Comparative Transcriptome Analysis of the Response of Two Lines of Rapeseed (Brassica napus L.) to Cold Stress (유채 두 계통에서 저온 스트레스에 반응하는 전사체 발현 비교 분석)

  • Rapeseed is a typical winter crop, and its freezing stress tolerance is a major feature for winter survival. Therefore, it is important to comprehend clearly the physical and molecular mechanisms of rapeseed under freezing stress conditions. This study investigates the physical and transcriptome changes of two rapeseed lines, 'J8634-B-30' and 'EMS26', under cold acclimation and freezing temperature treatments. The proline content of 'J8634-B-30' at 5 ℃ increased 8.7-fold compared to that before treatment, and there was no significant change in that of 'EMS26' RNA-sequencing analysis revealed 5,083 differentially expressed genes (DEGs) of 'J8634-B-30' under cold acclimation condition. Among the genes, 2,784 (54.8%) were up-regulated and 2,299 (45.2%) were down-regulated. The DEGs of 'EMS26' under cold acclimation condition were 5,831 genes, and contained 2,199 up-regulated genes (37.7%) and 3,632 down-regulated genes (62.3%). Among them, only DEGs annotated in the cold response-related signaling pathways were selected, and their expression in the two rapeseed lines was compared. Comparative DEGs analysis indicated that cold response related signaling pathways are proline metabolism and ABA (Abscisic acid) signaling. And ICE (Inducer of CBF expression) - CBF (C-repeat-binding factor) - COR (Cold-regulated) signaling were the significantly differentially expressed transcripts in the two rapeseed lines. The major induced transcripts of 'J8634-B-30' induced P5CS (Δ'-pyrroline-5-carboxylate synthetase), which is related to proline biosynthesis, PYL (pyrabactin resistance-like protein, ABA receptor) and COR413 (cold-regulated 413 plasma membrane 1). In conclusion, these result provide a foundation for understanding the mechanisms of freezing stress tolerance in rapeseeds. Further functional studies should be performed on the freezing stress-related genes identified in this study, which can contribute to the transgenic and molecular breeding for freezing stress tolerance in rapeseed.

Change of Gene Expression Pattern of Mycobacterium tuberculosis H37Rv Against Host Immune Response in Infected Mouse Lung (결핵균 H37Rv에 감염된 마우스의 폐에서 면역 반응에 대항하는 Mtb 유전자의 발현 변화)

  • Mycobacterium tuberculosis (Mtb) is one of the most successful pathogens to infect one third of world population. Th1-mediated immunity against Mtb infection is known as critical to express mycobacteriostatic function but it is not sufficient to resolve the infection. In this study, to verify the possibility Mtb itself change the gene expression to survive against host immune response, expression pattern of selected H37Rv genes, 16S rRNA, acr, fbpA, aceA, and ahpC, during the course of infection was measured with absolute quantitation method using real-time RT-PCR. The total number of transcripts of 16S rRNA increased during the course of infection, which was coincide with the increasing CFU. The total number of fbpA transcripts per CFU, which encode typical secreted Mtb antigen, Ag85A, increased for 10 days of infection before decreasing. The number of transcripts of acr per CFU, which encode heat shock protein, ${\alpha}$-crystallin, increased during the infection, and ahpC and aceA, they both are enzymes produced in oxidative stressful condition, increased for 20 days and then slightly decreased on day 30. These findings are one of survival strategy of pathogen evading host immune response lead to persistent infection inside host cells.

  • 본 논문의 목적은 우리들의 현실적 노면표지 형태가 그 강도, 종류 및 출현빈도 측면에서 드러내는 지속적 증대 내지 강화 경향성을 문제점으로 제기하면서, 이런 노면표지 형태와 운전행동 간의 관련성에 대한 심리학적 제반 이론들을 고찰해보고 아울러 여기서 상호 연관적 특성을 밝혀주는 하나의 적절한 모델을 개발해보고자 하는 데에 두었다 이 모델에서는 운전자의 사고예방이나 안전운행이 곡 필요한 수준의 노면표지 자극화(stimulation)를 통해 조성된 쾌적한 교통환경에서 보다 용이하게 가능하다는 점이 전제되고 있다. 우리가 인간행동을 총체적으로나 효과적으로 분석하는 데 있어서는 일반심리적이거나 행동주의적 제반 이론 관점이 다소 미흡하다는 점이 지적되었다. 반면에 보다 거시적이고 종합적인 통찰에 중점을 두고있는 형태주의 심리학이나 장 이론적 시각은 행동분석 시에 비교적 결실적인 것으로 밝혀졌다 노면표지 자극의 양상여하가 운전자에게 스트레스, 정보과부담, 과도한 심리적 각성상태 등으로 인해 부정적인 영향을 미친다는 점에서는 이런 문제점이 환경심리학적 고찰측면에서도 관심의 대상이 되고 있다. \"노면표지 조성화 양 관점 모델\"에서 시사되고 는 터이지만. 이 모델에서의 노면표지 형태가 이론적인 면에서 \"정서-인지적\" 인간관에 근거를 두고있어 궁극적으로 이것이 옳다면, 그것은 곡 필요한 최소한의 수준에 머물러야 한다는 것이다. 올바른 운전행동, 다시 말해 교통법규 준수 행동은 노면표지 양상 측면의 각종 자극적 강화대책보다도 오히려 실효성 있는 교통교육, 확실한 적발 단속과 엄중한 처벌대책에 상대적으로 훨씬 더 큰 비중을 두고있는, 바로 그런 운전자의 의식개혁을 통해 비로소 제대로 정착될 수 있기 때문이다. 이 모델의 타당성 여부는 후속적 실증연구를 통해 해명되어질 수 있을 것이다.

