Characteristics of the Graded Wildlife Dose Assessment Code K-BIOTA and Its Application (단계적 야생동식물 선량평가 코드 K-BIOTA의 특성 및 적용)

  • This paper describes the technical background for the Korean wildlife radiation dose assessment code, K-BIOTA, and the summary of its application. The K-BIOTA applies the graded approaches of 3 levels including the screening assessment (Level 1 & 2), and the detailed assessment based on the site specific data (Level 3). The screening level assessment is a preliminary step to determine whether the detailed assessment is needed, and calculates the dose rate for the grouped organisms, rather than an individual biota. In the Level 1 assessment, the risk quotient (RQ) is calculated by comparing the actual media concentration with the environmental media concentration limit (EMCL) derived from a bench-mark screening reference dose rate. If RQ for the Level 1 assessment is less than 1, it can be determined that the ecosystem would maintain its integrity, and the assessment is terminated. If the RQ is greater than 1, the Level 2 assessment, which calculates RQ using the average value of the concentration ratio (CR) and equilibrium distribution coefficient (Kd) for the grouped organisms, is carried out for the more realistic assessment. Thus, the Level 2 assessment is less conservative than the Level 1 assessment. If RQ for the Level 2 assessment is less than 1, it can be determined that the ecosystem would maintain its integrity, and the assessment is terminated. If the RQ is greater than 1, the Level 3 assessment is performed for the detailed assessment. In the Level 3 assessment, the radiation dose for the representative organism of a site is calculated by using the site specific data of occupancy factor, CR and Kd. In addition, the K-BIOTA allows the uncertainty analysis of the dose rate on CR, Kd and environmental medium concentration among input parameters optionally in the Level 3 assessment. The four probability density functions of normal, lognormal, uniform and exponential distribution can be applied.The applicability of the code was tested through the participation of IAEA EMRAS II (Environmental Modeling for Radiation Safety) for the comparison study of environmental models comparison, and as the result, it was proved that the K-BIOTA would be very useful to assess the radiation risk of the wildlife living in the various contaminated environment.

Studies on the Environmental Characteristics of the Breeding Ground in the Kogum-sudo, Southern Part of Korean Peninsula I. Seasonal Succession of Phytoplankton Population (거금수도내 양식어장의 해양환경특성 I. 식물플랑크톤 군집의 계절변동)

  • Field studies on the seasonal succesion of phytoplankton population were carried out at the 25 stations of the breeding ground in Kogum-sudo, Southern coast of Korean peninsula in Feburuary, April, August and October, 1993. Sixty four species belonging to 40 genera were identified. Predominant species were mainly centric diatoms throughout the four seasons, two centric diatoms, Skeletonema costatum and Thalassiosira sp. and a pennate diatom, Thaiassionema nitzschioides in the winter; two pennate diatoms, Thaiassionema nitzschioides and Asterionella kariana, and especially a dinoflagellate, Heterocapsa triquetra (station 10) in the spring, two centric diatoms, S. costatum and Chaetoceros diadema in the summer; and a centric diatom, Rhizosolenia alata and a pennate diatom, Bacillaria paxillifer in the fall. The main red tide organisms in the breeding ground were dinoflagellates, Prorocentrum dentatum, P. minimum, P. triestinum, Ceratium furro, Gymnodinium sanguineum, Noctiluca scintillans, H. triquetra, Scrippsiella trichoidea and a diatom S. costatum in the Kogum Sudo. Seasonal phytoplankton cell numbers were in a wide range between $8.8\times10^3$ cells/l and 1.4\times10^6$ cells/l; The seasonal average cell numbers were $12.2\times10^4\pm5.9\times10^4$ cells/l $(mean\;\pm\;standard\; diviation)$ in the winter, $3.3\times10^4\pm1.4\times10^4$ cells/l in the spring, $48.4X10^4\pm40.0\pm10^4$ cells/l in the summer, and $3.6\times10^4\pm1.9\times10^4$ cells/l in the fall, respectively.

