Preparation of Squid-Jeotkal with Pasteurized Red Pepper II. Shelf-Life Extension of Squid-Jeotkal (살균고춧가루를 이용한 오징어젓갈 제조 II. 양념오징어젓갈의 보존성 연장)

  • In this study, ohmic heating was applied for pasteurization of red pepper pow-der, and investigated its pasteurization effect. After pasteurization, seasoned squid-jeotkal was manufactured by using red pepper powder, and its quality changes were investigated. On com-paring sensory evaluation in squid-jeotkal by pasteurized and conventional red pepper powder during storage at 5, 16 and $25^{\circ}C$, quality changes in squid-jeotkal by the pasteurized red pepper powder. But quality change difference between two products were decrease with increasing stor-age temperature. Viable cell counts in two products stored at 5$^{\circ}C$ were increased slowly until 60 days. Viable cell counts in squid-jeotkal by conventional red pepper powder were increased up to ca. 10$^{8}$ CFU/g at 15, 25 $^{\circ}C$ after 30, 15 days, respectively, but that by pasteurized were ca. 10$^{8}$ CFU/g at $25^{\circ}C$ after 30 days. Changes in pH, VBN and NH$_2$-N in two products were shown difference significantly, but were decreased by increasing storage temperature. In squid-jeotkal by conventional red pepper powder, the main free amino acids were glutamic acid, leucine, glycine, aspartic acid and alanine, and these amino acids held 47.95% of the total free amino acid. But in squid-jeotkal by pasteurized red pepper powder, glutamic acid, glycine, aspartic acid, leucine and Iysine, and these amino acids held 57.58% of the total free amino acids.

Changes in Microbiological and General Quality Characteristics of Gamma Irradiated Kanjang and Shoyu (감마선을 조사한 간장의 미생물 및 일반품질 특성)

  • The effect of gamma-irradiation on the microbiological and general qualities of Kanjang (fermented soy sauce, Korean type) and Shoyu (fermented soy sauce, Japanese type) was studied. Samples were prepared, irradiated at 5, 10 and 20 kGy, and then stored at $25^{\circ}C$ for 18 weeks. The results showed that the Bacillus spp. was inactivated by 3 log cycles with the irradiation dose of 10 kGy and the number of Bacillus cells that survived from 10 to 20 kGy of gamma irradiation was decreased gradually during storage. Yeasts and Lactobacillus were nearly eliminated at 5 to 10 kGy of irradiation. The $D_{10}$ values of Bacillus, yeast and Lactobacillus in Shoyu were 2.67 kGy, 0.81 kGy and 1.30 kGy, in Kanjang were 2.75 kGy, 0.99 kGy and 1.47 kGy, respectively. The general quality of gamma irradiated Shoyu and Kanjang, such as total nitrogen, amino nitrogen, protease activity and pH were more stable than that of control during storage periods. Decolorization was observed just after irradiation, but the color was recovered during storage. The sensory evaluations showed that irradiated samples were more acceptable. Therefore, it was considered that gamma irradiation was effective for maintaining better quality of Shoyu and Kanjang during storage.

Studies on the Characteristics of Kefir Grains Collected from Korean (한국에서 수집된 케퍼 그레인의 특성에 대한 연구)

  • Kefir is a traditional fermented milk in Caucasusian area and is made mainly of milk fermented with lactic acid bacteria and yeasts. Six typical kefir grains were selected from ten kefir grains collected from different locals in Korea. Kefir grains were gelatinous in texture and had various shapes of villi, grapes, leaves, hulled millets, and towels. To investigate predominant microflora of kefir grains, SPC, MRS, M17, Rogosa, and APT agar media were used for viable cell count MRS, SPC, and Rogosa media were most acceptable for bacterial cell counts of the selected kefir grains. From one or two of the SPC agar plates which contained around 25∼50 colonies, all grown colonies were isolated and identified. Most predominant bacteria was identified as Lactobacillus fermentum by API 50 CHL kit. The proportions of Lb. fermentum and Lb. brevis among the total identified bacteria were around 41~88% and M4%, respectively. To select the best preservation method for kefir grains, refrigeration, freezing, and freeze drying were compared. Freeze drying was found most suitable for the preservation of kefir grains, based upon their acid-producing activities and production of off-flavors.

