Improvement of Storaging Ability of Waxycorn by Retort Pouch Technique (레토르트 파우치 방법에 의한 찰옥수수의 저장성 향상)

  • Retort pouch is widely used in food industry for a long-term preservation and safe food production. By applying retort pouch technique to waxycorn storage, the quality of waxycorn could be maintained and the storage expense could be saved during storage. Water activities(Aw) of retort waxycorn were below 0.80 except blanching treatment, and it is known that microbial propagation is subdued below 0.80. Commercial value of waxy corn was deteriorated when it was frozen quickly at $-40^{\circ}C$ before treating Retort due to obscurity of chromatocity, while the color change was not noticeable when it was treated with Retort at $121^{\circ}C$ for 10min. For all treatments, very small amounts of free sugar were detected, however, there were no significant differences between treatments. As storage period was longer, shelf lifes of waxy corn in control and waxy corn treated with blanching were more shortened when waxy corn was stored at $15,5^{\circ}C$ before Retort, while waxy corn with boiling treatment was not significantly different compared with that in storage in freezer.

Effect of Neurogranin Phosphorylation on Oxidative Stress by Hydrogen Peroxide in Early Onset of Batten Disease (과산화수소에 의한 산화스트레스가 영아형 바텐병에서 neurogranin의 인산화에 미치는 영향)

  • Early onset of Batten disease (EBD), one of the most lethal neurodegenerative storage disorders of childhood, is caused by inactivating mutations in the Ceroid Lipofuscinosis, Neuronal (CLN1) gene. Neurogranin, a calmodulin-binding protein, is expressed in the brain and participates in the protein kinase C (PKC) signaling pathway. While oxidative stress is the suggested cause of neurodegeneration in EBD, its molecular mechanism(s) remains obscure. In this research, we examined the levels of neurogranin in the brain mRNA of wild-type (WT) mice and EBD knockout (KO) mice, as well as the proteins. We also performed neuronal cultures to measure the expression levels of neurgranin and phosphorylated-neurogranin with or without oxidative stress inducers and anti-oxidants. Results showed that neurogranin in both EBD KO mice brain mRNA and protein extracts decreased in an age dependent manner. However, high amounts of phosphorylated-neurogranin were detected in the 6-month brain. This pattern was also confirmed by cultured neurospheres samples. Moreover, neurospheres treated with $H_2O_2$, an oxidative stress inducer, showed increased phosphorylated-neurogranin patterns. Interestingly, this pattern returned to normal status when treated with N-acetyl-L-cystein, an anti-oxidant, after $H_2O_2$ treatment was performed. Our results suggest that the phosphorylation of neurogranin is affected by oxidative stress status in EBD, and appropriate anti-oxidant treatment will relieve hyper-phosphorylation of neurogranin.

Effects of Plum Fruits Extracts at Different Growth Stages on Quinone Reductase Induction and Growth Inhibition on Cancer Cells (생육시기별 피자두 추출물의 암세포 증식 억제 효과 및 Quinone Reductase 유도 활성에 미치는 영향)

  • The plum (Prunus salicina L., cv. ‘Soldam’) fruits were harvested at different growth stages, and then extracted using 80% methanol, respectively. The methanol extracts of plum were investigated for their growth inhibition on 6 kinds of human cancer cells using MTT assay and for their activity to induce quinone reductase (QR) in murine Hepa1c1c7 cells. Among various methanol extracts of plum, the plum 1~4 (immature fruit), which thin out 10~25 days before final harvest, showed higher anticarcinogenic activity against 5 kinds of cancer cells than plum 5~9 (intermediate-mature and mature fruit). Especially, plum 1 and 2 were exhibited the strongest growth inhibiting activities to AGS, HepG2 and MDA cancer cells. Also the plum extracts induced the activity of QR, an anticarcinogenic marker enzyme, in Hepa1c1c7 cells while the induction of QR activities by adding plum extracts were shown to be a little difference depending on growth stages. These results suggested that methanol extracts of immature plum can be considered as an effective natural cancer chemoprevention materials.

