Effect of dietary changes from high-fat diet to normal diet on breast cancer growth and metastasis (고지방식이에서 일반식이로의 전환이 유방암의 성장 및 전이에 미치는 영향)

  • Purpose: It has been previously reported that breast tumor incidence, growth, and metastasis are stimulated by high-fat diet but reduced by caloric restriction. However, few studies have elucidated the effects of dietary change from a high-fat diet after breast cancer initiation. Therefore, in this study, we attempted to provide practical assistance to breast cancer prevention and management by investigating the effects of dietary change from a high-fat diet to normal diet on breast cancer growth and metastasis. Methods: The experimental animals were divided into 2 groups (high-fat diet control [HFC] group and diet restriction [DR] group) and consumed a high-fat diet for 8 weeks. 4T1 cells were transplanted into subcutaneous fat or tail vein to measure the growth and metastasis of breast cancer. The HFC and DR groups continuously ingested either high-fat diet or AIG-93G diet for 5 weeks or 3 weeks, respectively. Cell proliferation and apoptosis markers from tumor tissues were analyzed by Western blot analysis. The data were analyzed using the SPSS 25.0 package program. Results: The results show that the DR group significantly reduced breast tumor initiation, growth, and tumor tissue weight compared to the HFC group. The DR group suppressed tumor growth by decreasing proliferation and inducing apoptosis through down-regulation of Bcl-xL and up-regulation of caspase-3 activity. Furthermore, the DR group significantly reduced numbers of metastasized tumors in lung tissues. Conclusion: These results suggest that dietary change from a high-fat diet to normal diet decreased breast growth by reducing cell proliferation and inducing apoptosis and metastasis. Taken together, these results indicate that dietary change to a low-fat and balanced diet might suppress breast tumor growth and metastasis even after tumor diagnosis.

Undifferentiated Adenocarcinoma with Rhabdoid Features in the Stomach: A Case Report (횡문근양 미분화 위선암 1예 보고)

  • Rhabdoid tumor has been considered to be a rare subtype of Wilm's tumor with Rhabdomyoma features. Since rhabdoid tumor that developed in the kidney was described for the first time in 1989, it has been reported in the gastrointestinal tract, although this is rare. The appropriate treatment is radical resection, and the effect of adjuvant chemotherapy has not yet been reported on. The outcome of extra-renal rhabdoid tumor is different from renal rhabdoid tumor and the former shows a poor prognosis. Among extra-renal rhabdoid tumors, undifferentiated gastric adenocarcinoma with rhabdoid features is very rare and its prognosis is poor. A 63 years old male patient underwent total gastrectomy for a tumor that developed in the greater curvature of the gastric body and this was diagnosed as undifferentiated gastric adenocarcinoma with rhabdoid features, according to the histopathology. We experienced an undifferentiated gastric adenocarcinoma with rhabdoid features that was diagnosed by immunohistochemical staining and we report here on this case.

Dicumarol Inhibits PMA-Induced MMP-9 Expression through NQO1-independent manner in Human Renal Carcinoma Caki Cells (인간 신장암 Caki세포에서 dicumarol에 의한 PMA 매개 matrix metalloproteinase-9의 발현 억제 효과)

  • Dicumarol is a coumarin derivative isolated from sweet clover (Melilotus alba), and has anti-coagulant activity with the inhibitory activity of NAD(P)H quinone oxidoreductase1 (NQO1). NQO1 catalyzes the two-electron reduction of quinones to hydroquinones. Dicumarol competes with NAD(P)H for binding to NQO1, resulting in the inhibition of NQO1 enzymatic activity. The expression of matrix metalloproteinases (MMPs) has been implicated in the invasion and metastasis of cancer cells. The expression of MMPs is regulated by cytokines and signal transduction pathways, including those activated by phorbol myristate acetate (PMA). However, the effects of dicumarol on metalloproteinase (MMP)-9 expression and activity are not investigated here. This study investigated whether dicumarol inhibits MMP-9 expression and activity in PMA-treated human renal carcinoma Caki cells. Dicumarol markedly inhibited the PMA-induced MMP-9 mRNA expression and MMP-9 activity. NF-κB and AP1 promoter activity, which is important in MMP-9 expression, also decreased in dicumarol-treated cells. Furthermore, dicumarol markedly suppressed the ability of PMA-mediated migration in Caki cells. When the relevance of NQO1 in the dicumarol-mediated inhibitory effect on PMA-induced MMP9 activity was elucidated, knock-down of NQO1 with siRNA was found to have no effect on PMA-induced MMP9 activity, suggesting that the stimulating effect of dicumarol on PMA-induced MMP9 activity is independent of NQO1 activity. Taken together, the present studies suggested that dicumarol may inhibit PMA-induced migration via down-regulation of MMP-9 expression and activity.

