Characterization of the Biosurfactant-Producing Bacterium, Pseudoalteromonas sp. HK-3 Isolated from the Crude-Oil Contaminated Areas (원유로 오염된 지역으로부터 분리한 생물계면활성제 생산균주, Pseudoalteromonas sp. HK-3의 특성조사)

  • The purpose of this work was to investigate the characteristics of a biosurfactant-producing bacterium isolated from crude-oil contaminated soils. During the incubation of strain HK-3 with 1% crude-oil, bacterial growth pattern, the amount of biosurfactant production, and pH changes were monitored. In order to examine the effect of supplemented carbons on the production of biosurfactant, cultivation of HK-3 cells in BH media with different carbons (e.g. glucose, dextrose, mannitol, citrate, or acetate) revealed that the production of biosurfactant reached the maximal level at the 72 h incubation with mannitol, which the area of clear zone was measured to approximately 7.64 $cm^2$. Identification test using the BIOLOG system, morphology study based on scanning electron microscopy and the 16S rRNA sequence-based phylogenetic analysis assigned strain HK-3 to a Pseudoalteromonas species, designated as Pseudoalteromonas sp. HK-3 which was registered in GenBank as [FJ477041].

Biochemical Changes of Protein during the Senescence of Tobacco Leaf (담배잎의 노화과정에 따른 단백질의 생화학적 변화)

  • This experiment was conducted to obtain basic information of biochemical changes in the process of senescence by measuring the total RNA, protein, protease activity and electrophoretic pattern of protein in tobacco plant. The content of soluble protein increased by 15 days after leaf emergence and its level was not changed from 15 to 35 days after leaf emergence. The content of total RNA showed a maximum value at 15 days after leaf emergence and then decreased rapidly until 30 days after leaf emergence. The activity of protease of neutral fraction was higher than that of acidic fraction and rapidly increased up to the end of senescence after 50 days after leaf emergence. According to the analysis of electrophoresis, polypeptide band of 61kd was developed after 35 days after leaf emergence and increased by the end of senescence.

한국 작업자의 요통예방을 위한 작업하중 설계지침

  • 본 연구의 목적은 드는 작업에 있어서 한국 작업자의 적정하중을 구하여 중량물 안전기준을 정하는 것이다. 본 연구에서는 주어진 작업조건하에서 피실험자가 들어올릴 수 있는 적정하중을 결정하기 위 한 기준으로서 피실험자 자신에 의해서 인지되는 스트레스 정도를 이용하는 심리육체적 방법(Psychophysical method), 주어진 작업조건하에서 요추에 가해지는 압력을 이용하는 생체역학적 방법(Biomechanical method), 주어진 작업조건하에서 피실험자의 에너지 소모량을 이용하는 생리학적 접근법등이 이용되었다. 피실험자는 학생(n=3) 두개의 군으로 나뉘어진다. 실험에 들어가기 전에 피실험자에 대한 인체측정과 근력측정을 수행 하였다. 실험은 수직면에서 드는 빈도(1,2, 와 4회/분)와 드는 높이(0-8cm 와 47-102cm)를 조합한 6가지의 작업을 무작위로 선택하여 심리육체적 방법으로 각 작업에 대한 적정하중을 구하였다. 연구결과로서 피실험 자의 신체자료와 근력의 비교로부터 신체조건은 학생이 우수하였으나, 근력은 작업자가 우수하였다. 본 연구 에서 채택된 들기의 빈도가 변함에 따라 최대허용 하중은 변하였다. 작업자군에 대하여는 빈도가 증가할수록 최대허용 하중은 감소하였으며, 학생군에 대하여는 분당 1회와 분당 4회간의 유의한 차이가 있었다. 들기의 시작-종점의 변화에따른 최대허용 하중은 통계적으로 유의하지 않았다. 위의 두 결과는 들기의 빈도가 들기의 시작-종점보다 민감한 변수임을 가리키며 이는 미국 NIOSH의 결과와 일치한다. 심리육체적 방법을 사용하여 우리나라의 젊고 건강한 남성의 대부분(99%)을 고려하여 산출된 최대허용 하중은 미국 NIOSH 안전기준과 큰 차 이가 없음을 발견하였다. 또한 이 최대허용 하중은 인체역학적방법과 생리학적 방법을 사용하여 검토한 결과, 무리가 없는 수준임이 입증되었다. 본 연구의 결과를 토대로우리나라 젊고 건강한 남성에게 적합한 무게상수는 작업자군에 대하여 25.05kg, 학생군에 대하여 20.24kg 으로 나타나 이는 미국 NIOSH 안전기준과 대체로 일치함을 발견하였다.