Characteristics of Volatile Fatty Acids Release During the Hydrolysis of Rice Straw (볏짚의 가수분해과정중 유기산 생성 특성)

  • Objective of this study was to investigate the effects of pre-treatment of rice straw by sizes(3cm, milled), temperatures($35^{\circ}C\;and\;55^{\circ}C$), with/without NaOH treatment, and RPM on the characteristics of volatile fatty acids production. The concentration of total volatile fatty acids (TVFAs) was increased with the hydrolyzed time. Concentration of VFAs in milled rice straw was higher than that in 3 cm cut. With the alkali treatment, the concentration of TVFAs were sharply increased, which showed 3 times higher than non-treatment. Concentration of VFAs was high at 150 rpm at $35^{\circ}C$, and at 80rpm and 200 rpm at $55^{\circ}C$. Among them acetic acid was dominant, which showed the similar increase with TVFAs. It was also observed that in the case of fibrous material, the contents of cellulose and hemi-cellulose were reduced a little, but no change in lignin.

Seasonal Change in Inorganic Nitrogen Content in the Soil Profile of Urea-Fertilized Grassland (요소(尿素)(Urea)를 시용(施用)한 초지(草地)의 토양단면(土壤斷面)에서 무기태질소(無機態窒素) 함량(含量)의 계절적(季節的) 변화)

  • Field experiment was conducted to investigate the seasonal change in inorganic nitrogen content in grassland soil profile after urea application. Urea was applied at the levels of 0 (0N), 14 (14N), and 28 (28N) Kg N per 10a. Soil samples were taken at every 20 cm interval upto 100 cm soil depth in spring (May 26), summer (July 27), and autumn (October 18) and analysed for total and inorganic nitrogen ($NH_4-N$ and $NO_3-N$). The results obtained are as follows ; 1. In spring, the $NH_4-N$ content of ON treatment was higher than $NO_3-N$ content both in surface and subsoil. The urea application increasing both $NH_4-N$ and $NO_3-N$ contents in the surface soils and these contents decreased with soil depth. 2. In summer, increase in urea application rate elevated the $NO_3-N$ content in soil profile of 0 to 100cm and the content reached upto 42 ppm in the 28N treatment. 3. The seasonal difference in $NH_4-N$ content between summer and autumn was insignificant throughout soil profile. Soil $NO_3-N$ content in autumn were 7 and 14 ppm for 14N and 28N respectively, showing very low values compared with that of summer. 4. The ratio of inorganic nitrogen to total nitrogen increased with soil depth and with urea application rates.

The Effects of Pretreatment on the Lipid Oxidation of Korean Geoduck Muscle during Frozen Storage (코끼리 조개의 동결저장 중 지질의 변화에 미치는 전처리의 효과)

  • The effects of pretreatment on the lipid oxidation of korean geoduck muscle during storage at $-20^{\circ}C$ were investigated. In the lipid compositions of geoduck muscle, neutral, glycolipid, and phospholipid contents were $46.1\%,\;4.5\%\;and\;49.5\%$, respectively. And the content of phospholipid decreased to $39.9\%$ by blanching. In the fatty acids composition, $C_{16:0},\;C_{20:5}\;and\;C_{22:6}$ fatty acids were major portions and their contents showed, respectively, $10\%$ of total fatty acids amount or more. $C_{18:0},\;C_{16:1},\;C_{18:1},\;C_{14:0}\;and\;C_{20:1}$ fatty acids were also important fatty acids and their amounts were so much as $5\%$ or more. The polyunsaturated fatty acid contents were relatively high as $39.25\%{\sim}42.60\%$. POV in RM and VM samples increased gradually and showed 3.72meq/kg, or 3.00meq/kg after 90 days storage, respectively. However, in case of samples pretreated by blanching, there was slightly increase of POV throughout storage. From the results above, it was suggested that the pretreatment of blanching was effective in retardation of lipid oxidation for the longterm storage.

The Neutralization Treatment of Waste Mortar and Recycled Aggregate by Using the scCO2-Water-Aggregate Reaction (초임계이산화탄소-물-골재 반응을 이용한 폐모르타르와 순환골재의 중성화 처리)

