모의 방사성용액에서 옥살산에 의한 란탄족과 MA의 공침전

  • 알칼리 및 알칼리토금속(Cs,Rb,Ba,Sr), 전이감속원소(Zr,Fe,Mo,Ni,Pd,Rh), 란탄족 (La,Y,Nd,Ce,Eu.) 및 MA(Np,Am)등 17개 원소로 구성된 질산매질의 모의 방사성용액에서 옥살산에 의한 란탄족과 MA(Minor Actinide)의 공침전 연구를 수행하였다. 옥살산농도 0.5M에서 질산농도의 영향과 아스코빅산 첨가에 따른 원소들의 침전율이 조사되었다. 각 원소들의 침전율은 질산농도에 따라 약간 감소하였으나 란탄족과 MA는 99%이상 공침전되었다. 아스코빅산이 첨가되는 경우 Pd이 금속으로 환원침전되고 Mo.Fe,Ni.Ba의 경우는 침전율이 10∼20% 감소하는 것으로 나타났으나 기타원소들에 대해서는 영향이 나타나지 않았다. Pd의 환원침전은 질산농도 1.0M미만에서 일어났으며. 아스코빅산 농도가 0.01M∼0.02M 부근에서 최대로 나타났다. 하이드라진이 아스코빅산과 같이 첨가될 때 Pd의 환원침전을 억제하는 역할을 하였다.

Effects of Ascorbic Acid on the Proliferation of Subcultured Canine Chondocytes in Monolayer and Alginate Beads Culture (단순배양과 알긴산배양에서 개 연골세포의 증식에 있어 계대배양에 따른 아스코빅산의 영향)

  • Ascorbic acid has been used widely as a medium supplement to stimulate cell proliferation, but its effects on cell proliferation have not yet been elucidated, and no reports have analyzed effects on subcultured chondrocytes. Subcultured canine chondrocytes of passage one, two and four were cultured in monolayer and alginate beads with and without ascorbic acid. Cell proliferation was examined by 2,3-Bis(2-methoxy-4-nitro-5-sulfophenyl)-2H-tetrazolium-5-carboxanilide inner salt) (XTT) colormetric assay. Ascorbic acid stimulated cell proliferation significantly in both culture methods (p<0.05). The increased cell numbers by stimulation with ascorbic acid were significantly high in passage one cells compared to that of other passages. Differences in cell proliferative capacity by subculturing were not determined. These results suggest that ascorbic acid stimulated the proliferation of subcultured canine chondrocytes and enhanced it more in low-passage cells than in the other cells tested.

Oxalate Precipitation of Lanthanide and Actinide in a Simulated Radioactive Liquid Waste (모의 방사성용액에서 란탄족과 악티늄족원소의 옥살산침전)

  • The oxalate precipitation of lanthanide and actinide by oxalic acid was investigated in the simulated radioactive liquid waste, which was composed of 17 elements of alkali, alkaline earth(Cs, Rb, Ba, Sr), transition metal(Zr, Fe, Mo, Ni, Pd, Rh), lanthanide(La, Y, Nd, Ce, Eu) and actinide(Np, Am) in nitric acid solution. The effect of concentrations of nitric acid and ascorbic acid on the precipitation yield of each element in the simulated solution was examined at 0.5 M oxalic acid concentration. The precipitation yields of the elements were usually decreased with nitric acid concentration, nevertheless, the precipitation yields of lanthanide and actinide were more than 99%. Palladium was precipitated due to the reduction of Pd(II) into Pd metal by the addition of ascorbic acid in the oxalate precipitation and then, the precipitation yields of Mo, Fe, Ni, Ba decreased by 10~20% with concentration of ascorbic acid. The reductive precipitation of Pd(II) into Pd metal by the addition of ascorbic acid into the simulated radwaste occurred at below 1 M nitric acid concentration and its yield showed maximum at the ascorbic acid concentration of 0.01~0.02 M. The hydrazine suppressed the reductive precipitation of Pd by the ascorbic acid.

  • An activated carbon paste electrode was modified with the N-Hydroxysuccinimide(NHS) layer and applied to determine the dopamine in the presence of an excess ascorbic acid using square-wave voltammetry. The electrochemical properties of the modified electrode were examined in the solution containing dopamine/ascorbic acid using cyclic voltammetry(CV): the separation between the oxidation peaks of dopamine and ascorbic acid was largely dependent on the pH of the sample solution and became maximum at pH 4.0. Hence, the square-wave voltammetric determination of dopamine was carried out in a pH 4.0, 100mM phosphate buffer saline(PBS) containing 140mM NaCl. The detection limit and response slop were improved from $1.0{\mu}M\;to\;5.0\times10^{-2}{\mu}M\;and\;from\;0.93{\mu}A/{\mu}M\;to\;6.1{\mu}A/{\mu}M$, respectively, upon modification of the electrode surface by NHS.

