Effects of Protox Herbicide Tolerance Rice Cultivation on Microbial Community in Paddy Soil (Protox 제초제저항성 벼 재배가 토양미생물 군집에 미치는 영향)

  • BACKGROUND: Rice (Oryza sativa) is the most important staple food of over half the world's population. This study was conducted to evaluate the possible impact of transgenic rice cultivation on the soil microbial community. METHODS AND RESULTS: Microorganisms were isolated from the rhizosphere of GM and non-GM rice cultivation soils. Microbial community was identified based on the culture-dependent and molecular biology methods. The total numbers of bacteria, fungi, and actinomycete in the rhizosphere soils cultivated with GM and non-GM rice were similar to each other, and there was no significant difference between GM and non-GM rice. Dominant bacterial phyla in the rhizosphere soils cultivated with GM and non-GM rice were Actinobacteria, Firmicutes, and Proteobacteria. The microbial communities in GM and non-GM rice cultivated soils were characterized using the denaturing gradient gel electrophoresis (DGGE). The DGGE profiles showed similar patterns, but didn't show significant difference to each other. DNAs were isolated from soils cultivating GM and non-GM rice and analyzed for persistence of inserted gene in the soil by using PCR. The PCR analysis revealed that there were no amplified protox gene in soil DNA. CONCLUSION(S): These data suggest that transgenic rice does not have a significant impact on soil microbial communities, although continued research may be necessary.

A Simulation Study to Investigate Climatic Controls on Net Primary Production (NPP) of a Rugged Forested Landscape in the Mid-Western Korean Peninsula (기복이 심한 한반도 중서부 산림경관에서 기후가 순일차생산(NPP)에 미치는 영향에 대한 모사연구)

  • We have investigated microclimatic controls on the spatiotemporal variations of net primary production (NPP) of a rugged forested watershed using the process-based biogeochemical model (BIOME-BGC). To validate the model simulation of water and carbon cycles at the plot scale, we have conducted field survey over deciduous broadleaf forest (DBF) and evergreen needleleaf forest (ENF) since 2000. The modeled values of soil temperature, soil moisture and soil respiration showed high correlation with those from the field measurements. The modeled seasonal changes of NPP showed high correlation with air temperature but no significant correlation with water related parameters. The precipitation frequency turned out to be the best climatic factor to explain the annual variation of NPP. Furthermore, NPP of ENF was more sensitive to precipitation frequency than that of DBF. With changes in vegetation cover and topography, the spatial distribution of NPP was of great heterogeneity, which was negatively correlated with the magnitude of NPP. Despite the annual precipitation of 1,400mm, NPP at the study site was constrained by the amount of water available for the vegetation. Such a modeling result should be verified by the field measurements.

Characteristics of the Newly Developed Rain Shelter for Grapevine and Growth of 'Campbell Early' (신개발 포도 비가림 시설의 특성과 '캠벌얼리'의 생육)

  • Fruit cracking and vine leaf spot of grapes tend to occur when the plants were directly exposed to rain under outdoor culture. Rain shelter facility can be an alternative method to prevent the cracking and disease of grape, but it also has some limitations in practical usages. We designed rain shelter facility which can completely shut out the rain and ventilate naturally, and it was upgraded to meet the standards of disaster prevention against snow and wind load. The newly developed rain shelter has two-story roof structure, and the $2^{nd}$ floor roof was equipped over $1^{st}$ floor roof at a distance of 40cm. For natural ventilation and water proof, the upper roof protruded about 50cm from the ridge of a $1^{st}$ floor roof. The various tests were carried to examine such as grape quality, brown spot and fruit cracking of Campbell Early under the conventional and the newly developed rain shelter facility which was built about $100{\ss}{\check{S}}$. In comparison of temperature between the conventional and the newly developed rain shelter facility when outside temperature was more than $34^{\circ}C$, the inside temperature was recorded as $40.7^{\circ}C$ and $37.4^{\circ}C$, respectively. There was no significant difference between the two facilities when outside was below $32^{\circ}C$ The quality such as soluble solids and marketable fruit was increased, and fruit cracking of grapes and vine leaf spot also drastically diminished in the newly developed rain shelter.

