Isolation of the Bacteria Strains with Highly Active β-glucosidase from Traditional Wine and Use them for the Synthesis of Non G lucoside Product in Plant (식물유래 비배당체 활성 성분 증대를 위한 고활성 β-glucosidase 생산 균주 선별 및 특성 규명)

  • 식물 내에는 배당체 형태의 생리활성 물질들이 다량 존재하고 있으며, 비배당체로 전환하였을 경우 생리활성이나 투과성이 증대된다는 연구들이 보고되어 있다. 따라서 본 연구에서는 고활성 β-glucosidase 활성을 갖는 균주를 선별함으로써 식물 내의 비배당체 활성 성분을 증대시키는 데 활용하고자 한다. 균주는 전남지역 제조된 막걸리부터 분리하였다. 분리된 균주는 성장 속도 등 산업적으로 활용될 수 있는 가치를 평가하여 11종 22점의 균주를 분리하고, 16S rRNA분석을 통해 균주를 동정하였다. 동정된 균주를 이용하여 β-glucosidase의 활성을 평가하여 고활성 균주를 1종을 선별하였다. 선별된 균주는 다른 균주에 비하여 상대적 활성이 57% ~ 99%정도 활성이 높았으며, 공시 균주와 비교하였을 때도 상대 활성이 높은 았다. 선별된 균주의 특성을 규명하기 위하여 glycerol, L-arbinose를 비롯한 49가지의 탄수화물의 대사 활성을 확인해본 결과 25가지의 탄수화물 대사 활성을 가지고 있는 것을 알수 있었다. 또한 Lipase, β-glucosidase 19가지의 다양한 효소활성을 확인해본 결과 8종의 효소활성이 있음을 확인할 수 있었다.

  • This study evaluated the effectiveness of biostimulating agent in contaminated coastal sediment. The study was conducted via column tests in coastal sea wherein two separate columns were employed for two different polymers used and another column for a blank. The biostimulating agent was made by mixing sea sediment with biostimulants viz acetate, nitrate, sulfate. The biostimulating agent was then rolled into balls, dried and coated with either Cellulose Acetate (CA) or Polysulfone (PS) to control the release of the biostimulants. The pH was around 7.6~8 for 4 months while COD, TP and TN were significantly lower in the column containing biostimulating agents. Heavy metal(Fe, Zn, Cd, Cr, Pb, Cu) was converted to stable forms and PS coated biostimulating agent had a high efficiency of heavy metals distribution.

  • The biological activity of monoglycerides, diglycerides, triglycerides and free fatty acids were investigated by the rice inclination test that is specific bioassay for brassinosteroids. The experimental results revealed that some diglycerides, triglycerides and free fatty acids as well as monoglycerides showed positive response. ${\alpha}-Monoolein$ and diolein as active principles showed higher activity than indole-3-acetic acid at the same concentration. From the results, it can be confirmed that the active principles are related to olein which especially have a high activity.

  • The antimicrobial activty of leek (Allium tuberosum) was investigated against 17 strains of microorganisms. Methanol extracts of leek showed the growth inhibition effects on the wide range of microorganisms including gram positive bacteria, gram negative bacteria and yeasts. The extracts were analysed by using solvent fractionation, silica gel adsorption chromatography, Sephadex LH-20 column chromatography, TLC, silica gel partition chromatography and HPLC techniques. Six components whose molecular weights range from 200 to 400 were confirmed to have the antimicrobial activity.

  • For the further development of thelepin analog as antibiotic agents, we undertook the synthesis of spirobenzopyranone derivative ${\underline{5}}$ as thelepin analog by oxidative phenol coupling. The spirobenzopyranone analog ${\underline{5}}$ showed high activity against Bacillus subtilis (IFO 3108) in $5\;{\mu}g/disc$.

Enhanced Bioremediation of Phenanthrene Using Biosurfactant (생물계면활성제를 이용한 Phenanthrene의 생물학적 처리)

