Distribution of bacterial population and activities at the tidal flat in southern area of Ganghwa Island, Korea (강화도 남단 갯벌에서의 세균수 및 활성의 분포)

  • Vertical distribution of bacterial population and activities were compared between the muddy and sandy sediment of the tidal flat located in southern area of Ganghwa Island. The average of bacterial number and activities in mud flat was slightly higher than that in sand flat. Bacterial number was markedly increased at the sandflat during the investigation period, which seemed to be the result of increase of mud content. The number and activities of microorganisms in mudflat was comparable to that of the mangrove sediments. There was no differences in bacterial number with the depth, but the aminopeptidase activity and bacterial productivity were decreased markedly within 3~5cm depth of sediment due to the availability of carbon sources and molecular oxygen in pore water. The number and activities of microorganisms in mudflat did not correlated with the distribution of benthic microalgae; however, those in sand flat were closely correlated with the distribution of benthic microalgae. The distribution of marsh plant in the mudflat area might be the reason of the difference.

Determination of Intracellular ATP of bacteria on the surface of Chicken (Bioluminescence방법에 의한 계육표면에 존재하는 세균수 측정)

  • Determination of bacterial adenosine triphosphate (ATP) based on luciferin-luciferase bioluminescene reaction was applied to the measurement of bacteria on the surface of chicken as an alternative rapid method. The light yield was proportional to the concentration of ATP giving a straight line within a range of $10^{-10}\;to\;10^{-6}M$. The bacteria isolated from the surface of chicken were identified as Escherichia coli, Hafnia alvei, Pseudomonas putida and Aeromonas hydrophila. When the ATP contents of each bacteria were determined by bioluminescence reaction and compared, there was no significant difference (r = 0.95). The Patterns of growth curves for E. coli look very similar, when the bacterial growth was monitored by ATP content and viable cell count. When bacterial ATP of each samples collected every 2 days during storage for 14 days at $4^{\circ}C$was determined and compared with viable cell count, it gave a good correlation (r = 0.95, n = 32).

Bacteria on Granular Activated Carbon for Tap Water Purifier (정수용 입상활성탄상의 세균)

  • To investigate the bacterial growth on granular activated carbon (GAC) for the purification of tap water, fixed bed columns with GAC were installed and operated at an empty bed contact time (EBCT) of 1 min$\pm$0.08 min. There was no bacterial breakthrough in the spring. However, the bacterial concentrations of effluent (($10^3$ CFU/ml) were higher than that of the influent ($10^2$ CFU/ml) after 10 day operation in summer. More bacteria were enumerated near the entering point of the tap water, while the bacterial activities were similar throughout the columns. Different bacterial species were detected on coal- and plant-based GAC, although the dominant genus was the same as Acinetobacter.

Bacterial Community of Free-living and Aggregated Bacteria at Thawing Period in Lake Baikal (해빙기 바이칼호에서 부유세균과 Aggregates에 부착한 세균의 군집구조)

  • Fluorescent in situ hybridization (FISH) with rRNA-targeted oligonucleotide probes was used to compare the community structures of free-living and aggregated bacteria at thawing period in Lake Baikal. Targeted groups were Eubacteria, $\alpha$-, $\beta$-, $\gamma$- proteobacteria groups, Cytophaga-Flavobacterium group and Planctomycetales. Total bacterial numbers of free-living bacteria were ranged from $0.2{\times}10^6\cells{\cdot}ml^-1$ to $3.2{\times}10^6\cells{\cdot}ml^-1$, which were decreasing with depth, while the aggregated bacterial numbers were dramatically increasing from $0.4{\times}10^4 to 3.3{\times}10^4 \cells{\cdot}ml^-1$ with depth. The ratios of EUB probe binding cells to DAPI counts were ranged from 52.3 to 74.1% in free-living bacteria, and from 39.6 to 66.7% in the aggregated bacteria, respectively. Community structures of the aggregated bacteria were very different from each free-living bacteria at every depth. At 25 m depth, where the chlorophyll a concentration was highest, both structures were quite different from those of surface layers, rendering the fact that the community structures might be affected by phytoplankton. The vertical profile of community structure of aggregated bacteria is particular. The proportion of $\beta$-proteobacteria group was increasing with depth and it was 51.8% at 100 m, but the dominant group was $\gamma$-pro-teobacteria group at 250 m. Taken together, the biodiversity and succession of aggregated bacteria are quite different from free-living bacteria.

Vertical Variation of Total Bacterial number in Daechung Reservoir (대청호에서 세균의 수직적 분포에 관한 연구)

  • To define the ecological roles of bacterial community in Daechung Reservoir. Vertical variation of total bacterial numbers were estimated at MAN site for one year with DAPI staining. The bacterial numbers were counted, environmental factors were monitored at the surface, -5m, -10m, -15m and -25 m of MAN sited and tried to explain the vertical bacterial numbers with environmental factors. The bacterial number was $1.6-1.7.0 x10^6$ cells/ml at the surface, $2.3-11.0x10^6$ cells/ml at 5 m depth, $1.2-1.4.0x10^6$ cells/ml at 10 m depth, $1.4-15.0x10^6$ cells/ml at 15 m depth and $1.4-1.3.0x10^6$ cells/ml at 25 m depth. The Mean bacterial number at the surface was more that those at any other depth. The explanation of the vertical total bacterial numbers with environmental factors were suggested.

