Distribution and Physiological Characteristics of Yeasts in Traditional and Commercial Kochujang (재래식 및 개량식 고추장 효모의 분포 및 생리특성)

  • To investigate the yeast florae in the traditional and commercial Kochujang, computer identification systems, Vitek, API kit and conventional identification methods were used. Yeast florae of each process were compared and their typical physiological characteristics were also tested. Various process intervals yielded 330 colonies, which resulted in 11 species 184 strains classified. They were identified into Candida glabrata C. guilliermondii. C. humicola. C. rugosa, C. zeylanoides, Cryptococcus uniguttulatus, Pichia farinosa, Rhodotorula glutinis, Saccharomyces cerevisiae and Zygosaccharomyces rouxii. The strains of Candida, Pichia, Saccharomyces and Zygosaccharomyces were existing in both processes. In case of commercial process, the maximum distribution of Z. rouxii and S. cerevisiae were 33% at 15 day fermentation and 13% at 21 day, respectively. The distribution of Candida spp. was gradually decreased throughtout the fermentation period from 40% to 10%. In the traditional process, the maximum distribution of Z. rouxii and S. cerevisiae were 53% after 3 months and 26% after 7 months, respectively, S. cerevisiae and Z. rouxii showed distintive growth pattern at the high concentration of glucose and sodium chloride and played important roles in both processes of fermentation. Physiological tests revealed that only two major yeasts. S. cerevisiae and Z. rouxii, showed vigorous carbon dioxide formation under the tested conditions.

The Preparation of Yogurt from Egg White Powder and Milk Products (난백분말과 유제품을 이용한 요구르트의 제조)

  • Gel-type yogurt was prepared from egg white powder (3%, w/v), glucose (2%, w/v) and four kinds of milk products (4%, w/v). The effects of milk product on acid production and growth of Lactobacillus were studied. The effects of milk product on sensory property and volatile aroma compounds were also studied. Acid production by L. acidophilus at 24 hr in samples containing milk product was significantly lower than that by L. acidophilus in milk (p<0.05). The sample containing casein produced less acid than the other samples. Number of viable cells of L. acidophilus at 24 hr in milk and samples containing milk product was $2.0{\times}10^{9}/mL$ and $5.0{\times}10^{8}{\sim}8.0{\times}10^{8}/mL$, respectively. Sensory property of the samples containing milk product was lower than that of milk yogurt (reference). However, sensory property of the sample containing casein was not significantly different from that of milk yogurt (p<0.05). The sample containing whey powder showed lower sensory score than other samples. Though the composition of volatile aroma compounds was slightly different from sample to sample, gas chromatographic analysis detected acetone, ethanol, diacetyl and acetoin in samples fermented by L. acidophilus.

  • This experiment was conducted to determine the responses of some pine species to acidity of cultured soils, and absorption behavior of alkaline elements and to observe the growth characteristics during afforesting period. The results were summarized as follows. 1. Total dry weight indicating growth volume was not much influenced by the acidity of cultured soils (pH 4~8) in all of the 4 species studied. Pinus koraiensis showed more dry weight than any other species, which indicated statistically significant difference. 2. T-R ratio of dry weight was not much affected by the acidity of cultured soils (pH 4~8) and the difference of cultured soils (pH 4~8) and by species. Growth phase also did not show remarkable change through the experiment period. 3. The average sodium content ratio in the plant body had no correlation with cultured soils (pH 4~8), bue significant differences were recognized among species statistically. 4. It was found that sodium content ratio was not much influenced by the cultured soils (pH 4~8), even when the tops and roots were separately analyzed. But, very significant difference was found among the species. Pinus koraiensis especially, showed greater volume growth maintaining low sodium content ratio than any other species, which indicates that Pinus koraiensis has unfavorable characteristics for growing on the seaside.

Quality Characteristics of Sikhae Made with Monascus Anka Rice (홍국쌀로 제조한 식혜의 품질특성에 관한 연구)

  • This study investigates the quality characteristics of Sikhye, a Korean traditional drink, made with different amounts of Monascus anka rice, or red yeast rice, and the results are as follows. During the saccharification process of red yeast rice, there was not significant change in pH, and its sugar content increased. It revealed that the optimal hours of saccharification for making Sikhye were four or five hours, which showed the highest in $^{\circ}Brix$. PH decreased significantly with increasing amounts of red yeast rice, and there was no change in sugar content and reducing sugar. Chromaticity L values decreased with increasing amounts of red yeast rice, and a value, b value and the turbidity increased. DPPH free radical scavenging activity of the samples showed significant differences in the samples with high scores as the amounts of red yeast rice increased. As a result of measuring the total microorganism number of the sample stored at $4^{\circ}C$, storability was improved with increasing amounts of red yeast rice. Preference was high in the order of 40%, 50%, 30%, 20%, 0%, and 10%, and the optimal amount of red yeast rice was less than 40-50%. Adding red yeast rice showed high scores in the sensory test, showing red color in Sikhye. In addition, its antioxidative activity effect and microbial growth inhibitory activity were considered to improve storability and preference.

