Application of Gamma Irradiation for the Microbiological Safety of Fried-Frozen Cheese Ball (냉동치즈볼의 미생물학적 안전성 확보를 위한 감마선 조사기술의 이용)

  • This study was conducted to sanitize fried-frozen cheese ball by gamma irradiation. Total aerobic bacteria and yeasts and molds counts were 4.4 and 2.8 log CFU/g in non-irradiated sample, respectively. Microorganisms were decreased with increase of irradiation dose $(D_{10}=1.25\;kGy)$, and were not detected in samples irradiated at 3 kGy or more $(<10^2\;CFU/g)$. $D_{10}$ value for Escherichia coli (KCTC 1682) was 0.25 kGy. TBA (2-thiobarbituric acid) values were increased as irradiation dose was increased, but there was no significant difference between non-irradiated and irradiated samples at 3 kGy or less (p<0.05). The results of sensory evaluation showed that there was no significant difference between non-irradiated and irradiated samples at 3 kGy or less, the sensory scores were decreased with irradiation at 5 kGy or more (p<0.05). These results indicated that gamma irradiation at 3 kGy was considered to be an effective treatment to ensure the microbiological safety of fried-frozen cheese balls without any sensorial change, even though further studies should be investigated to reduce detrimental effects induced by irradiation.

  • Mutual growth limitation was observation when the two antagonistic fungi was come in contact with each other. Brown line was formed 2day after contact with Trichoderma spp., and then, green spores formed overnight. The laccase activity of L. edodes was stimulated when this fungus wsa co-incubated with Trichoderma spp. for a few days in liquid media. In sawdust-rice bran nixtures, outstanding broun line developed when the two antagonistic fungi co-cultured. The pH of the substrates changed from 5.5 to 4.5 after overgrowth, suggesting a difference in the degradation ability and the preference of the two fungi for the lignocellulose material.

  • Two experiments were conducted to study the nature of protein degradation in fish muscle postmortem, first one with English sole (Paraphyrus vetulus) followed by another with rockfish (Sebastodes spp.). In the first one, proteolysis was measured by the increase of amino-N in gutted fish during storage in ice and in the homogenates prepared from fish of different ice storage during $20^{\circ}C-incubation$. In order to test the possible involvement of fish muscle a cathepsin, a portion of each homogenate sample was exposed to 0.5 Mrad of gamma radiation to destroy viable microorganisms prior to the incubation. Proteolysis was not detected until viable count reached a level above $10^7$ cells per gm fish flesh, corresponding to 31 days of ice storage. Even if fish flesh were mechanically disrupted by means of homogenization and subsequently incubated at $20^{\circ}C$, proteloysis attributable to muscle cathepsin was not detected. In the second with rockfish muscle aseptically prepared from freshly killed fish, the samples were inoculated with a proteolytic strain of fish spoilage Pseudomonad or irradiated at 0, 0.5 and 3.0 Mrad. The four samle groups were stored at $0-2^{\circ}C$ to compare the spoilage pattern of sterile and non-sterile muscle. In sterile muscle both total-N (extracted in 0.5M KCl) and amino-N $(soluble\;in\;70\%\;ethanol)$ declined slightly while the inoculated muscle showing increase in parallel with the increase of number of inoculated bacterium. The results indicate that proteolysis is a part of normal fish spoilage and the onset of proteolysis is delayed until viable count reaches its maximum level. Contribution of fish muscle cathepsin to protein degradation in white flesh fish muscle post-mortem is nil.

Characterization of Tobacco mosaic virus Isolated fromSolanum tuberosum ‘Chubak’ in Korea (감자 '추백' 에 발생한 Tobacco mosaic virus 의 특성)

  • An isolate of Tobacco mosaic virus (TMV) was isolated from potato cultivar ‘Chubak’ showing vein clearing and mild mosaic at Namhae in Korea. This isolate, TMV-St, was differentiated from other tobamoviruses based on biological properties, serological relationships and nucleotide sequence analyses of coat protein genes. TMV-St caused typical symptoms on four indicator plants as compared to the tobamovirus of TMV-U1, Pepper mild mottle virus (PMMoV), and Tomato mosaic virus (ToMV), which caused economic losses in Solanaceous vegetables, tomato, pepper, and eggplant. Remarkably, the TMV-St induced distinctly different symptom of systemic chlorotic spots on Chenophodium murale. On C. murale, Gomphorena globosa, and Nic-otiana rustica, the four viruses were classed by the virulence of systemic or local infections. In serological test TMV-St antiserum showed a precipitation line with each other tabamovirus. The CP gene of TMV-St contain 477 nucleotides, and the nucleotides sequence was the most similar to that of TMV-U1.

