1,3-Propanediol Fermentation using the by-Products from Fat Industry (글리세롤을 함유한 유지산업 부산물의 1,3-propanediol 발효)

  • 1,3-Propanediol as a bifunctional organic compound could be used in polymerization reactions producing polyesters and polyurethanes. Byproduct containing high concentration of glycerol from fat industry was used to produce 1,3-propanediol in lower production cost as well as waste treatment. In this study, various attempts were made to increase 1,3-propanediol production under different conditions using Klebsiella pneumoniae ATCC 15380. The conversion yield and byproduct formation were influenced significantly by the fermentation pH and temperature. The optimal glycerol and nitrogen concentration for 1,3-propanediol production were found to be 25 a/L and 1%(w/v), respectively. The formation of 1,3-propanediol was optimal at pH 6.0 and temperature $35^{\circ}C$. 1,3-Propanediol production from byproduct from 2.5% glycerol was lower than that of 2.5% commercial glycerol and amounted only to 9.84 a/L from byproduct, while to 12.13 a/L from commercial glycerol.

  • 농산 부산물인 볏짚을 이용하여 수송연료인 휘발유를 대체할 수 있는 바이오에탄올을 생산하기 위한 전단계인 전처리와 당화에 대한 연구를 수행하였다. 볏짚은 농산 부산물 중에서 비교적 발생량이 많으며, 목질계 바이오매스에 비해 소프트하기 때문에 전처리가 용이하다는 장점이 있다. 전처리 방법으로는 여러 가지가 있으나, 본 연구에서는 증기폭쇄에 의한 전처리 방법을 채택하였다. 당화실험은 생물학적 방법인 효소당화를 하였다. 전처리 실험결과 온도가 낮고 처리 시간이 짧을수록 고형물 및 삼성분에 대한 수율이 높은 것으로 나타났다. 삼성분 중의 Glucan 수율은 $180^{\circ}C$ 실험에서 68-71%를 보였으며, $200^{\circ}C$ 실험에서는 59-64%를 보였다. Xylan은 거의 대부분이 소실되어 Xylan을 회수하는 전처리로써는 적합하지 않은 방법으로 나타났다. 반면 Lignin은 거의 제거가 되지 않았다. 당화 실험결과 $180^{\circ}C$에서 전처리한 폭쇄재는 당화 수율이 50%에도 미치지 못하였으나, $200^{\circ}C$에서 전처리한 폭쇄재는 70% 내외를 보여 주었다. 특히 분쇄 세척 멸균한 효소당화 수율은 90%가 넘는 높은 수율을 보였다. 전처리 수율과 당화수율을 종합하여 수율을 분석한 결과 $200^{\circ}C$ 전처리 실험조건에서 효소당화시 미처리 시료와 분쇄 세척 시료보다 멸균까지 실시한 조건에서 Glucan의 종합수율이 55% 내외로 월등히 높은 것으로 나타났다. 증기폭쇄에 의한 볏짚의 전처리는 Glucan의 수율이 낮아 손실이 많은 것으로 나타났으며, 이를 보완할 수 있는 전처리 조건 혹은 다른 전처리 방안이 요구된다.

  • 지난 30여 년간 반도체 제조 공정 중 FAB공정에서 칩 수율 모델의 개발과 적용은 반도체생산 계획 및 조업 관리를 위해 반도체 제조사들에게는 중요한 관리 대상이 되어 왔으며 제조업체들은 다양한 수율 모델들을 각 업체의 조건에 맞게 채택, 적용하여 왔다. 집적 기술의 발전은 반도체 칩의 크기에도 변화를 가져와 웨이퍼상의 결점들이 형성하는 클러스터를 설명할 수 있어야 했으며 칩 면적의 증가는 새로운 수율 모델을 개발케 하였다. 본 논문은 반도체 제조 공정에 대한 고찰과 수율 계산에 영향을 미치는 결점의 클러스터 효과 및 결점 크기를 중심으로 하는 치명 확률에 대하여 살펴보고, 포아송 모델에서 파생된 대표적인 칩 수율 모델들에 대한 설명과 칩 면적의 변화에 따른 각 모델별 수율 계산 비교 및 반도체 수율의 이용에 대하여 기술한다.

In-vitro Production of Glutathione Using Yeast ATP Regeneration System and Recombinant Synthetic Enzymes from Escherichia coli. (효모의 ATP 재생산계와 대장균 유래의 재조합 생산효소를 이용한 in vitro 글루타치온 생산)

  • An ATP regeneration system was used for the production of glutathione which was synthesized by a sequential action of ${\gamma}$-glutamyl-cysteine synthetase and glutathione synthetase. The synthetases above were produced in the recombinant E. coli (TG1/pDG7) with the highest specific production yield of 31 mg glutathione/g wet cell. Bakers yeast was considered to have economically a better ATP regeneration system although the glutathione production yield was lower than that of acetate kinase. It was also observed that the ATP regeneration system of bakers yeast was superior to that of Saccharomyces cerevisiae ATCC24858. The yield of glutathione production with bakers yeast was 36% with the ATP concentration of 5 mM. To avoid the cysteine limitation during the early phase of glutatione production, an extra cysteine was added at 2 hours after reaction and the production yield increased 1.91 times. The effectiveness of bakers yeast as an ATP regeneration system was proved by several sets of extra feeding experiments. The product inhibition by glutathione above 14 mM was also observed.

