닭 수정란 미세조작이 배자의 쌍자 형성에 미치는 영향

  • 조류는 발생학적 특성상 수정과 초기 배발생을 제외한 거의 대부분의 개체 발생 과정이 난각 속에서 진행된다. 또한 배자가 발생하는 동안 배자의 뼈를 구성하는 석회성분의 거의 대부분은 난각에서 유래한다. 난각 석회성분은 배자유래의 융모성 뇨막(chorioallantoic membrane; CAM)의 모세혈관을 통하여 배자로 이동하다. 배자 발생후기로 가면 융모성 뇨막은 전체 난각과 거의 대부분 밀착하여 배자 발생 후기에 필요한 많은 양의 석회성분을 난각에서 배자로 공급하게 된다. 그러므로 한 개의 난각 속에서 두 개의 배자가 정상적으로 발생되기란 거의 불가능하다.   • 프리마틴(freemartin)이라 함은 반추동물들 중 특히 소에서 나타나는 간성(intersex)형태로 이성 쌍자의 경우 태어난 암컷의 대부분이 정상적인 자성 생식기의 발육 및 발생양상을 지니지 못하여 생식불능상태로 되는 것으로 알려져 있다. 최근 실용화된 수정란이식이나 쌍자유기 등의 기술은 현재 일반 농가에 까지 정책적으로 확대 보급되어 산업화되고 있는 실정이나 불행히도 이러한 기술들을 이용하여 생산된 개체들 중 상당수가 이성쌍자로 이들 중 암컷 개체들은 거의 freemartin으로 생산되어 실지 노력에 비해 경제적 효과들을 크게 반감시키고 있다. 뿐만 아니라 생산된 freemartin 개체들에 대하여 조기 진단 없이 다만 형태적으로 암컷으로 판단하여 번식우로 사양관리 함에 따른 경제적 손실이 매우 크다 할 수 있겠다.

Studies on Embryo Cryopreservation and Twinning by Embryo Transfer of Korean Native Cattle II. Transfer of Bisected Embryos and Production of Twin Calves (한우 수정란의 동결보존 및 쌍자생산에 관한 연구 II. 이분 수정란의 이식과 쌍자 생산)

  • This study was carried out to enhance the efficiency of Korean Native cattle embryos and establish the techniques for producing the twin calves. Bisected embryos without zona pellucida which were divided by simple method not using holding pipette or whole two embryos were transferred to recipients.The pedigrees of monozygotic twin calves produced by transfer of bisected pair embryos were identified. The results obtained were as follows ; The average successful bisection rate was 89.16%. The embryos of blastocyst stage (91.66%) were bisected successfully at significantly (P<0.05) higher rate, compared with the morula stage embryos (86.66%). The average survival rate of bisected embryos following 24 hours culture was 59.02%. The survival rate of morula stage embryos (62.50%) was significantly (P<0.05) higher than that of blastocyst stage embryos (55.5%). For the production of monozygotic twin calves, ten pairs of flesh or frozen demi-em- lymphocytes antigen, the twin calves produced by transfer of bisected pair embryos of Korean Native cattle were identified in pedigrees and confirmed as monozygotes.

  • 뇌전위 측정에 의해 흥분 뉴런군의 위치를 추정하는 source localization problem은 Evoked Potential 해석법에 있어서 매우 중요한 의미를 갖는다. 이번 논문에서는 EEG실험에서의 전극 배치가 S/N(signal to noise ratio)과 추정 오차 사이에 어떤 영향을 미치는 가를 Monte Carlo 시뮬레이션으로 조사하였다. 머리 모델은 3중 구각 모델을 사용하였고 이론 이용하여 forward problem을 계산하였다. 쌍극자 파라미터를 minimization 하는 문제는 simplex method를 이용하여 계산하였다. 컴퓨터 시뮬레이션 결과, 특이한 점은 전극의 밀도와 입체각에 의해 쌍자 파라미터 오차가 변화했다는 사실이다. 이것은 곧바로 전극 배치와 연관이 된다. 실제 EEG 실험에서 전극 배치를 어떻게 했는가에 따라 그에 따른 오차가 변화한다. 이러한 오차의 원인을 제거하기 위해서 새로운 전극 배치를 모델링하여 기존의 전극 배치와 비교해 보았다. 그 결과 전극 밀도와 입체각에 대한 오차를 크게 줄일 수 있었다.