Effects of UV-B Radiation and Water Stress on Hardening Phase Growth of Container-Grown Betula platyphylla Seedlings (자작나무 콘테이너묘(苗)의 경화단계(硬化段階) 생장(生長)에 미치는 UV-B 와 수분(水分)스트레스의 효과(效果))

  • This study was carried out to investigate the possibility of supplemental UV-B application to the hardening phase of container-grown Betula platyphylla seedlings. The containerized seedlings were grown in a growth chamber for four months and then treated with UV-B(UV-$B_{BE}$ $3.2KJ\;m^{-2}\;day^{-1}$ and $5.2KJ\;m^{-2}\;day^{-1}$) radiation and water stress regime(irrigation in one week interval) for four weeks. The differences in growth and physiological responses of the seedlings before and after the treatments were analyzed. UV-B radiation and water stress reduced height growth and leaf dry mass accumulation of the seedlings. The root collar diameter growth was reduced by UV-B radiation but increased by water stress. The reduction in leaf dry weight by UV-B radiation and water stress reduced T/R ratio of the seedling. The reduction in T/R ratio was the most apparent by water stress. Chlorophyll index observed by a chlorophyll meter was the lowest in the $5.2KJ\;m^{-2}\;day^{-1}$ of UV-B radiation, and those in the $3.2KJ\;m^{-2}\;day^{-1}$ and water stress were similar. UV-B radiation and water stress reduced both water content in the seedlings and leaf water potential, and increased leaf osmatic pressure. The water content of leaves and shoots was reduced more rapidly by UV-B radiation than by water stress treatment. In conclusion, growth responses and physiological changes in water relation by supplemental UV-B radiation which was applied to the hardening phase of container-grown Betula platyphylla seedlings were similar results to the water stress treatment.

New Columnar Apple Variety 'Tinkerbell' for Pollenizer (사과 무측지성 수분수용 신품종 '팅커벨')

  • This research was performed to breed new columnar-typed apple variety for pollinator in apple orchard. We cross-fertilized 'Hongro' that was an ovary parent and 'Maypole' a pollen parent in 2003. We gathered seeds in 2003 and sowed after breaking of dormancy in 2004. Seedlings was been growth and development till flowering. After fruit setting, We investigated characteristics of growth and fruit including mating compatibility from 2010 to 2012, then finally selected '03-2-95' named 'Tinkerbell' in 2012, registered as new variety on Korea Seed & Variety Service in 2015. The full bloom stage of 'Tinkerbell' was later 2 days then of 'Maypole'. Tree habit of 'Tinkerbell' was alike to of 'Maypole', for example weak tree vigor, columnar type. But there were many different characteristics between 'Tinkerbell' and 'Maypole'. Fruit shape of 'Tinkerbell' was ovoid whereas of 'Maypole' was ellipsoid. Fruit of 'Tinkerbell' was bigger, smoother skin, no waxy bloom than of 'Maypole'. Fruit of 'Tinkerbell' was more delicious than of 'Maypole' because of having higher soluble sugar content and lower titratable acidity. 'Tinkerbell' was excellent pollinator in apple orchard. Because of having high fruit setting rate with major apple varieties and a narrow tree width that was advantageous as pollinator in apple high density system.

The Analysis of Forest Successional Trend by Species Replacement Model in the Natural Forest (천연림의 수종 대치 작용 모델에 의한 산림천이 경향 분석)

  • The successional status and potential natural vegetation were examined in the natural deciduous forest in Mt. Chombong area. The examination was based on the subsequent process of generation replacement by understory saplings for the dominant canopy trees within 106 20mx20m square sample plots. The transition matrix model, which was modified from mathematical theory of Markov chain, was employed to analyze the successional status of the study forest. The model suggests that study forest is still seral, and it is considered to be more than 500 years away from the steady state or climax in terms of species composition. The simulations predict a remarkable decrease in the proportion of species composition of the present dominant Quercus mongolica and Kalopanax pictus from current 42.6% and 8.1% to less than 13.3% and 0.5%, respectively, at the steady state. On the contrary, the proportions of Abies holophylla, Acer mono, Fraxinus mandshurica, Tilia amurensis, and Acer pseudo-sieboldianum will increase at the steady state. The change of predicted composition ratio was generally coincide with the result of tolerance index to be compared with the study model. The hypothesis and sensitivity of the model were also discussed to evaluate the applicability to the real situation. The overall results indicated that the present dynamics of the forest must reflect the seral state due to previous disturbance mainly by human related interference.

