이 글에서는 나노스케일의 직경을 갖는 섬유를 빠른 생산속도로 제작할 수 있는 전기방사공정(electrospinning process)에 대한 개요와 조직공학용 지지체(tissue engineering scaffold)로의 응용을 위한 제조방법에 대해 소개하고자 한다. 세포의 증식, 분화 등의 생물학적 활동에 기반한 조직공학 및 조직재생 분야에서는 일시적 또는 영구적으로 세포가 부착하여 생장할 수 있는 지지체(scaffold)의 활용이 필수적이다. 세포가 이상적으로 성장할 수 있는 지지체를 제작하기 위해서는 세포의 부착 특성, 화학적/물리적/구조적 성장 환경 등이 고려되어야 한다. 따라서 이상적인 세포 성장 환경을 구현하기 위해 실제 세포 주변의 미세환경(microenvironmenr)조건을 모사하는 연구가 많이 이루어지고 있다. 세포외기질(extracellular matrix)이라고 하는 나노크기의 직경을 갖는 섬유기반의 세포 주변 환경을 모사하는 방법의 하나로 전기방사 공정이 '90년대에 들어 활용되기 시작하였다. 현재까지도 전기방사를 이용하여 제작되는 나노섬유는 공정조건 및 재료를 다양하게 응용하여 조직의 물리 화학적 특성을 잘 반영할 수 있는 장점이 있어 조직공학용 지지체로서 광범위하게 활용되고 있다.
This study was designed in the fabricated poly (L-Lactic-co-${\varepsilon}$-Caprolactone) (PLCL) scaffold using chitosan-alginate hydrogel, which would be more suitable to maintain the biological and physiological functions continuing three dimensional spatial organizations for chondrocytes. As a scaffold, hydrogels alone is weak at endure complex loading within the body. In this study, we made cell hybrid scaffold constructs with poly (L-Lactic-co-${\varepsilon}$-Caprolactone) (PLCL) scaffold and hydrogels to make a three-dimensional composition of cells and extracellular matrix, which would be a mimic of a native cartilage. Using a particle leaching technique with NaCl, we fabricated a highly-elastic scaffold from PLCL with 85% porosity and $300-500{\mu}m$ pore size. A mixture of bovine chondrocytes and chitosan-alginate gel was seeded and compared with alginate as a control on the PLCL scaffold. The cell maturation, proliferation, extracellular matrix synthesis, glycosaminoglycans (sGAG) production and collagen type-II expressions were better in chondrocytes seeded in chitosan-alginate hydrogel than in alginate only. These results indicate that chondrocytes with chitosan-alginate gel on PLCL scaffolds provide an appropriate biomimetic environment for cell proliferation and matrix synthesis, which could successfully be used for cartilage repair and regeneration.

Recently, three-dimensional scaffolds and nanofibers are being developed for bone tissue regeneration. In this study, we fabricated a hybrid scaffold using a nano-micro precision deposition system. The fabrication process involved the application of the solid freeform fabrication (SFF) technology and electrospinning. The hybrid scaffolds were combined using micro scaffolds and nanofibers. The nanofibers were deposited on each layer of the micro scaffolding using the electrospinning process. The micro scaffolds were fabricated using the SFF technology at a temperature of $100^{\circ}C$, pressure of 650 kPa, and scan velocity of 250 mm/s. Nanofiber fabrication was conducted by means of electrospinning using the flow rate, solution concentration, distance from the tip to the collector (TCD), and voltage. The nanofibers were fabricated using a flow rate of 0.1 ml/min, voltage of 5 kV, TCD of 1 mm, and 10 wt% of solution concentration. MG-63 cells were seeded into the hybrid scaffold for the purpose of its evaluation.