  • Certain parts of 190 kinds of medicinal herbs and 171 kinds of original materials of food were extracted by methanol. The extracts were tested their microbial inhibition activities against several food spoilage microorganisms, Micrococcus luteus, Bacillus subtilis, Bacillus cereus, Listeria monocytogenes, Pseudomonas aeruginosa, Salmonella typhimurium, Escherichia coli, Saccharomyces cerevisiae, Candida albicans, Penicillium citrinum, Aspergillus flavus and Aspergillus niger. The methanol extracts of Cornus officinalis, Evodia officinalis, Glycyrrhiza glabra, Salvia miltiorrhiza. Schizandrae fructus, Coptidis rhizoma, aroma hop and bitter hop were shown inhibitory effect on certain species of gram(+) bacteria. Aroma hop and bitter hop were shown inhibitory effect on certain species of gram(-) bacteria. The methanol extract of Salvia miltiorrhiza exhibited a strong antibacterial activities. It was purified by solvent fractionation, silicagel column chromatography, prep. TLC, prep. HPLC. The purified active substance was identified as cryptotanshinone by EIMS, $1^H-NMR,\;{13}^C-NMR$ and DEPT. Cryptotanshinone showed a strong antibacterial activity against gram positive bacteria $(MIC\;:\;3.91{\sim}62.50\;{\mu}g/mL)$. Especially, this compound was the most strong activity against Bacillus subtilis $(MIC\;:\;3.91\;{\mu}g/mL)$.

Antimicrobial Effect of Ampelopsis brevipedunculata Extracts On Food Spoilage or Foodborne Disease Microorganism (개머루덩굴 추출물의 식품부패 및 병원성 미생물에 대한 항균효과)

  • This study was performed to investigate the antimicrobial effects of Ampelopsis brevipedunculata ethanol-extract on food spoilage or foodborne disease microorganism. The antimicrobial activity of Ampelopsis brevipedunculata extract was determined by a paper disc method. The extract exhibited growth inhibiting activities in a concentration dependent manner on 7 species of microorganisms. The extract of Ampelopsis brevipedunculata showed the highest antimicrobial activity against Escherichia coli O157:H7. The growth inhibitory effects of the extract at the concentration of 250, 500, 1,000, 2,000 mg/L on food poisoning microorganism were determined against Salmonella typhimurium, Yersinia enterocolitica, Escherichia coli O157:H7 and Staphylococcus aureus. The growth of microorganisms was not affected by the extract at the concentration up to 250 mg/L, but was significantly (p<0.05) inhibited by the extract at the concentration higher than 500 mg/L. The results in the present study demonstrate antimicrobial effects of Ampelopsis brevipedunculata ethanol-extract against foodborne pathogens, suggesting that Ampelopsis brevipedunculata could be an effective natural antibacterial agent in food.

The Effects of Food Additives on the Shelf-life of Low-salted Myungran-jeot (식품첨가제에 의한 저염 명란젓의 보존 효과)

  • The biggest Problem of low-salted Jeot-gal is the reduction of its shelf-life. Chitosan, glucono $\delta-lactone, $ and sykeeper were added to extend the shelf-life of low-salted Myungran-jeot, and various chemical and microbiological analyses were carried out during fermentation at $10^{\circ}C.$ Sykeeper inhibited the increase in pH, and production of lactic acid, VBN, TMA, TBA, and the microbial growth, whereas enhanced the production of amino-N. But, chitosan and glucono $\delta-lactone$ had no significant difference from the control. The estimated shelf-life periods of low-salted Myngran-jeot fermented at $10^{\circ}C$ for control, chitosan, glucono $\delta-lactone, $ and sykeeper were about 14, 12, 16, and 19 days, respectively.

Antimicrobial Effect of Methanol Extracts from Some Medicinal Herbs and the Content of Phenolic Compounds (약용식물 추출물에 대한 항미생물 활성 검색과 폴리페놀 함량)

  • Methanol extracts were prepared from 32 medicinal herbs of the extracts were tested their microbial inhibition activities against food borne pathogens and/or food poisoning microorganisms, food-related bacteria and yeast. Methanol extracts of Cinnamomum cassia, Paeonia suffruticosa, Alnus japonica, Eugenio caryophyllata and Illicium verum exhibited antimicrobial activity for the microorganisms tested, except lactic acid bacteria and yeast. Minimum inhibitory concentrations(MIC) were about 5 mg/mL for Bacillus subtilis, Staphylococcus epidermidis and Pseudomonas aeruginosa. Cell growth of lactic acid bacteria was inhibitied, but greatly on Leuconostoc mesenteroides. The phenolic compound contents were 10.98 mg/g, 10.31 mg/g, 8.55 mg/g and 6.69 mg/g in Thea sinensis, Eugenia caryophyllata, Alnus japonica and Artenisia capillaris, respectively. Antimicrobial activity appeared to be related to phenol compound content in medicinal herbs. The methanol extracts of medicinal herbs could be suitable for the development of a food preservative.