  • The batch and column experiments were performed to overcome the limitation of the neutralization process using the $scCO_2$-water-recycled aggregate, reducing its treatment time to 3 hour. The waste cement mortar and two kinds of recycled aggregate were used for the experiment. In the extraction batch experiment, three different types of waste mortar were reacted with water and $scCO_2$ for 1 ~ 24 hour and the pH of extracted solution from the treated waste mortar was measured to determine the minimum reaction time maintaining below 9.8 of pH. The continuous column experiment was also performed to identify the pH reduction effect of the neutralization process for the massive recycled aggregate, considering the non-equilibrium reaction in the field. Thirty five gram of waste mortar was mixed with 70 mL of distilled water in a high pressurized stainless steel cell at 100 bar and $50^{\circ}C$ for 1 ~ 24 hour as the neutralization process. The dried waste mortar was mixed with water at 150 rpm for 10 min. and the pH of water was measured for 15 days. The XRD and TG/DTA analyses for the waste mortar before and after the reaction were performed to identify the mineralogical change during the neutralization process. The acryl column (16 cm in diameter, 1 m in length) was packed with 3 hour treated (or untreated) recycled aggregate and 220 liter of distilled water was flushed down into the column. The pH and $Ca^{2+}$ concentration of the effluent from the column were measured at the certain time interval. The pH of extracted water from 3 hour treated waste mortar (10 ~ 13 mm in diameter) maintained below 9.8 (the legal limit). From XRD and TG/DTA analyses, the amount of portlandite in the waste mortar decreased after the neutralization process but the calcite was created as the secondary mineral. From the column experiment, the pH of the effluent from the column packed with 3 hour treated recycled aggregate kept below 9.8 regardless of their sizes, identifying that the recycled aggregate with 3 hour $scCO_2$ treatment can be reused in real construction sites.

Changes of Flavor Components in Chewing Gum during Storage by Gas Chromatography/Mass Spectrometry (GC/MS에 의한 저장중 츄잉껌 향기성분 변화연구)

  • The changes of amount in flavor components of chewing gum during storage of various relative humidity were studied by GC/MS. The volatile components in chewing gum were extracted by sample preperation and 15 volatile components were identified by NIST/NBS library searching. According to the period of storage, quantities of major volatile components extracted by tetrahydrofuran-methanol solutions were determined by using tetradecane as internal standard. Until 4 weeks, comparing with starting peak area ratio, the decrease of chewing gum flavor components were rapid at 33, 75 than 53 R.H(%). And after 16 weeks, amount of chewing gum flavor at all storage conditions similarly decreased to $45{\sim}49%$ of the initial amount.

  • 청정 에너지원으로 높은 잠재력을 가지고 있는 가스하이드레이트는 상업적 기술개발이 미확보된 상태이다. 현재 전 세계적으로 가스하이드레이트 개발 및 생산에 관한 연구가 활발히 진행되고 있으며 이에 대한 기초자료로서 가스하이드레이트가 함유된 퇴적층의 물성자료가 필요하다. 특히, 현장 시료에 대한 물성 측정은 향후 가스하이드레이트 개발 및 생산 계획을 수립하는데 있어서 매우 중요하다. 탄성파 측정 결과는 다른 물성 들에 비하여 하이드레이트 함유 시료의 성형과정에 큰 영향을 받는다. 또한 그 외의 실험 경계조건과 취득 자료의 처리 과정에도 매우 민감하게 반응한다. 따라서 측정을 하는 과정은 물론 측정 후 자료의 활용 과정에서 다양히 고려해야 할 점들이 있다. 본 연구에서는 인공 모래를 이용하여 다양한 조건에서 탄성파 속도를 측정한 후 그 결과를 토대로 하여 기존의 연구 결과와 비교하여 음파 측정연구 시 고려해야 할 기술적 사항 들을 정리해 보았다. 실험에 사용된 장비는 고압의 퇴적층을 모사할 수 있는 압력셀과 메탄과 염수 주입에 사용되는 유체 주입장비, 하이드레이트 형성을 위한 온도조절장비, 자료 획득 장비로 구성되어 있다. p파 속도는 음파 송수신장비를 사용하였다.

Development of the Sample Pretreatment Technique using Microwave for Analysis of Insecticide Imidacloprid Residues (마이크로파를 이용한 잔류 살충제 Imidacloprid 분석용 시료전처리 기술개발)

  • This study was conducted to elucidate extraction efficiency by microwave technique in comparison with sonication technique for extraction of insecticide residue in pear. In the analysis of the extraction efficiency of microwave for a pear spiked with imidacloprid, the extraction efficiency by microwave power of 300 W with extraction temperature of $80^{\circ}C$, heating time of 1 to 3 minute was shown to be similar with the extraction time 20 minutes by sonication. The optimal condition. in consideration of economical condition and treatment time, for microwave extraction of imidacloprid in the pear were 300 watts of power supply, $100^{\circ}C$ of extraction temperature, 1 minute of heating time and 10 mL of acetone volume. A new microwave vessel was developed to rapidly process the sample of the insecticide imidacloprid residues in the pear. This vessel was designed to include a reaction chamber and a filtration equipment, and a gathering chamber. The system could curtail a pretreatment time to 21 minutes than sonication and 7.9 minutes than the previous microwave vessel.