Safety Test of Brown Rice Expressing Arabidopsis Calcium Transporter by Feeding Trial in Mice (애기장대 칼슘수송체를 발현하는 형질전환 현미의 생쥐 식이를 통한 안전성 평가)

  • Previously, we found that the transgenic rice plants over-expressing the Arabidopsis $H^+/Ca^{2+}$ antiporter CAX 1 (accession no. U57411) gene accumulated 2.7 to 7.5-fold more calcium in the T3 rice grains as compared to those of control. To examine physiological safety of the $T_3$ rice grains, the effect of the $T_3$ brown rice on change in levels of body weight and white blood cells was compared with that of the control Ilpum brown rice by feeding trial in mice. During the feeding trial for one month, there was no significant difference between two mice groups, which were fed by the $T_3$ brown rice or Ilpum brown rice. There were no detectable differences in their effects on immune functions including plaque-forming unit, peritoneal macrophage number, and NK-cell activity. In addition, biochemical analysis of the blood failed to exhibit any difference between two mice groups. Together, these results suggested that the $T_3$ brown rice, which was produced from a genetically modified organism (GMO), might be safe and possess a potential to be applicable as calcium-fortified feed or food. Long-term safety of the $T_3$ brown rice, however, remains to be elucidated.

Glucocorticoid Regulation of Gene Expression in Hippocampal CA3 and Dentate Gyrus (글루코코티코이드 호르몬에 의한 뇌해마의 CA와 Dentate Gyrus 부분의 유전자 발현 변화)

  • Glucocorticoids (GCs) alter metabolism, synaptogenesis, apoptosis, neurogenesis, and dendritic morphology in the hippocampus. To better understand how glucocorticoids regulate these aspects of hippocampal biology, we studied gene expression patterns in the CA3 (Hippocampal pyramidal cell field CA3) and dentate gyrus (DG). Litter-matched Lewis inbred rats treated for 20 days with either 9.5 mg per day sustained-release corticosterone or placebo pellets were compared with high-density oligonucleotide microarray analysis (Rat Neurobiology U34 Arrays, Affymetrix). In placebo-treated rats, 32 genes were expressed at greater levels in CA3 than DG, whereas 3 genes were expressed at great levels in DC than CA3. Regional differences were also apparent in corticosterone-induced changes in the hippocampal transcriptome. Six genes in CA3 and 41 genes in DC were differentially regulated by corticosterone. As per the glucocorticoid effects on gene transcription in the brain, forty three of these genes were upregulated, and 4 genes were downregulated. Genes differentially expressed in hippocampus included those for 13 neurotransmitter proteins, 5 ion channel related proteins, 4 transcription factors, 3 neurotrophic factors, 1 cytokine, 1 apoptosis related protein, and 5 genes involved in synaptogenesis. Interestingly, GCs can have suppressive effects on brain BDNF mRNA transcription, one of the neurotrophic factors. These results indicate the diversity of targets affected by chronic exposure to corticosterone and highlight important regional differences in hippocampal neurobiology.