In tissue engineering, a scaffold is a three-dimensional(3D) structure that serves as a template for regeneration the functions of damaged tissues or organs. Among materials for scaffolds, polycaprolactone(PCL) and ${\beta}$-tricalcium phosphate(${\beta}$-TCP) are biodegradable and biocompatible. In this study, we fabricated 3D PCL, blended PCL (60 wt %)/${\beta}$-TCP (40 wt %), and pure ${\beta}$-TCP scaffolds by a multi-head scaffold fabrication system. Scaffolds with a pore size of $600{\pm}20{\mu}m$ was observed by scanning electron microscopy. The effects of 3D PCL, blended PCL (60 wt %)/${\beta}$-TCP (40 wt %) and pure ${\beta}$-TCP scaffolds were analyzed by evaluating their mechanical characteristics. In addition, in an in-vitro study using osteoblast-like saos-2 cells, we confirmed the effects of 3D scaffolds on cellular behaviors such as cell adhesion and proliferation. In summary, the 3D blended PCL (60 wt %)/${\beta}$-TCP (40 wt %) scaffold was found to be suitable for human cancellous bone in terms of its the compressive strength, biocompatibility, and osteoconductivity. Thus, blending PCL and ${\beta}$-TCP could be a promising approach for fabricating 3D scaffolds for effective bone regeneration.
Scaffolds of tissue engineering should be biocompatible and biodegradable for cell attachment, proliferation and differentiation. In the various scaffold fabrication, 3D printing technique can make the three dimensional scaffold with interconnected pores for cell ingrowth. Polycaprolactone (PCL) is biodegradable polyester with a low melting temperature and has been approved by the Food and Drug Administration (FDA). In this study, PCL scaffold was fabricated by 3D bioprinting system and surface modification of PCL scaffold was controlled by NaOH treatment. Morphological change and wetability of NaOH-treated scaffold were observed by SEM and contact angle measurement system. The remnant of PCL treated with NaOH was measured by ATR-FTIR. In vitro study of scaffolds was evaluated with WST-1 and ALP activity assay. NaOH treatment of PCL scaffolds increased surface roughness, hydrophilicity, cell proliferation and osteogenic differentiation. These results indicate that NaOH-treated PCL scaffold made by 3D bioprinting has tissue engineered potential for the development of biocompatible material.

Poly(lactide-co-glycolic acid) (PLGA) has been widely used in the drug delivery and tissue engineering applications because of its good mechanical strength and biodegradation profile. However, cell attachment to the scaffold is low compared with that on fibrin although cells can be attached to the polymer surface. In this study, PLGA scaffolds were soaked in cells-fibrin suspension and polymerized with dropping fibrinogen-thrombin solution. Cellular proliferation activity was observed in PLGA/fibrin-seeded costal cartilage cells (CC) on 1, 3, and 7 days using the MTT assay and SEM. The effects of fibrin on the extracellular matrix (ECM) formation were evaluated using CC cell-seeded PLGA/fibrin scaffolds. The PLGA/fibrin scaffolds elicited more production of glycosaminoglycan (GAG) and collagen than the PLGA scaffold. In this study, fibrin incorporated PLGA scaffolds were prepared to evaluate the effects of fibrin on the cell attachment and proliferation in vitro and in vivo. In this result, we confirmed that proliferation of cells in PLGA/fibrin scaffolds were better than in PLGA scaffolds. The PLGA/fibrin scaffolds provide suitable environment for growth and proliferation of costal cartilage cells.