Effects of Simulated Acid Rain on Growth and Antioxidant System in French Marigold (Tagetes patula L.) (인공산성비가 만수국(Tagetes patula L.)의 생육 및 항산화 작용에 미치는 영향)

  • This study was conducted to investigate the effect of simulated acid rain (SAR) on growth and antioxidant system in french marigold (Tagetes patula L.). Plants were subjected to four levels of SAR (pH 5.6, 4.0, 3.0, 2.0) in the growth chambers for 2 weeks. SAR decreased both plant height and plant dry weight of french marigold. As the pH levels decreased from 5.6 to 2.0, the content of MDA highly increased linearly. The ratios of dehydroascorbate/ascorbate and oxidized glutathione/reduced glutathione were significantly increased with decreasing pH levels. The enzyme (superoxide dismutase, ascorbate peroxidase etc.) activities of the plant affected by SAR were increased as the pH decreased. Based on the results, SAR caused oxidative stress in french marigold and resulted in significant reduction in plant growth. Biochemical protection responses might be activated to prevent the plant from damaging effects of oxidative stress generated in SAR.

A Study on Stress Responses of Korean-American (한국인의 스트레스 반응양상 -미국이민 한국인을 대상으로-)

  • Immigration of Koreans to the United States has increased since the 1960's. Adjusting to life in the United States produces a great deal of stress for immigrants. Despite better economic opportunites, many see the U.S. culture as threatening to their family and cultural values. Differences in culture, language, expectations and social behavior can lead to misunderstandings between health care providers and clients. These misunderstanding can leaf to frustration on the part of each. The ultimate result of this is that often Korean-immigrants do not get their health care needs met and stress response symptoms can lead to disease if there is no appropriate care. To determine the health care needs and concerns of Korean-Americans, a health needs assessment is needed. Appropriate and adequate information about the health care needs of these individuals is important as it relates to American policy changes allowing greater numbers of immigrants to enter the U.S. The purpose of this prospective study was to describe Korean-American stress response. This study focused on the primary presenting problems for which subjects reported having sought care. These included a variety of stress-related symptoms, including peripheral manifestations, cardiopulmonary symptoms, central-neurological symptoms, gastrointestinal symptoms, muscle tension, habitual patterns, depression, anxiety, emotional irritability and cognitive disorganization. Of the 300 subjects who entered the study, 80% (N=223) completed the questionnaire in full. Demographically, the percentage of females and males was 50% each and they ranged in age from 20 to 69 years. Ninety percent of the subjects were highly educated, 25% owners of business, 25% white collar professionals, 15% employed in sales or as skilled /unskilled labor, 30% had no occupation : and 5% were housewives or students. The SOS inventory is designed to quantify self-perception of behavioral, cognitive, and physiological components of the stress response. It consisted of 94 items divided into 10 subscales. The result of this study are as follows : The total mean 505 of all subjects (N=223 was .8129 ; the mean 505 for male(N=114) was .7665 and for females, (N=108) .8594. The level of symptoms for central-neurologic and muscle tension was higher for than for males. The highest stress response of all subjects was emotional irritability symptoms(1.0644) : the lowest stress response of all subjects was peripheral manifestation symptoms.

  • The author conducted a literature review to better characterize the current state of knowledge regarding the relationship between psychological and behavioral factors and cardiovascular disease. This review focus on several problems : Hypertension, coronary artery disease such as myocardial infarction and angina pectoris. We describe model for understanding the relation-ship of psychological factors to the disease, review the results of relevant research studies and provide recommendation for further research.

  • Chronic obstructive pulmonary disease (COPD) is a chronic progressive disease, characterized by irreversible airflow limitation, with a partially reversible component. The pathological abnormalities of COPD are associated with lung inflammation, imbalances of proteinase and antiproteinase, and oxidative stress, which are induced by noxious particles and gases in susceptible individuals. The physiological changes of COPD are mucus hypersecretion, ciliary dysfunction, airflow limitation, pulmonary hyperinflation, gas exchange abnormalities, pulmonary hypertension, cor pulmonale and systemic effects. The airflow limitation principally results from an increase in the resistance of the small conducting airways and a decrease in pulmonary elastic recoil due to emphysematous lung destruction. This article provides a general overview of the pathophysiology of COPD.