  • 휴대폰 전자파로 인해 두통, 불면증, 단기간의 기억력 소실 등의 신경 생리학적 증상을 호소하는 휴대론 전자파 과민증 (EHS, Electromagnetic Hypersensitivity) 환자들이 늘어나고 있다. 그러나 휴대폰 전자파 과민반응의 원인이 막연한 불안감 때문인지 혹은 전자파 노출로 인한 증상 인지에 대하여 확실하게 규명이 되지 않고 있다. GSM 방식의 휴대폰의 경우 전자파 노출 시 EHS 군을 대상으로 혈압, 맥박수 변화 등의 과민 반응에 대한 연구들이 수행되었으나 일반인과 EHS 군의 실험-대조군 연구로 생체신호, 자각증상 및 전자파 인지 여부에 대한 복합석인 연구와 CDMA 휴대폰 EHS 연구는 거의 없는 실정이다. 본 연구예서는 일반인 6명과 EHS 6명을 대상으로 30분간 300mW의 CDMA 휴대폰 전자파에 노출시 교감신경의 흥분을 나타내는 맥박, 호흡수, HRV(heart rate variability의 변화를 측정하였다. 그 결과, 일반인과 EHS 군 모두 전자파 노출여부에 따라 맥박, 호흡수, HRV의 변화가 없었다.

The author reviewed the physiology of menopause and discussed the physiological mechanism and biological management of vascular flushing and menopausal depression. The above symptoms are hard to distinguish from those of psychiatric disorders.

Changes of the Excitability of the α-Motor Neuron by Taping (테이핑에 의한 α-운동 신경원 흥분도 변화)

  • The application of tape is being widely used for treatment of the musculoskeletal disorders and injury prevention. The techniques of the tape application has been relied on empirical evidence in preference to the neurophysiological evidence. Thus, the mechanism of taping has to be elucidated further. In this study, we assessed the effect of the elastic and non-elastic tape upon the gastrocnemius ${\alpha}$-motor neuron excitability using the gastrocnemius H-reflex. The amplitude of the M-wave and H-reflex were measured across three conditions: before tape application, with tape and with the tape removed. No significant changes of the excitability of the ${\alpha}$-motor neuron were obtained across three condition, either in the elastic and non-elastic tape. This results were quite different with other recent studies, which needs to be explored further.

The Transmission Electron Microscopic Study on the Alteration of Filtration Barrier in Aged Rat Kidney (흰쥐 콩팥여과관문의 노화 변화에 관한 투과전자현미경적 연구)

  • The filtration barrier of kidney consists of endothelial cell, glomerular capillary, glomerular basement membrane, mesangial matrix, and podocyte. In aged rats, the morphological changes were shown in various parts, including the glomerulus. These changes were thickening of basement membrane and mesangial matrix, crescent formation of glomerular capillary, deformity of foot processes, glomerular sclerosis and obsolescence. But these glomerular morphologies are partial images or few serial images analysis. In this study, we examined the morphological alteration of glomerulus in the young and aged rats by light microscopy, transmission electron microscopy and three dimensional reconstruction. We were found in aged rat glomerulus, expansion of urinary space and mesangial matrix, thickening and degrading of glomerular basement membrane, decreasing in podocyte foot processes, fragmentation of podocytic nucleus membrane. These observations indicate that may provide useful data for investigating the pathogenesis of age-related dysfunction of kidney.

Relationship between Heart Rate Variability (HRV) and Serum Cortisol Level to Life Stress (생활 스트레스에 대한 심박변이와 혈청 코티졸과의 관계)

  • Purpose The physiological and biochemical responses of healthy men and women to life stress were measured in order to investigate the correlation between these two responses under the normal situation. Materials and Methods The population of the research is some randomly chosen health college students located in Daejeon City during the period from August to September 2014, and the sample of 94 students. Heart rate variability (HRV), blood pressure (BP) were selected as physiological stress indices and serum cortisol level was used as a biochemical stress biomaker. The data which is collected used SPSS19.0 programs and frequency and percentage and T-test, correlation. Results Male showed the significant higher value of serum cortisol level (p<0.05), BP (p<0.05), and LF/HF ratio of HRV (p<0.05) than female. The difference of BP between correlated significantly with serum cortisol level (p<0.05). The LF/HF ratio of HRV also correlated significantly with serum cortisol level (p<0.05) Conclusion We suggest that LF/HF ratio of HRV and BP may be good indices for the assessment of life stress.