Preparation of Ag Fine Particles from Aqueous Silver Solution by Reduction Reaction (Ag 水溶液으로부터 還元反應에 의한 Ag 微粒子의 製造 硏究)

  • A study on the preparation of Ag fine particles was performed through a reduction reaction using ascorbic acids as a reductant, which is one of the indispensable processes for the recycling of silver-bearing wastes. Silver nitrate solution in the range of 10~120 mmole/l was used and Tamol NN8906 or PVP was also used as a dispersant in the preparation of Ag fine particles size analyze, SEM, and TEM to determine the particle size and morphology of them. As a result, the reduction reaction of silver ions with ascorbic acid reached equilibrium within 10 min. It was found that about 60% excess of ascorbic acid was required in order to reduce completely silver ions in the solution. The particle size distribution of Ag particles prepared through the reduction reaction showed typically biomodal or trimodal distribution. Especially, initial Ag concentration in the solution, the type and amount of dispersant added during the reduction reaction played an important role in determining the mean particle size of Ag particles.

Fungal Development, Respiration and Activity of Oxidative Enzymes in Rice Plants Inoculated with Pyricularia oryzae in Both Compatible and Incompatible Combinations (벼 도열병균에 감염된 친화 및 비친화 조합 벼에서의 균생장, 호흡 및 산화효소 활성)

  • Appressorial formation of Pyricularia oryzae on leaves showed no marked difference between highly resistant Tongil and susceptible Norm No. 6. Race N-2 of the blast fungus penetrated directly into motor cells of susceptible cultivar Norm No. 6, later extensively spreading hyphae were developed, while in the cultivar Tongil, after penetration, no further hyphal extension resulted. In discoloration of infected tissues, the highly resistant cultivar Tongil not only discolored rapidly, but also the percentage of discolored cells was higher than the susceptible cultivars, Jinheung and Norm No. 6. The respiratory rate, was generally higher in infected tissue than in healthy tissue. No significant difference in the respiration rate of resistant Suwon No. 180 was not found between the infected and healthy leaf tissue, whereas, in susceptible Jinheung, a marked increase in respiratory rate was caused by blast infection. The respiratory rate increased at the appearance of the first visible symptom in all cultivars resistant or susceptible. Higher peroxidase activity was found in infected tissues as compared with healthy tissue. Peroxidase activity increased in resistant and susceptible reactions. Particularly, in resistant reaction, the increase of the activity was more pronounced. In highly resistant reaction, there was no difference in peroxidase activity between healthy and infected tissues. Ascorbic acid oxidase, hydroquinone oxidase and catechol oxidase had the same trend as the peroxidase. In contrast, activity of catalase rather decreased in leaf tissues infected with compatible races of the fungus.

In Vitro Antioxidant Activity of Alnus firma Extracts (사방오리(Alnus firma) 추출물의 in vitro 항산화 활성)

  • This study evaluated the antioxidant activity of the extract and fractions of Alnus firma. Alnus firma had the highest total phenolic content ($452.80{\pm}7.01{\mu}g$ gallic acid equivalents/mg) in a methanol (MeOH) fraction and the highest total flavonoid content ($112.29{\pm}11.14{\mu}g$ rutin equivalents/mg) and antioxidant capacity ($936.23{\pm}0.07{\mu}g$ ${\alpha}$-tocopherol equivalents/mg) in an ethylacetate (EA) fraction. The antioxidant activities of various solvent extract fractions of Alnus firma were evaluated using various antioxidant assays, including ${\beta}$-carotene-linoleate assay, reducing power assay, 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging assay, metal chelating activity assay, superoxide anion radical scavenging assay, and lipid peroxidation inhibition assay using the ferric thiocyanate method. These activities were compared with those of ascorbic acid, butylated hydroxyanisole (BHA), gallic acid (GA), butylated hydroxytoluene (BHT), and ${\alpha}$-tocopherol. First, at a $250{\mu}g/ml$ concentration, the EA and MeOH fractions of A. firma showed 92.43% and 89.20% DPPH radical scavenging activity, respectively. Second, $50{\mu}g/ml$ of the EA fraction exhibited 72.49% superoxide anion radical scavenging activity, a little greater than the same dose of GA (60.88%). Finally, 0.5 and 1 mg/ml of the EA fraction showed 73.45% and 73.29% inhibition of peroxidation in the ${\beta}$-carotene-linoleic acid system, respectively. The decreasing order of reducing power was EA fraction > n-butanol (BuOH) fraction > dichloromethane (DCM) fraction > n-hexane (HX) fraction. The results obtained in the present study indicated that Alnus firma can be used as an easily accessible potential source of natural antioxidants.
