Fabrication of TiO2 Impregnated Stainless Steel Fiber Photocatalyts and Evaluation of Photocatalytic Activity (TiO2 담지 스테인리스 강 섬유 광촉매 제조 및 광촉매 활성 평가)

  • $TiO_2$ impregnated stainless steel fiber photocatalysts ($TiO_2/SSF$) were fabricated to overcome inherent problems of powdery $TiO_2$ photocatalysts in water treatment. Adhesion strength of the impregnated $TiO_2$ was examined using an ultrasonic-cleaner. Photocatalytic activity was evaluated through decomposition experiment of methylene blue and formic acid. Bactericidal efficiency was evaluated through sterilization experiment of E. Coli and Vibrio Vulnificus. Adhesion strength of the impregnated $TiO_2$ was so high that more than 95% was left over even after the treatment in an ultrasonic-cleaner for 30 min. Methylene blue and formic acid were decomposed as much as 60% and 38% of the initial concentration and more than 99.9% of E. Coli and Vibrio Vulnificus were killed after 1 hour exposure to the prepared photocatalyst under UV irradiation. In the case of decomposition of formic acid, decomposition ratio increased if oxidants were added. Especially the decomposition ratio increased as high as 80% when hydrogen peroxide was added as an oxidant.

  • 수중방전을 환경분야에 적용하기 위한 플라즈마 부상법이 개발되었다. 플라즈마 부상법은 물 속에서 발생시킨 플라즈마가 가지고 있는 주요특성 중 물리적 특징인 쇼크웨이브, UV조사, 버블생성 등과 화학적 특징인 OH라디칼 및 염소산화물 생성 등을 이용하여 물 속에 존재하는 용존성 및 입자성 물질을 부상분리 기법으로 제거하는 공법이다. 유기물을 제거하는 기작으로는 침전, 여과, 분해 등이 있고, 이를 구현하기 위한 공정으로 중력침강법, 부상분리법, 멤브레인법, 미생물법 등이 있다. 이 중에서 가압공기부상법은 침강법에 비해 부지면적을 적게 소모하고 처리시간이 50% 이상 감소되는 특징이 있다. 가압공기부상법은 물 속에 공기를 과포화시킨 후 노즐을 통해 재분사할 때 발생하는 압력차에 의해 미세기포가 발생함을 이용하여 유기물을 분리하는 공법이다. 그러나, 가압용 장비 및 반송수가 필요하고, 미생물분리는 불가능한 단점이 있다. 이에 본 연구에서는 미생물살균과 유기물 분리가 동시에 일어나는 플라즈마를 이용한 부상분리기법을 개발하였다. 본 연구에서는 난분해성 용존유기물인 휴믹산 100 mg/L의 플라즈마 공기부상법에 의한 제거능을 확인하였다. 용존성 휴믹산을 입자성 물질로 전환하여 플록을 형성시키고자 알루미늄설페이트(Al2(SO4) $3{\cdot}18H2O$)를 100 mg/L 주입하였고, 침출수와 같이 염도가 높은 물을 모사하고자 35 g/L의 염화나트륨을 첨가한 상태에서 방전을 실시하였다. 방전에 사용된 전원은 EESYS사에서 제작한 펄스형 고전압 전원장치를 사용하였고 최대 15 kW의 출력 중 6 kW의 전력을 인가하였다. 전극 한 개는 2 mm 텅스텐봉을 세라믹튜브로 감싼 구조로 총 사용전극은 28개이다. 전극 한 개당 대략 200 Watt의 전력이 소모되며 이 때 최대의 버블이 생성됨을 확인하였다. 전극 1개에서 생성되는 버블의 부피는 14 mL/min 로 측정되었다. 버블의 크기는 평균 70 um이고 가압공기부상법에서 최적공기크기로 제시하고 있는 40~80 um 의 버블은 약 80% 가량 생성된다. 본 연구에서 사용된 반응시스템에서의 물의 높이는 약 500 mm 이고 전체 40 L의 수조가 3개의 벽으로 분리되어 4개의 수조로 분리되었다. 각 수조는 하부에 7개의 전극을 포함하고 있다. 플라즈마 발생시 생성되는 기포는 약 1분 방전 후에 포화농도에 도달하며 방전종료 후 약 4분간 수체 내에 남아있게 된다. 이를 공정에 적용하여 1분 방전 및 4분 휴지의 순서로 플라즈마를 인가하였다. 휴믹산 용액의 유량을 2 lpm 으로 운전하였을 때 최종 처리율은 94% 이고 이때의 대장균 살균능은 99%이다.

