Antimicrobial Effect of Lonicerae Flos Extracts on Food-borne Pathogens (식중독 유발세균의 증식에 미치는 금은화 추출물의 항균효과)

  • Lonicerae Flos was extracted with methanol and successively fractionated with petroleum ether, chloroform, ethyl acetate, and methanol to investigate their antimicrobial effects against food-borne pathogens and food spoilage bacteria using paper disc method. Ethyl acetate extracts of L. Flos showed highest antimicrobial activity against Shigella dysenteriae. Synergistic effect in inhibition was observed when L. Flos extract was mixed with Artemisa capillaris extract as compared to using each extract alone. Growth inhibition curves were determined using ethyl acetate extracts of L. Flos against Staphylococcus epidermidis and S. dysenteriae. Ethyl acetate extract of L. Flos had antimicrobial activity against S. dysenteriae at 3,000 ppm, retarding growth of S. dysenteriae up to 12 hr.

  • 본 연구는 식품의 이온화 조사 처리기술로 얻을 수 있는 경제적 이득을 정량적으로 보여줄 수 있는 자료를 마련하기 위한 것이다. 이온화 조사가 허용되었거나 앞으로 허용 가능한 농산물의 수확 후 관리 손실 비용을 분석하고, 가공식품의 저장성 향상에 의한 경제적 이득에 대하여 조사하였으며, 식품의 이온화 조사로 예방 할 수 있는 식중독 사고의 정도와 그 사회 경제적 비용절감을 추산하였다. 신선식품의 연간 수확 후 손실 비용은 감자, 마늘, 양파, 고추에 대해 총 1조 1,251억 원으로 추산되었다. 현재 이온화 조사가 허가된 품목(다류, 장류, 조미식품, 드레싱)의 유통기한 초과로 인한 폐기 손실액은 899억 원으로 조사되었다. 현재 이온화 조사가 허용되지는 않았으나 향후 조사품목을 확대하였을 때 포함될 수 있는 품목(식육과 알 가공품, 어육가공품, 건포류, 축산가공품)의 유통기한 초과에 의한 폐기손실액은 총 1,931억 원으로 조사되었다. 식품의 이온화 조사로 예방 가능한 식중독의 보건 사회적 비용을 추산한 결과 이온화 조사로 사멸효과가 큰 E. coli O157, Campylobacter jejuni, Salmonella spp.에 의한 식중독 감소 효과로 연간 880억 원의 보건 사회적 비용 감소효과를 가져올 것으로 보여지며, 전체 세균성 식중독으로 확대하여 분석 한 결과 1,790억 원의 비용 감소효과를 가져올 것으로 추산되었다. 이온화 조사 처리기술은 식중독 예방에 의한 보건의료비의 절감 기능과 아울러 식품산업의 유통손실 절감과 식량 유실을 막는 효과적인 방법이므로 식량안보적 차원에서 적극 활용되어야 할 것으로 판단된다.

An Analysis of Epidemiological Investigation Reports Regarding to Pathogenic E. coli Outbreaks in Korea from 2009 to 2010 (최근 2년간(2009-2010) 우리나라 병원성 대장균 식중독 역학조사 보고서 분석)

  • Recently pathogenic E. coli is one of the main foodborne pathogens resulting in many patients in Korea. To understand the characteristics of pathogenic E. coli outbreaks in Korea, the epidemiological investigation reports of pathogenic E. coli outbreak in 2009 (41 reports) and in 2010 (27 reports) were collected in the web site of the Korea Centers for Disease Control and Prevention, reviewed and analysed in this study. The main places of the pathogenic E. coli outbreaks were food catering service area (64.8%) and restaurants (25.0%). The main type of the pathogens were EPEC (44.7%) and ETEC (34.2%). EAEC and EHEC was responsible for 10.5 and 9.2%, respectively. Eight of 68 outbreak cases were caused by more than 2 types of pathogenic E. coli which implicates the complicated contamination pathways of pathogenic E. coli. The incidence rate of pathogenic E. coli was $33.6{\pm}30.5%$ and the main symptoms were diarrhea, stomach ache, nausea, vomiting, and fever etc. The two identified food sources were identified as frozen hamburger pattie and squid-vegetable mixture. To improve the food source identification by epidemiological investigation, food poisoning notification to the agency should not be delayed, whole food items attributed the outbreak should be collected and detection method of the various pathogenic E. coli in food has to be improved. In conclusion, the characteristics between the EHEC outbreaks in the western countries and the EPEC or ETEC outbreaks in Korea needs to be distinguished to prepare food safety management plan. In addition, the development of the trace back system to find the contamination pathway with the improved detection method in food and systemic and cooperative support by the related agencies are necessary.