  • Background : Nucleic acid hybridization has become an essential technique in the development of our understanding of gene structure and function. The quantitative analysis of hybridization has been used in the measurement of genome complexity and gene copy number. The filter hybridization assay is rapid, sensitive and can be used to measure RNAs complementary to any cloned DNA sequence. Methods : The authors assessed the accuracy, linearity, correlation coefficient and specificity of the hybridization depending on the added dose(0, 1, 5, and $10{\mu}g$) of non-specific rat spleen RNA to hybridization of surfactant protein A mRNA. Filter hybridization assays were used to obtain the equation of standard curve and thereby to quantitate the mRNA quantitation. Results : 1. Standard curve equation of filter hybridization assay between counts per minute (X) and spleen RNA input (Y) was Y=0.13X-19.35. Correlation coefficient was 0.98. 2. Standard curve equation of filter hybridization assay between counts per minute (X) and surfactant protein A mRNA transcript input (Y) was Y=0.00066X-0.046. Correlation coefficient was 0.99. 3. Standard curve equation of filter hybridization assay between counts per minute (X) and surfactant protein A mRNA transcript input (Y) after the addition of $1{\mu}g$ spleen RNA was Y=0.00056X-0.051. Correlation coefficient was 0.99. 4. Standard curve equation of filter hybridization assay between counts per minute (X) and surfactant protein A mRNA transcript input (Y) after the addition of $5{\mu}g$ spleen RNA was Y=0.00065X-0.088. Correlation coefficient was 0.99. 5. Standard curve equation of filter hybridization assay between counts per minute (X) and surfactant protein A mRNA transcript input (Y) after the addition of $10{\mu}g$ spleen RNA was Y=0.00051X-0.10. Correlation coefficient was 0.99. Conclusions : Comparison of cpm/filter in a linear range allowed accurate and reproducible estimation of surfactant protein A mRNA copy number irrespective of the addition dosage of non-specific rat spleen RNA over the range $0-10{\mu}g$.

Trace Metal Contents in Seaweeds from Korean Coastal Area (한국 연안 해조류의 미량금속 함량)

  • In order to assure the safety of the seaweeds, we measured the contents of the trace metals in the seaweeds harvested from Korean coastal area. We collected 620 marine algal samples from fourteen areas, and then analyzed the trace metals such as Hg, Cd, Cr, Cu, Mn, Ni, Pb and Zn. The contents of Hg, Cd, Ni and Mn were higher in the seaweeds collected from Chungnam Taean area. The contents of Cr and Cu were higher in the seaweeds collected from Yeongdeok and Tongyeong area, respectively. And Pb and Zn were highly detected from the seaweeds of Ulsan area. The mean levels of trace metals were high in the order of Zn $(48.02\pm41.20{\mu}g/g)\;>\;Mn (34.63\pm38.95{\mu}g/g)\;>\;Cu(6.29\pm6.52{\mu}g/g)\;>\;Ni(2.17\pm4.00{\mu}g/g)\; >\;Cr(1.61\pm2.35{\mu}g/g)\;>\;Pb(1.28\pm2.37{\mu}g/g)\;>\; Cd(0.67\pm0.75{\mu}g/g)\;>\;Hg(0.02\pm0.02{\mu}g/g)$. Zn was consistently most abundant metal in all samples, followed by Mn and Cu, which are very important for human health. There were higher contents of Hg and Cd in brown algae, the contents of Cr, Cu, Mn, Ni and Pb in green algae, the content of Zn in red algae among them. Additionally, significant linear correlations were observed between a total 28 different pairs of metals, of them, the correlation factor between Cr and Ni showed the highest (r=0.374, p<0.001). The trace metal contents of seaweeds varied with habitats and kinds of samples, nevertheless they showed very safe level in edible seaweeds tested. Scytosiphon lomentaria, which has been known as a little use of food, have shown a clear selectivity for Cr and Pb. Among non-edible seaweeds, Sargassum thunbergii for both Cd and Mn, Lomentaria hakodatensis for Ni and Grateloupia prolongata for Zn have shown a clear selectivity, respectively. Therefore, these seaweeds could be useful as bioindicator for each trace metal pollution.

Trends of mushroom science and mushroom industry (버섯과학과 버섯산업의 동향)

  • World production of mushrooms has been increasing 10-20% every year. Recently, Pleurotus eryngii and P. nebrodensis are very popular as new mushroom species for cultivation. Two kinds of mushrooms, Gumji (Ganoderma) and Soji, were described in old book of Samguksagi (History of the three kingdoms; 1145) in Koryo-dynasty. Many kinds of mushrooms were also described in more than 16 kinds of old books during Chosun-dynasty in Korea. One hundred and sixty commercial strains of 25 species in mushrooms were distributed to cultivators. By the way, only 8 varieties of them have registered variety protection. Mushroom industry as important export products developed from 1960 to 1980. Production of mushrooms as food was 181,828 metric tons valued at 800 billion Korean won in 2003. Isolated and identified substances from mushrooms are promising antifungal, antiinflammatory, antitumor, antiviral (anti-HIV), antibacterial & antiparasitic, antidiabetic, immunomodulating, kidney tonic, hepatoprotective, nerve tonic, and sexual potentiator. These substances can also be used for blood pressure regulation and effective against cardiovascular disorders, hypocholesterolemia & hyperlipidemia, and chronicbronchitis. Mushroom products including pharmaceuticals, tonics, healthy beverages, functional biotransformants, and processed foods have also became available on the markets. Compost and feed can likewise be made from mushroom substrates after harvest. The mushroom industry is already one of the fastest growing investment sectors in Korea. By the way, there is a need to strain improvement for variety protection, advanced cultivation technology at low cost for growers, and control of demand and supply for marketing in order to more upgrade development of mushroom industry in the future.