The Change Analysis of Plant Diversity in Protected Horticulture of Agricultural Ecosystems (시설원예단지 조성이 농업생태계의 식생다양성에 미치는 영향 분석)

  • Although the ecosystem extends numerous functions for the benefit of humankind, construction of horticultural facilities can potentially lead to the degeneration of some of these functions owing to the expansion of impervious regions and loss of habitats. Thus, this study aimed to examine the effect of construction of horticultural facilities on plant biodiversity. Analysis of the vegetation distribution characteristics showed that horticultural facilities had significantly lower plant diversity than did rice paddies. Hence, the proposed approach involved low-impact development, arrangement of habitat space, plant preservation, restricted use of chemical fertilizer, habitat creation, and reduced preservation cost. Lifestyle analysis suggested the importance of developing favorable environments for the growth of annual plants and aquatic plants. In all, 20 species of naturalized plants belonging to 10 families were identified. Furthermore, the proportion of these naturalized plants was higher in glass greenhouses and multi-span greenhouses, suggesting the advantages of employing natural soil mulching while avoiding concrete mulching. Statistical analysis was performed to validate the results, which suggested that impervious regions be converted to natural soils. Collectively, the findings of this study are expected to be used for establishing policies for the construction of eco-friendly and ecological horticultural facilities; this may aid the maintenance of sustainable agricultural landscapes and large-scale development of the reclaimed lands.

Distribution of Cadmium, Copper, Lead, and Zinc in Paddy Soils around an Old zinc Mine (가학광산 주변 논토양의 카드뮴, 구리, 납 및 아연 함량 분포)

  • This study was carried out to provide information for establishing counter measures of soil pollution through analysis of Cd. Cu, Pb, and Zn in paddy soils and brown rice. Cadmium, Cu, Pb, and Zn contents in soils were analyzed and distribution maps for these heavy metals were prepared. Heavy metal contents in brown rice were also measured. Average contents of Cd, Cu, Pb, and Zn in surface paddy soils extracted with 0.1 N HCl were 7.4, 35.8, 98.9, and $118.8mg\;kg^{-1}$, respectively. These were 9 times (Cu) to 50 times (Cd) higher than the background level of heavy metals in unpolluted paddy soils in Korea. The contents of Pb and Zn were lower than those measured in 1980, whereas Cd content did not decrease. The levels of heavy metal contamination in paddy soils may not affect growth or yield of rice plant, however, Cd contents indicated a level of serious concern to humans. The average contents of Cd, Cu, Pb, and Zn in brown rice were 0.38, 2.38, 1.31 and $22.31mg\;kg^{-1}$, respectively.

Community Structure and Species Composition of Pinus densiflora for. erecta Forest in Mt. Cheonchuk (천축산 일대 금강소나무림의 군집구조 및 종조성)

  • This study was conducted to analyze community structure and species composition of Pinus densiflora for. erecta Stand in Mt. Cheonchuk (653 m). Field survey was carried out from June to September in 2013. 74 plots (20×20 m) were set up, 5 herb layer plots (3×3 m) were constructed for each plot, and there, Diameter at Breast Heigh t(DBH), height, environmental factor, annual growth were measured. Vascular plants were surveyed as following; 66 family, 165 genus, 211 species, 2 sub species, 29 variety, 6 form 248 taxa. Results of cluster analysis for P. densiflora for. erecta forest, 3 communities were divided into; Quercus mongolica (P-1), Quercus variabilis (P-2) and Quercus aliena-Stephanandra incisa (P-3). There were significant environmental factors that organic layer, annual growth, CEC, total total nitrogen, organic matter and pH for each community. As a result of DCA, P-1 and P-2 were distributed large range of environmental factors but relatively limited in P-3. Distributions of herb layer were affected by sand, cation exchange capacity, silt and total nitrogen. Results of MRPP test for herb layer communities, it was significantly analyzed (A=0.003, P<0.008). Species diversity index was highly recorded in P-3 and influenced by cation exchange capacity, total nitrogen, annual growth in consequence of NMS analysis.

