Studies on the Identification of the Anthocyanins in Plum (자두 Anthocyanin의 구조확인(構造確認)에 관(關)한 연구(硏究))

  • The anthocyanins in Santa rosa plum (widely growing var. in Korea) were extracted with 0.5% methanolic HCl and purified with Amberlite IRC-50 cation exchange resin. The individual pigments were separated by paper chromatography. The two pigments were identified as cyanidin-3-monoglucoside and cyanidin-3-xyloglucoside based on the following evidence: Rf values in various solvents, partical acid hydrolysis, stepwise hydrolysis, sugar moiety, alkaline degradation, and absorption spectra in the visible region. Cyanicin-3-monoglucoside was the abundant pigment of the plum.

Conversion Patterns of Yellow Pigment from Gardenia jasminoides by Staphylococcus epidermidas and Lactobacillus plantarum (Staphylococcus epidermidas와 Lactobacillus plantarum에 의한 치자황색소의 변환양상)

  • The Gardenia jasminoides yellow pigment and converted pigments were completely separated by Amberlite XAD-4 column chromatography. These Pigments were gel filtrated on Sephadex LH-20 column chromatography. The characteristics of absorption spectra of eluate and fractionated pigments were investigated. The pigment converted by Lactobacillus plantarum showed a single blue color with an absorption peak at 588 nm and its molecular size was bigger than that of crocetin. The pigment, converted by Staphylococcus epidermidis, Showed blue-green color, which was composed of yellow color with an absorption peak at 418 nm and blue color at 588 nm. Molecular size of the yellow pigment was smaller than crocetin and that of blue color.

Separation and Purification of Fructo-oligosaccharides by an Ion-Exchange Resin Column (이온교환수지탑을 이용한 Fructo-oligosaccharides의 분리 및 정제)

  • Separation of pure fructo-oligosaccharides from the mixed solution of glucose, sucrose and fructo-oligosaccharides was studied using a cationic ion-exchange resin column. Optimum separation conditions, i.e., temperature, feeding rate and the ratio of column vs. diameter were evaluated, which were found to be $85^{\circ}C$, $0.25h^{-1}$ and 30, respectively. At the optimized separation conditions, high-purity fructo-oligosaccharides up to 96% were obtained and the total recovery yield was about 66% after four cycles. After the chromatographic separation, purification to remove the salts and color in pure fructo-oligosaccharides solution was successfully conducted using the mixed-bed of cationic and anionic ionexchange resin columns.

  • Claviceps purpurea PRL 1980 produces a fluorescent reddish brown pigment in the alkaloid production medium. When D,L-tryptophan $[side\;chain-3-^{14}C]$ was administered into the production medium, the radioactive pigment and 5-hydroxytryphan were isolated from the cultures. Conversion of tryptophan to 5-hydroxytryptophan in vivo was shown by an isotopic trapping procedure. 5-hydroxytryptophan isolated from the cultures contained appreciable radioactivity and was recrystallized to constant specific radioactivity. The injection of the $^{14}C-labelled$ 5-hydroxytryptophan showed an incorporation of radioactivity into brown pigment significantly higher than that of tryptophan. The brown pigment produced by Claviceps purpurea PRL 1980 seems to be derived from tryptophan through 5-hyrdroxytryptophan.

Purification of $\beta$-glucanase from Bacillus subtilis Using Chromogenic Substrate (색소기질을 이용한 Bacillus subtilis의 $\beta$-glucanase 정제)

  • Bacillus subtilis K-4-3, which produces considerable amount of $\beta$-glucanase was selected among extracellular $\beta$-glucanase-producing bacteria isolated from soil. $\beta$-glucanase was purified by ammonium sulfate fractionation, Sephadex G-100 gel filtration and DEAE-sephacel ion exchange chromatography. The purified enzyme revealed a single band by polyacrylamide gel electrophoresis and SDS-polyacrylamide gel electrophoresis. Its molecular weight was estimated to be 17000 dalton by SDS-polyacrylamide gel electrophoresis. The optimum pH and temperature of the purified $\beta$-glucanase were 7.0 and $50^{\circ}C$, respectively. The enzyme was strongly inhibited by 1.0mM of $Fe^{3+}$, and activated by 1.0mm of $Li^{}47+$. The absence of glucose after thin layer chromatography of reaction products revealed that the purified enzyme contains no cellobiase or laminarinbiase activity. The loberation of ki, tri-and tetra-saccharide as reaction products can be explained by endoaction of the enzyme.