Studies on the Production of Twins in Cattle (소의 쌍자 생산술 개발에 관한 연구)

  • Induction of twinning was attempted by transfer of two whole- or demi-embryos in Holstein cows and heifers. Cows were superovulated with follicle stimulating hormone(FSH) administered twice daily in intra-muscular injection. On day of 6.5 to 7 post-estrus, embryos were collected nonsurgically, Normal morulae and early balstocysts were obtained from superovulated cows. The embryos were hisected with a micro-blade made from a razor. Twenty seven pairs of half embryos were successfully produced and 23 pairs of half embryos were transferred to recipients( Group A). Twenty cows were treated with low unit of FSH (3mg${\times}$2,2mg${\times}$2 ; Group B1, 2mg${\times}$2, 1mg${\times}$2, 1mg${\times}$2 ; Group 32) to induce double or triple ovulations. The cows of Group B1 and B2 were Inseminated artificially(A1) at following estrus. Twenty four heifers were bred by A1 and received an additional embryo into the uterine horn contralateral to the corpus luteum(CL) 6 days later(Group C). One embryo was transferred into each uterine hem of 16 heifers 6 days after estrus(Group D). Fourteen heifers were received two embryos into the uterine horn ipsilateral to the CL at day 6 of estrous cycle(Group I). Pregnancy rates at 60 days in Group A, B1, B2, C, D and E were 34.8%, 70.0%, 60.0%, 66.7%, 62.5% and 57.1%, respectively. Twinning rates were 8.7%, 20.0%, 10.0%, 16.7%, 18.7% and 21.4%, respectively. The present experiments demonstrate that FSH treatment( Group 31) and ipsilateral transfer( Group I ) of two whole-embryos are more useful methods to produce twins than the others.

  • In vitro fertilization(IVF) derived morula and blastocyst embryos were bisected by a simple method and cultured in vitro without zona pellucida And also bisected embryos were frozen-thawed and cultured in vitro) to evaluate the survival rate. The results obtained were as follows : The average number of grade I or II immature follicular oocytes recovered by slicing method per ovary was 11.9 from 142 ovaries. Fundamental Parameter 법에 의한 만장굴용암 쌍자석주의 형광X선분포

  • Cheju Island, which was formed by volcanic activity, is an oval in its shape with the major axis 80km and the minor axis of 40km. The island holds in its heart Mt. Hanala rising 1,950m above the sea. Petrological study of this volcanic island has been made actively by Sang-Man Lee, Chong-Kwang Won and Moon-Won Li. The chronological measurements of the island by Chong-Kwan Won and Moon-Won Lee showed that it is composed of Sanbangsan trachytes and Backlokdam trachytes(25,000 year ago). These reports are based on the chemical analysis and the rediometric chronological measurements on the ground. However, there has been no reports about the inside of caves. We made an (composition) analysis of the inside of Manjang Cave by the fundamental parameter method in X-ray fluorescence spectrometry. The fundamental parameter method in X-ray fluorescence spectrometry is nondestructive analysis. and it enables us to make the values processed by a computer. The results obtained by this methods are as follows: SiO$_2$(49%), $Al_2$O$_3$(17%), Fe$_2$O$_3$(13%), CaO(8.1%), MgO(5.5%), Na2O(3.6%), TiO$_2$(2.1%), $K_2$O(0.86%), P$_2$O$_{5}$(0.28%), and MnO(0.20%) respectively. The data obtained by the fundamental parameter method in X-ray fluorescene was compared with the data provided by Chong-Kwan Won and Moon-Won Lee. Our measurement was made by K-Ar-method in cooperation with T. ITAYA. The samples are of 30,000-420,000 years ago. The composition of the values of our underground analysis with the existing values obtained by the analyses on the ground produced new data about Cehju volcanic island.d.