Effect of Homofermentative and Heterofermentative Lactic Acid Bacteria on the Quality and Aerobic Stability of Silage : Meta-Analysis (Homo형 발효 및 Hetero형 발효 젖산균의 사용이 사일리지의 품질 및 호기적 안정성에 미치는 효과 : 메타분석)

  • In the silage quality, the process of fermentation and use of micro-organism as starter cultures are crucial. The present study investigated the effect of homo and heterofermentative lactic acid bacteria as a starter culture strain on the silage quality and its aerobic stability using meta-analysis. Total 19 studies were used in the analysis. According to the strains of lactic acid bacteria used, experimental groups were assigned to heterofermentative (HEL), mixture of hetero and homofermentative (HHO), homofermentative (HOL) and various homofermentative (HOM). In each experimental group, standardized result of the treatment (with inoculant) was compared with the control (without inoculant) using risk ratio calculation method. The effect of experimental group was also compared. Different starter culture strains did not influence on silage pH and propionic acid production. However, lactic acid and acetic acid production were significantly (p<0.05) influenced by the start culture strains. HOM showed the greatest lactic acid production and HEL was the lowest. In contrast, HEL showed the greatest in acetic acid production and HOM and HOL were the lowest. In terms of ammonia nitrogen, HHO produced more (p<0.05) than the rest of the treatment. In crude protein and water soluble carbohydrate contents, there were no significant effects among treatments. Different strain of lactic acid bacteria significantly influenced on the effect of aerobic stability (p<0.05). Use of heterofermentative showed greater aerobic stability and the use of homofermentative lactic acid bacteria.

Control effects of some systemic insecticides to small brown planthopper, Laodelphax Striatellus (Fallen), on barley (맥전세대 애멸구에 대한 몇가지 침수성 살충제의 방제효과)

  • The control effects of four systemic insecticides $Disyston^{(R)}$ (disulfoton) 5G, $Temik^{(R)}$ (aldicarb) 15G, $Ortran^{(R)}$ (acephate) 5G and $Curaterr^{(R)}$ (carbofuran) 3G, on small brown planthopper, Laodetphax striatellus (fallen), were studied by applying them in the furrow of spring barley field. Two different application times; April 19 and 25, and three different dosage rates were used. The results are as follows: 1. No significant difference in the mortalities among the developmental stages were observed with the treated barley on April 25, when various life stages had infested on the cutting of the treated barley in the laboratory. The residual periods were 19 days for Disyston and 31 days for Temik. 2. With field experiments, highly effective control effects were obtained with Disyston and Temik. The residual period was longer with the treatment on April 19 than with those on April 25 for Disyston, while they were reverse for Temik. 3. The effective doses were 2kg/10a for Disyston, while the effectiveness lasted more than 40days with dose of 1kg/10a for Temik. 4. From above results, Disyston and Temik could effectively reduce the population of small brown planthopper in the barley field, which is the effective reproductive sources of the population in the rice field.

Development of Seed Culture Using Soybean for Mass Production of Lovastatin with Aspergillus terreus ATCC 20542 Mutant (대두를 이용한 Lovastatin 대량생산용 Seed Culture의 제조기술)

  • Lovastatin (Mevinolin, Monacolin K) is a well-known drug for the therapy of hypercholesterolemia. It is an important fungal secondary metabolite as it inhibits 3-hydroxy-3-methylglutaryl-coenzyme A reductase (HMG-CoA reductase, EC 1.1.1.34) which catalyzes a major rate-limiting step in the biosynthesis of cholesterol. Both soybeans and black soybeans with Aspergillus terreus ATCC 20542 mutant were used as the seed culture for the mass production of lovastatin. The production of lovastatin in soybean seed culture of Asp. terreus was twofold compared to that of black soybean seed culture. The effect of two different vessels (petri dish and Erlenmeyer flask) on lovastatin production was also studied. The production of lovastatin on petri dish was tenfold to that of Erlenmeyer flask. Furthermore, the most lovastatin production on rice bran was achieved when the soybean seed culture was treated by heat shock at $30^{\circ}C$ for 1 hour, representing 82% of HMG-CoA reductase inhibition in the koji extract. We estimated that the heat treated soybean seed culture could be a new method for the mass production of lovastatin.