Clinical Analysis of Metastatic Tumors of Bone - Survivorship Analysis after Bony Metastasis - (전이성 골종양에 대한 임상적 분석 - 골전이 후의 생존분석 -)

  • Purpose : To analyze the clinical behaviors and survivorship of metastatic bone tumors. Materials and Methods : One hundred and ninty-eight metastatic bone tumors had been diagnosed from January 1982 to December 1998. Age and sex distribution, primary cancer types. metastatic sites, duration from diagnosed of primary tumors to bony metastases and survivorship were analysed. Results : Mean age was 57(24~86) years old. Lung(32.3%) and breast(16.2%) cancers were two most common primary foci. The spines was the most common site of metastases especially lumbar region(38%). Survivorship analysis was done in one hundred and fifteen patients who had been followed up. The mean survival period was 15.3 months. The survivorship of hepatoma(7.1 Mons), lung(8.72 Mons) and renal cell(4.8 Mons)cancers was relatively shorter and breast cancer(54.1 Mons) longest. Conclusion : The mean age of metastatic bone tumors of this study was older than the past reports. The axial skeletons especially spine was predominant metastatic site. The survivorship of metastatic bone tumor decreased sharply as time goes by, so early diagnosis is clue for longer survival after bony metastases.

Flow Directions and Source of the Dongmakgol Tuff in the Cheolwon Basin, Korea (철원분지 동막골응회암의 유향과 공급지)

  • The Dongmakgol Tuff is a stratigraphic unit which is composed of voluminous ignimbrites in the Cheolwon basin. The ignimbrites belong to pumice-rich vitric tuffs that show eutaxitic to parataxitic fabrics from fiamme or pumice clasts. They are almost densely welded and strongly flattened, but often parallel aligned and stretched. Also they exhibit flow indicators such as flow lineations, imbrications, tensional cracks and boudins from their alignment and/or elongation, and lithic and pumice clasts show lateral grading in their average maximum diameter. Flow direction map from the lineations, asymmetric structures and lateral grading diagram indicate that the Dongmakgol Tuff has a source from its southwestern part near a boundary between southern Dongmakri and northern Gomunri, and is considered that the ignimbrites took emplacement processes of laminar flows during the final stage of flowage and the flow lineations are from the result of shear stress during that times.

Morphology of Utricles and Maturing Period in Codium fragile(Suringar) Hariot (청각, Codium fragile(Suringar) Hariot의 포낭의 형태 및 성숙시기에 관하여)

  • The present work was carried out to investigate the maturing period and the morphology of utricle Codium fragile collected from April 1984 to March 1985 in the coast of Chumgmu southern of Korea. Ranges of water temperature and salinity during the survey period were $7.5\~27.0^{\circ}C$ and $32.9\~34.5\%_{\circ}$, respectively. The gametangia were produced throughout the year. Mature gametangia were present on plants collected from June to November, whereas most of plants collected from January to May occured as the elongated utricles with both mature and immature gametangia. As for the sexuality, most of plants were dioecious and some monoecious, and there were more male gametophytes rather than female gametophytes. Some plants collected during winter months of December to February were observed as thalli with both brunted and elongated utricles, and male and female gametangia occured together on a single utricle.

Papillary Thyroid Carcinoma in Renal Allograft Recipients (신장이식후 발생한 유두상 갑상선암)

  • Purpose:The chronic use of immunosuppressive therapy in transplant recipients can increase the long-term risk of carcinoma. The aim of this study was to determine the incidence, biological behaviors, and treatment outcomes in PTC(papillary thyroid carcinoma) in renal allograft recipients. Material and Methods:The present study examined the incidence and biological behavior of PTCs in RA recipients. A total of 1,739 RA patients treated between January 1986 and December 1999 were followed-up for a median 137(84-238) months. During the follow-up period, 129(7.4%) recipients were identified as having posttransplant malignancies. Of those, 12(0.7%) had PTCs, and these comprised six male and six female patients with a median age of 41(23-57) years. Results:Nine cases(incidentalomas) were diagnosed based on ultrasonography(US) screening. Eight of those nine were TNM stage I, and two of the three clinical carcinomas were TNM stage IVa. During a median follow-up of 94(18-159) months, two(16.7%) PTC patients developed loco-regional recurrence, but no patients showed distant metastasis. Posttransplant PTC showed no gender bias, and was often associated with aggressive lymphatic metastasis. However, most incidentalomas showed a favorable treatment outcome. Conclusion:In conclusion, routine surveillance of the thyroid gland using US screening is recommended to ensure early detection, treatment and favorable prognosis in RA patients with PTC.