Identification of Spoilage Bacteria Isolated from Aseptic Packaged Cooked Rice and Application of Acidic Electrolyzed Saline Solution as Water-for-Cooked Rice (무균포장밥으로부터 분리된 부패 미생물의 동정 및 전해산화수의 취반수로서의 이용 효과)

  • In this study, it was investigated that the isolation and identification of spoilage bacteria from contaminated aseptic packaged cooked rice, the potential for application of acidic electrolyzed saline solution (AESS) as water-for-cooked rice and the microbiological safety of AESS-based cooked rice. Five strains of Bacillus subtilis and a B. cereus strain among the total six isolates were partially identified by biochemical method and by Microbial Identification System (MIS). The bactericidal effect of AESS was similar as 0.1% NaOCl and 70% ethanol solution, or better than that. All of the test microorganisms except Bacillus spp. that were exposed to AESS for five seconds were destructed. The effect of AESS against Bacillus spp. was much better than that of the two solutions and all of them were destructed or inhibited on exposure for five minutes. The pH value of cooked rice prepared using AESS was in the range of 3.6 to 4.3 and was not almost changed through the storage period. Various concentrations of cell suspension of Bacillus isolates were inoculated to cooked rice, which were prepared with tap water and AESS, and stored at $37^{\circ}C$ for two weeks. The result was shown that the bacteria in tap water-based cooked rice appeared normal cell growth, while they were completely repressed in AESS-based cooked rice.

  • 선박이 배출하는 안정수(ballast water)는 외부로부터 유해 생물들이 유입되어 전파해 오는 주요경로로써 해양환경의 매우 중요하고 위험한 일종의 하나이지만, 이에 대한 효과적인 처리방법은 아직까지도 개발되지 못하였다. 그러나 최근 강 전리방전을 이용하여 고 밀집 산소와 물분자를 고농도 수산자유기(OH: hydroxyl radical)로 전리, 활성입자를 발생시켜 신속히 확산시키면 넓은 범위에서 비교적 낮은 농도로 유해성 침입 생물을 잔류물 없이 저렴한 비용으로 살균제나 촉매제의 사용 없이 소멸시켜 처리하는 효과적인 새로운 녹색방법을 제안하였다. 또한, 수산기는 강 산화제로써(산화환원 전위는 2.80 eV), 적조생물을 신속, 효과적으로 사멸시켜 잔유물과 오염물 발생 없이 이상적으로 해양적조현상을 처리할 수 있는 활성물질이다. 고출력 강 전리장치를 활용하면 수산기 활성제의 발생 농도를 Sr104 이상으로 얻을 수 있으므로, 해양적조처리에 요구되는 문턱 값 농도(~l$\times$$10^{-6}$)를 충족시킬 수 있으며, 이 경우 적조생물 소멸처리시간은 불과 10 sec 내외이므로 선박 안정수 처리문제와 함께 적조발생의 난문제를 해양동력학적으로 동시에 해결할 수 있는 효과적인 기술이다. 실험결과로부터 시간당 1 k톤의 활성물질을 발생하는 수산기활성제 제조장치의 경우, 약 4$\times$$10^2$ $\textrm{km}^2$/h의 적조해면을 처리할 수 있으며, 그 비용은 약 US$l,000 정도에 상당하므로, 적조에 따른 경제손실과는 비교될 수 없는 저렴하고 효과적인 방법이다. 활성물질의 생성시간과 가공시간은 불과 수십 $\mu\textrm{s}$ 및 수 sec 에 불과하므로, 1 kton/h 용량의 수산기활성제 제조장치의 환산소비동력은 약 200 kW이고, 장치의 체적은 10~30 ㎥의 공간으로 충분하므로, 소형선박으로 상당면적의 적조피해를 효과적으로 해결할 수 있다.

