Characteristics of Autographa californica Nuclear Polyhedrosis Virus in Spodoptera exigua Cell Line. (파밤나방 세포주에서 Autographa californica 핵다각체병 바이러스의 감염 특성)

  • To study the usefulness of Se301 cells, which is originated from Spodoptera exigua and has susceptibility to the Autographa californica NPV (AcNPV), as a host for the AcNPV-based expression vector system, we compared the characteristics of AcNPV in Se301 and Sf-21 cells. The symptom by viral infection was similar in both of cells, but the ratio of polyhedra released from the cell was higher in Se301 cells than in Sf-21 cells. The overall PIB productivity of AcNPV was similar in both cells but the size of polyhedra was larger in Se301 cells. While the polyhedrin expression efficiency was about 2.4 times higher in Se301 cells than in Sf-21 cells, the viral growth was higher in Sf-21 cells. These results suggested that Se301 cell is very useful in the AcNPV-based expression system as a host.

  • 어류에서 바이러스성 질병이 중요하다는 인식이 증가하고 있으며 어류바이러스의 연구를 위한 세포주의 발달이 활성화되고 있다. 하지만상대적으로 해산유래의 세포 주 수는 많지 않다. 특히 유럽과 우리나라 남ㆍ서해안에서 고급어종인 감성돔은 림포 시스티스병, 이리도바이러스병 등의 바이러스성 질병이 발생하고 있다. 본 연구는 감성돔에서 유래한 세포주 개발을 목적으로 하였으며, 감성돔의 비장에서 유래한 세포를 초대배양 및 계대배양 하여 새로운 세포주를 확립하였으며, 일련의 실험을 실시하여 확립된 세포주의 특성을 확인하고 바이러스에 대한 감수성을 검사하였다.   • The effect of extraceflular matrix (ECM) protein on the neuronai differentiation of SI(-N-SH and IMR-32 human neuroblastoma cell lines was examined. When ceils were cultured on the laminin/collagen coated plate for 7 days, the extensive neurite outgrowth was observed In IMR-32. To address the reason why IMR-32 cell llne did not respond to ECM proteins, the ECM mediated early signalling mechanisms were analysed in both SK-N-SH and IMR-32. When cells were plated on the laminin/collagen coated plates, tyrosine phosphorylated proteins were Increased within an hour In both of these cells. Moreover, the foaal adhesion IlInase (FAK) was tyrosine phosphorylated in both of these two cell lines. These results suggest that the ECM mediated early signalling mechanism was normal in IMR-32 cell line. The expression of both NSE and Bcl-2 was increased by ECM treatment in SK-N-SH. However, these components were not changed by ECM In IMR 32 cells to ECM component Is likely due to the abnomality of the transcriptional regulation mechanism which Is responsible for the neuronal differentiation.

Establishment and Characterization of Clonal Cell Lines from Zebrafish, Danio rerio (제브라피쉬(Danio rerio) 배아로부터 동형세포주 확립)

  • Three types of clonal cell lines were isolated according to their size and phenotype from the adherent cell populations in long-term liquid cultures from the embryonic fibroblast cells of Zebrafish, Danio rerio. All kind of cell lines were well proliferated. The size and number of clonal cell lines derived colonies from stable embryonic cells were significantly increased in the presence of NAC and A2P conditioned medium from the cell lines. The stable cell lines and clonal cell lines were cap-able of well proliferation in vitro. These cell lines have been maintained in continuous culture without change in characteristics. A majority of the clonal cells (80%) was shown a normal chromosomal complement (50 chromosomes, 2N) in according with FACs analysis. Majority of cells were positive to vimentin staining and none of them were positive for nestin and Oct -4 by immunocytochemistry. These results indicate that the clonal cell lines obtained from cultured cells are fibroblasts and may be extremely useful in genetic manipulation for further nuclear transfer and fish cloning.