Breeding System and Allozyme Genetic Diversity of Deutzia paniculata Nakai, an Endemic Shrub in Korea (고유종 꼬리말발도리의 생식특성과 동위효소 유전다양성)

  • Deutzia paniculata is an endemic species, which is geographically restricted within southern part of Korea. Four populations of D. paniculata were sampled across its natural range, from the smallest population, Mt. Dalum, which held less than 100 individuals, to the largest, Mt. Unmum, over 3,500 individuals. Artificial pollination study showed that D. paniculata had an obligate outcross breeding system. Major pollinators were two bee species, Lasioglossum exiliceps and Allograpta balteata (de Geer). The breeding system and patterns of allozyme variation of D. paniculata were investigated to understand the population biology and to explain on reserve designs and management proposals relevant to this species. D. paniculata held relatively low genetic variation at the eight allozyme loci surveyed. Measures of genetic variation in this species alleles per locus ($A_s=1.33$), proportion of polymorphic loci (P=23.85%), and expected heterozygosity ($H_{es}=0.110$) were similar to values reported for endemic species. Mt. Dalum population (DAL) was composed with one clone based on allozyme data. Individuals of D. paniculata were frequently included in root connected clusters. Population genetic structure between and within four populations was probably the result of shrinking effective population size and the extinctions of intervening populations. For the conservation of genetic diversity, maximum number of different genotype need to be protected based on genetic structure and mating system.

Information Requirements, IS Planning Typology, and IS Planning Effectiveness : An Empirical Study (정보요구, 정보시스템 기획유형, 그리고 정보시스템 기획 효과성에 관한 실증적 연구)

  • 정보시스템 기획(IS planning)은 경영환경의 변화로부터 야기되는 조직의 정보요구 를 제대로 파악함으로써 새로운 정보시스템의 구축이나 실행과정에 반영하는 중요한 과업중 의 하나이다. 하지만, 아직까지 정보요구를 반영한 정보시스템 기획유형에 관한 연구나 정 보시스템 기획의 효과성에 관한 영향에 대한 실증적인 연구가 없었다. 그리하여, 본 연구에 서는 기업의 경쟁력 강화 목표를 달성하기 위해 필요한 정보시스템의 효율적인 관리와 활용 을 위하여 정보시스템 기획에 관한 상황 이론적 접근을 시도하였다. 본 연구는 과업환경을 중심으로 발생하는 정보요구의 유형에 따라 정보시스템 계획을 어떻게 추진하는 것이 바람 직한 것인지를 파악하고, 정보시스템을 기획 시 정보요구 수준과의 적합성(fitness)에 따라 정보시스템 기획의 효과성은 어떻게 달라지는 지를 파악하고자 한다. 이를 위하여 본 연구 에서 사용된 Construct와 변수들은 기존의 정보시스템 기획관련 문헌을 이용하였다. 본 연 구를 위하여 국내 매출액 1000대 기업을 대상으로 설문지를 우송하였으며, 그 중 187부가 회수되었다. 자료분석의 결과, 정보요구 수준과 정보시스템 기획유형간의 관련성은 긍정적 으로 관련되어 있었으며, 정보요구 수준에 따른 기획의 효과성에는 차이가 없었으나, 정보 시스템 기획유형에 따른 기획의 효과성은 유의한 차이가 있었다. 특히, 정보요구 수준과 정 보시스템 기획유형간의 적합성에 의한 정보시스템 기획의 효과성에 대한 t-test결과는 적합 한 조직(fit organization)보다는 적합하지 않은 조직(unfit organization)의 성과가 더 높은 것으로 나타났다.성 형질전환 계통의 식물체 내에서의 바이러스 증식 및 이동과 관련된 저항성 기작, 여러 가지 PVY 계통에 대한 저항성 유무, 수량, 생육 특성 및 주요 화학 성분 함량 등을 발표하고자 한다.-glucose로 구성된 다당류 이었다. 아미노산은 Asp 및 Glu의 산성 아미노산과 Ala, Leu 등의 함량이 높게 나타났으며, 비알칼리 추출물에서 Ser과 Thr의 함량이 높게 나타났다. 다당류 T-AS는 평균 분자량이 2,000 kD와 12kD에서 주 peak를 나타냈으며, 수용성 분획의 평균 분자량은 12kD이고 비수용성 분획은 36~2,000 kD의 평균 분자량 분포를 갖는 것으로 나타났다. IR과 NMR 분석 결과 890 cm-1에서 흡수 peak를 나타내어 $\beta$-(1,3)0glucan과 $\beta$-(1,6)-glucan의 구조를 갖는 다당류로 확인 되었다. T-AS 분획은 C:H:O:N의 함량비가 38.9:5.7:49.6:1.84%이며, 이 물질의 융점은 163 $^{\circ}C$로 연한 갈색을 나타낸다. 분리된 GLG의 항암활성 기전 규명을 위해, in vivo 항암실험, 항보체 활성능, 항체 생성능, serum protein 분비능, 대식세포의 탐식능과 활성능 및 세포간 물질 분비 등의 상관관계를 조사하였다. 다당류 GLG 분획물들 가운데 항보체의 활성이 높았던 분획은 sarcome 180에 대한 항암 활성이 높게 나타났다. 다당류 T-AS의 보체 활성화 기작은 classical과 alternative complement pathway의 양 경로를 통해 활성화 되었다. Comparative Analysis to Damage Reduction of Host Plant by Applying a Mating Disruptor of the Oriental Fruit Moth, Grapholita molesta in Two Different Cultivation Environments of Apple Orchard (재배환경이 상이한 사과원에서 복숭아순나방(Grapholita molesta) 교미교란제 처리에 의한 기주 피해 억제 효과 비교)

