Effect of Repeated Induction of Superovulation on Ovulation Rates and In Vitro Development of Embryos in Rabbit (토끼에서 반복적인 과배란유도가 배란율과 난자의 체외 발육율에 미치는 영향)

  • 토끼에서는 수정란 이식과 같은 기본적인 번식공학적 방법의 효율성이 아직 생쥐와 같은 실험동물에 비해 떨어지고 있어 생물공학적인 기술을 응용하는데 큰 어려움이 있다. 특히 유전자 이식에 의한 형질전환 토끼의 생산과 같은 생물공학적인 기술을 실용화하는데 효율이 높은 수정란이식 기술의 개발이 필수적이라고 할 수 있다. 본 연구에서는 토끼에서 수정란 이식 기술의 첫 단계인 과배란 유도를 효율적으로 이용할 수 있는 방법을 정립하기 위해 반복적인 과배란 유도가 배란율 및 수정란의 질적인 면과 양적인 면에 미치는 영향을 조사하였다. 연구방법으로는 FSH와 HCG를 사용하여 과배란을 유도하였고 2.5 개월의 반복처리간격으로 3번의 반복적인 과배란 처리를 한 후 반복처리에 따른 배란율과 배란된 난자의 형태학적 상태, 배양에 의한 발생 능력 상태 등을 감소하였으며 배란수의 변이도 커지는 경향을 나타내었다(첫번째, 32.6(+-)2.5; 두번째 28.7(+-)3.7; 세번째 20.99(+-)3.8). 제 2극체의 돌출, 전핵의 형성, cummulus cell의 존재등에 의한 회수된 난자의 형태학적 관찰에 의한 방법으로 난자를 분류한 결과 과배란의 반복수가 증가함에 따라 다양한 모양의 난자가 회수되어 배란이 광범위한 시간대에 일어나고 있음을 나타내었다. 또한 과배란의 반복적인 유도에 의해 난소의 혈포수는 증가하였으나 채란된 난자의 채외배양에 의한 발육율에는 차이가 없었다. 그러므로 과배란의 반복적인 유도는 공란토의 난소반응에는 영향을 미쳤으나 난자의 질에는 영향을 미치지 않았음을 나타낸다.

In Vivo Embryo Production and Embryo Transfer in Hanwoo and Jeju Black Cattle Using CIDR (CIDR를 이용한 제주 한우 및 흑우의 체내 수정란 생산과 이식)

  • The objectives of this study was to produce valuable offsprings of Hanwoo and Jeju black cattle using in vivo embryo production and embryo transfer techniques during 5 years ($2001{\sim}2005$) in Jeju. Two hundred and eighty six Hanwoo and sixty nine Jeju black cattles, at random stages of the estrous cycle, received a CIDR. Seven days later, the animals were superovulated with a total of 400 mg pFSH ($Folltropin^{(R)}-V$) administered twice daily in constant doses (each 50 mg) over 4 days. On the 6th administration of FSH, CIDR was withdrawn and 25 mg $PGF_{2{\alpha}}$ was administered. Cows were artificially inseminated thrice after estrous detection at 12 hr intervals. The cows received $250{\mu}g$ GnRH at the time of 2nd insemination. Embryos were recovered 7 or 8 days after the 1st insemination. Recipients were synchronized with donors by insertion of a CIDR for 7 days and administration of 25 mg $PGF_{2{\alpha}}$ at the time of CIDR removal. The collected embryos were transferred to 1,219 recipients by 6 transfer persons. The mean numbers of total ova and transferable embryos from Hanwoo and Jeju black cattle donors were 7.4 and 4.7, respectively The number of transferable embryos differed between Hanwoo (5.0) and Jeju black cattle (3.5, p<0.05), while that of total ova did not differ. Repeated superovulation treatments decreased (p<0.05) the ratio of numbers of the flushed animals vs. superovulated animals in Jeju black cattle, and the numbers of total ova and transferable embryos in Hanwoo. More transferable embryos were collected at summer (5.6) than winter (2.9, p<0.01). The mean pregnancy rate was 40%. The pregnancy.ate was affected by transfer year (2001<2004, p<0.05) and transfer person ($33.0{\sim}41.9%$, p<0.01), while not by donor (embryo) breed. These results showed that in vivo embryo preduction and embryo transfer techniques using CIDR for Hanwoo and Jeju black cattle donors as well as recipient, regardless of their estrous cycle, may enable a stable embryo production and recipient preparation.