Granulite xenoliths in porphyroblastic gneiss from Mt. Jiri area, SW Sobaegsan massif, Korea (소백산육괴 서남부 지리산지역의 반상변정질 편마암에서 산출되는 백립암질 포획암)

  • Mafic granulite xenoliths are found in precambrian porphyroblastic gneiss of the Mt. Jiri area, SW Sobaegsan massif, Korea. The xenoliths are rounded to ellipsoidal in shape, 50-100 cm in length and coarse-grained with granoblastic and foliated texture. The xenoliths consist of orthopyroxene, garnet, biotite, plagioclase, quartz, ilmenite and secondary orthoamphibole. Orthopyroxene is mostly resorbed and rimmed by coronitic orthoamphiboles. Garnets occur as porphyblasts and are zoned with higher pyrope content in cores than in rims. Geothermo-barometry results yield conditions of about $800-850^{\circ}C$, 6 kb and $500^{\circ}C$, 4 kb for early and retrograde stages of equilibration, respectively. According to available geochronological data, it is suggested that the granulite facies metamorphism occurred prior to 2.1-1.9Ga and that the area was superimposed by the high-grade (over $600-700^{\circ}C$) metamorphism between 1.9-1.7Ga, followed by cooling during uplift.

Changes in Cell Proliferation During the Development of Renal Cell Tumors Induced by N-Nitrosomorpholine in Rats (흰쥐에서 신장암이 발생하는 동안 세포분열속도의 변화)

  • Sequential changes in cell proliferation during the development of epitherial kidney tumors induced in rats were investigated by autoradiographic determination of the $^3H$-thymidine-labeling index. Renal cell tumors were induced in male Sprague-Dawley rats by oral administration of N-nitrosomorpholine at the concentration of 120 mg/l in the drinking water for 7 weeks. At different times between 12 and 34 weeks after withdrawal of the carcinogen (stop model) animals were sacrificed. According to cytological criteria, neoplastic lesions were classified into clear cell, acidophilic cell, basophilic cell and oncocytic tumors. The labeling index was found to be increased in all types of preneoplastic tubules as compared to their corresponding original tubules. A much stronger elevation of cell proliferation was ocurred during the development of renal cell tumors from preneoplastic tubules. Of four tumor types, acidophilic cell tumor showed the highest labeling index while oncocytoma exhibited the lowest proliferative activity. These findings are in good accordance with the clinical observations that acidophilic cell tumors have a worse prognosis than oncocytoma. The data presented in this study suggest that the individual proliferation rates may be an objective biological marker of kidney tumor aggressiveness.

Role of Iridin Isolated from Iris koreana Nakai on Doxorubicin-induced Necrosis in HK-2 Cells, and Effect on Cancer Cells (노랑붓꽃에서 분리된 Iridin의 독소루비신 유도 HK-2 세포 괴사에 대한 역할 및 암세포에 대한 작용)

  • Doxorubicin is a anti-cancer drugs that interferes with the growth and spread of cancer cells in human body. Doxorubicin is used to treat different types of cancers that affect the ovary, thyoid and lungs, but induced side effect such as nephrotoxicity and cardiotoxicity. Thus, we investigated that the effect of iridin on doxorubicin-induced necrosis in HK-2 cells, a human proximal tubule cell. To confirm effect of iridin on doxorubicin-induced necrosis, HK-2 cells are treated with $10{\mu}M$ doxorubicin and $80{\mu}M$ iridin. $80{\mu}M$ iridin reduced $10{\mu}M$ doxorubicin-induced necrosis, the mitochondrial over activation and caspase-3 activation. However, iridin reduces anti-cancer effect of doxorubicin such as PARP1 and caspase-3 activation, checkpoint proteins (CDK4 and CDK6) in NCI-H1129 cells (Human non-small cell lung cancer cell). In HCT-116 cells (Human colorectan cancer cell), iridin do not increased protein expression of CDK4 and CDK6 decreased by doxorubicin. Results indicate that treatment of iridin was diminished doxorubicin-induced necrosis in HK-2 cells. However, iridin was decreased anti-cancer effect of doxorubicin on NCI-H1229, but not HCT-116. Thus, further experiment are required to iridin treatment on various cancer cells and animal models because effect of iridin different cell type.
