Transcriptional Modulation of Metabolism-Related Genes in Brackish Water Flea Diaphanosoma celebensis Exposed to Mercury (수은 노출에 따른 기수산 물벼룩의 대사 관련 유전자의 발현 양상)

  • Mercury (Hg) is a major concern in marine environment because of their bioaccumulation and biomagnification properties, and adverse effects to aquatic organisms at even a trace amount. However, little information on the effects of Hg, compared to other heavy metals, is available in marine small crustaceans. Here, we investigated the transcriptional modulation of metabolism-related genes in the brackish water flea, Diaphanosoma celebensis after exposure to sublethal concentration (0.2, 0.4, 0.8 ㎍/l) of HgCl2 for 48 h. Relative mRNA expression levels of five detoxification enzyme-coding genes (cytochrome P450; cyp360A1, cyp361A1, cyp4AP3, cyp4C122, and cyp370C5) and six digestive enzyme-coding genes [alpha amylase (AMY), alpha amylase related protein (AMY-like), trypsin (TRYP), chymotrypsin-like protein (CHY), lipase (LIP), pancreatic lipase-related protein (PLRP)] were analyzed using quantitative real time reverse transcription polymerase chain reaction (qRT-PCR). As results, Hg increased the mRNA level of cyp370C5 (clan2) and cyp4AP3 (clan4) in a concentration dependent manner. A significant increase in TRYP mRNA was also concentration-dependently observed after exposure to Hg. These findings suggest that cyp370C5 and cyp4AP3 play a key role in Hg detoxification in D. celebensis, and Hg can affect energy metabolism by modulating the transcription of digestive enzyme. This study will provide better understanding the molecular effects of Hg in marine small crustacean.

Cellular Responses of Salmonella typhimurium Exposed to Green Tea Polyphenols (녹차폴리페놀에 노출된 Salmonella typhimurium의 세포반응)

  • The purpose of this study was to examine the cellular response of Salmonella typhimurium exposed to tea polyphenols (TPP) extracted from Korean green tea (Camellia sinensis L.). TPP showed a dose-dependent bactericidal effect on S. typhimurium. Analysis of cell membrane fatty acids of S. typhimurium cultures treated with TPP identified unique changes in saturated and unsaturated fatty acids, while scanning electron microscopic analysis demonstrated the presence of perforations and irregular rod forms with wrinkled surfaces in cells treated with TPP. Two-dimensional polyacrylamide gel electrophoresis of soluble protein fractions from S. typhimurium cultures showed 16 protein spots increased by TPP. These up-regulated proteins including proteins involved in antioxidants and chaperons, transcript and binding proteins, energy and DNA metabolism were identified by peptide mass fingerprinting using MALDI-TOF. These results provide clues for understanding the mechanism of TPP induced stress and cytotoxicity on S. typhimurium.

Effect of PCBs on the Primitive Gonad and Kidney Development of the Larval Stage of Olive Flounder, Paralichthys olivaceus in Culture Farm (넙치, Paralichthys olivaceus 자어의 원시생식소와 신장의 발달에 미치는 Polychlorinated Biphenyls (PCBs)의 영향)

  • The primitive gonad development in Paralichthys olivaceus was classified into (ⅰ) primordial germ cell (PGC), (ⅱ) genital ridge type (GRT), (ⅲ) primitive gonad type I (PGT I) and (ⅳ) primitive gonad type II stages. In the control group, PGC was recognizable on the 3rd days after hatching, and the primodial gonad after 24th days, while it was around the 22nd day after hatching in the group exposed to PCBs 3.0 $\mu\textrm{g}$/L for 30 days. Likewise, the progress of kidney development was recognized in four stages; unitubular type of mesonephric duct (UTMD), the branched mesonephric duct (BMD), convoluted tubule formation (CTF) and glomerulus appearing (GA) stage. It was structurally completed between the 25th and 30th day after hatching in two groups. There was no significant difference (p>0.05) in the time scale of development of gonad, and kidney between control and the PCBs - exposed group.

  • These experiments were performed to investigate the effects of sublethal doses of fungicides (validamycin A 3 Sp and neoasozin 6.5 Lq) and insecticides (diazinon 34Ec and decamethrin 1Ec) on the possible ways to the resurgence of brown planthoppers (BPH) (Nilaparvata lugens $St{\aa}l$). The possible factors tested in this experiment were fecundity of BPH, longevity, egg-hatchability, amount of honeydew excreted, population build-up following the pesticide treatments in the next generation, and hopperburn rate of potted rice plants. Among the insecticides tested, the applications of diazinon and decamethrin in rice fields may induce the BPH resurgence and accelerate the hopperburn of rice plants through stimulating the fecundity, increasing the adult longevity, promoting the feeding activity and causing the positve population build-up of the BPH

Acute and Chronic Toxicity of Phenol to Mysid, Archaeomysis kokuboi (곤쟁이, Archaeomysis kokuboi에 미치는 phenol의 급성 및 만성독성)