단기 숙성 생햄의 냉장 및 실온저장 중 미생물 및 관능학적 품질

  • 본 연구는 단기 숙성형 생햄의 식중독균 오염 상태와 냉장 및 실온 저장 중 미생물 및 관능학적 품질 변화를 살펴보기 위하여 실시되었다. 생햄의 초기 총균수는 3.06 log CFU/g이었으며 저장 말기까지 균 수 증가는 미미하였다. 주 종균은 유산균과 Staphylococcus 균이었다. 10과 $25^{\circ}C$의 저장 온도에서 생햄의 저장 수명은 각각 75와 45일 정도로 예상되었다. 본 실험에서 생햄 원료 및 저장 중 단지 $25^{\circ}C$에서 21일 저장된 시료에서 C. perfringens가 검출되었고 다른 시료에서는 비가열육제품의 위생 기준으로 명시된 6가지 식중독균은 검출되지 않았다. 종합적으로 판단하건데 현실적으로 비가열 제품의 경우 국내 식중독 미생물에 대한 음성 기준을 만족시키기는 어렵고 외국 기준에 비하여 너무 엄격하므로 관련 기준의 완화가 요구된다.

Genetic diversity of Clostridium perfringens form food-poisoning outbreak in Gyeonggi-do, 2013-2014 (경기도 식중독에서 분리된 Clostridium perfringens의 유전적 특성 분석)

  • Clostridium perfringens is both a ubiquitous environmental bacterium and a major cause of human gastrointestinal disease, and C. perfringens food poisoning ranks among the most common gastrointestinal diseases in developed countries. 120 isolates of C. perfringens were obtained from food-poisoning outbreaks in 2013~2014, Gyeonggi-do. Using PCR, all 120 isolates were identified as C. perfringens type A. Of the tested isolates, 49 isolates carried the cpe gene, 71 isolates carried the cpb2 gene. The outbreak cases of cpb2 and cpe /cpb2 genes were 7 and 7, whereas the outbreak cases of cpe-gene were 2. The epidemiological relationship between C. perfringens isolates has previously been investigated chiefly by pulsed-field gel electrophoresis (PFGE) and multi-locus sequence typing (MLST). The genetics relatedness of the isolates raged from 53.5-100% and 75 district PFGE type were observed. The PFGE results revealed a wide genetic diversity among the 64 cpb2 carrying isolates (except 7 isolates), while 46 cpe-carrying isolates (except 3 isolates) showed a high genetic similarity. The MLST analysis revealed that 14 cpe isolates (cpe-chromosomal isolates) belong to a distinct cluster that is significantly distant from all the other cpb2 isolates (cpe-plasmid carrying and cpe-negative isolates). The isolates carrying a cpb2 appear to be rarely related, and are more variable than chromosomal cpe isolates. The results suggest that the cpe-positive outbreak isolates showed close genetic relation, whereas the cpb2-positive isolates revealed a wide genetic diversity.