  • Soft X-ray photography was used to study some characters of seeds, such as fullness, damage by blight and harmful insects and degree of mechanical damages, degree of development of embryos and endosperms, size and position of cotylendons, existence and position of twin seed. Thirty nine species of woody plants (31 angiosperms and 8 gymnosperms)and four species of herbs were examined in this study. The results showed that through the mechanism of the selective sensitivity of seed structure of soft X-ray, seed characters mentioned above could be recognized regardless of size of seeds and thickness of seed coat. We concluded that it is possible to use soft X-ray photography reliably to find out the various characters of seeds.

  • This study was carried out to establish the techniques for producing the calves of genetically superior Korean Native cattle by transfer of frozen-thawed embryos. The effects of some factors related to embryo recovery following superovulation and pregnancy rate following transfer of frozen-thawed embryos were evaluated. Also calving state was investigated. The results obtained were as follows ; The mean number of total and transferrable embryos recovered per superovulated cow was 8.72 and 4.90, respectively, from a total of 72 superovulations using 34 donor cows. There were no significant differences in the number of total or transferrable embryos recovered per superovulated cow between products of follicle stimulating hormone (FSH), years, seasons, and collection numbers. The pregnancy rate was found 44.44% following transfer of frozen-thawed embryos of Korean Native cattle to a total of 180 recipient cows including 82 Angus, 27 Charolais, 62 Hereford and 9 Korean Native cows. The pregnancy rate was significantly (P<0.05) higher in the transfer of excellent (42.99) and good embryos (40.17%), compared with fair (5.90%) grade embryos. And the pregnancy rate was significantly (P<0.05) higher in the transfer of embryos of morula stage (43.86%) than blastocyst stage (15.51%). But there were no significant differences in pregnancy rates between natural and induced estrus estrus asynchrony of 1 days, breeds, and parities of recipient cows. The normal calving rate of 80 pregnant cows following transfer of frozen4hawed em-bryos was 87.5% and the other 10 pregnant cows showed abortion during the period from pregnancy diagnosis at 50~60 days to calving. The average gestation length of normally delivered recipients was 288.50 days and the average birth weight of 70 calves born was 24.22 kg. The gestation length was significantly (P<0.05) shorter in the recipients delivering female calves (286.70 days) than males (289.39 days). But there were no significant differences in gestation tength and birth weight of calves born between the recipient breeds.

Study on Production of In Vitro Embryos and Twin Calves by Embryo Transfer in Korean Native Cattle (한우 체외수정란의 생산과 이식후 쌍자 생산에 관한 연구)

  • The objectives of this study were performed to increase the efficiency of the culture conditions of embryos produced in vitro, and to assess the developmental potential after transfer of those embryos into recipients. The mean number of folliclular oocytes recovered from an ovary was 10.7. The rates of maturation and fertilization in Grade I oocytes were significantly (P<0.05) higher than Graden and III. Developmental rate into blastocyst in the culture group of TCM-199 with BOEC were significantly higher (P<0.05) than the groups of TCM-199 and conditioned medium (24.7% vs. 12.4% and 18.2%). The survivability of post-thawed blastocysts equilibrated for 3 min in EFS solution was significantly (P<0.05) lower than l0 for 1 and 2 min (32.1% vs. 82.9% and 73.3%). Significantly higher (P<0.05) survival rate in blastocysts was seen after freezing than in morulae stage embryos. Out of all 105 recipients, 49 (46.7%) were confirmed in pregnant. On pregnancy of cattle, 48 calves were born from 40 recipients. The ratio of twin and single calves was 30.5% (32/40 and 7.6% (8/40), respectively. However, the others composed of abnormal, as judging as 6 (12.2%) for abortion and 3 (6.1%) for stillbirth during the pregnant period.
