An immunohistochemical study of the endocrine cells in the stomach of the Korean hedgehog(Erinaceus korean us) (고슴도치 위점막의 내분비세포에 관한 면역조직화학적 연구)

  • The gastric endocrine cells of the Korean hedgehog, Erinaceus korean us were studied immunohistochemically. Seven kinds of endocrine cells-, gastrin-, somatostatin-, 5-HT-, glucagon-, BPP-, motiIin-and GIP-immunoreactive cells- were identified in this study. The chracteristic findings of the regional distribution and relative frequency of them were examined. Gastrin-immunoreactive cells were very numerously detected only in the pyloric region. Somatostatin-immunoreactive cells were more numerous in the pyloric region than in the cardiac and fundic regions. 5-HT-immunoreactive cells were more numerous in the cardiac and pyloric regions than in the tundic one. Glucagon-immunoreactive cells were found few or rarely in the fundic and pyloric regions. BPP-imunoreactive cells were numerously distributed in the pyloric region, moderately in the fundic region and few in the cardiac region. Motilin-immunoreactive cells were found rarely or few in the fundic and pyloric regions. GIP-immnuoreactive Cells were detected onIy in the pyloric region.

An Immunohistochemical Study on the Endocrine Cells in the Gastrointestinal Tract of the Mandarin Fish (Siniperca scherzeri) (쏘가리의 위장관 내분비세포에 관한 면역조직화학적 연구)

  • The regional distribution and relative frequency of neurohormonal peptides-producing cells were demonstrated in the gut of the stomach teleost, the Mandarin fish, Siniperca scherzeri Steindachner, using 7 types of specific antisera raised against mammalian regulatory peptides. The gastrointestinal tract of the Mandarin fish was divided into three portions from proximal to distal, stomach, small intestine and large intestine. Cells showing immunoreactivities against regulatory peptides were situated in the epithelial lining, between epithelial cells, and gastric or intestinal gland regions with various frequencies along with gastrointestinal tract. Mast of immunoreactive cells in the epithelial lining portion were generally spherical or spindle shape having long cytoplasmic process that were reached to the lumen (open type cell) while cells showing round in shape (closed type cell) were found in the gastric gland of the stomach occasionally. Serctonin-, samatostatin-, gastrin-, cholecystokinin (CCK)-8- and human pancreatic polypeptide (HPP)-immunoreactive cells were observed in this study. However, no insulin- and glucagon-immunoreactive cells were found. Serotonin- and somatostatin-immunoreactive cells were restricted to the stomach regions with moderate and numerous frequencies, respectively. Gastrin-immunoreactive cells were demonstrated in the stomach and small intestinal portions with a few and moderate frequencies, respectively and CCK-8-immunoreactive cells were restricted to the small intestinal portions with moderate frequency. In addition, HPP-immunoreactive cells were demonstrated in the stomach and small intestine with numerous frequencies, respectively. In conclusion, the distribution and relative frequency of these immunoreactive cells in the gastrointestinal tract of the Mandarin fish shows peculiar patterns compared to those of other stomach and/or stomachless teleost.

  • The regional distributions and relative frequencies of some gastrointestinal endocrine cells in the 8 portions (fundus, pylorus, duodenum, jejunum, ileum, cecum, colon and rectum) of the gastrointestinal tract of ICR mouse (ICR) with immunohistochemical method using 7 types of specific antisera against somatostatin, serotonin, glucagon, cholecystokinin (CCK)-8, secretin, pancreatic polypeptide (PP) and gastrin. In this study, somatostatin-, serotonin-, glucagon-, CCK-8-, secretin- and gastrin-immunoreactive (IR) cells were identified. Most of these IR cells in the intestinal portion were generally spherical or spindle in shape (open-typed cell) while cells showing round in shape (close-typed cell) were found in the stomach regions occasionally. Their relative frequencies were varied according to each portion of gastrointestinal tract. Somatostatin-IR cells were demonstrated throughout whole gastrointestinal tract except for large intestine. Serotonin-IR cells were detected throughout whole gastrointestinal tract and they were most predominant endocrine cell types in this species of mouse. Glucagon-IR cells were restricted to the fundus and rectum with moderate and a few frequencies, respectively. CCK-8-IR cells were observed in the pylorus, duodenum and ileum with numerous, moderate and rare frequencies, respectively. Secretin-IR cells were restricted to the duodenum and ileum with a few and rare frequencies, respectively. Gastrin-IR cells were restricted to the pylorus with numerous frequency. However, no PP-IR cells were found in this study. In conclusion, some peculiar distributional patterns of gastrointestinal endocrine cells were found in the ICR mouse compared to those of other mammals.

  • Using immunohistochemical technique, three endocrine cell types were observed in the GIT of the Korean salamander, Hynobius leechii. BPP-immunoreactive cells were distributed from the pyloric region to the small intestine. A moderated number of somatostatin-immunoreactive cells were restricted to the pyloric region. 5-HT-immunoreactive cells were demonstrated throughout the GIT, reaching a peak in the pyloric region. The presence of BPP- and 5-HT-immunoreactive cells are identified for the first time in Hynobius leechii.

