Prediction of Sucrose Hydrolysis Rate using Equivalent Time at A Reference Temperature under Regular Temperature Fluctuations (규칙적인 온도변화에서 표준온도 상당시간을 이용한 Sucrose 가수분해속도의 예측)

  • A simple approximate model using equivalent time at a reference temperature ($t_{eq}$) was derived to predict quality changes caused by temperature fluctuations. The validity and effectiveness of this model have been assessed with experimental data of sucrose hydrolysis. Kinetic parameters of sucrose hydrolysis were estimated by one step method using equivalent time at a reference temperature with linearly increasing temperature profile. Sucrose hydrolysis was a first order reaction, and the activation energy was 25.84 kcal/mol. The extent of sucrose hydrolysis of liquid model system under accelerated test with sinusoidal temperature fluctuations were determined. The proposed model yielded accurate prediction with the correlation coefficient in the range of $0.92{\sim}0.99$.

EROSION OF TOOTH ENAMEL BY ACIDIC DRINKS AND REMINERALIZATION BY ARTIFICIAL SALIVA (산성 음료에 의한 법랑질의 침식과 인공타액에 의한 재광화)

  • The purpose of study was to assess the influence of acidic drinks on the erosion of tooth enamel and the effect of fluoridated saliva on the remineralization. Twenty five drinks were sampled. The erosive potential and remineralizing effect were measured by the tooth surface microhardness test. The pH of most drinks were below pH 5.5. Reduction rates of enamel surface hardness by the flavored carbonated drink were 16.90%, 25.11%, 35.10%, and 41.62% after 5, 10, 30, and 60 minutes of demineralization, and recovery rates by remineralizaing solution were 61.52%, 67.96%, 72.13% and 75.93% after 1, 24, 48, and 72 hours of remineralization, respectively. The results suggest that the most drinks in the markets have the potential to erode the teeth and that erosion occurs fast but remineralization proceeds slowly.

  • Recently, mobile-type analysis system when emergency patient can check his blood condition in the transportation or vessel has being required. Under the present system, it takes lots off-time to know the blood analysis result of patient, so, it may lead to a dangerous situation. But, in this study, analysis time makes to fall down to $1{\sim}min$. to emergency treatment patient more quickly, and you can check the information by wireless PC through Bluetooth base. It is able to measure in about 7minutes from 25minutes of warming-up time in existing facilities by testing temperature peculiarity curve, to operate system without error even minute temperature change by adoption DUAL heating. And Bluetooth base was adopted to reduce power consumption and be able to hospital networking by keeping and sending analyzed data when it needs.

Biodegradation of Kerosene by Pseudomonas aeruginosa K14 (Pseudomonas aeruginosa Kl4를 이용한 등유(Kerosene)의 생물학적 분해)

  • In this study, we isolated 32 strains of kerosene degrading bacteria from oil contaminated soil by enrichment culture. Isolates were screened for kerosene degradation efficiencies and K14 were selected which had the highest removal efficiency for 1,000 mg/L of kerosene. K14 were identified as Pseudomonas aeruginosa by morphological, biochemical test and 16S rDNA analysis. The optimal culture condition were determined as initial inoculated cell concentration, 1.0 g/L; substrate concentration, 1,000 mg/L; temperature $30^{\circ}C$; pH 7. When we enforced batch test in this condition, K14 degraded 72% of kerosene with 1,000 mg/L during 72 hr. And, at low concentration (200 mg/L), K14 degraded 95.8% of kerosene during 48 hr. As a result, kerosene biodegradation by Pseudomonas aeruginosa K14 could be useful for clean up of groundwater and soil contaminated with crude oil.

Effects of Acid Hydrolysis on Isoflavone of Defatted Soybean Flour (산가수분해시 산의 농도가 탈지대두박의 isoflavone에 미치는 영향)

  • The effect of acid hydrolysis on isoflavone contents of defatted soybean flour (DSF) was investigated in this study. Isoflavone analyzed were daidzein, genistein, glycitein, daidzein, genistein and glycitein with using HPLC. The DSF suspension was heated at $95^{\circ}C$ for 4 hour with addition of HCl (0.25-3.00 N) and analyzed isoflavone. The results showed that the total isoflavone was increased as the HCl concentration increased up to 1.0 N HCl during heating, indicating conversion of glucoside isomers of isoflavone to its aglycons. However, higher concentration than 1.0 N HCl caused a relatively fast decrease in isoflavone after 4 hour of heating. These results were probably caused by degradation of isoflavone by high concentration of acid during heating. The glucosides of isoflavone were steadily decreased while its aglycons were rather increased during $1.0{\sim}2.0$ hour of heating.

