hydrate is one of the most widely used sedative agents to control the difficult to treat young age group in the dental clinic. We are often frustrated to see the patient still awake and cry with agitation even after far more than the normal onset time. In such a case, the patient has to be rescheduled for another sedation visit with different agents and/or routes which greatly disappoints the guardians. This case report presents a sedative regimen that can possibly help the clinician complete scheduled treatment without postponement. We have tried sleep induction with Enflurane (1-1.5vol%) for 60 seconds to 37 patients of those who failed to respond properly to the dose(70mg/kg)of oral Chloral hydrate. The average age and weight of the patients was 34.3 months(22-43mo.) and 14.9kg(11-21kg) respectively. It is suggested that sleep induction with low dose Eflurane produced a stable state during dental treatment with respect to vital sign and behavior. Evidence of adverse effect was not detected or reported during and/or after the procedures.

  • Due to fast industrialization and reduced agricultural population, there has been increased farm mechanization to reduce the labor requirement. In rice production, mechanical transplanting has been increasingly popular due to the heavy labor requirement in hand transplanting and development of convenient transplanters. For mechanical transplanting, rice seedlings is grown in boxes with limited soils under super dense population, thus short period of exposure to unfavorable temperature and poor water management would cause severe damage to rice seedlings such as seedling damping off and the similar physiological disorders. Several chemicals were evaluated for their effectiveness to control the disease and disorders, and other effects as plant growth stimulants. 3-hydroxy-5-methylisoxazole-a soil fungicide, Metalozyl-a fungicide which controls mildew, SF 8002-the composite of above two chemicals, and Isoprothiolane-a fungicide which controls rice blast were found to be effective controling seedling damping off and similar physiological disorders, and improvement of physiological characteristics of the seedlings such as the amount of bleeding sap, rooting ability, negative geotrophism, and regrowth after cutting. The results indicated that the chemicals will be very effective raising healthy seedlings even under unfavorable environments by the improvement of physiological activities of seedlings and preventing seedling damping off and the similar physiological disorders.

  • Oriental mineral medicines are single or mixture of more than one mineral species or rock/fossil which are used to treat disease. Mineral medicines remove harmful or useless substances to decrease toxicity and secondary effects, and cause the manufacture of medical compounds with increased efficacy. The extraction test is an accepted in vitro system to predict the bioaccessibility of major and minor elements from mineral medicine. It incorporates gastrointerstinal tract parameters representative of a human body that including stomach and small intestinal pH which are the same as digestion condition. The bioaccessibility of a mineral medicine is the fraction that is soluble in the gastrointestinal environment and is available for absorption. Reaction path modeling in the human body can predict digestion with gastric fluid as well as absorption in the small intestine, existence in body fluids and reaction progress of the exhaust process according to pH conditions in body. Also reaction path modeling can predict bioavailability, which is equal to existence rate in the body and the form and amount of a medicine in the body after intake. The study results from predicating the existence form mineral medicines in the body, and proving the effective ingredient using bioaccessibitily and human risk assessment, suggest these that should be necessary data for new medicine development.

  • This paper was intented to find out the effect of graft of Paeonia moutan scion into Paeonia albiflora stock on the yield and the pharmaceutical components of the root-Paeoniflorin, Albiflorin, Oxypaeoniflorin and Paeonol. The roots of Paeonia albiflora(PA), Paeonia moutan(PM) and Paeonia albiflora grafted in Paeonia moutan (M/A), were collected after being cultivated for three years in Yoenki, Chungnam, Korea. The following results were obtained ; 1. The growth characteristics of the underground part of M/A were similar to those of PA. 2. The yield of M/A was 3-5 times as that of PA in appearance. 3. The amount of Paeoniflorin in M/A was 0.2-0.5% higher than that in PA. The content of paeonol in M/A was not increased by grafting of paeonia moutan with high concentration of paeonol.

Study of the Amount and Share of Drug Cost in the Total Medical Fee under Medical Insurance Scheme (의료보험(醫療保險) 의약품(醫藥品)의 요양취급기관(療養取扱機關) 종별(種別), 약효군별(藥效群別), 상병별(傷病別) 사용(使用)에 관(關)한 조사연구(調査硏究))

  • To grasp the idea about how drugs are used under Medical Insurance Scheme, the amount and share of drug cost in the total medical fee have been reviewed and analyzed for different types of patients (in-patient out-patient), medical institutions and frequently seen diseases and following findings were revealed. In 1986, drug cost took 32.78% of total medical fee for in-patients and 32.98% for out-patients averaged over 30% share as a whole. When drug cost per case in 1980 be indexed to 100, it has shown steady growth to become 200 for in-patients and about 150 for out-patients in 1986. The contribution of drug cost to the total medical fee is, regardless of patient type-in-patients and out-patients, the highest in University hospitals and followed by General hospitals, Hospitals and Clinics in decending order That for the most frequent 10 diseases came out the highest,79 a with the essential benign hypertension of out-patients in the General hospitals, 61% for the gastric ulcer of out-patients in Hospitals and 33% for the female genital diseases of out-patients in Clinics. The drug cost of oral formula was contributed the most, 7.93% by cardiovascular agents followed by hepatic detoxicants(5.47%) and out-patients(4.93%), and that of injectable formula was contributed the most by antibiotics(24.17%), followed by protein amino-acid preparations(6.19%). The order of drug usage by specialty for the in-patients was the highest with internal medicine followed by general surgery and E.N.T, and that for the out-parients was in the order of Internal medicine, neuropsychology and Ob/Gy. This study revealed that the drug dependency was characteristically different to specialty. In view of the fact that drug cost on average exceeds over 30% of total medical fee, proper drug administration appears to be vitally important for the stabilization of the financial standing of the Medical Insurance Scheme. As a consequence, drug usage guidelines including antibiotics usage shall be established first of all and the voluntary participation for the regulation of drug usage and propagation of the guidelines to medical institutions are strongly coerced.