  • This study was carried out 1) to investigate the pH effect on solubilization of phenanthrene by biosurfactant in aqueous system and 2) to evaluate the pH effect on the biodegradation rate of phenanthrene in the presence and the absence of the biosurfactant by phenanthrene degraders. Tween 80, which is a chemically synthesized surfactant, showed greater solubilizing capacity than rhamnolipid. The solubilization capacity can be expressed as a MSR(molar solubilization ratio=moles of organic compounds solubilized per mole of surfactant). The calculated MSR of Tween 80 and rhamnolipid were 0.1449 and 0.0425 respectively. The kinetic study of phenanthrene solubilization by rhamnolipid showed that solubilization mechanism could reach equilibrium within 24 hours. Addition of 240 ppm rhamnolipid solution, which concentration is 4.3 times of Critical Micelle Concentration(CMC), caused 9 times solubility enhancement compared to water solubility. The highest solubilities were detected around a pH range of 4.5-5.5. Changes in apparent solubility with the changes in pH are possibly related to the fact that the rhamnolipid, an anionic surfactant, can form different structures depending on the pH. Two biodegradation experiments were performed in the absence and the presence of rhamnolipid, with the cell growth investigated using a spread plate method. The specific growth rates at pH 6 and 7 were higher than at the other pH, and the HPLC analysis data, for the total phenanthrene loss, confirmed the trends in the $\mu$(specific growth rate) values. In presence of rhamnolipid, maximum $\mu$ values shifted from around pH 5 which showed maximum enhancement of solubility in the abiotic experiment, compared to the $\mu$ values obtained without the biosurfactant. In this study, the increase in the observed specific grow rate(1.44 times) was not as high as the increase in solubilization(5 times). This was supported by the fact all the solubilized phenanthrene is not bioavailable to microorganisms.

Cultural Characteristics of a Biosurfactant-Producing Microorganism Pseudomonas aeruginosa F722 (Biosurfactant 생산균주 Pseudomonas aeruginosa F722의 배양특성)

  • Productivity of biosurfactant (rhamnolipid) by Pseudomonas aeuginosa F722 was investigated in the several culture conditions and culture composition. Biosurfactant production by P. aeuginosa F722 was amounted to 0.78 g/l as the result of the nitrogen sources and carbon sources without investing of optimum conditions. As for that one was investigated, biosurfactant production by P. aeruginosa F722 was amounted to 1.66 g/l. Biosurfactant production increased twofold because the composition of a modified C-medium was investigated efficiently. $NE_4$Cl or $NaNO_2$ inorganic nitrogens and yeast extract or trypton organic nitrogens were effective, but others inorganic nitrogens and organic nitrogens tested were not efficient far biosurfactant production by P. aeruginosa F722. The optimum concentration of $NH_4$Cl; inorganic nitrogen and yeast extract; organic nitrogen were 0.05% and 0.1%, respectively. In various carbon sources, others with the exception of hydrophobic property substrate (n-alkane) and hydrophilic property substrate (glucose, glycol) were not found to be effective fur biosurfactant production, and 3.0% was better in yield than other concentration of glucose. This yielded C-to-N ratios between 17 and 20. In our experiment, the highest biosurfactant production by P. aeruginosa F722 were observed in 5 days cultivation, containing glucose 3.0%, $NH_4$Cl 0.05%, and yeast extract 0.1% and C-to-N ratio was 20. Optimal pH and temperature for biosurfactant production were 7.0 and $35^{\circ}C$, respectively. Under the optimal culture conditions with glucose, biosurfactant production was amounted to 1.66 g/l. Velocity of biosurfactant production and strain growth increased after nitrogen depletion. The average surface tension of 30 mN/m after the 3 days of incubation under optimal culture condition was measured by ring tensionmeter.

Characterization of the Biosurfactant-Producing Bacterium, Pseudoalteromonas sp. HK-3 Isolated from the Crude-Oil Contaminated Areas (원유로 오염된 지역으로부터 분리한 생물계면활성제 생산균주, Pseudoalteromonas sp. HK-3의 특성조사)

  • The purpose of this work was to investigate the characteristics of a biosurfactant-producing bacterium isolated from crude-oil contaminated soils. During the incubation of strain HK-3 with 1% crude-oil, bacterial growth pattern, the amount of biosurfactant production, and pH changes were monitored. In order to examine the effect of supplemented carbons on the production of biosurfactant, cultivation of HK-3 cells in BH media with different carbons (e.g. glucose, dextrose, mannitol, citrate, or acetate) revealed that the production of biosurfactant reached the maximal level at the 72 h incubation with mannitol, which the area of clear zone was measured to approximately 7.64 $cm^2$. Identification test using the BIOLOG system, morphology study based on scanning electron microscopy and the 16S rRNA sequence-based phylogenetic analysis assigned strain HK-3 to a Pseudoalteromonas species, designated as Pseudoalteromonas sp. HK-3 which was registered in GenBank as [FJ477041].