  • The pancreatic endocrine cells of the cat-shark, S. torazame, were studied using immunohistochemical method. Five kinds of endocrine cells (glucagon-, somatostatin-, insulin-, 5-HT-and BPP-immunoreactive cells) identified in this study. The chracteristic findings of the distributions of five immunoreactive cells were as follows. Glucagon-immunoreactive cells were detected as clustering group in the epithelia of the interlobular duct and singly the pancreatic acini, respectively. Insulin -immunoreactive cells were moderately observed in the epithelia of the interlobular duct or in the periphery of the islet. Somatostatin-immunoreactive cells were distributed in single or mass groups in the epithelia of the interlobular duct and the exocrine gland of the pancreas. A very few 5-HTimmunoreactive cells were seen in the periphery of the islet and the acini of the pancreas-BPP-immunoreactive cell was singly located in the periphery of the pancreatic islet, but GAS/CCK-and Chromogranin-immunoreactive cells were not found in this study.

태아 십이지장 내분인포군집의 형태 및 면역조직화학적 특성

  • 임신 26주, 27주의 생명력이 없는 한국인 태아 2예를 대상으로 십이지장 점막에서의 장내분비세포군집(일부 일본인 학자들이 명명한 "Segi's cap")의 출현, 형태 및 구조를 관찰하고. 이들 세포군집에서 내분비세포들의 존재 및 면역조직 화학적 특징들을 관찰하고자, 파라핀조직절편과 냉동박절 후 hematoxvlin과 eosin염색. Azan염색, Comori법, 그리고 면역조직화학염색 표본을 제작, 관찰하여 다음과 같은 결과들을 얻었다 세포들의 군집 형태는 장 내강을 향해서는 오목한 형태였고, 기저막을 향해서는 볼록한 형태였다. 군집을 이룬 세포들은 중층을 이루었고. 술잔세포도 장 내캉목에서 관찰되었다 또한. 내강쪽에 위치한 세포들의 경우 미세융모들의 줄무의가장자리는 관찰되지 않았다 Leuenkephalin, somatostatin, substance p, vasoactive intestinal polypeptide 및 5-HT 항혈청을 이용하여 면역조직염색한 결과 somatostatin 양성반응세포와 5-HT 양성반응세포만이 관찰되었다. 이들 somatostatin 양성반응 세포와 5-HT 양성반응세포들은 대개가 원추형의 세포들로서 개구형을 이루고 있음을 관찰한 수 있었다.관찰한 수 있었다.

The Immunological Position of Fibroblastic Reticular Cells Derived From Lymph Node Stroma (림프절 스트로마 유래 Fibroblastic Reticular Cell의 면역학적 위치)

  • Lymph nodes (LNs) are crucial sites where immune responses are initiated to combat invading pathogens in the body. LNs are organized into distinctive compartments by stromal cells. Stromal cell subsets constitute special niches supporting the trafficking, activation, differentiation, and crosstalk of immune cells in LNs. Fibroblastic reticular cells (FRC) are a type of stromal cell that form the three-dimensional structure networks of the T cell-rich zones in LNs, providing guidance paths for immigrating T lymphocytes. FRCs imprint immune responses by supporting LN architecture, recruiting immune cells, coordinating immune cell crosstalk, and presenting antigens. During inflammation, FRCs exert both spatial and molecular regulation on immune cells through their topological and secretory responses, thereby steering immune responses. Here, we propose a model in which FRCs regulate immune responses through a three-part scheme: setting up, supporting, or suppressing immune responses. FRCs engage in bidirectional interactions that enhance T cell biological efficiency. In addition, FRCs have profound effects on the innate immune response through phagocytosis. Thus, FRCs in LNs act as gatekeepers of immune responses. Overall, this study aims to highlight the emerging roles of FRCs in controlling both innate and adaptive immunity. This collaborative feedback loop mediated by FRCs may help maintain tissue function during inflammatory responses.

Immunocytochemical Studv of the Newe Growth Factor Receptor in the Neuron and its Organelles of the Adult Rat Basal Forebrain Nuclei (흰쥐 전뇌 기저부 핵의 신경세포와 그 세포내 소기관에서 신경성장인자 수용체에 대한 면역세포화학적 연구*)