A survey on status of quality and risk assessment in dentifrices and mouthwashes (치약제 및 구중청량제의 품질 실태 조사 및 안전성 평가)

  • The quality of the products was investigated by analyzing fluorine content, pH, preservatives and tar colors in 31 dentifrice products (6 items for children) and 15 mouthwash products (2 items for children) marketed. It was intended to provide correct information to consumers by checking whether the standards and product indications match. As a result of measuring the fluoride concentration, 26 dentifrice and 15 mouthwash products contained from 48 to 1,472 ppm and from 85 to 225 ppm, respectively. Fluorine detection rates of dentifrice and mouthwash products were 83.9 and 83.3 %, respectively showing similar levels. Of the 41 fluoride-detected dentifrice and mouthwash products, 40 were 90.7~109.8 % of the displayed amount and suitable for the fluorine content standard of 90.0 to 110.0 %, but one dentifrice was found to be inappropriate at 36.3 % of the content indicated on the product. The pH of the dentifrice was 5.1~9.4, and the mouthwash was 4.2~6.2, which met all standards. As a result of simultaneous analysis of the concentration of six preservatives, benzoic acid was detected the most in 15 cases with a 30.6 % detection rate, sorbic acid was detected in 9 cases (detection rate of 18.4 %), and all four types of methyl p-hydroxybenzoate, ethyl p-hydroxybenzoate, propyl p-hydroxybenzoate, butyl p-hydroxybenzoate were not detected. As a result of analyzing the concentration of 10 types of tar colors, six types including red40, yellow4, yellow5, yellow203, green3, and blue1 were detected in a total of 9 cases (2 dentifrices and 7 mouthwashes) with blue1 being the most frequently detected. Detected fluorine concentration, added preservatives and tar colors were consistent with the product markings and it was well written on product packaging. The detected preservatives and tar colors were at a safe level due to low risk compared to Acceptable Daily Intake.

Biochemical Properties and Application of Bacteriocins Derived from Genus Bacillus (Bacillus속 세균 유래 박테리오신의 특성과 응용)

  • Bacteriocins are antimicrobial peptides synthesized on ribosomes, produced by bacteria, that inhibit the growth of similar or closely related bacterial strains. Since the discovery of nisin, many bacteriocins with unique structures and various modes of antibacterial activity have been described, and genes encoding production, secretion, and immunity have been reported. Nisin is one of the bacteriocins applied in cheese, liquid eggs, sauces and canned foods. Many of the bacteriocins of the genus Bacillus belong to lantibiotics, which are modified peptides after translation. Other genus Bacillus also produce many non-lantibiotic bacteriocins. Bacteriocins of the genus Bacillus are sometimes becoming more important because of their broader antibacterial spectrum. Bacteriocins are considered attractive compounds in the food and pharmaceutical industries to prevent food spoilage and growth of pathogenic bacteria. Bacteriocins can be used as biological preservatives in a variety of ways in the food system. Biopreservation refers to extending shelf life and improving safety of foods using microorganisms and/or their metabolites. The demand for new antimicrobial compounds has generated great interest in new technologies that can improve food microbiological safety. Applications of bacteriocins are expanding from food to human health. Today, many researchers are shifting their interest in bacteriocins from food preservation to the treatment of bacteria that cause infections and antibiotic-resistant diseases. This exciting new era in bacteriocin research will undoubtedly lead to new inventions and new applications. In this review, we summarize the various properties and applications of bacteriocins produced by the genus Bacillus.

A Survey of the Safety in Seasoned Dried Fishes (조미건어포류의 안전성에 대한 조사연구)

  • A survey of food preservatives, sulfur dioxide and microbial contamination levels was conducted on 73 seasoned dried fishes(41 squids, 17 filleted fish, 6 Alaska pollack, 9 others) collected from domestic markets and internet. Among preservatives, only sorbic acid was detected in 25 samples including 23 seasoned dried squids, 1 seasoned filleted fish and 1 seasoned dried Alaska pollack with a range $0.01{\sim}0.92g/kg$. Sulfur dioxide was detected from one sample(seasoned dried squid) and its content was 10.5 mg/kg. All samples were contaminated by total aerobic bacteria, and the degree was $3.00{\sim}6.45\;log_{10}$ CFU/g(average level was $5.09\;log_{10}$ CFU/g). Escherichia coli, Salmonella spp. and Staphylococcus aureus were not detected in all samples. On the basis of the results above, most of the seasoned dried fishes were appropriate for Korea food standard, yet biological hygiene control will be needed constantly.