The Seasonal Variation of Active Bacterial Abundance in Lake Soyang (소양호에서 활성세균수의 계절적.수직적 변화)

  • Vertical and temporal variations of active and total bacterial abundance were monthly estimated in Lake Soyang from April 1999 to January 2000. The number of total and respiring bacteria was determined directly under microscope by AODC and CTC methods, respectively. The number of total and active bacteria varied from $2.1{\times}10^5 to 3.1{\times}10^6 $,$cells{\cdot}ml^{-1}$ and $1.8{\times}10^4 to 8.0{\times}10^5 $,$cells{\cdot}ml^{-1}$, respectively. The proportions of respiring bacteria to total cell ranged from 3.7 to 44.2% : The proportions was the highest in November 1999 and the lowest in December 2000. The specific activity of${\beta}$-glucosidase divided by total bacteria was$1.6{\times}10^5\;amol{\cdot}cell^{-1}{\cdot}hr^{-1}$in August and$1.4{\times}10^5\;amol{\times}cell^{-1}{\times}hr^{-1}$in September while the specific activity divided by CTC active bacteria was about$3.6{\times}10^5\;amol{\cdot}cell^{-1}{\cdot}hr^{-1},\;24.0{\times}10^5\;amol{cdot}cell^{-1}{cdot}hr^{-1}$. The specific activity of active bacteria in September was 6.7 times higher than that of August. By these data of active bacteria, the new information of aquatic ecosystem was unveiled.

Evaluation of Bacteriological Safety in Deep Confined Aquifer (심층피압지하수에서 세균군집의 분석에 의한 분변성 오염 평가)

  • The study on the bacterial distribution was performed to evaluate the bacteriological safety in 4 deep confined aquifers drilled for drinking. All the investigated aquifers were oligotrophic, howevec bacterial densities and activities and the number of indicator bacteria in C-2 boreholes were much higher than those in other boreholes. Fecal pollution was observed in 3 boreholcs except C-1 borehole. The ratio of gram positive bacteria of the C-2 borehole was much lower than those 01 another boreholes. The numbers of total colifonns, fecal coliforms, and fecal Slreptococcus in the C-2 boreholes were 61, 33, 55 CFU (250 ml)$^{-1}$

  • Bioluminescence technique was applied to ginseng powders. ATP bioluminescence can be used as a rapid method that can be implemented for microbiological monitoring of contaminated ginseng powders. The RLU (relative light units) of ATP was proportion to bacterial CFU (colony forming units) when in high contaminated ginseng powders ($\geq$ about 1.0$\times$10$^4$CFU/g). However, when in low contaminated ginseng powders ($\times$10$^4$CFU/g), the RLU was not proportion to CFU, respectively.

  • 장내세균의 구성과 각종 질환의 상관관계에 대한 연구가 활발히 수행되고 있다. 대장관련 질환 뿐 아니라, 당뇨, 비만 등 대사성 질환, 심지어 정신질환에까지 장내세균 구성의 영향이 미치는 것으로 밝혀졌다. 이에 다양한 천연 소재를 활용하여 각종 동물질환모델에서 생리활성을 검토할 때, 장내세균의 구성을 분석하여 그 상관관계를 밝힐 필요가 있다. 장내세균의 구성에 따라 질환의 양상이 달라질 수 있으며, 치료에 영향을 줄 수 있다. 동물실험을 수행할 때에도 실험동물 간 장내세균의 구성이 다르면 질환의 유도 또는 천연소재에 의한 치료 패턴이 달라질 수 있음을 가정하여 본 연구를 수행하였다. 본 연구에서는 EtOH와 LPS를 이용하여 간장해 동물모델을 구축하고, 장내세균 구성을 T-RFLP법으로 분석하여 그 상관관계를 검토하였고, tracheloside 등의 천연물을 투여함으로써 성분의 대사 패턴을 확인하였다. 이 결과는 천연소재의 간장해 개선 연구를 수행하기 위한 기초 결과로 활용될 수 있다.

Seasonal Variation of Heterotrophic Bacteria of the Marine Ranching Ground of Tongyeong Coastal Water, Korea (통영 바다목장 해역의 종속영양세균의 계절적 변화)

  • Surface and bottom sea water samples were harvested from the 5 stations in the marine ranching ground of Tongyeong coastal water from year 2000 to 2002. Cell number of heterotrophic bacteria was determined by using the plate counting method to explore the variation of the cell population of heterotrophic bacteria. Sea water samples collected in summer (in July and August) contained much larger number of heterotrophic bacteria than those harvested in spring, autumn and winter. Heterotrophic bacteria were usually more abundant in surface sea than in bottom sea water. However the reverse was true for sea water collected in December 2001 and February 2002 due to suspended solids accumulating more abundantly in seabed area because of the slower convective current of the sea water in winter. Number of heterotrophic bacteria did not have a strong relationship with frequency of typhoon indicating that the path and powerfulness of the typhoon, localized torrential downpour and temperature variation accompanying the typhoon should be considered all together at the same time as well as the frequency of typhoon to explain clearly the variation of cell number of heterotrophic bacteria. The dominant species isolated from the marine ranching ground of Tongyeong were identified to be Pseudomonas fluorescens, Pseudomonas stutzeri, Acinetobacter lwoffii and Sphingomonas paucimobilis.
