Studies on the Content of Lectin in Korean Mistletoe according to the Host Tree Species and Characterization for Its Application to the Quality Control (한국산 겨우살이 숙주별 렉틴 함량과 지표물질로서의 특성 조사)

  • Traditionally, mistletoe is known as an effective anti-cancer medicinal plant, and lectin is recognized as a major component with cytotoxic and immuno-stimulant activity in mistletoe. A Korean mistletoe lectin (KML) has specificity to galactose and galactosamine and is distinguish from European mistletoe lectin (EML). When we examined the concentration of lectin in mistletoe originated from five different types of host trees, the result indicate that the lectin concentration is variable depending on the host tree. Noticeably, mistletoe from chestnut tree contains ten folds higher lectins than that of an oak tree. We also tested the concentration of KML and crude extract (KM-110) of Korean mistletoe that shows 90% cytotoxicity in L5178Y-ML25 lymphoma cell. In addition, the cells show 90% and 70% viability by the treatment of two neutralizing antibodies of KML, 9H7-D10 and 8B11-2C5 neutralization effect with two monoclonal antibodies of KML, 9H7-D10 and 8B11-2C5. Therefore, the result expected that the mistletoe contain some other cytotoxic components except lectin. Finally, the production of $TNF-{\alpha}$ and IL-6 by RAW 264.7 cells stimulated with lectin free-crude extract (LFKM-110) following neutralization by 9H7-D10 monoclonal antibody shows higher than that of lectin containing-crude extract (KM-110). These results suggest that the Korean mistletoe lectin ha a great potential to be developed as therapeutic agent of cancer.

Production of hGM-CSF from Cell Suspension Culture of Transformed Lettuce Using Agrobacterium-mediated Transformation System (Agrobacterium을 이용한 형질전환 상추의 세포 현탁배양으로부터 hGM-CSF의 생산)

  • Lettuce (Lactuca sativa) was transformed with Agrobacterium tumefacience LBA4404 containing human granulocyte macrophage colony stimulating factor (hGM-CSF) gene to produce in cell suspension cultures. Cell suspension culture was established using callus from transgenic lettuce plant. Integration of hGM-CSF gene into plant chromosome was confirmed through genomic PCR and Southern blot analysis. In addition, Northern blot analysis indicated the expression of the introduced hGM-CSF gene in transformed lettuce. The recombinant hGM-CSF was expressed in transgenic cell cultures derived from transgenic plants as a yield of about 149.0 $\mu\textrm{g}$/L in culture filtrate, which was determined by ELISA. These results demonstrated that transformed lettuce cell suspension cultures could be used as a production system of therapeutic proteins such as hGM-CSF.

Isolation and Characterization of Plant Pathogen that Cause Soft Rot Disease in Napa Cabbage (배추무름병 원인균 분리 및 특성 연구)

  • In order to establish in vitro infection model for research of plant pathogen based on tissue softening disease in napa cabbage, eighty independent bacterial strains were isolated from the softened napa cabbage tissues. Eight bacterial isolates were primarily screened with the generation of reproducible tissue softening disease to fresh napa cabbages within 24${\sim}$48 hours after inoculation. Through various microbiological biochemical and morphological examinations, three Gram (-) isolates which harbor independent biological properties were finally chosen, and named as RBI, RB2 and RB6. Collective results obtained from API 20E test and analyses of VITEK 2 COMPACT and nucleotide sequences of 165 rRNA of each isolate proposed that isolates RBI and RB2 are close to the Erwinia carotovora subsp. odorifera, and RB6 is close to the Erwinia carotovora subsp. carotovora. These isolates grew optimally at $30^{\circ}C$ with neutral pH culture condition. The isolates caused softening tissue disease with dose-dependent manner regardless of pre-surface damages of napa cabbage. Minimum dose to cause soft rot disease for RBI, RB2 or RB6 were $8.0{\times}10^8$ CFU/mt $10^9$ CFU/ml or $4.7{\times}10^6$ CFU/ml respectively. These isolates caused tissue softening disease to eggplant, paprika and napa cabbage out of 14 different tested vegetables, indicating that these isolates damages specific plant tissues. The bacterial isolates obtained in this research and in vitro plant infection model will be adapted in the understanding of the mechanism of pathogenesis by plant pathogen.