Production of Calves Following Transfer of Sexed Hanwoo Embryos and Hanwoo Embryos Cultured In Vitro (한우 체외 수정란 및 성 감별 수정란 이식에 의한 송아지 생산)

  • This study was carried out to examine the efficiency of biopsy methods, and the pregnancy rate, calving and abortion rates, gestation length and birth weight of Hanwoo calves following transfer of fresh, frozen and sexed Hanwoo embryos produced in vitro. The survivability of biopsied embryos was 80.0 and 90.0% using aspiration and punching methods at 24 h after culturing, respectively. The ratios of male and female embryos were 42.1 and 52.6%, respectively, and the percentage of sex unidentified was 5.3%. Pregnancy rates was not significantly different between hCG and control group (46.4 vs. 38.5%), fresh and frozen embryos (41.3 vs. 35.0%), and sexed and IVP embryos (27.5 vs. 41.2%) (p>0.05). Calving and abortion rates of IVP and sexed embryos were not significantly different in calving (85.0 vs. 87.0%) and in abortion (15.2 vs. 13.3%) (p<0.05). Gestation length of IVP and sexed calves were 281.3 and 288.2 days in female and 283.0 and 282.3 days in male, and the birth weight of IVP and sexed calves were 23.6 and 25.0 kg in female and 24.6 and 23.8 kg in male, respectively. There were no difference in gestation length and birth weight between IVP embryos and sexed embryos (p>0.05). Administration of hCG to recipients did not improve the pregnancy rate following transfer of Hanwoo embryos produced in vitro and sexed embryos. Although the production of calves derived from sexed Hanweoo embryos cultured in vitro can be obtained, the efficiency of sexed calves production need to be improved in biopsy methods and pregnancy rate. Further study should be focused on the improvement of pregnancy rates for commercial application of embryo transfer.

Production of Korean Native Cow from Mongolian Cow following Transfer of Vitrified Blastocyst (Mongolian 수란우에 한우 동결수정란의 이식 후 산자 생산)

  • The purpose of this study was to investigate the comparison of viability of bovine blastocysts following glass micropipette (GMP) vitrification and the possibility of production of Korean Native Cow ("Hanwoo,"Bos taurus coreanae) following embryo transfer into Mongolia cows (Bos taurus mongolian). The embryos of Korean Native Cow were produced by IVMFC or superovulation methods in Korea, cryopreserved by GMP vitrification, and subsequently trans-ported to Mongolia. The recipient cows were synchronized using a CIDR plus and prostaglandin $F_2\alpha$($PGF_2\alpha$) treatment. To produce in vivo embryos, seven cows were superovulated using FSH and PGF$_2$/sub $\alpha$/ treatment. A total of 64 blastocysts ( $9.1\pm2.94$ per cow) were collected. In vitro embryos were produced using a defined culture system which cleaved in 80.1% ova (174/217), and developed to blastocyst stage embryos of 40.8% (71/174). The post-thaw survival rate of in vivo blastocysts (93.7%; 45/48) was significantly higher than that of in vitro blastocysts (82.5%; 52/63, P<0.05). Embryo transfer was carried out using 8 Mongolian recipient cows and 2 post-thaw blastocysts per recipient. Five of 8 recipients were found pregnant at Day 60 but one abortion occurred by Day 240. Two of offspring were produced from the Mongolian cows at 275 days after embryo transfer. These results indicated that a GMP vitrification method could be used as a cryopreservation technique for in vivo or in vitro bovine blastocysts and produced effectively a Korean Native Cow following embryo transfer into a Mongolian recipient cow.