STUDIES ON THE IDENTIFICATION OF Pleioblastus Simoni IN KOREAN BAMBOOS (Pleioblastus Sinomi Nakai (해장죽)의 동정의 관한 연구)

  • Studies on the anatomical observation of Bamboos distributed in East-South Asia for systematic botany were scarecely reported except for the morphological studies on Bamboos, which was investigated by Dr. Hiroshi Usui, present author and others. Recently it has been certain that it could be hardly classify some species of bamboo family without this work. For this reason the author reported the anatomical studies on Korean bamboos in 1959, and furthermore the author intended to identify the two species of Pleioblastum Simoni Nakai in Korean bamboos by studying the characteristics of internal structure and qualitative assay of free amino acid containg in Pleioblastus Simoni Nakai(1), (2). In view of fact that there is some differences between two species, it is likely to be identifiable completely different species one another. Furthermore, a lot of characteristics surveyed by the author were as follow: It was reassumed that two species cultivated under same conditions for three years were different alternately in the species of Pleioblastus Simoni Nakai. At the same time, these studies could clarify some evolutional processes of bamboo family. On the other hand, it was found that two species had not only the obvious difference in aerenchyma, the size of thickwalled parenchyma and bulliform cell, but also the internode of Pleioblastus Simoni Nakai(1) is longer than the other, the speed of growing is more rapid, the leaf of former is narrow and that of latter is wide. The free amino acids containg in each species were found as 18 kinds, especially Pleioblastus Simoni Nakai (2) had not Histidine in spite of containg in Pleioblastus Simoni Nakia(1). From the characteristics and the experiments described above, it seemed to the author that Pleioblasus Simoni Nakai (1) which was growing at the region of Ulsan in Korea.

Fermentation of Kanjang, Korean Soy Sauce, in Porosity-Controlled Earthenwares with Changing the Mixing Ratio of Raw Soils (흙배합비를 달리하여 기공율이 조절된 담금용기 항아리에서의 간장 발효)

  • This study focuses on investigating the effect of porosity-controlled earthenware on fermentation of kanjang, Korean soy sauce. Porosity of fermentation vessel was controlled by changing the mixing ratio of raw soils at manufacturing earthenware. Earthenwares contented 0%, 40% and 60% of the mixture of red brown soil and powdered soil (1 : 1), respectively. The more contents of the mixed soil, the more porosity in earthenware. During fermentation of kanjang in porosity-controlled earthenwares at $30^{\circ}C$ for 4 months, physical, chemical, microbiological and sensory quality attributes were monitored. Compared to other containers, kanjang in the earthenware that had 0% mixed soil showed less water loss, salt content and pH. It also produced higher total acidity, protease activity, total nucleotide, and microbiological changes which included total aerobic bacteria, lactic acid bacteria and yeast. Total nitrogen and free amino acids in kanjang did not show the consistency with the mixed soil contents of fermentation containers, which may take more effect of other factors as water loss than the porosity of vessels. However, the percentage of glutamic acid in total free amino acids was a little higher in the earthenware that had 0% of mixed soil than other containers. These positive physicochemical, microbiological changes also resulted in higher sensory quality.

Effects of Carbon Substrates on Exopolysaccharide Production by Enterobacter sp. (Enterobacter sp. 의 다당 생산에 미치는 탄소원 기질의 영향)

  • The effects of carbon sources for exopolysaccharide production during batch cultivation of an Enterobacter sp. isolated from the composter were investigated. The highest amount of exopolysaccharide was obtained when lactose was used as carbon source. Lactose in medium was converted into glucose and galactose. Glucose was metabolized fast and was completely consumed, but about $20\%$ of lactose was accumulated as galactose. On the other hand, enzyme activity was about $350\~450$ unit with the increase of lactose concentration. Thus, it was considered that the exopolysaccharide might be produced in the course of that lactose was hydrolyzed into glucose and galactose by $\beta-galactosidase$ with respect to that enzyme activity on lactose hydrolysis was accorded to the exopolysaccharide production. When glucose and galactose were added to lactose medium, respectively, it could be considered that glucose was as a repressor and galactose was as a inducer for $\beta-galactosidase$ synthesis even though the mechanisms were not elucidated. The increase of lactose concentration was almost ineffective to the specific growth rate $(0.133\~0.151\;hr^[-1})$ but showed the difference in the biomass content. The higher carbon source concentration, the more residual sugar remained. It was assumed that the optimum lactose concentration for exopolysaccharide production was $30\~70g/L.$ On the other hand, it was considered that the nitrogen acted as growth limiting nutrients to the cell growth. In the cases of 30 and 70 g/L of the fixed carbon concentrations, the increase of the nitrogen sources concentration caused a remarkable increase within the range of $0.059\~0.225\;hr^{-1}$ and $0.141\~0.237hr^{-1}$ of the specific growth rate, respectively, while there was no significant difference in biomass.
