Background : The intrapleural hypofibrinolysis is caused by mainly excessive concentration of pleural plasminogen activator inhibitor-1 antigen(PAI-1 Ag), which binds tissue type plasminogen activator. In pleural inflammation induced by sclerosing agents for pleurodesis, levels of pleural PAI-1 antigen increase in relation to decreasing D-dimer levels. It has been known that the pleural mesothelial cells have the capability of secreting PAI-1 Ag in response to inflammation in vivo. Therefore, we estimated whether pleural inflammation changes the balance between fibrinolytic and coagulative properties in exudative pleural effusions. Method : The thirty cases was included in our study. We determined the pleural levels of glucose, lactic dehydrogenase(LDH), pH and the counts of white blood cell(WBC), polymorpho leukocyte(PMN), lymphocyte as the parameters of pleural inflammation and cellular components of pleural fluid. The plasma level of fibrinogen in fluid and the neutrophil count in blood were determined. The levels of D-dimer, PAI-1 Ag and thrombinantithrombin III complex(TAT) were determined by ELISA(Behring, Marburg, Germany). Result : The causes of pleural effusion were as following : tuberculous in 14 cases, malignant in 10 cases and parapneumonic in 6 cases. The levels of pleural D-dimer, PAI-1 Ag and TAT was significantly higher than that of plasma(p<0..001). The severity of pleural inflammation did not correlated with pleural D-dimer, PAI-1 Ag, TAT and their plasma levels. But the level of pleural TAT correlated with pleural WBC and lymphocyte count. Conclusion : We found that the severity of pleural inflammations did not correlated with pleural D-dimer, PAI-1 Ag, TAT and the possibility of local production of PAI-1 antigen is present.
본 연구는 섬유소 분해효소 유전자 Cel D(Cellulase Digestion)가 도입된 형질전환 돼지 생산을 통하여 섬유소 함유 사료의 이용 효율을 증대시키고, 나아가 장기이식동물 및 고가의 의료용 단백질 생산가축을 개발하기 위한 원천기술 확보에 있다. 섬유소분해 유전자의 크로닝 및 조직 특이적 발현벡터를 개발하기 위하여, 우선 소의 위중 제4위 내의 미생물로부터 전체 유전자를 분리하였고, 이렇게 작성된 DNA library에서 섬유소분해 관련 유전자인 약 2.0 kb의 Cel D유전자를 크로닝하였으며, 췌장 특이적 발현 프로모터(rat elastase I: 약 200bp)를 크로닝한 후, 미세주입용 형질전환 재조합 벡터를 구축하기 위하여 rElastase I 프로모터 하류에 섬유소 분해 유전자(Cel D)를 연결하여 약 3.0 kb 크기의 재조합 벡터를 준비하였으며, 재조합 유전자를 1세포기 수정란 전핵내에 미세주입 하기 위해 Sal I과 BglII를 이용 유전자 단편을 만들었다. 구축된 유전자를 미세주입하기 위한 수정란을 회수하기위해 총68두의 돼지를 4-5두씩 분리사육하면서 발정동기화 및 과배란 유기를 위해 PG600, Altrenogest, FSH, hCG를 투여하였으며 hCG투여후 약54시간에 외과적 방법에 의해 총 1,359개의 수정란을 회수하였고, 이중 미세주입가능한 1세포기 수정란은 1,296개로 두당 평균 15.9개 였다. 1,296개의 1세포기 수정란 중에서 재조합 유전자(rE I-CelD)가 미세주입된 660개의 수정란을 32두의 수란돈에 외과적 방법에 의해 이식하였으며, 이식되어진 모돈 13두가 분만하여 40.6%의 임신율을 나타내었다. 이렇게 분만된 13두에서 총 65두(암:33두, 수:32두)의 자돈이 생산되었으며, 형질전환 여부를 판명하기 위해 자돈의 꼬리조직으로 부터 genomic DNA를 추출하고 PCR 검정을 실시하였다. PCR 검정 결과, 섬유소 분해 유전자가 도입된 자돈은 5두 이었으며, 그 결과를 Table에 나타내었다.(Table Omitted) Table 1 에서와 같이 섬유소 분해효소유전자가 형질전환된 자돈은 65마리 중 5마리로 7.69%의 형질전환율을 나타내었으며, 5마리의 자돈중 2두(암:1두, 수:1두)는 분만 후 즉시 폐사되었으며 2두(암:1두, 수:1두)는 86일령 그리고 14일령에 폐사하여 현재 1두(암)가 생존하여 섬유소 분해 사양 실험 중에 있다.