Resistance to Cisplatin Renders High Metastatic Potential in Human Non-Small Cell Lung Cancer Cell Line (Cisplatin 내성을 보이는 비소세포폐암 세포주에서의 전이 능력 증가)

  • 배경: Cisplain과 같은 세포돗성 약제에 대한 내성은 폐암 치료 실패의 중요한 원인이다. 이러한 항암제에 대한 내성의 발생기전은 복잡하고 아직 완전히 알려져 있지 않지만 불량한 예후의 원인으로 생각된다. 특히 약제 내성이 발생한 환자의 경우 기존의 종양의 급속한 성장뿐 아니라 새로운 전이 병소가 급속히 발생 및 진단됨은 약제 내성을 가진 종양이 전이에의 용이성을 획득하는게 아닌가 의심케한다. 이를 규명하기 위해 Cisplatin에 내성을 지닌 비소세포폐암 세포주 H460/CISm이 전이 능력을 Cisplatin에 민감한 비소세포폐암 세로주 H460과 비교하고자 한다. 대상 및 방법: 약제 내성 세포주를 확보하기 위하여 H460세포에 cisplatin을 점차적으로 증가시켜 처리한 후 배양하였다. H460 세포와 H460/CIS 세로에서의 혈관신생인자와 성장관련인 자의 발현양상, gelatin zymography 분석 그리고 in vivo 실험으로 nude 마우스에서의 자발적 전이 능력의 차이를 비교하였다. 결과: H460 세포를 이식한 마우스에 폐에서는 종양이 형성되지 않았으나 H460/CIS세포를 이식한 마우스 10마리중 8마리에서 종양이 형성되었다. 또한 H460/CIS 세포주에서 전이 관련 유전자로 알려진 angiopoietin-1, vascular endothelial growth factor, basic fibroblast growth factor, matrix metalloproteinase 2 등이 더 발현되었고, 전이의 침습성을 유발하는 gelatinase의 활성이 증가된 것을 확인할 수 있었다. 결론: 본 여구 결과를 통해 cisplatin에 내성을 가진 비소세포폐암세포에서 전이 능력이 증가될 수 있다고 여겨지며 이러한 사실을 토대로 초기 비소세포폐암 환자의 수술 전 항암약물요법의 타당성에 대해서 이야기 하기 위해서는 많은 임상적 연구가 필요하리라 생각된다.

Arrest of Cell Growth by Inhibition of Endogenous Reverse Transcription Activity in Cancer and Somatic Cell Lines (사람의 암세포주 및 정상세포주에서 역전사 효소의 억제에 의한 세포 성장의 제한)

  • The present study assessed the cytotoxic effects on cell growth and senescence in human cancer (A-549, AGS, HCT-116, MDA-MB-231, and U 87-MG) and normal (MRC-5 and mesenchymal stem cells) cell lines treated with efavirenz (EFA), an inhibitor of non-nucleoside reverse transcriptase (RTase). Following EFA treatment, the half-maximal inhibitory concentration (IC50) values were approximately 15 µM, and the IC50 value was significantly (p<0.05) lower in the cancer cell lines, compared to normal cell lines. After determining the IC50 values against EFA, each cell line was treated with 15 µM EFA for up to one week. Significant (p<0.05) decreases in endogenous RTase and telomerase activity were observed in the cancer cell lines. RTase and telomerase activity were absent or detected at very low levels in both EFA-untreated and treated MRC-5 and MSC normal cells. The cell doubling time (CDT) was also significantly (p<0.05) prolonged by the decreased cell growth rate in the EFA-treated cancer cell lines compared to the untreated cell lines. Furthermore, EFA-treated cancer cells displayed a high number of cells with a high intensity of senescence-associated ß-galactosidase activity (SA-ß-gal activity), compared to the untreated cells. The present study showed that inhibition of RTase activity induces cellular senescence and arrests cell growth in human cancer cell lines; however, normal cell lines showed greater tolerance against EFA. RTase treatment could offer optional chemotherapy for cancer treatment in human cancer cell lines with high RTase activity.

Inhibitory effect of Smilacis Glabrae Rhizoma on nitric oxide production in the macrophage cell line RAW 264.7 (토복령(土茯笭)이 RAW264.7 대식세포주에서 산화질소 억제에 미치는 영향)