Improvements in the Marine Environmental Survey on Impact of Seawater Qualities and Ecosystems due to Marine Sand Mining (바다모래 채취 시 해수 수질 및 생태계 영향에 대한 해양환경조사 개선 방안)

  • We reviewed investigation status on turbidity plume in the statement of marine environmental survey(2008 to 2012) associated with marine sand extraction projects. The survey statement from seven marine sand extraction sites (extraction area of Southern EEZ, extraction area of Western EEZ, relocation zone in the Western EEZ, sea area under jurisdiction of Taean-gun, sea area under jurisdiction of Ansan City, and two discrete sea areas under jurisdiction of Ongjin-gun) in the nearshore and offshore of Korea showed that in situ observations were carried out for the dispersion and transport of suspended sediments on two areas (One is a extraction area in the EEZs, the other is an area of coastal sites). However, sampling station and range have not been selected considering physical, geographical factors (tide, wave, stratification, water depth, etc.) and weather conditions (wind direction and velocity, fetch, duration, etc). Especially turbidity plumes originating from three sources, which include suspended sediments in overflow(or overspill) discharged from spillways and reject chutes of dredging vessel, and resuspended sediments from draghead at the seabed, may be transported to a far greater distance outside the boundary of the extraction site and have undesirable impacts on the marine environment and ecosystem. We address that behaviour of environmental pollutants such as suspended solids, nutrients, and metals should be extensively monitored and diagnosed during the dispersion and transport of the plume. Finally we suggest the necessity to supplement the current system of the sea area utilization consultation and establish the combined guidelines on marine sand extraction to collect basic data, to monitor cumulative effects, and to minimize environmental damages incurred by the aftermath of sand extraction.

Brucite Treatment to Reduce Phosphorus Release from Polluted Sediments (퇴적물로부터 인 용출 저감을 위한 Brucite 처리)

  • Lab-scale batch experiments using several 25-L transparent acrylic reactors were conducted to develop optimum capping materials that can reduce phosphorus released from polluted sediments. The sediment used in the experiment was very fine clay(8.8 $\Phi$ in mean grain size), and organic carbon($C_{org}$) content was as high as 2%. Four kinds of batches with different capping materials Brucite($Mg(OH)_2$), Sea sand($SiO_2$), Granular-gypsum($CaSO_4{\cdot}2H_2O$), Double layer(brucite+sand), and one control batch were operated for 30 days. Phosphorus fluxes released from bottom sediments in the control batch were estimated to be 14.6 $mg{\cdot}m^{-2}{\cdot}d^{-1}$, while 9.5 $mg{\cdot}m^{-2}{\cdot}d^{-1}$, 5.2 $mg{\cdot}m^{-2}{\cdot}d^{-1}$, 4.2 $mg{\cdot}m^{-2}{\cdot}d^{-1}$, and 3.1 $mg{\cdot}m^{-2}{\cdot}d^{-1}$ in the batch capped with Sea sand, Granular-gypsum, Double layer, and Brucite, respectively. The results obtained from lab-scale batch experiments show that there were 70% reduction of phosphorus for some materials such as Brucite, Double layer(brucite+sand), and whereas sea sand only about 35%. The pH range of surface sediment to which Brucite was applied showed about $8.0{\sim}9.5$ in the weak alkaline state. This effect can prevent liberation of $H_2O$. The addition of gypsum into the sediment can reduce the progress of methanogenesis because of fast early diagenesis and sufficient supply of $SO_4^{2-}$ to the sediments, stimulate the SRB highly. Therefore, the application of Brucite and Gypsum can reduce phosphorus release from the sediment as a result of formation of $Mg_5(OH)(PO_4)_3$, pyrite($FeS_x$), and apatite-mineral.

Comparative Evaluation of the VITEK 2 System and Species-specific PCR Methods for the Detection of Vibrio Species Isolated from Shrimp (새우에서 분리된 Vibrio species 동정을 위한 VITEK 2 system방법과 species-specific PCR방법 비교 평가)