In vitro digestibility of filefish, protein was substantially decreased by fiber constituents in the follow-ing order : pectin (9.97%), gum karaya (7.03%), sodium alginate (6.12%),and cellulose (1.52%). The order of reduction by fibrous residues from vegetables ranked as follows : sea tangle (12.36%), Ro-maine lettuce (11.12%), perillar leaf (8.96%), and green pepper (5.15%). The inhibitory effect of the dietary fibers towards filefish protein digestion, expressed as soybean trypsin inhibitor equivalents, in-creased with added levels, but the inhibition differed with the sources of dietary fibers. Sea tangle and sodium alginate were most active in decreasing the concentration of essential amino acid from filefish protein hydrolysis. Sodium alginate exerted an inhibitory effect on the activity of trypsin, but the other fiber constituents did not have an inhibitory potency on trypsin and bacterial pretense (Streptomyces griceus). Results supported that dietary fiber components may reduce protein digestibility through the interaction of dietary fiber components with filefish protein.
A filamentous microorganism was isolated from completely rotten wood for the production of cellulolytic enzyme. The Trichoderma sp. FJ1 produced a large amount of cellulolytic enzymes, such as CMC, xylanase, ${\beta}-glucosidase$, and avicelase. For the production of the enzymes, when cellulolsic wastes were used as carbon sources of strain FJ1, rice straw showed higher enzyme activities than sawdust and pulp. The activities of CMC, xylanase, ${\beta}-glucosidase$, and avicelase were 2.95, 5.89, 0.45, and 0.12 U/ml in use of rice straw, respectively. To enhance production of the enzymes, the mixture substrate of rice straw and commercial cellulosic materials was investigated as carbon sources. The highest activities of CMCase, ${\beta}-glucosidase$, and avicelase were found in the mixture of rice straw and avicel, particularly rice straw:avicel (50:50), and the highest xylanase was obtained in the mixture ratio of 71:29. Bacto peptone addition of 0.1% showed enhanced production of the cellulolytic enzymes in which the activities of CMCase, xylanase ${\beta}-glucosidase$, and avicelase were 19.23, 27.18, 1.28, and 0.53 U/ml, respectively. The production of the enzymes using rice straw was efficiently induced in present of avicel and pulp containing high content of cellulose. Consequently, the filamentous microorganism, strain FJ1 utilized various cellulosic wastes as carbon sources and cellulases productivities were excellent compared to those of others strains reported previously, suggesting that the strain FJ1 will be expected as a favorable candidate for biological saccharification of cellulosic wastes in further.

In particular, a medium composed of rice straw (0.5%, w/v) and pulp (0.5%, w/v) yielded larger cellulolytic enzymes: CMCase 24.3 unit/ml, xylanase 38.7 unit/ml, $\beta$-glucosidase 1.5 unit/ml, and avicelase 0.6 unit/ml. The filamentous microorganism, strain FJ1 utilized various cellulosic wastes as carbon sources and will be expected as a favorable candidate for biological saccharification of cellulosic wastes.

Fish paste was prepared to enhance physiological functions by adding 2.5, 5 and 10% dietary fiber isolated from ascidian (halocynthia roretzi) tunic. Hardness, adhesiveness, gumminess, chewiness and shear force of the fish paste were increased with addition of the dietary fiber. Water activity and Hunter's color values of the fish paste were not significantly changed by addition of the dietary fiber. Results of sensory evaluation indicated that no difference was observed in color, texture and overall acceptance (p<0.05). However, the fish paste with 5% dietary fiber scored the highest and was generally preferred by sensory panels.
Response surface methodology (RSM) was used for optimization the fraction conditions of xylose from pepper stem with dilute sulfuric acid. The independent variables were acid concentration in the range of 1.134 to 2.866%, reaction temperatures in the range of 142.68 to $177.32^{\circ}C$, and hydrolysis time in the range of 6.34 to 23.66 min. were studied. The dependent variables were xylose yield from pepper stem, and the production of by-products, for example, furfural, acetic aicd, HMF etc. Experimental results had a good match with statistical result. The maximum xylose yield obtained in this experiment was 71% concentration.