Citric Acid Production Using Encapsulated Aspergillus niger (Aspergillus niger 고정화 캡슐을 이용한 구연산 생산 특성)

  • The encapsulatpd A. niger grew up inside the capsule and mycelia penetrated through the pore of the capsule membrane. The mycelia on the capsule wall became loose when the carbon source and oxygen were deficient in the medium. On the contrary, the production rate increased and mycelia made a lump tightly when the carbon source and oxygen were sufficient. Namely, number of proper capsule of unit volume in the medium was existed. The phenomenon which was swelled of capsule membrane in cultivation could prevented by adding CaCl2 into the medium. According to the time adding CaCl2 into the medium, the production rate of citric acid was influenced. In case of adding CaCl2 into the medium at 7th day cultivation, the production yield of citric acid was increased about 40 percent higher than that of adding CaCl2 initially. The production yield of citric acid using encapsulated A. niger of flask culture was influenced with oxygen supply. The production yield of citric acid ($\Delta$p/$\Delta$s) of the flask culture was increased 3.88 time by using T-flask instead of parafilm sealed flask. Therefore, the productivity and consumption rate concerning production which was taken carbon source were increased when oxygen supply was sufficient. The production of citric acid using encapsulated A. niger was increased average 30 percent higher than that of bead in between 6th and 13th day cultivation.

  • Two stage fermentations of glucose/xylose mixture which is similar composition with rice straw hemicellulose hydrolysate were performed by Candida mogii ATCC 18364. In first stage, glucose was consumed rapidly for cell growth in aerobic condition (2 vvm, 300 rpm), then D-xylose was used for xylitol production in semi-aerobic condition (1 vvm, 300 rpm). After 4 days of fermentation, about $24\;g/{\ell}$ xylitol was produced with a yield of 0.58 g/g and volumetric productivity of $0.25\;g/{\ell}{\cdot}h$. To improve the xylitol yield by reduction of xylose consumption for cell growth and maintenance, D-glucose was continuously supplemented during the second stage of fermentation. By D-glucose feeding of $6.8\;g/{\ell}{\cdot}$ day, xylitol was produced up to $29\;g/{\ell}$ with a yield of 0.8 g/g and volumetric productivity $0.30\;g/{\ell}{\cdot}h$ which are 1.2-1.3 times higher than those obtained without D-glucose feeding.

  • A serum-free media for CRIP/MFG-LacZ retrovirus production was developed and applied to continuous packed-bed culture system. The serum-free media developed by fractional factorial design contains indulin ($10\mug/mL$), transferrin ($5\mug/mL$), BSA (4 mg/mL), EGF (25 ng/mL), and linoleic acid ($10\mug/mL$). Operation of continuous packed-bed reactor using Fibra-cel enabled the packging cell to stably maintain retrovirus titer for about 1 week. The optimal operation conditions for dilution rate and temperature were 0.67(h(sup)-1) and $32^{circ}C$, respectively. Using this media, the retrovirus titer(cfu/mL) in the packed continuous culture was about 50% that of continuous culture with serum containing DMEM media.

  • The effect of biomass concentration of Saccharomyces cerevisiae ATCC 24858 on specific growth rates, biomass yields, ethanol yields and productivity in the batch fermentation of rotary shaker was investigated. The specific growth rate decreased according to the increase in the biomass density and finally became zero at a biomass concentration, 55g/L. The ethanol yield $Y_{p/s}$ represented a constant value, 0.43, regardless of the change of biomass concentrations. However, the biomass yield $Y_{x/s}$ showed a trend to diminish in values with augmentation of biomass density and ultimately to reach zero at 55g/L of biomass concentration. The ethanol productivity increased linearly with biomass concentration so that, in case of initial sugar concentration, 170g/L, the productivity for 55g/L of biomass density rose up to 30g/L$\cdot$hr for all the batch fermentations. And also the ethanol concentration inhibiting completely the growth was verified 95g/L by applying experimental data to Luong's equation.

  • The effects of nitrogen sources(sodium glutamate and peptone) on the production of xanthan gum were investigated. The fermentation using sodium glutamate as a nitrogen source is longer than that of peptone. In the initial nitrogen concentration of 0.4-1.0g/L, Bs was about 2.0 and ${\beta}$s was 1.2. The optimal yields were obtained when the carbon source/nitrogen concentration was 10-16. The fermentation time and product yields in the fermentation medium of mixed nitrogen source [sodium glutamate-N(0.75g/L)+peptone-N(0.25g/L)] were similar to those of peptone.

  • 차세대 반도체 산업은 제조공정의 미세화, 복잡화, 다단계화 등이 심화되고 이로 인해 제조원가의 절감을 위한 단위 Wafer당 보다 많은 칩을 생산할 수 있는 450mm 웨이퍼의 도입을 반드시 필요로 한다. 본 논문에서는 클러스터 툴을 주로 사용하는 450mm 웨이퍼 수율관리 개선 시스템의 구현방향과 상세 기능과 각 모듈에 대한 연구를 수행하였으며, 450mm 웨이퍼 생산체제 하에서 필요한 수율관리시스템인 RTFMD 시스템을 제안 하였다.