  • Objectives : 본 연구의 목적은 제습, 해독, 통리관절등의 효능이 있는 토복령이 RAW264.7 대식세포주에서 lipopolysaccharide(LPS)로 처치하여 생성되는 nitric oxide(NO)와 prostaglandin $E_2$($PGE_2$)에 억제작용이 있는지 관찰하고자 하는 것이다. Methods : 토복령이 RAW264.7 대식세포주에 세포독성이 유무를 관찰하기 위하여 농도별 MTT assay를 수행하여 세포생존율을 측정하였다. 또한 LPS로 염증유발된 RAW264.7 대식세포주에서 토복령의 농도별 처치가 NO 및 $PGE_2$ 생성억제에 미치는 영향을 관찰하고자 NO 및 $PGE_2$ assay를 수행하였다. Results : 토복령의 농도별 처치가 RAW264.7 대식세포주에서 세포독성을 유발하는지 MTT assay로 측정한 결과 세포독성은 관찰되지 않았으며, 토복령은 LPS처치로 인하여 증가된 NO 및 $PGE_2$ 생성을 통계학적으로 유의하게 감소시킴을 관찰하였다. Conclusions : 본 연구를 통하여 토복령은 RAW264.7 대식세포주에서 LPS로 유도된NO 및 $PGE_2$ 생성을 효과적으로 억제시킴으로써 추후 염증질환의 치료제로서 가능성을 확인하였다.

  • The present report aimed at evaluating the effect of bisphenol A(BPA) and diethylstilbestrol(DES) on Leydig or Sertoli cell-lines. To identify the differences in the susceptibility to BPA upon different cell-types, assay of the cell viability was done on TM3(Leydig cells) and TM4(Sertoli cells) cell-lines. The result indicates that Sertoli cells are more sensitive to low dose of BPA than Leydig cells. Also, the BPA- or DES-treated Sertoli cells showed a reduction of phospholipase D(PLD) activity identically. According to the confirmation of the mRNA expression of fas receptor and fas ligand in the BPA-treated cells, fas/fasL system activated by BPA will deliver the apoptosis signal onto Sertoli TM4 cells. However, Fas/FasL system was not activated in the DES-treated cells unlike the BPA-treated cells.

  • 목적 : 본 연구는 계혈등 약침액이 자궁경부암 세포주 SNU-17에서 세포 사멸 효과가 있는지 알아보고자 하였다. 방법 : 자궁경부암 세포주 SNU-17에서 세포 사멸의 변화를 관찰하기 위해서 MTT cytotoxicity assay, DAPI staining, TUNEL assay, RT-PCR analysis 방법을 이용하였다. 결과 : 세포독성 검사에서 계혈등 약침액은 자궁경부암 세포주 SNU-17에 농도 의존적으로 세포독성을 나타내었다. 이러한 계혈등 약침액의 세포독성이 세포사별로 인한 것인지 다른 기전에 의한 것인지 알아본 결과 계혈등 약침액에 의한 세포독성은 DAPI staining 과 TUNEL assay에서 세포사멸의 특정적인 소견들을 나타내었다. 계혈등 약침액이 Bax, Caspase-3의 발현에 미치는 영향을 RT-PCR로 관찰한 결과 계혈등 약침액은 Bax, Caspase-3의 발현을 증가시켰다. 결론 : 이상의 결과 계혈등 약침액이 자궁경부암 세포주 SNU-17에서 세포 사멸을 야기하여 자궁경부암의 치료에 유용할 것으로 사료된다.

Selection of Acifluorfen-tolerant Eastern Black Nightshade (Solanum ptycanthum Dun) and the Expression of This Tolerance in Regenerated Plants and Their Progeny (제초제 Acifluorfen 저항성 세포주 선발 및 분화된 식물체와 그 후대에서의 저항성 발현)

  • Acifluorfen-tolerant cell lines of S, ptycanthum were isolated by stepwise selection using suspension culture. Growth of unselected line was completely inhibited at $0.5\;\mu\textrm{M}$, while some selected lines grew at $8\;\mu\textrm{M}$ acifluorfen. After subculturing on acifluorfen-free medium for 4 passages, six of the eleven cell lines screened and maintained their tolerance to $2\;\mu\textrm{M}$ acifluorfen. The regeneration capacity of selected cell lines in Solanum ptycanthum differed depending on the tell line. The acifluorfen tolerance of the somarclones regenerated from acifluorfen-tolerant cell lines differed depending on the somarclone. When plants were heated with $16\;\mu\textrm{M}$ acifluorfen, unselected control plane had over 75% phytotoxicity Many selected cell lines had less phytotoxicity than the seed-grown control plants. Tolerance to acifluorfen was inherited to the self-pollinated progenies. The inheritance patterns differed depending on the clone. Acifluorfen tolerance was inherited as a semidominant trait. Other segregation patterns were also observed. acifluorfen tolerance was recessive and acifluorfen sensitivity was dominant.
