Effects of Topping Methods on Yield and Major Agronomic Characteristics in Hibiscus manihot L. (적심방법이 닥풀[Hibiscus manihot L.]의 주요형질과 수량에 미치는 영향)

  • To investigate the effects of topping method on the yield and major agronomic traits of three varieties differed in plant type of Hibiscus manihot the experiments were carried out Mokpo Branch Station of Crop Experiment Station, in 1993. Fresh weight of root, dry weight of culm, dry weight of capsule and dry matter ratio on root were highest in the long culmed variety, and at the plots with topping of growing point in main culm and branches(Treatment No.3). Considering from the above results, optimum topping method seemed to be topping of growing point in main culm and branches in the long culmed variety.

Varietal Differences in Growth and Development Affected by Temperature in Barley (대맥품종의 오도에 따른 생육반응)

  • In order to get informations for breeding early barley cultivars, the thermosensitivity and low temperature growth ability about the breeding materials vernalized were tested at the high '||'&'||'amp; low temperature and field conditions. The temperature coefficient (Q10), ratio and thermosensitivity for plant height were various among cultivars versus temperature treatments. The early cultivars had greater Q10 values and insensitive responses to temperature changes. The Q10 and thermosensitivity for the time of flag leaf unfolding were various among cultivars, but the coefficient of variation in the thermosensitivity was greater, so the character would be more useful for investigating the varietal differences. The Q10 and thermosensitivity for plant height were not significantly correlated with Q10 and thermosensitivity for the time of flag leaf unfolding. The periods from heading to flowering were 2 to 4 days at the high temperature plot, but 1 to 11 days at the low temperature plot, showing much greater variations. In general the cultivars with greater difference of heading time between the high and low temperature plots were Suwon 218, SB 76588, Oweolbori, and Jogangbori, showing the sensitive thermophase and early heading.

Effects of Flower Color and Culture Periods on the Growth Characteristics and Quality Differences in Chinese bellflower (Platycodon grandiflorum) (길경(桔硬)의 화색(花色) 및 재배년차(栽培年次)에 따른 생육특성(生育特性)과 품질(品質) 차이(差異))

  • These studies were conducted to know the any differences in growth characteristics and root quality due to flower color and culture periods in Chinese bellflower. White flowered lines had tendency of increasing in growth characters such as stem numbers, capsule numbers and seed yield per plant, plant height, and root characters such as root weight, root length and root diameter even though statistically not significant. No differences were observed in saponin and essential oil contents between them. Three years old plants had more stems and capsules per plants, root weight and root length than two years old plants while no difference in saponin and essential oil contents between them. Saponin content was gradually decreased while essential oil content was clearly lowed in the roots from plants cultured more than 3 years even though no differences in stem and capsule numbers per plant and plant height.

Optimization of particle gun-mediated transformation system in Cymbidium (유전자총을 이용한 형질전환 심비디움 식물체 생산체계 최적화)

  • This study is conducted to develop an efficient transformation system via particle bombardment with PLBs (Protocorm-like bodies) in Cymbidium. For this, pCAMBIA3301 vector which carries a herbicide-resistant bar gene and gus gene as a reporter gene was used for transformation with Cymbidium cultivars 'Youngflower ${\times}$ masako' line. To select transformants, proper concentration of herbicide, PPT (phosphinotricin), should be determined. As a result, 5 mg/l of PPT was selected as a proper concentration. Further, proper conditions for particle bombardment were determined to obtain a high frequency of transformation. Results showed that 1.0 ${\mu}g$ of DNA concentration, 1,100 and 1,350 psi for helium gas pressure, 1.0 ${\mu}m$ of gold particle and 6 cm of target distance showed the best result for the particle bombardment experiment. Also, pre-treatment with combination 0.2 M sorbitol and 0.2 M mannitol for 4 hrs prior to genetic transformation increased the transformation efficiency up to 2.5 times. Using transformation system developed in this study, 3.2 ~ 4.0 transgenic cymbidium plants can be produced from 100 bombarded PLBs on average. Putative transgenic plants produced in this system confirmed the presence of the bar gene by PCR analysis. Also, leaves from randomely selected five transgenic lines were applied for Basta solution (0.5% v/v) to check the resistance to the PPT herbicide. As a result, three of them showed resistance and one of them showed the strongest resistance with the maintenance of green color as non-transformed plants showed. Using this established transformation system, more genes of interests can be introduced into Cymbidium plants by genetic transformation in the future.