  • 신경성장인자 수용체(nerve growth factor receptor, HGFr)의 소재를 휜쥐 전뇌 기저부 핵들의 신경세포와 그 세포내 소기 관에서 연역세포화학적 방법으로 관찰하였다. NGFr에 면역반응을 보이는 신경세포들은 내측중격, 수직 및 수평대각선 브로카대, 거대세포 시삭전핵 그리고 Meynert 기저핵에는 다수 미상핵-피각과 복부담창구에는 소수 관찰 되었다 NGFr에 면역반응을 보이는 신경세포들은 형태학적으로 3가지 형 즉, 1) 난형(또는 원형). 2) 방추형, 3) 삼각형(또는 다각형)으로 구분되었다 내측중격은 주로 난형의 세포로 구성되었으며(91.2%), 수직 및 수평대각선 브로카대, 거대세포 시삭전핵 및 Meynert 기저 핵에는 난형의 세포가 높은 율로 구성되었으나, 방추형과 삼각형 세포들도 내측중격에서보다는 많았다 특히 복부담창구에는 다른 핵들에 비하여 방추형세포(25%)들이 높은 출현율을 보였다 일반적으로 이들 세포의 크기는 삼각형세포가 제일 컸으며, 방추형세포가 그 다음, 그리고 난형 세포가 제일 작았다 전자현미경적 관찰에서 0.05% triton X-100을 처리한 조직중 Meynert 기저핵을 관찰한 결과. Golgi체, multivesicular body 및 소포체들이 N6Fr에 면역반응을 보였으며. trion X-100을 처리하지 않은 조직에서는 단지 수평대각선 브로카대의 신경세포 원형질 막에서만 약한 면역반응을 보였다 위의 결과로 미루어 NGFr은 조연소포체에서 합성되어. Golgi체에서 농축되고, multivesicular body를 통하여 원형질막에 위치하게 되며, 원형질막에서 NGFr은 외래성의 NGF와 복합체를 형성한후, 궁극적으로는 Iysosome의 형태로 세포체 안으로 들어 가는 것으로 추정된다.

Immunohistochemical and Ultrastructural Characterization of the Choline Acetyltransferase-immunoreactive Nerve Cells in the Diagonal Band of Broca of the Rat Basal Forebrains (흰쥐의 전뇌 기저부 대각 Broca대에서 Choline Acetyltransferase 면역반응 신경세포에 대한 면역조직화학 및 미세구조)

  • This study was performed to investigate the immunohistochemical and ultrastructural characterization of the choline acetyltransferase (ChAT)-immunoreactive nerve cells in the diagonal band of Broca of the rat basal forebrains, utilizing techniques of immunohistochemical and immunocytochemical microscopy. The ChAT-immunoreactivities were shown within neuronal cell bodies and processes by the light micoscope. According to cell shape and ratio of long axis vs short axis of cell body, the ChAT-immunoreaclive nerve cells in both vertical and horizontal limbs of the diagonal band of Broca were classified into 6 types. at the light microscopic level; round, oval, elongated, fusiform, triangular and polygonal types. As a result of the electron microscopic observation, the ChAT-immunoreactivated products appeared on the outer nuclear envelope, membranes of rough endoplasmic reticula (rER), free ribosomes and polysomes. Each cell type was subdivided into subtype I and II according to the several criteria such as volume of cell body, nuclear size relative to the cytoplasm, kinds and distribution of cell organelles and numbers and sorts of synapses. The subtype I of immnunoreactive nerve cells had large cell body and a small nucleus showing shallow indentations of nuclear evelope. In this subtype I with abundant cytoplasm, rER were well differentiated. Their long cisternae were parallelly ditributed and lamellated. One or two lamellar bodies and nematosomes were observed. The subtype II cell had small cell body and a large nucleus with deep indentations of nuclear envelope. In this subtype II with small cytoplasm, the rER were irregularly distributed and the lamellar body and nematosome were not found. A few axosomatic synapses in the subtype I and II were shown to be symmetric or asymmetric. The ratios of the symmetric synapse to the asymmetric one were investigated to be 1 : 2 and 1 : 4 in the subtype I and II, respectively. The axodendritic ones were almost asymmetric. But, the fusiform and triangular immunoreactive nerve cells were shown only to be subtype I. According to observations in this study, it is considered that the ultrastructural characterization in the 2 subtypes of each cell type may reflect the differences of the metabolic activities and projecting distances to the target cells.

  • The regional distribution and relative frequency of the pancreatic polypeptide (PP)-immunoreactive cells in the pancreas of the Korean native goat (Capra hircus) were studied by immunohistochemical methods (PAP methods) using specific antisera against to PP during developmental stage, fetus, neonate, 1-month-old, 6-month-old and adult. The different regional distribution and relative frequency of PP-immunoreactive cells in the pancreas of Korean native goat were observed during development. In the exocrine portions, they were detected in the all ages and distributed in the interacinar regions. PP-immunoreactive cells were increased from neonate to 1-month-old but thereafter decreased with developmental stage. In the pancreatic duct, PP-immunoreactive cells were demonstrated from 1-month-old but they were decreased with developmental stages in these regions. These cells were observed in the subepithelial connective tissue of the pancreatic duct with a few frequencies in 1-and 6-month-old. However, they were located in the interepithelial cells of the pancreatic duct in the adult with rare frequency. In the endocrine portion (pancreatic islets), PP-immunoreactive cells were observed from neonate, and the regional distribution of PP-immunoreactive cells from 1-month-old was divided into two patterns : dispersed 1) in the marginal regions of the pancreatic islets with moderate or a few frequencies, and 2) in the whole pancreatic islets (in a case of 1-month-old) or in the central cores (in a case of 6-month-old and adult) with numerous frequency. In conclusion, the different regional distribution and relative frequency of PP-immunoreactive cells in the pancreas of the Korean native goat were observed during development. It is suggested that the changing of feeding habits and physiological conditions during different developmental stages may cause these differences.