Studies on Host Plants, Development, and Distribution within Plants of the Greenhouse Whitefly, Trialeurodes vaporariorum(Westwood) (온실(溫室)가루이의 숙주식물(宿主植物), 발육(發育) 및 주내분포(株內分布)에 관(關)한 연구(硏究))

  • This study was conducted to investigate the host plants in greenhouse, developmental periods at different temperatures and distribution patterns on cucumber of the greenhouse whitefly, Trialeurodes vaporariorum(TV) in $1984{\sim}1985$. Host plants of 39 species belonging 27 families only in the greenhouse were recorded in 1984. Longevity of adult was 26.8 days and number of eggs per female was 305 eggs at $25^{\circ}C$. The eggs, larval and pupal periods were 8.2, 8.3, 7.5 days respectively at $25^{\circ}C$. The threshold temperature of development from egg to larva was $8.8^{\circ}C$. Survival rates from eggs to adults at $22^{\circ}C,\;25^{\circ}C,\;30^{\circ}C$ were 70.3%, 58.0% and 66.4%, respectively. Adults and eggs were found only on the apical leaves of cucumber plants within 40 days after transplanting. On the other hand, larva distributed on the middle leaves and pupa on the lower leaves.

Characterization of Anionic Peroxidase Induced by Low Host-Specific Elicitor in Suspension Cultures of Rose (Rosa sp.) (장미(Rosa sp.) 현탁배양세포에서 숙주 특이성이 낮은 Elicitor에 의해 유도되는 Anionic Peroxidase의 특성)

  • Whereas cationic extracellular peroxidases (PODs) were observed in the suspension cultures of rose (Rosa sp. L. cv Pual's scarlet) grown under normal conditions, new anionic isozymes were induced within 24 hr by the treatment of low host-specific elicitor (10 mg glucan/L media) prepared from yeast cell wall. Prominent anionic (pI 6.1) and cationic POD (pI 8.4) were purified and characterized to understand the physiological role of the enzymes. Both enzymes were purified (ca.200 fold) by the ammonium sulfate precipitation, ion exchange chromate-graphy and gel filtration chromatography. The Km values of the purified anionic POD for ferulic acid and $\textrm{H}_2\textrm{O}_2$ were 4.64 mM and 0.72 mM, whereas those of the cationic POD were 1.38 mM and 0.48 mM, respetively. The activity of the anionic POD as NADH oxidase was twice higher than that of cationic POD. The NADH oxidation in the anionic POD fraction was inhibited by 60% on the addition of 0.1 mM coniferyl alcohol, while that in the cationic fraction was inhibited by 15%.

Occurrence Ecology of Ricania sp. (Hemiptera: Ricaniidae) and Selection of Environmental Friendly Agricultural Materials for Control (갈색날개매미충(신칭, Ricania sp.)의 발생생태와 친환경 방제자재 선발)

  • An outbreak of Ricania sp. occurred in the Kurye, Jeonnam area in 2011. This outbreak damaged many kinds of fruit trees such as Cornus, Persimmon and Chestnut. This experiment was conducted to survey the occurrence ecology of Ricania sp. such as host plants, oviposition characters, morphological characters and life cycle, as well as to select environmental friendly control agents. Ricaina sp. host plants included 51 species such as 32 xylophytes, and 19 herbaceous plants. Ricaina sp. preferred Cornus officinalis, Diospyros kaki, Castanea crenata, Eucommia ulmoides, Styrax japonicus for oviposition. Adults laid eggs on new inner twigs with 28.8 eggs per egg-mass. Egg size was 1.24 mm(length), 0.55 mm(width) in an oval shape. Nymphs molted four times. Every nymph stage had an x shape of yellow or white beeswax around the anus. Overwintered eggs of Ricania sp. hatched from the mid May to early June. Nymphal periods were from mid May to mid August and adults appeared from mid July but spawning began in mid August. Ricania sp. damaged new twigs by oviposition and retarded growth by sucking nutrients and producing a sooty mold. Sophora and natural plant extracts were effective environmentally friendly agricultural materials used to control the nymph and adult Ricania sp. Mortality was > 80%.

