  • 최근, 물이 인류의 생존을 위협하는 문제로 인식되며 중요시되고 있으며, 이는 식수에 대한 국민의 생존권 및 건강권 뿐 아니라 청정하지 못한 수질에서 어획 또는 양식되는 수산자원의 안전성과도 결부되어 중요하게 인식되고 있다. 이는 중금속 뿐 아니라 세균이나 바이러스 등이 먹이연쇄를 통하여 건강을 위협할 수 있기 때문이다. 이로부터 다양한 수처리 기법이 개발되고 있으며, 그 중 하나인 화학적 산화법은 수중 오염물을 화학적으로 산화시켜 생분해성이 좋아지게 하거나 흡착법으로 쉽게 제거될 수 있는 최종 또는 중간 생성물로 변화시키는 방법으로서 염소, 오존, 과산화수소, 과망간칼륨 등이 통용되고 있다. Sporicidal Efficacy of a Fumigation Disinfectant Composited to Ortho-phenylphenol Against Spores of Clostridium Perfringens (Ortho-phenylphenol을 주성분을 하는 훈증소독제의 Clostridium perfringens 아포에 대한 살아포 효과)

  • This study was performed to evaluate the sporicidal efficacy of a fumigation disinfectant containing 20% ortho-phenylphenol against Clostridium perfringens (C. perfringens) spores. In this research, efficacy test of fumigant against C. perfringens spores was carried out according to French standard NF T 72-281. C. perfringens spores working culture suspension number (N value), all the spore numbers on the carriers exposed with the fumigant (n1, n2, and n3), the number of bacterial spore suspensions by pour plate method (N1), the number of bacterial spore suspensions by filter membrane method (N2) and the mean number of bacterial spore recovered on the control-carriers (T value) were obtained from the preliminary test. In addition, the reduction number of C. perfringens spores exposed with the fumigant (d value) was calculated using T value, the mean number of bacterial spore in recovery solution (n'1) and the mean number of bacterial spore on carriers plated in agar (n'2). N value was $4.3{\times}10^7spores/mL$, and n1, n2, and n3 were higher than 0.5N1, 0.5N2 and 0.5N1, respectively. Additionally, T value was $4.9{\times}10^5spores/carrier$. In the sporicidal effect of the fumigant, the d value was 4.52log reduction. According to the French standard for the fumigant, the d value for the effective sporicidal fumigant should be over than 3log reduction. The results indicated that Fumagari $OPP^{(R)}$ had an efficient sporicidal activity against spores of C. perfringens, then the fumigant can be applied to disinfect food materials and kitchen appliances contaminated with bacterial spores.

  • This study was performed to evaluate the bactericidal efficacy of Fumagari OPP$^{(R)}$, fumigation disinfectant, containing 20% ortho-phenylphenol against Staphylococcus aureus (S. aureus). N value was $4.0{\times}10^8$ CFU/mL, and n1, n2, and n3 were higher than 0.5N1, 0.5N2 and 0.5N1, respectively. Additionally, T value was $3.4{\times}10^6$ CFU/mL. In the bactericidal effect of the fumigant, the d value was 6.43 logCFU/mL. According to the French standard for the fumigant, the d value for the effective bactericidal fumigant should be over than 5 logCFU/mL. The results indicated that Fumagari OPP$^{(R)}$ had an effective bactericidal activity against S. aureus, then the fumigant can be applied to disinfect food materials and kitchen appliances contaminated with pathogenic bacteria.

Bactericidal Efficacy of a Fumigation Disinfectant Containing Paraformaldehyde Against Salmonella Typhimurium

In this study, the efficacy test of a fumigant against S. Typhimurium was carried out according to French standard NF T 72-281. The S. Typhimurium working culture suspension number (N value), all bacteria numbers on the carriers exposed to the fumigant (n1, n2, and n3), the number of bacterial suspensions by the pour plate method (N1), the number of bacterial suspensions by the filter membrane method (N2), and the mean number of bacteria recovered on the control carriers (T value), were obtained from the preliminary test. In addition, the reduction number of S. Typhimurium exposed to the fumigant (d value) was calculated using the T value, the mean number of bacteria in the recovery solution (n'1) and the mean number of bacteria on carriers plated in agar (n'2). The N value was $5.5{\times}10^8$ colony forming units (CFU)/mL, and n1, n2, and n3 were higher than 0.5N1, 0.5N2 and 0.5N1, respectively. Additionally, the T value was $3.5{\times}10^6CFU/carrier$. In terms of the bactericidal effect of the fumigant, the d value was 5.25. According to the French standard for fumigants, the d value for an effective bactericidal fumigant should be greater than 5. The results indicated that the fumigant containing 35% paraformaldehyde had an efficient bactericidal activity against S. Typhimurium, and, therefore, can be used to disinfect food materials and kitchen appliances contaminated with foodborne bacteria.

Inactivation of Avian Influenza Viruses by Alkaline Disinfectant Solution (알칼리성 소독액에 의한 조류인플루엔자바이러스 불활성화)

  • Avian influenza viruses cause a considerable threat to humans and animals. In this study, we investigated whether alkaline disinfectant solution can inactivate H5N1, H3N2, H6N1, and H9N2 subtypes of avian influenza virus. When H5N1, H3N2, H6N1, and H9N2 avian influenza viruses were treated with alkaline solution diluted with PBS (pH 7.2) prior to infection into MDCK cells, alkaline disinfectant solution (at dilutions up to $10^{-2}$) completely inactivated all avian influenza subtypes tested. To confirm the inactivation of avian influenza viruses by alkaline disinfectant solution, we used an immunofluorescence assay with influenza A anti-nucleoprotein antibody and FITC-labeled secondary antibody to stain MDCK cells infected with avian H9N2 influenza viruses. No staining was observed in MDCK rells infected with H9N2 viruses that were pre-treated with a $10^{-2}$ dilution of alkaline disinfectant solution, while strong staining was observed in MDCK cells infected with H9N2 viruses without pre-treatment. Our results indicate that alkaline solution could help to control avian influenza viruses including the highly pathogenic H5N1 subtype.