  • Lithospermum erythrorhyzon has been used as a red fooddye and traditional Chinese medicine to treat wounds, skin diseases and burns. Platelet activation plays an important role in thrombosis and haemostasis. Here, we studied the inhibition of platelet activation and its active compound from the root of Lithospermum erythrorhyzon. Its ethyl acetate extract inhibited the aggregation of washed rabbit platelets induced by collagen or thrombin. Five naphthoquinone pigments , shikonin , acetylshikonin , is obutylshikonin, $\alpha$-methyl-n-butylshikonin and $\beta$,$\beta$-dimethylacrylshikonin were isolated by means of high pressure liquid chromatography. The structures were determined by comparison of their proton nuclear magnetic resonance spectra. The potency of their inhibition was in the following order : $\beta$,$\beta$-dimethylacrylshikonin$\geq$$\alpha$-methyl-n-butylshikonin>isobutylshikonin>acetylshikonin>shikonin. It is suggested that the size of the aliphatic hydroxy group of shikonin is important for the enhancement of potency.

  • Safety of yellow pigment produced by Monascus sp. CS-2 was evaluated. Acute oral toxicity, pyrogen test, and histamine test, as well as antimicrobial activity were determined. The results obtained were; LD oral in mice was 132.5 mg/20 g, pyrogen test in rabbit was 5 mg/kg, and histamine test in cat was 10 mg/kg. Also the pigment was particularly sensitive to Bacillus subtilis (ATCC 6633), Sarcina lutea (ATCC 9341) and Staphylococcus aureus (ATCC 6538 P), whereas not sensitive to Pseudomonas pyosyanea (ACTC 10490), Bacillus var. mycoides (ATCC 11778), Bordetella bronchiseptica (ATCC 4617) and Staphylococcus epidermidis(ATCC 12228).

  • In this research chromoproteins, namely phycoerythrin and phycocyanin in Korean laver are isolated and purified and their characteristics and the contents of amino acid are investigated. Three kinds of laver, Yonpyungdo product, Jindo product, and Kohung product are used in the experiments. The results are as following: 1) 0.68g of phycoerythrin and 0.72g of phycocyanin are isolated from 50g of laver. 2) when observed by microscope (1,000 times amplification), the crystals of phycocrythrin have the shape of small needles and phycocyanin that of platelets. 3) Absorption spectra of purified phycoerythrin shows strong absorption peaks at 546, 526, and $496m{\mu}$,and phycocyanin shows strong absorption peaks at 610 and $546m{\mu}$. After the phycoerythrin and phycocyanin were kept at $4^{\circ}C$ for 24 and 48 hours. The peaks of phycoerythrin are slightly shifted and those of phycocyanin are decreased. 4) In amino acid analysis, sixteen kinds of amino acids are detected in phycoerythrin and phycocyanin. Amino acids found in the greatest amounts in phycoerythrin are glutamic acid, alanine, glycine and leucine. Essential amino acids are found to exist in phycocyanin in greater amounts than in phycoerythrin.

  • A simple, rapid, efficient method is for extraction of 13 synthetic water-soluble food colours (Tartrazine, Amarnth, Indigo carmine, New coccine, Sunset yellow FCF, Allura red AC, Eosine, Fast Green FCF, Brilliant Blue FCF, Erythrosine, Acid red, phloxine, Rose Bengal) by polyamide resin and for their quantitative by high performance liquid chromatography (HPLC). Colours (coal-tar dyes) were extracted with polyamide resin and then determinated by HPLC. The HPLC conditions using a reverse phase partition type column $(Nova-pak\;C_{18})$, photodiode array (PDA) detector and 1% Ammonium acetate / 60% acetonitrile in water as eluent, were acceptable for various kinds of colorants. By the use of the proposed method, a survey of coal-tar dyes was carried out on 20 samples and that were detected $4.76{\sim}133.47\;ppm$.
