Effect of Salinity on the Bacterial Community in the Sewage Treatment System (하수 처리 과정에서 염분이 세균 군집에 미치는 영향)

  • For elucidating the effect of salinity to the effect of wastewater treatment, the heterotrophic bacterial numbers, total bacterial numbers, and the bacterial community structure by FISH method were analyzed. The total bacterial numbers were not significantly changed by the salinity. But the heterotrophic bacterial numbers and bacterial community structures were drastically changed by the increase of salinity. In case of 1% salinity, the heterotrophic bacterial numbers and structure were slightly changed comparing to those of contol. In case of 2% and higher salinities, the numbers of heterotrophic bacteria and the proportions of Eubacteria, Proteobacteria $\alpha$-group, $\rho$-group and Cytophaga-Flavobacterium groups were deceasing. By these results, the salinity stress to bacterial community in waste water treatment was unveiled, and for sustaining the waste water treatment system, the salinity should be lower than 1%.

  • In this study, we integrated 40 kinds of aerobic bacteria that has a higher appearance rate and data for experts and educations, and constructed the aerobic bacterial images database. Constructed system is useful to culture of bacteria for novice without expert's heuristics and deep knowledge and to decrease time and money for handling of patient's examination results. Moreover, it can add new bacterial information in database and contribute to raise a medical quality and it is useful to support a expert's technology in department of laboratory medicine.

Microbiological Evaluation of Foods and Kitchen Environments in Childcare Center and Kindergarten Foodservice Operations (보육시설과 유치원 급식의 식품 및 환경 미생물의 오염도 평가)

  • Whereas the numbers of childcare centers and kindergartens are increasing rapidly, systematic management to control the food safety of foodservice operation is not yet well established. Samples from 12 centers in Seoul and Gyeonggi Province were collected to assess the microbiological quality of 32 raw materials, 24 cooked foods, 76 food-contact surfaces (knives, cutting boards, dish towels and gloves), 17 employees' hands and 12 air-borne bacteria. The microbiological analyses were performed for aerobic plate counts (APC), Enterobacteriaceae, E. coli and 7 pathogens (B. cereus, C. jejuni, C. perfringens, L. monocytogenes, Salmonella spp., S. aureus, and V. parahaemolyticus). Among raw materials, E. coli ($1.39{\sim}2.08\;\log\;CFU/g$) were detected in 4 out of 6 meats and 7.46 log CFU/g of APC in tofu. High enterobacteriaceae levels of 4.23, 5.14 and 4.19 log CFU/g were found in cucumber salad, steamed spinach with seasonings and steamed bean sprout with seasonings, respectively. No pathogens were found in all samples except for C. perfringens detected from raw spinach and raw lotus root. Only APC and enterobacteriaceae were found in food-contact surfaces. Two of the 23 knives and three of the 24 kitchen boards showed over 500 CFU/$100\;cm^2$ of APC; also, APC levels (5.03 to 5.44 log CFU/g) were detected in 4 of the 12 dish towels. Only one glove showed Enterobacteriaceae (2.44 log CFU/glove) contamination. Enterobacteriaceae were found in 2 employees' hands ($2.37{\sim}4.44\;\log\;CFU$/hand) among the 16 employees. The contamination levels of air-borne bacteria were shown unacceptable in two (2.25 and 2.30 log CFU/petri-film/15 min) out of the 12 kitchen areas. These results suggest that the microbiological hazards in some foods and environments are not well controlled and thus a guideline should be provided to ensure the food safety in childcare center and kindergarten foodservice operations.

  • 병원성미생물을 분리${\cdot}$동정하는 방법으로 선택배지를 이용하는 검사방법은 4${\sim}$7일의 시간이 소요되는 단점이 있으며, 기존의 PCR방법은 대부분 단일 병원성 미생물만을 검출할 수 있는 반면 본 연구에서 사용된 시스템은 6종류의 세균성 식중독균을 한 번의 분석으로 확인할 수 있었기 때문에 비용과 분석시간을 대폭 절감하는 효과가 있었다. Fig. 1은 6종류의 세균성 식중독균이 혼합된 시료에 각각의 단일 프라이머를 이용하여 특이성을 확인한 결과이며, Fig. 2는 멀티플렉스 프라이머를 이용하여 각각의 세균성 식중독균 열 추출 시료에 대한 특이성을 확인한 결과이다. PCR 산물들이 사다리(ladder) 형태로 증폭됨으로써 동시에 6종류의 세균성 식중독균을 용이하게 확인할 수 있었다(Lane 1${\sim}$Lane 6). 또한 3종류의 세균성 식중독균이 혼합된 열 추출시료에서도 특이적인 PCR 증폭 산물들을 탐지할 수 있었다(Lane 10 & Lane 11).

Distribution of bacterial biomass in the water column of Soyang lake (소양호 수중 생태계에서의 세균 생체물질량의 분포)

  • Microbiological parameters such as bacterial biovolume and biomass in Soyang Reservoir was statistically analyzed with the physico-chemical enviromental factors. Analysis of correlation and multiple regression showed that temperature affects most of microbiological parameters. Variations of total bacterial number, total bacterial biovolume and saprophyte number were highly correlatd with the concentrations of chlorophyll a and pheophytin a. Bacterial production by the $^{3}H$-thymidine incorporation rate was largely affected by Seston. It suggests that microbiological factors such as bacterial biovolume and bacterial biomass were controled by the concentration of seston and distribution of phytoplankton which acts as carbon and energy source for the bacterial community in the water column of Soyang Reservoir.