Microbial Activity Analysis for the Selectively Sterilizing of Government-controlled Bulk Public Archives (대량 공공 기록물의 선별적 소독을 위한 미생물 활성도 분석 연구)

  • An archive is a collection of documents or records. Currently, most archived documents are made of paper. Paper is susceptible to biological damage and deterioration due to its material properties. To control the biological damage, treatment with chemical disinfectants and control of the storage environment are often used. In government-controlled bulk public archives, all documents are chemically sterilized before storage. However, an extremely large quantity of public records have been produced, and storage space and conservation management are gradually reaching their limits. In this study, 60 species of microbes were identified using a genetic method. We successfully applied the adenosine triphosphate (ATP) bioluminescence method to detect microbial contamination on paper documents. A calibration curve of the ATP bioluminescence as a function of the microbe quantity was obtained, and the microbial activity on non-sterilized paper archives from 1951 was analyzed using an ATP luminometer. It was found that the microbial activity was suppressed or reduced in climate-controlled storage environments at $22^{\circ}C$ and 55% relative humidity. We anticipate that these results will be used to establish selective sterilization systems for government-controlled bulk public archives.

Analysis of Trace Levels of Lodinated Trihalomethanes in Water Using Headspace - GC/ECD (Headspace - GC/ECD를 이용한 수중의 미량 요오드계 트리할로메탄류 분석)

  • Trihalomethanes (THMs) are formed as a results of the reaction of residual chlorine, used as a disinfectant in drinking water, with the organic matter in raw water. Although chlorinated and brominated THMs are the most common disinfection byproducts (DBPs) reported, iodinated THMs (I-THMs) can be formed when iodide is present in raw water. I-THMs have been usually associated with several medicinal or pharmaceutical taste and odor problems and is a potential health concern since they have been reported to be more toxic than their brominated and chlorinated analogs. Currently, there is no published standard analytical method for I-THMs in water. An automated headspace-gas chromatography/electron capture detector (GC/ECD) technique was developed for routine analysis of 10 THMs including 6 I-THMs in water samples. The optimization of the method is discussed. The limits of detection (LOD) and limits of quantification (LOQ) range from 12 ng/L to 56 ng/L and from 38 ng/L to 178 ng/L for 10 THMs, respectively. Matrix effects in river water, sea water and wastewater treatment plant (WWTP) final effluent water were investigated and it was shown that the method is suitable for the analysis of trace levels of I-THMs, in a wide range of waters. The method developed in the present study has the advantage of being rapid, simple and sensitive.

Efficacy of Sanitizers Due to the Changes of Contact Time and Temperature (사용시간 및 온도조건 변화에 따른 살균소독제의 유효성)

  • The bactericidal efficacy of three common sanitizers (100 or 200 ppm of sodium hypochlorite, 100 or 200 ppm of n-alkyl($C_{12}-C_{18}$)benzyldimethyl ehloride, and 50 or 100 ppm of peroxyacetie acid) against Escherichia coli ATCC 10536 and Staphylococcus aureus ATCC 6538 was studied using the suspension test method at various exposure temperatures (4~$40^{\circ}C$) and times(1~60min) under both dirty and clean conditions, respectively. During the suspension tests, sodium hypochlorite (200 ppm) showed higher bactericidal activity than the other sanitizers under clean conditions, with 5 log reductions against E. coli as well as S. aureus in the first 1 min of treatments at $4^{\circ}C$, However, the efficacy of sodium hypochlorite decreased markedly under dirty conditions due to its susceptibility to interfering substances. The efficacy of the n-alkyl($C_{12}-C_{18}$)benzyldimethyl chloride increased considerable as the exposure temperature and time increased. The bactericidal efficacy of the n-alkyl($C_{12}-C_{18}$)benzyldimethyl chloride might be less effective on low temperature, however, the longer time the sanitizer is in contact, the more effective the sanitization effect. Treatment with peroxyacetic acid (100 ppm) showed at least 5 log reduction against E. coli and S. aureus for 5 min at $4^{\circ}C$ under both clean and dirty conditions. The efficacy of the peroxyacetic acid was not much altered by interfering substances and aflected by changes in temperature or time.

  • This study was conducted to investigate the effect of barley tea with roasted grains and barley tea with a tea bag on the reduction of chlorination by-product(CBP) levels in chlorinated drinking water. Since the concentrations of six volatile compounds of eight CBPs were blow their respective detection limits after 10 minute heating, two nonvolatile CBPs dichloroacetic acid and trichloroacetic acid, and total chlorine were compared between tap water and two kinds of barley tea. No significant differences were observed in the relative changes of the amounts of the above three items, and new peaks which were not found in the original water appeared in the chromatograms of gas chromatograph/electron capture detector(GC/ECD). Thirty three organic compounds were identified in the extracts of barley tea with roasted grains, barley tea with a tea bag, corn tea, and Cassia tora seed tea which were prepared with distilled/deionized water, using gas chromatography/mass selective detection(GC/MSD). Exclusive of fatty acids, most of the compounds were aromatic compounds such as phenols, furans, and pyrroles.