  • Vibrio is a genus of Gram-negative, curved, halophilic, and non-spore-forming bacteria. Some of the Vibrio species, such as V. cholerae and V. parahaemolyticus, often contaminate seafood products and occasionally cause human diseases when the seafood products are ingested. A total of 24 Vibrio strains were isolated from shrimp samples on Thiosulphate citrate bile salt sucrose (TCBS) media in this study. All of the 24 isolates were confirmed to belong to the genus Vibrio by using 16S rRNA gene sequence analyses. Vitek 2 system and species-specific polymerase chain reaction (PCR) methods were used to further identify a total of 29 Vibrio strains at the species level, including the 24 shrimp Vibrio isolates and five Vibrio reference strains. The specificities of the two methods to identify Vibrio strains at the species level were compared in this study. The species-specific PCR method was designed to detect five different Vibrio species, such as Vibrio cholerae, Vibrio parahaemolyticus, Vibrio vulnificus, Vibrio alginolyticus, and Vibrio mimicus. From the 24 Vibrio shrimp isolates, the Vitek 2 system method could identify 15 (62.5%) strains as Vibrio species and 7 (29.2%) strains as non-Vibrio species, but could not identify the rest 2 (8.3%) strains. But species-specific PCR method could identify 16 (66.7%) strains as Vibrio species and could not identify the rest 8 (33.3%) strains. Among the 24 Vibrio shrimp strains, these two methods could unanimously identify 7 (7/24, 29.2%) strains (2 V. parahaemolyticus, 4 V. alginolyticus, and 1 V. mimicus). Considering that such different identification results were obtained using the two different methods in this study, identification method for Vibrio species must be carefully chosen.

Atmospheric Aerosol Monitoring Over Northeast Asia During 2001 from MODIS and TOMS data (MODIS와 TOMS자료를 이용한 2001년 동북아시아 지역의 대기 에어로졸 모니터링)

  • The spatial and temporal variations of aerosol optical depth (AOD) over Northeast Asia regions have special importance in the aerosol research for estimation of aerosol radiative forcing parameters and climate change. Aerosol optical and physical properties (AOD and ${\AA}$ngstrom parameter) have been investigated by using Moderate Resolution Imaging Spectroradiometer (MODIS) and Total Ozone Mapping Spectrometer (TOMS) Aerosol Index (AI) to estimate aerosol characteristics over the study region during 2001. Additionally, aerosol characteristics over the Korean peninsular during Aerosol Characteristic Experiment in Asia (ACE-Asia) Intensive Observation Period (IOP) have been investigated by using satellite observations. The results showed that the daily-observed aerosol data indicate seasonal variations with relatively higher aerosol loading in the spring and very low during the winter. The typical Asian dust case showed higher AOD (>0.7) with lower Angstrom exponent (<0.5) and higher AI (>0.5) that is mainly due to the composition of coarse particles in the springtime. Mean AOD for 2001 at 4 different places showed 0.65$\pm$0.37 at Beijing, 0.31$\pm$0.19 at Gosan, 0.54$\pm$0.26 at Seoul, and 0.38$\pm$0.19 at Kwangju, respectively. An interesting result was found in the present study that polluted aerosol events with small size dominated-aerosol loading around the Korean peninsular are sometimes observed. The origin of these polluted aerosols was thought to East China. Aerosol distribution from satellite images and trajectory results shows the proof of aerosol transport. Therefore, aerosol monitoring using satellite data is very useful.

The Damage Assessment, Construction Point of Time and Deterioration Diagnosis and Conservation Maintenance of Stone Statues Around the Stone Pagoda in Mireuksaji Temple in Iksan (익산 미륵사지 석탑 석인상의 조영시기와 훼손도 진단 및 보존관리)

  • The stone statues in the site of Mireuksaji Temple(Iksan, South Korea) were created after the stone pagoda was built in 639. They, transitional statues between animal sculptures and human shaped statues made from the late Goryeo dynasty until the early Joseon dynasty, were set up at the four corners of the stone pagoda by way of guardians. In the case of three statues, their surfaces were denudated and their iconographies have been indiscernible. However, the one in the southwest clearly shows its iconography. It is inferior in properties to the other three statues in the northwest, the northeast and the southeast respectively, but on the other hand its iconography has been well maintained. The reason is related to exposure to harmful environments; specifically, the retaining wall, built around the stone pagoda in the 17th century, had the southwest statue inside and could naturally worked as a buffer against harmful environments. As a result, for about 400 years there has been difference in weathering conditions between the three stone statues and the southwest statue, which brought denudation, the consequent indiscernibleness of iconography and biological invasion to the three statues, notwithstanding superior properties(northwest statue:$176kgf/cm^2$, northeast statue:$109kgf/cm^2$, southeast statue:$273kgf/cm^2$). In contrast, the southwest statue significantly shows its iconography with black contaminants and granule decomposition, despite inferior properties($133kgf/cm^2$). Defenseless exposure to external environment is not recommended for the stone statues, because it is hard to preserve the extant iconography. Herein lies the application of the data on microclimate around Mireuksaji Temple. As regards the weathering zone in which the stone statues are located, Conservation increases in acidity and frequency as years go by, Hereat, in the approach to the Conservation of stone statues, the first consideration needs to be morphological historicity rather than geographical location.