Genetic Analysis of Quantitative Trait for Flowering Habits by Diallel Crosses in Sesame (참깨의 이면교잡에 의한 유한형 양적형질 유전분석)

  • The Gene action for eight quantitative characters related to the plant type was estimated using diallel cross among three different plant types of sesame (Sesamum indicum L.) in 2001. The parental varieties used for diallel cross were Ahnnam and Yangbaeck as indeterminate type, ksan22 as semi-determinate type, and dt-45 and Suwon131 as determinate type. In variance and covariance analysis (Wr-Vr) for eight characters the mean square of array except for capsule length, 1,000 seed weight were significant, which suggest that varieties involved in diallel set cross may have epitasis. Complete dominance was observed in the flowering periods, and ratio of matured grains while partial dominance was observed in the plant height, no. of capsules per plant, and no. of branch per plant. Broad sense heritability for flowering periods, plant height, no. of capsules per plant, and no. of branch per plant ranged from 0.91 to 0.99. Narrow sense heritability for flowering periods, capsule length, ratio of matured grain, 1,000 seed eight were 0.18 to 0.34, and plant height, no. of capsules per plant and no. of branch per plant were ranged 0.77 to 0.81.

Growth and Yield of Atractylodes japonica Koidz. Affected by Mulching Methods (멀칭 방법이 삽주의 생육 및 수량에 미치는 영향)

  • Plant growth should be affected by mulching method. The study was done to examine the effect of different mulching methods on growth and yield of Atractylodes japonica Koidz. The experiment was done by six different mulching methods; no mulching, rice straw, transparent polyethylene (PE), black PE, two combined treatments of rice hull and transparent PE or black PE. Green shoot and related characters, growth and yield of early and late stages were measured on May 8, June 8 and October 26, respectively. Green shoot fresh weight was the highest in transparent PE mulching, while number of leaves, shoot and stem lengths were the greatest in rice straw mulching of the methods. In its early stage, rice straw mulching had the fewest number of leaves, roots, stems, the smallest width of leaves, and the least fresh weights of shoots and rhizomes. Combined mulching of black PE and rice hull showed the highest values in root-related characters; number of roots, fresh weights of rhizomes and roots. In its late stage, the treatment effects shown in the early stage disappeared owing to high temperature of mid-summer so that there was no significant difference between the mulching treatments. Yield per unit area and marketable yield were the highest in rice straw mulching, and combined mulching of black PE and rice hull.

Effect of Sulfur Dioxide and Hydrogen Fluoride on Rice Plant Growth in Industrial Estate (공업단지 주변에서의 아황산가스 및 불화수소가 수도 생육에 미치는 영향)

  • The study was performed to investigate the effects of gaseous emission of sulfur dioxide and hydrogen fluoride on the growth of rice plant under stressed field conditions. This study is specifically dealt with multiple effects of sulfur dioxide and hydrogen fluoride on various plant growth indicators such as leaf damage, weight of grain, panicles per hill, spikelets per panicle and percent fertility. It appears that there is a good correlation between ambient concentrations of sulfur oxides and sulfur contents found in leaves with an average correlation coefficient of 0.868 within a 1% significance level. A better multiple correlation was found between percent leaf damage and sulfur and fluorine contents found in leaf with a significance of 1% level. The correlation coefficient ranges from 0.807 to 0.978 with an average being 0.922. An evaluation of data observed has demonstrated that both panicles per hill and percent fertility are significantly affected by air pollutants. As expected, hydrogen fluorides have more effects than sulfur oxides. It is, however, interesting to note that spikelets per panicle has slightly been affected while no indication of effects on l000-grain weight has been observed. This may lead to a conclusion that a reduction in yield of rice under polluted field conditions may have more been caused by the diminution panicles per hill and percent fertility rather than by the diminution of spikelets per panicle and grain weight.