Influence of CGMMV Infection Times on Growth and Quality of Watermelon and Cucumber (CGMMV 감염시기가 수박과 오이의 생육 및 품질에 미치는 영향)

  • We investigated the effect of infection time of CGMMV on the growth and quality of watermelon and cucumber plants. The effect (damages by CGMMV) was estimated on the watermelon where CGMMV had been inoculated at different growth stages, vegetative (transplanting stage, vegetative growth stage) and reproductive growth stage (fruiting stage and fruit hypertrophy stage). In the case of cucumber, CGMMV was inoculated at transplanting stage and Erst flowering stage, respectively. When watermelon was infected with CGMMV at vegetative growth stage, vine length, internode length, leaf area, and fruit weight of the plants largely decreased compared with control plants, while the infected plant growth was not very different from control plants when it was infected at reproductive growth stage. Brix of the fruit of watermelon also decreased when the plants was infected with the virus earlier than fruiting stage. The occurrence of 'Pisubag', internal discoloration and decomposition of watermelon fruits, tended to be increased as earlier infection time with CGMMV In the case of cucumber infection time with CGMMV did not influence earlier growth of the plants, but did later growth showing that plant height, vine length, internode length, number of leaf, leaf wide, and leaf length of the plants decreased as infection time became to be earlier.

Detection of CGMMV from Commercial Cucumber Seed and Resistance Test of Cultivars (오이 종자로부터 CGMMV의 검출과 품종 저항성 검정)

  • To analyze the relationship between CGMMV occurrence and seed contamination by cucumber cultivars, cucumber cultivars of CGMMV infected field and diseased degree were investigated in cucumber main cultivation regions of Jeonnam province from 1999 to 2002. While the diseased degree was different by years and cultivars, CGMMV occurred for 4 years in 'Janghyeongnakhap', for 3 years in 'Jangjukcheongjang', and for 2 years in 'Gyeoulsali' and 'Gyeoulnagi'. CGMMV was detected in seeds of 'Janghyeongnakhap' and 'Jangjukcheongjang' by ELISA test, and seed of 'Jangjukcheongjang' showed positive reaction in bioassay. As a result of resistant test of 34 cultivars to CGMMV, all cultivars showed mosaic symptoms in pot test but only 'Hanboksamcheok' showed mild mosaic symptom in field test.

An Improved Method for Testing Pathogenicity of Pseudomonas syringae pv, actinidiae Causing Bacterial Canker of Kiwifruit (참다래 궤양병의 간편한 병원성 검정법 개발)

  • This research was conducted to develop a simple and effective method for pathogenicity assay of the causal agent of bacterial canker on kiwifruit. The developed method is a modified version of syringe-infiltration method that is used in the assay fer the hypersensitive response assay. Bacterial cell suspensions in 50 mM potassium phosphate buffer(pH 7.5) were infiltrated using a plastic syringe with 25G needle into primary leaves of five-year-old kiwifruit. Typical symptoms of bacterial canker were observed five days after infiltration. Symptoms developed on the leaves were detected in these inocula that treated above 10$^4$cfu/ml or above. Using this technique, host range of Pseudomonas syringae pv. actinidiae and three other plant pathogenic pseudomonads were investigated for 25 different plant species. The various symptoms were showed depend-ing on different plant species and inoculated pathogen combinations. This method has the advantage that symptoms can be showed faster compared to other methods and high humid conditions are not required.

Influence of Weather Condition at Heading Period on the Development of Rice Bacterial Grain Rot Caused by Burkholderia glumae (출수기 기상환경이 세균성 벼알마름병 발생에 미치는 영향)

  • The relationships between the weather conditions and the occurrence of rice bacterial grain rot caused by Burkholderia glumae during the rice heading period, were examined by analying the data accumulated from 1992 through 2000 and by conducting greenhouse and field experiment to develop a model far forecasting the disease. The disease severely occurred in 1994, 1995, 1998, and 2000, when it was high in temperature and rainfall during the heading period of middle-late rice varieties. While it occurred weakly in 1993 was high in rainfall and low temperature and it was reversely in 1997. When treated under wetting condition (above the 24-hour)after inoculation at heading period, the infection rate increased as the inculum concentration increased, it was 86.1% at 10$^{8}$ cfu/ml. When under drying condition, the disease of 12.5% occurred only at $^{8}$ cfu/ml. On the other hand, 1,000 grain weight and the percentage of ripened grains remarkably decreased as the infection rate increased.