Control Efficacy of a New Fungicide Fludioxonil on Lettuce Gray Mold According to Several Conditions (발병 조건에 따른 fludioxonil의 상추 잿빛곰팡이병 방제효과)

  • Fludioxonil is derived from the antifungal compound pyrrolnitrin produced by Pseudomonas pyrrocinia and classified as a reduced-risk fungicide by the US EPA. The efficacy of fludioxonil for the control of lettuce gray mold caused by Botrytis cinerea was evaluated under several conditions such as growth stages of host, inoculum concentrations, and amounts of potato dextrose broth (PDB) included in spore suspension of B. cinerea. At 4-leaf stage of lettuce plants, fludioxonil applied at 2 ${\mu}g$/ml was more effective for the control of gray mold than at 5- and 6-leaf stages. However, fludioxonil at more than 10 ${\mu}g$/ml provided similar control activity in all growth stages of lettuce tested. The fungicide (10 and 50 ${\mu}g$/ml) also gave excellent control of gray mold on lettuce seedlings inoculated with spore suspensions of B. cinerea ($2.5{\times}10^5$ to $2{\times}10^6$ spores/ml). But, control efficacy of fludioxonil (2 ${\mu}g$/ml) was negatively correlated with inoculum concentration. Addition of PDB in spore suspension of B. cinerea resulted in higher disease severity than non-treated control. By inoculating spore suspension including 0.5% PDB, the fungicide gave the most control activity on the disease, followed by 1% and 2% PDB. The results suggest that fludioxonil has potential to control gray mold of lettuce, but the fungicide at a concentration having moderate activity may represent low control efficacy on the disease under some conditions.

Development of Predictive Models of Listeria monocytogenes in Fresh-Cut Fruits and Vegetables (신선편의 냉장·냉동 과채류에서 Listeria monocytogenes의 예측모델 개발)

  • Processing fresh produce into fresh-cut products increases the risk of bacterial growth and contamination by breaking the exterior barrier of produce. Our objective in this study was to develop predictive models of Listeria monocytogenes in the fresh-cut salad, fresh-cut pineapple, and frozen mango. Predictive growth and survival models were developed to predict the change of L. monocytogenes populations in the fresh-cut salad (4, 10, 12, 13, 17, 25, and 36℃), fresh-cut pineapple (4, 10, 17, 25, 30, and 36℃), and frozen mango (-2, -10 and -18℃) as a function of temperature. The growth of L. monocytogenes in fresh-cut salad and pineapple was observed at above 13℃ and 10℃, respectively. The growth of L. monocytogenes in pineapple was faster than in salad. The delta value of L. monocytogenes in frozen mango increased as the storage temperature decreased. The results indicate that L. monocytogenes behave differently according to the physicochemical properties of fresh-cut fruits and vegetables. Since L. monocytogenes grow and survive well in refrigerated and frozen conditions, management programs and preventive controls for the processing of fresh-cut produce should be effectively implemented to enhance the safety of fresh-cut fruits and vegetables at retail markets.

The Yield Loss Due to the Brown Plant hopper, Nilaparvata lugens Stal, in Rrelation to the Growth Stages of the Rice (벼의 생육단계별 벼멸구 피해가 수량에 미치는 영향)

  • This study was carried out to analyst the damage due to the brown planthopper, Nilapalvata lugens Stal, at different stages(maximum tillering, booting, heading, and milking) of rice. The results are as follows: The population density of the migrating and the first generations of the brown planthoppers seemed not to be large enough to cause significant effects on the plant height and the number of tillers, while the second and the third generations caused significant reduction in 1000 grain weight(Yl) and the filled grain percentage$(Y_2)$. They were very closely related to the population density(X) of tile second generation of the BPH; $$Y1(g)=25.602-0.0213X(r=-0.5205^{**})$$ $$Y2(\%)=76.97-0.1370X(r=-0.4820^{**})$$ The relationship between rice yield (Yg) and the accumulated total density (X) of BPH per hill by the time of harvest; $$Y(g)=24.694-0.0576X(r=-0.6959^{**})$$ Since the source of the outbreak BPH is the first generation, it nay conclude that the detailed information on the population status of the first generation; density, age components, ratio of the brachypterous form, are very important in relation to management of the pest.