  • Carbohydrate-free caseinomacropeptide (CMP) was isolated from the sweet whey powder by a precipitation method using 12% trichloroacetic acid. The yield of carbohydrate-free CMP was 2.7 g from 100 g sweet whey powder. The electrophoretic pattern and the amino acid analysis of CMP showed that isolated CMP was quite pure, indicating the precipitation with 12% trichloroacetic acid was very effective for isolating carbohydrate-free CMP from the sweet whey powder. Trypsin, covalently immobilized on pore glass beads by carbodiimide (EDC) method, was 20mg per 1g glass beads. CMP was almost completely hydrolyzed by soluble trypsin in 24hr, but not by immobilized trypsin. The tryptic hydrolysates were fractionated on a Bio-Gel P 4 column $(1.5{\times}120\;cm)$and separated peptides were tested for their capacities to inhibit platelet aggregation using a aggregometer. The hydrolysate obtained from CMP after 24hr digestion by immobilized trypsin showed the highest activity.

Effect of Whole Soy Flour on the Properties of Wet Noodle (대두분말의 첨가가 제면특성에 미치는 영향)

  • Addition of whole soy flour(WSF) to wheat flour(WF) was studies for its effects on dough and noodle characteristics. The WF used was medium grade of strength and WSF was a fine flour of 350 mesh. The addition ratio of WSF to. n was up to 20%. The dough properties and textural properties of wet and cooked noodles were measured with using Farinograph, Amylograph and Rheometer. Farinogram data showed the minimum dough development time and stability at 6% addition of WSF. Amylograph data of initial pasting temperature and time were increased while the maximum and final viscosity and setback were decreased as the WSF added more to W The initial pasting temperature of 2.0∼6.0% WSF added flour were comparable to 100% WF eventhough viscosity was almost half of WF. The water absorption capacity was increased from 81.6% to 92.3% at 6.0% WSF and then decreased as the addition ratio of WSF increased. The extensibility of dough showed a maximal value at 8.0% WSF addition. Strengthness and hardness of wet and cooked noodle were increased to the heighest measurement as the WSF added up to 6.0% followed by a steady decrease thereafter. It was also found that WSF addition resulted an increase in adhesiveness of wet noodle and decrease in hardness and strengthness of cooked noodle.

Detection of TNF-alpha in Serum as the Effect of Corticosteroid to the Myocardial Protection in Cardiopulmonary Bypass (체외순환시 스테로이드의 심근보호효과에 관한 혈청내 TNF-alpha 측정의 의의)

  • Proinflammatory cytokines such as tumor necrosis factor-$\alpha$(TNF-$\alpha$) have been implicated in myocardial and organ dysfunction associated with postperfusion syndrome. We tested the hypothesis that cytokine productions are depressed by preoperative cortiosteroid injection for cardiopulmonary bypass(CPB) and the postoperative courses will be better than without steriod pretreated cases. Cardiac surgery was performed in randomized blind fashion for 20 patients from June 1996 to September 1996. In the steroid group(n=10), corticosteroid(dexamethasone 1 mg/kg) was injected 1 hour before anesthetic induction, but in the control group(n=10), nothing was injected. Each of groups were sampled 11 times as scheduled for TNF-$\alpha$ bioassays. We have checked EKG, cardiac enzymes(CPK, LDH with isoenzyme), WBC count preoperative day, one day and three days after operation. Viatal signs were continuously monitored for three postoperaive days. In the postoperative period three patients in the control group had elevated body temperature and four patients had hypotension that required considerable intravenous fluid administration. But steroid injected patients showed normal body temperture and acceptable blood pressures without supportive treatment. CPK enzymes rose in control group higher than steroid group at postoperative 1st and 3rd day(CPK; 1122$\pm$465 vs 567$\pm$271, 864$\pm$42 vs 325$\pm$87), and CPK-MB enzymes rose in control group higher than steroid group at postoperative 1st day(106.4$\pm$115.1 vs 29.5$\pm$22.4)(P=0.02). Arterial tumor necrosis factor-$\alpha$ rose during cardiopulmonary bypass, peaking at 5 minutes before the end of aortic cross clamping(ACC-5min) in steroid group(11.9$\pm$4.7 pg/ml), and 5 minutes before the end of cardiopulmonary bypass(CPB-5min) in control group(22.3$\pm$6.8 pg/ml). The steroid pretreated patients had a shorter period of time in respirator suport time, ICU stay day, hospital admission day. We conclude that corticosteroid suppress cytokine production during and after cardiopulmonary bypass, and may improve the postoperative course through inhibition of reperfusion injury such as myocardial stunning and hemodynamic instability.