Effect of Agrimonia Pilosa Ledeb. Extract on the Growth of Food-Borne Pathogens (선학초 추출물이 식중독 유발세균의 증식에 미치는 영향)

  • This study was performed to investigate the antimicrobial effect of the Agrimonia pilosa Ledeb. extracts against food-borne pathogens. First, the Agrimonia pilosa Ledeb. was extracted with methanol at room temperatures, and fractionation of the methanol extracts from Agrimonia pilosa Ledeb. was carried out by using petroleum ether, chloroform, and ethyl acetate, and methanol respectively. The antimicrobial activity of the Agrimonia pilosa Ledeb. extracts was determined using a paper disc method against food-borne pathogens and food spoilage bacteria. The petroleum ether extracts of Agrimonia pilosa Ledeb. showed the highest antimicrobial activity against Pseudomonas aeruginosa. The synergistic effect has been found in combined extracts of Agrimonia pilosa Ledeb. and Perillae folium as compared to each extracts alone. Finally, the growth inhibition curve was determined using ethyl acetate extracts of Agrimonia pilosa Ledeb. against Bacillus Cereus and Salmonella Enteritidis. The petroleum ether extract of Agrimonia pilosa Ledeb. showed strong antimicrobial activity against Bacillus Cereus at the concentration of 4,000 ppm. The 4,000 ppm of petroleum ether extract from Agrimonia pilosa Ledeb. retarded the growth of Bacillus Cereus more than 24 hours and Salmonella Enteritidis up to 36 hours. The petroleum ether extracts of Agrimonia pilosa Ledeb. has been shown the antimicrobial effect against Bacillus Cereus and Salmonella Enteritidis. (Korean J Nutrition 38(2): 112~116, 2005)

Survival of Food-borne and Pathogenic Microorganisms in Hot Spring Water (온천수에서 식중독 및 병원성 미생물의 생존 양상)

  • This study was conducted to investigate the effects of hot spring water against the survival of food-borne and pathogenic microorganisms. Staphylococcus aureus, Bacillus cereus, and Escherichia coli, which are food-borne microorganisms, Candida albicans and Trichophyton mentagrophytes, which are skin disease pathogens, and Helicobacter pylori, gastritis inducing microorganism, were tested. The content of fluoride in tested hot spring water is 14.1 mg/L, which is higher than the standard of safe for drinking water 1.5 mg/L, but the results on 48 other items were up to the standard. Hot spring water didn't show the bactericidal effects against food-borne microorganisms, C. albicans, and H. pylori tested. However, the viable cell populations of B. cereus and T. mentagrophytes were decreased, which were depends on the temperature of hot spring water. From these results, we confirmed that hot spring water didn't show the bactericidal effects against food-borne microorganisms, skin disease pathogens, and gastritis inducing microorganism, but the growth of some microorganisms were inhibited by high temperature ($41^{\circ}C$).

Analysis of the Recovery Rate of Food-borne Pathogens according to Sample Preparation Methods in Animal Origin Foods (축산식품 중 전처리 방법에 따른 식중독균 회수율 분석)

  • This study was performed to evaluate and establish a sample preparation method for the detection of food-borne pathogens in animal origin foods. Ham, yogurt, and Korean beef inoculated with Escherichia coli O157:H7, Staphylococcus aureus, and Salmonella Typhimurium, were tested for the effects of diluent composition, processing time, and proportion of diluent to sample. The diluents used were peptone water (PW), Saline solution (SS), Butterfield's phosphate buffered dilution water (BPD), and Buffered peptone water (BPW). The processing time periods considered for the samples were 30, 60, 90, 120, and 300 sec, and the proportions of diluent to samples tested were 1:2, 1:4, 1:9, and 1:19. Yogurt and beef showed the highest number of bacteria when treated with BPW (p < 0.05). However, ham showed no significant difference between the treatments with four different diluents. Optimum proportions of diluent to ham, yogurt, and beef were 1:9, 1:2, and 1:4, respectively. The processing time of 120 sec was chosen as optimum, because it showed the best recovery rate in all sample types. In this manner, detection of food-borne bacteria with the selected optimal conditions was indicated by a recovery rate of more than 85%. These data suggest that an appropriate diluent composition and diluent volume should be used depending on the type of sample, which would thereby increase the accuracy of detecting food-borne bacteria in animal origin foods.