  • This study demonstrates a variability in efficacy of mating disruption against the oriental fruit moth, Grapholita molesta, populations infesting apples cultivated in differential environmental conditions. Throughout the growing seasons, trap catches of G. molesta male moths and damaged leaf and fruit were examined to evaluate the efficacy of a commercialized mating disruptor. Treatment of the mating disruptor significantly reduced the trap catches of male moth in treated plot, compared to those of the untreated plot. This reduced trap catches were significantly correlated with leaf and fruit damage. Moreover, the highest host damage occurred in June just after the highest overwintering adult peaks in both plots. The treatment of mating disruptors in Chungsong effectively disrupted the overwintering population in April and May, resulting in no noticeable host damage during the following growing seasons. However, there was a marked difference in host damage between two plots, especially in late seasons. Variation in the efficacy of mating disruption technology in terms of host damage appeared to be related with nearby pheromone-untreated orchards, which may result in the immigration of gravid females.

Analysis of Environment Factors in Pleurotus eryngii Cultivation House (새송이버섯 재배사의 환경요인 분석)

  • Pleurotus eryngii(King oyster) is one of the most promising mushrooms produced on the domestic farms. The quality as well as quantity of Eryngii is sensitively affected by micro climate factors such as temperature, relative humidity, $CO_2$ concentration, and light intensity. To safely produce high-quality Eryngii all the year round, it is required that the environmental factors be carefully controlled by well designed structures equipped with various facilities and control systems. At the commercial mushroom cultivation house(A,B), this study was carried out to find out reasonable range of each environmental factor and yield together with economic and safe structures influencing on the optimal productivity of Eryngii. this experiment was conducted for about two-month from Nov. 11, 2002 to Dec. 30, 2002 in Eryngii. cultivation house-A, B. Ambient temperature during the experiment period was not predominantly different from that of a normal year. The capacity of the hot water boiler and the piping systems were not enough. Maximum air temperature difference between the upper and lower growth stage during a heating time zone was about 2~3$^{\circ}C$. The max. and min. relative humidity were ranged approximately 60~100%, and average relative humidity was ranged approximately 80~100%. And $CO_2$concentration increased until maximum 1,600~1,800 ppm with the passing growing period. The illuminance in cultivation house was widely distributed from 20lx to 160 lx in accordance with position, and it was maintained lower than the recommended illuminance range 100~200 lx. The average yield per bottle was about 67~85g. But the optimal productivity will be evaluated by considering the quality and quantity of mushroom production, energy requirements, facility construction and management cost, etc.