Functional characterization of a CCCH type zinc-finger protein gene OsZF2 by ectopic overexpression of the gene in rice (과발현 형질전환벼에서 CCCH type zinc-finger protein 유전자 OsZF2 기능 분석)

  • We have previously isolated a CCCH type zinc-finger protein gene, OsZF2 (Oryza sativa Zinc Finger 2), from the cold-treated rice cDNA library. To investigate the potential role of OsZF2, transgenic rice lines over-expressing OsZF2 under the control of CaMV 35S promoter have been developed through Agrobacterium-mediated transformation. Elevated level of OsZF2 transcripts was confirmed by RNA gel blot analysis in transgenic rice. Under the 100 mM NaCl condition, the transgenic rice showed significantly enhanced growth rate in terms of shoot length and fresh weight, implicating that OsZF2 is likely to be involved in salt response of rice. In the field condition, however, the transgenic rice showed a dwarf phenotype and flowering time was delayed. Genome expression profiling analysis of transgenic plants using the 20K NSF rice oligonucleotide array revealed many up-regulated genes related to stress responses and signaling pathways such as chaperone protein dnaJ 72, salt stress-induced protein, PR protein, disease resistance proteins RPM1 and Cf2/Cf5 disease resistance protein, carbohydrate/ sugar transporter, OsWAK kinase, brassinosteroid LRR receptor kinase, and jasmonate O-methyltransferase. These data suggest that the CCCH type zinc-finger protein OsZF2 is a upstream transcriptional factor regulating growth and stress responsiveness of rice.

Responses of Bacteria to TNT: Cells′Survival, SDS-PAGE and 2-D Electrophoretic Analyses of Stress-Induced Proteins (TNT에 대한 세균의 반응기작: 생존율, 스트레스 유도단백질의 SDS-PAGE 및 2-D 전기영동 분석)

  • The cellular responses of soil-borne bacterium, Pseudomonas sp. HK-6 to explosive 2,4,6-trinitrotoluene (TNT) were examined. Two stress shock proteins (SSPs), approximately 70-kDa DnaK and a 60-kDa GroEL were found in HK-6 cells in response to TNT. Analyses of SDS-PAGE and Western blot using anti-DnaK and GroEL revealed that SSPs were induced in HK-6 cells exposed to 0.5 M of TNT far 6-12 hrs. The maximum induction of proteins was achieved at 8-hr incubation point after HK-6 cells'exposure to TNT. Similar SSPs were found to be induced in HK-6 cells by heat shock (shift of temperature, from $30^{\circ}C$ to $42^{\circ}C$) or cold shock (shift of temperature,$30^{\circ}C$ to $4^{\circ}C$).2D-PAGE of soluble protein tractions from the culture of Pseudomonas sp. HX-6 exposed to TNT demonstrated that approximately 450 spots were observed on the silver stained gels ranging from pH 3 to pH 10. Among them, 12 spots significantly induced and expressed in response to TNT were selected and analyzed. Approximately 60-kDa protein, which was assumed highly expressed on the gel, was used for amino acid sequencing. N-terminal microsequencing with in-gel digestion showed that N-terminal sequence of the TNT-induced protein, <$^1XXAKDVKFGDSARKKML^17$, shared extensive similarity with $^1XXAKDVKFGDSARKKML^17$, N-terminal sequence of (P48216) GroEL of Pseudomonas putida.

Transcriptional Analysis Responding to Propanol Stress in Escherichia coli (대장균에서 프로판올 스트레스에 관한 전사분석)

  • We compared the transcriptome in response to propanol stress in wild-type and propanol-resistant mutant Escherichia coli using the DNA microarray technique. The correlation value of RNA expression between the propanol-treated wild type and the untreated-one was about 0.949, and 50 genes were differentially expressed by more than twofold in both samples. The correlation value of RNA expression between the propanol-treated mutant and the untreated one was about 0.951, and 71 genes in two samples showed differential expression patterns. However, the values between the wild type and mutant, regardless of propanol addition, were 0.974-0.992 and only 1-2 genes were differentially expressed in the two strains. The representative characteristics among differentially expressed genes in W3110 or P19 treated with propanol compared to untreated samples were up-regulation of hest shock response genes and down-regulation of genes relating to ribosome biosynthesis. In addition, many genes were regulated by transcription regulation factors such as ArcA, CRP, FNR, H-NS, GatR, or PurR and overexpressed by sigma factor RpoH. We confirmed that RpoH mediated an important host defense function in propanol stress in E. coli W3110 and P19 by comparison of cell growth rate among the wild type, rpoH disruptant mutant, and rpoH-complemented strain.