In vitro 상에서 골조직을 원활하게 재생하기 위해서는 3차원 지지체를 이용한 세포 배양과 세포 배양 시 세포의 형태와 기능을 유지/향상시키기 위한 인체 내 미세 환경 재현은 필수적이다. 따라서 본 연구에서는 뼈 성분과 유사한 생체 활성 물질인 hydroxyapatite (HA)와 생분해성 고분자인 poly $\varepsilon$-caprolactone (PCL)를 복합재료로 이용하여 내부 연결성이 우수한 골조직 재생용 3차원 지지체를 제작하였으며, 골 재생 능력 향상을 위하여 인체내 골조직의 기계적 미세 환경을 체외에서 구현한 새로운 형태의 perfusion bioreactor system을 개발/적용하였다. 또한 본 연구에서 개발된 perfusion bioreactor system의 생물학적 평가를 위해 MG63 (osteoblast like cell, 한국 세포주 은행)과 New Zealand White Rabbit에서 분리한 중간엽 줄기세포를 골조직 재생용 3차원 지지체에 파종하였다. 48시간 동안 안정화 후 perfusion bioreactor system을 이용하여 기계적 자극을 파종된 세포에 인가하였으며, 배양 기간 동안 세포의 증식 확인 및 형태학적 관찰을 실시하였다. 본 연구 결과, perfusion bioreactor system을 이용하여 기계적 자극을 인가한 실험군에서 세포의 증식 및 활성도가 대조군에 비해 우수함을 확인 할 수 있었다. 따라서, perfusion bioreactor를 이용한 세포 배양은 세포의 활성 향상 및 골조직 재생에 도움이 될 것으로 사료된다. 차후 perfusion bioreactor를 이용한 다양한 패턴의 자극이 골재생 능력 및 중간엽 줄기세포의 골 분화능에 미치는 영향에 대한 연구가 필요할 것으로 사료된다.
In this report, we present an inverse opal scaffold that can enhance the chondrogenic differentiation of human adipose-derived stem cells (hADSCs) without drug, gene, or cytokine supplement. Inverse opal scaffolds based on poly(D,L-lactide-co-glycolide) were formed with uniform $200{\mu}m$ pores. Due to uniform pore sizes and well-controlled interconnectivity of inverse opal scaffold, hADSCs were allowed to distribute homogeneously throughout the scaffolds. As a result, high cell density culture with scaffold was possible. Since the hADSCs cultured in inverse opal scaffolds were subjected to limited supplies of oxygen and nutrients, these cells were naturally preconditioned to a hypoxic environment that stimulated the up-regulation of hypoxia-inducible factor-$1{\alpha}$ (HIF-$1{\alpha}$). As a result, apoptotic activity of hADSCs until 3 weeks after initial cell seeding was significantly reduced and chondrogenic differentiation related molecular signal cascades were up regulated (transforming growth factor-beta, phosphorylated AKT, and phosphorylated p38 expression). In contrast, hADSCs cultured with small and non-uniform porous scaffolds showed significantly increased apoptotic activity with decreased chondrogenic differentiation. Taken together, inverse opal scaffold could potentially be used as an effective tool for improving chondrogenesis using stem cells.
Biomedical scaffold for tissue regeneration was fabricated by one of rapid prototyping processes, bioplotting system, with a biodegradable and biocompatible poly($\varepsilon$-carprolactone)(PCL). Through dynamic mechanical test, it was observed that the PCL scaffold manufactured by the bioplotting process has the superior mechanical properties compared to the conventional scaffold fabricated by a salt-leaching process, and the plotted scaffold could be employed as a potential scaffold to regenerating hard and soft tissue. The plotted scaffold was consisted of porous structures. which were interconnected with each pore to help cells be easily adhered and proliferated in the wall of pore tunnels, and metabolic nutrients can be transported within the matrix. By using the plotting system, we could adjust the pore size, porosity, strand pitch, and, strand diameter of PCL scaffolds, which were important parameters to control mechanical properties of the scaffolds, and consequently we could determine that the mechanically controlled scaffolds could be used as a matching scaffold for any required mechanical properties of the target organ. The fabricated 3D PCL scaffold showed enough possibility as a 3D biomedical scaffold, which was cell-cultured with chondrocytes.

Biodegradable polymers have been used extensively as scaffolding materials to regenerate new tissues and the ingrowth of tissue have been reported to be dependent directly of the porosity, pore diameter, pore shape, and porous structure of the scaffold. In this study, porous poly (L-lactide-co-glycolide) (PLGA) scaffolds with five different pore sizes were fabricated to investigate the effect of pore sizes for AF tissue regeneration. Cellular viability and proliferation were assayed by MTT test. Hydroxyproline/DNA content of AF cells on each scaffold was measured. sGAG analyses were performed at each time point of 2 and 6 weeks. Scaffold seeded AF cells were implanted into the back of athymic nude mouse to observe the difference of formation of disc-like tissue depending on pore size in vivo. We confirmed that scaffold with $180{\sim}250{\mu}m$ pores displayed high cell viability in vitro and produced higher ECM than scaffold with other pore sizes in vivo.