  • The total and heterotrophic bacterial distributions, compositions and alkaline phosphatese actibities were analyzed in Lake Soyang from Sep. 1987 to Aug. 1988. The heterotrophic bacteria was small portion, 0.07-2.63% of total bacterial number which ranged from $3.2{\times}10^{5}$ to $3.2{\times}10^{6}$ cells/${\mu}\ell$. The composition of bacterial community was less diverse in summer and at the fish farm site and Peridinium blooming site. Pseudomonas and Flavo bacterium were the dominant genera in all sites. The highest proportion and activity of alkaline phsophatase was appeared in Flavobacterium, while Pseudomonas was the most predominant group.

  • Methods for the measurement of aquatic bacteria can be divided into two groups. The first group of these methods is based on the 'replicon' concept that live bacterial cells, when diluted and transferred to a suitable medium, produce colonies. These methods distinguish living from dead bacteria, but they massively underestimate bacterial numbers. The second group of enumeration methods uses visual counting technique using specific apparatus such as a microscope. These methods are generally direct and simple, but it is very hard to distinguish between live and dead bacteria and between small particle and bacteria. Recently developed technique in staining methods has provided a reliable method of visual determination of aquatic bacteria. This uses epifluorescence microscopy to measure the total bacterial population. In order to present the fluorescence microscopy as a new methodology for the determination of bacterial numbers in water supplies, data were obtained from chlorine and monochloramine doses added to samples. Total counts by fluorescence microscopy were compared with standard plate count method. The total number of bacteria in water supplies can be determined with fluorescence microscopy. This technique allows better resolution of small bacteria and differentiation of particle from bacteria. Chloramine was found to persist longer in natural waters and prevent bacterial regrowth.

Disease monitoring of Alaska pollock (Gadus chalcogrammus) based on growth stages (명태 (Gadus chalcogrammus)의 성장 단계별 질병 모니터링)

  • The Alaska pollock (Gadus chalcogrammus) belongs to the family Gadidae; it is a cold water fish, and has been developed as a novel aquaculture species in Korea. In this study, we describe ongoing surveillance for aquatic animal pathogens based on growth stages. We investigated bacterial flora in rearing water, and monitored pathogens; we also analyzed histopathological traits of abnormal fish. In rearing water, the total bacterial counts were $2.1{\times}10^3cfu/mL$ and Vibrio spp. (52%) were predominant in the larvae stage. In the juvenile and adult stages, the total bacterial counts were $3.4{\times}10^3$ and $3.2{\times}10^2cfu/mL$, respectively (with Pseudomonas sp. as the predominant species; 90% and 52%). This result revealed that the bacterial flora in rearing water changed depending on the feeding types. No virulent-bacteria or problematic viruses (VHSV, viral hemorrhagic septicemia virus; NNV, nervous necrosis virus; MBV, marine birnavirus) were detected from outwardly healthy fish using either culture or PCR assay. Some juveniles (less than 5%) had gas bubbles on the gill lamellae, degeneration of the corneal epithelium, and choroid gland degeneration, suggesting that these symptoms were caused by external injury and secondary infection by opportunistic bacteria. Disease management is important to cope with disease emergence in the novel aquaculture species Alaska pollock.

Storage Characteristics of Irradiated Pacific Oysters, Crassostrea gigas (감마선에 쬔 참굴의 저장성)

  • 산란기 직전의 참굴(Crussostrea gigas)을 통상 방법으로 개각하여 0.3 및 2.0 Mrad의 감마선에 각기 쬔 후 $0^{\circ}C$의 온도하에 저장기간중 번식하는 총 세균수와 pH 변화를 측정하고 부패상을 관찰함으로씨 방사선을 쬔 참굴의 저장성을 조사하였다. 자외선을 쬔 무균해수에 개각하지 않고 정화시킨 굴과 보통 해수에 수용하였던 굴을 비교하여 정화 처리에 의한 세균 제거의 효율을 조사하였으나 4-5일의 장기간 처리에도 불구하고 별다른 세균제거 결과를 볼 수 없었다. 이것은 굴이 보유하고 있는 본래의 총 세균수가 적기 때문이라고 추측된다. 감마선을 쬐지 않은 굴은 저장 15일째에 부패했음이 완연했으나 0.3 Mrad의 감마선을 쬔 굴은 35일후 까지도 시큼할뿐 별로 변질되지 않았다. 2.0 Mrad의 감마선을 쪼인 굴은 관능상의 선도 유지에 있어서 0.3 Mrad로 쬔 굴보다 휠씬 나빴고 off-odor가 심했다. 굴 저장중의 pH 변화로 선도를 판정함은 그 신빙성이 극히 희박하였고 선도의 저하와 함께 pH가 내려간다는 일반적인 인식과는 달리 어류부패의 경우와 같이 저장 초기에는 약간 하강하나 부패의 진행과 함께 상승함이 확인되었다.