  • Acute and chronic toxicity of phenol on the mortality, long-term survival and respiration rates of the mysid, Archaeomysis kokuboi adult and juvenile were examined. This experiment was condurted by static bioassay procedure with the different salinity at $25^{\circ}C$ In lethal test, the test animals were exposed to 6 different phenol concentrations to determine $LC_{50}$ and I$LT_{50}$ (median lethal concentration and time) values. The $LC_{50}$ values with the exposure time for the mysid adult ranged from 31.31ppm to 1.49ppm phenol and for the mysid juvenile ranged from 6.90ppm to 0.26ppm in all experimental groups. Mortality was increased with the decrease of salinity, The $96hr-LC_{50}$ values at 16, 24 and $32\%o$ salinity for the mysid adult were 1.49, 2.71 and 4.53ppm phenol, white the values for the mysid juvenile were 0.26, 0.56 and 0.71ppm, respectively. The ratios of $96hr-LC_{50}$ values for the mysid adult to those for the mysid juvenile at 16, 24 and $32\%p$ salinity were 5.73, 4.84 and 6.38, respectively. The mysid juveniles were more sensitive to phenol than the mysid adults. Compared $LT_{50}$ values for the mysid adult with those for the mysid juvenile, the $LT_{50}$ values for the mysid adult ranged from 384.7 to 29.0 hours at 1.7-127ppm phenol concentrations and for the mysid juvenile ranged from 132.2 to 18.7 hours at 0.5~6.Oppm phenol concentrations. The lowest $LT_{50}$ values for the mysid adult and juvenile were showed at the combination of the highest experimental concentration of phenol and the lowest experimental salinity. The mysid juveniles showed lower $LT_{50}$ values than those of adults. The chronic effects of phenol on the mysid at the sublethal effective concentration of phenol were lower in the $32\%o$ salinitr group than 16 or $24\%o$ salinity groups. Oxygen consumption rates of the mysid adult were decreased with the increase of phenol concentration and exposure time, and decreased significantly in lower salinity at the same concentration or phenol.

  • Background : Severe acute lung injury(ALI), also known as the adult respiratory distress syndrome(ARDS), is a heterogenous nature of dynamic and explosive clinical synrome that exacts a mortality of approximately 50%. Endotoxin(ETX) is an abundant component of the outer membrane of gram-negative bacteria capable of inducing severe lung injury in gram-negative sepsis and gram-negative bacterial pneumonia, which are among the most common predisposing causes of ARDS. The influx of PMNs into airway tissue is a pathological hallmark of LPS-induced lung injury. And there is a substantial evidence suggesting that cytokines are important mediators of lung injury in gram-negative sepsis. However, the kinetics of phagocytes and cytokines by an exact time sequence and their respective pathogenic importance remain to be elucidated. This study was performed to investigate the role of phagocytes and proinflammatory cytokines in ETX-induced ALI through a time course of changes in the concentration of protein, $TNF{\alpha}$ and IL-6, and counts of total and its differential cells in BALF. The consecutive histologic findings were also evaluated. Method : The experimental animals, healthy male Sprague-Dawley, weighted $200{\pm}50g$, were divided into control- and ALI- group. ALI was induced by an intravenous administration of ETX, 5mg/kg. Above mentioned all parameters were examined at 0(control), 3, 6, 24, 72 h after administration of ETX. $TNF{\alpha}$ and IL-6 cone. in BALF were measured by a bioassay. Results : The protein concentration and total leukocyte count(TC) in BALF was significantly increased at 3h compared to controls(p < 0.05). The protein conc. was significantly elavated during observation period, but TC was significantly decreased at 72h(p < 0.05 vs. 24h). There was a close relationship between TC and protein cone. in BALF(r = 0.65, p < 0.001). The PMN and monocyte count was well correlated with TC in BALF, and the correlation of PMN(r = 0.97, p < 0.001) appeared to be more meaningful than that of monocyte(r = 0.61, p < 0.001). There was also a significant correlation between protein cone. and PMN or monocyte count in BALF(PMN vs. monocyte : r = 0.55, p < 0.005 vs. r = 0.64, p < 0.001). The count of monocyte was significantly elavated during observation period though a meaningful reduction of PMN count in BALF at 72h, this observation suggested that monocyte may, at least, partipate in the process of lung injury steadly. In this study, there was no relationship between IL-6 and $TNF{\alpha}$ cone., and $TNF{\alpha}$ but not IL-6 was correlated with TC(r = 0.61, p < 0.05) and monocyte(r = 0.67, p < 0.05) in BALF only at 3, 6h after ETX introduced. In particular, the IL-6 cone. increased earlier and rapidly peaked than $TNF{\alpha}$ cone. in BALF. In histologic findings, the cell counts of lung slices were increased from 3 to 72h(p < 0.001 vs. NC). Alveolar wall-thickness was increased from 6 to 24h(p < 0.001 vs. NC). There was a significant correlation between the cell counts of lung slices and alveolar wall-thickness(r= 0.61, p < 0.001). This result suggested that the cellular infiltrations might be followed by the alterations of interstitium, and the edematous change of alveolar wall might be most rapidly recovered to its normal condition in the process of repair. Conclusion : We concluded that although the role of PMN is partly certain in ETX-induced ALI, it is somewhat inadequate to its known major impact on ALL Alveolar macrophage and/or non-immune cells such as pulmonary endothelial or epithelial cells, may be more importantly contributed to the initiation and perpetual progression of ETX-induced ALI. The IL-6 in ETX-induced ALI was independent to $TNF{\alpha}$, measured by a bioassay in BALF. The early rise in IL-6 in BALF implies multiple origins of the IL-6.