Studies on Agronomical Characteristics of Rice Varieties Recommended during 1910-1980 in Korea (수도 품종변천에 따른 유용형질의 특성변이에 관한 연구)

  • Heading behaviour of native japonica (GI) and Japanese varieties (GII) grown during 1910-1920s were thermo-sensitive, home bred varieties during 1930s (GIII) - 1950s (GIV) were photo-sensitive and semi-dwarf indica derived varieties released since 1971 (GV) were controlled by basic vegetative phase. Number of leaves on main culm was increased with varietal improvement and culm length was gradually shortened as variety improved. GV variety had the shortest plant height among the groups. The first internode length was about 30cm in all groups and difference of 2nd-4th internode length was caused shortening the culm length. Panicle length and diameter of panicle neck were shortened and became thinner as variety developed from GI to GIV, however, GV had longer panicle and thick panicle neck, respectively. Number of panicles/hill and straw weight/hill increased and became heavier as variety improve from GI to GIV but GV had moderate number of panicle/hill and relatively light straw weight. Number of spikelet/panicle has been reduced with varietal improvement but GV had the biggest number. Grain yield/hill also increased as variety improved, however, it was decreased as transplanting season postponed and GV had shown the most sensitive difference. Top dry matter weight at heading stage and at 20 days after heading (20 DAH) had no difference among the groups except GI which was the smallest. Average grain weight at 20 DAH was heavier in the order of GI > GII, GV > GIV > GIII. It had a little variation in GI & GII while that of other variety group showed significant decrease as transplanting delayed. Harvest index (HI) of GV was the highest at 54% for the earlier transplanting and GIII had the lowest HI. Difference of HI became conspicuous for the later transplanting, the varieties bred later had the lower HI when transplanting of rice delayed. At the earliest transplanting applied (May 15), the grain yield was higher in the order of GI > GIV > GIII > GII > GI, however, that of Tongil type variety (GV) was the lowest for the latest transplanting (June 29). Interrelationship among the agronomical characteristics and grain yield was discussed.

Geographical Variations of Sargassum thunbergii Morphology in Korea (한국산 갈조식물 지충이의 지리적 형태변이)

  • In this study, we investigated the effect of geographical variations on Sargassum thunbergii morphology to make a taxonomic reconsideration about infraspecific taxa in this species. In order to examine the morphological characteristics of S. thunbergii, total 27 matured and morphologically intact thalli were collected from the east, west and south coast in spring 2011. Interestingly, it was observed that the species populations on the west coast were characterized by short, thin and coarse thallus, and soft texture. However, the populations on the east coast showed thicker thallus, larger leaf and vesicle, and tougher texture. Thallus height of S. thunbergii was found to be similar at both east and west coast. Further, the height of the thallus and lateral branch of the species populations residing south coast were highest whereas the size of leaf and vesicle are shortest, in comparison with east and west coast species. Although morphological characteristics of the north east coast populations corresponded to the original description of S. thunbergii f. latifolium, we could not find exact morphological features and diagnostic characters to distinguish form in S. thunbergii. These results indicated that it is not the optimal characteristics to identify infraspecific form in this species. In contrast, morphological variations may signify the adaptation of this species to local environmental factors. Thus, we recommend that intraspecific morphological variation of S. thunbergii should be carefully used to identify infraspecific taxa.

Bioconversion of Citron oil by Co-Culture of E. coli EC3, EC4, and EC6 (E. coli 형질전환주의 공동배양에 의한 유자정유의 생전환)

  • E. coli transformants EC3, EC4. and EC6. harboring citron oil degrading pathway genes, were co-cultured in M9 media with citron oil as a sole carbon source at 28$^{\circ}C$. Each co-culture(EC3+EC4, EC3+EC6, EC4+EC6 and EC3+EC4+EC6) showed three to four times higher cell growth than each transformant single culture. Microbial conversion products from the co-cultures were determined by GC-MS. Linalool. 4-terpineol and ${\alpha}$-terpineol were the major common products from co-cultures. Various minor products also were detected and important in flavor characteristics of cultures.