  • 인간의 정서를 분류하려는 노력은 오래전부터 있어왔다. 시각 및 청각 자극을 사용한 심리생리적 정서연구는 최근에 들어 활발하게 이루어지고 있으나, 촉각 자극을 사용한 정서연구는 찾아보기 힘들다. 본 연구는 피부에 가해지는 다양한 자극에 따라 나타나는 정서와 관련된 뇌파특징을 측정하고자 하는 기초연구이다. 물리적 특성이 다른 다섯가지의 직물자극을 20명의 대학생 피험자(20-25세)에게 무선적으로 제시하였다. 각 자극의 제시시간은 1분이었으며, 자극간 간격은 3분이었다. 각 자극제시후 매자극후 피험자는 제시자극에 대한 느낌을 주관적 정서척도로 평정하였다. 뇌파는 양쪽 귓볼(A1, A2)을 기준전극으로 하여 좌.우반구의 전두엽(F3, F4), 두정엽(P3, P4), 후두엽 (O1, O2)에서 측정되었고, 측정된 뇌파는 FFT방법으로 분석되었다. 각 피험자의 주관적 평가의 결과에 따라 가장 쾌한 자극과 가장 불쾌한 자극이 제시되었을 때 측정한 뇌파의 분석결과를 비교하였다. 쾌자극 제시시의 뇌파는 불쾌자극 제시시보다 slow alpha(8-10Hz)파의 출현량은 적었다. 결론적으로 직물촉각자극에 의해 유발되는 심리적 반응에 특정적인 뇌파반응이 존재하며, 이는 촉각에 의해 유발되는 주관적인 쾌.불쾌정서를 객관적으로 측정하기 위한 지표로서 뇌파특징을 사용할수 있음을 의미한다.

Reactive Ion Etching을 이용한 PTFE 발수특성

  • 최근, 자연의 기능성 표면을 모사하여 우리 생활에 응용하기 위한 연구가 활발하다. 초-발수 특성을 가지는 대표적인 예인 연꽃잎은 마이크로-나노 크기의 거친 미세돌기(papillae)를 가지고 있으며 그 위에 낮은 표면 에너지를 가지는 왁스(wax)가 발달 되어 항상 깨끗한 상태를 유지한다. 본 실험에서는 이를 모사하여 RIE (Reactive Ion Etching)방법을 이용하여 기판인 Poly silicon wafer를 Sf6가스를 사용하여 Metal mash로 거칠기를 만들어 주었고, RF-magnetron sputtering 장치를 사용하여 $6{\times}10^{-3}$ Torr의 진공도에서 낮은 표면에너지를 가지는 PTFE (polytetrafluoroethylene)를 증착하여 표면 구조와 발수특성에 대하여 조사하였다. SSME(Surface shape measurement equipment)측정결과 0.24~0.36 um RSa 값이 측정되었고, 12 uL의 Di-water로 접촉각을 측정 한 결과 RIE 10분 처리를 한 기판 위에 PTFE를 3분 증착하였을 때 가장 높은 $153^{\circ}$의 초-발수 특성이 나타났으며, 4주의 시간이 지났을 때에도 접촉각이 유지가 되었다. XPS 측정결과 초-발수 표면에서 나타나는 CF2와 CF3 피크 값이 측정되었다. Reactive Ion Etching을 이용한 PTFE 발수 특성은 방수, 스마트 윈도우, 자가세정(Self-Cleaning), 디스플레이 표시장치, 김서림 방지(Anti-Fogging), 대전방지 코팅 등에 다각적으로 응용 가능할 것이라 사료된다.