  • Potassium iodide (KI) TRO sensor for measuring total residual oxidant which is an index for disinfection and neutralization in ballast water has been constructed. The results of UV absorption wavelength and absorbance test in various TRO concentrations show high linear correlation ($r^2=0.9825$) at 350 nm wavelengths and it can be possible to visualize numerical value of TRO concentration (0.1~7.0 mg/L). The result of comparison test according to TRO concentration between the KI based TRO sensor system and the DPD based TRO sensor system showed identical trend. Overall results indicated that the KI TRO sensing system could be applied for continuous TRO concentration measurement in ballast water treatment system.

Inhibitory Effect of Chlorine Dioxide on Phenoloxidase Activation of the Indianmeal Moth, Plodia interpunctella (화랑곡나방(Plodia interpunctella)의 페놀옥시데이즈 활성화에 대한 이산화염소의 억제 효과)

  • Phenoloxidase (PO) is an oxidizing enzyme and plays crucial roles in insect immunity and cuticle sclerotization. High oxidizing activity of chlorine dioxide gives effective control activities against microbes and insect pests. These allowed us to assess any inhibitory activity of chlorine dioxide against PO with respect to insect immunity. PO activities of the Indeanmeal moth, Plodia interpunctella, was detected in both hemocytes and plasma. Upon bacterial challenge, PO activity was significantly increased especially in plasma. However, the immune challenge coupled with chlorine dioxide treatment did not enhance PO activity. When different chlorine dioxide concentrations were incubated with activated PO by immune challenge, they did not inhibit the activated PO. These results indicate that chlorine dioxide suppresses PO activity by inhibiting PO activation.

Detection of Vibrio vulnificus in Fish Farm and Bactericidal Methods on this Bacteria (가두리 양식장의 Vibrio vulnificus 검출 및 제어 방법)

  • Detection of Vibrio vulnificus in fish farm and searching for the bactericidal methods on this bacteria were studied. To detect this microorganism in sea water, mud, fish and mussels, selective isolation methods and detection of vvhA gene were used from January to October,2000. V. vulnificus was detected from May when the water temperature was over $17^{\circ}C$. From June to September, higher than $19^{\circ}C$, this bacteria could be isolated from most of the samples. Freezing and refrigerating did not inhibit the growth of V. vulnificus. Citric acid did not show the bactericidal effect, but more than 500 mg/l of EDTA did. With the aid of UV and photocatalyst, $TiO_{2}$ showed bactericidal effect after 15 minute treatment. Photocatalytic system consisted of glass bead coated with $TiO_{2}$ and UV illumination showed bactericidal effect on V. vulnificus at the turnover rate of 0.2/min.

Attempted Synthesis of Carboxin Derivative through Ring Expansion Reaction on Solid Phase (고체상에서 환팽창 반응에 의한 카르복신 유도체의 합성시도)

  • Solid phase synthesis of 16, which is a derivative of the first systemic fungicide, carboxin 1 was described. Reaction of 1,3-oxathiolane derivative with solid resin of 4-hydroxy-3-nitrobenzophenone 6 gave 9 in 82% yield. Oxidation of sulfur in the solid 1,3-oxathiolane 9 by MCPBA followed by a ring expansion reaction under the acid catalyst afforded the corresponding dihydro-1,4-oxathiin derivative 12. Treatment of the solid 1,3-oxathiolane 9 with p-methoxyaniline resulted in 1,3-oxathiolane 14, 1,3-oxathiolane sulfoxide 15, dihydro-1,4-oxathiin 16, and acetoacetanilide derivative 17 in 41%, 35%, 14%, 10% yields, respectively.

Changes of Polyphenols and Polyphenol Oxidase Active Bands during Apple Wine Fermentation (사과주(酒) 발효과정중(醱酵過程中) Polyphenol성(性) 물질(物質) 및 Polyphenol Oxidase 활성(活性) Band의 변화(變化))

  • To examine enzymatic browning of apple wine, changes of active bands of polyphenol oxidase (EC 1.10.3.1) as well as polyphenol substances related to browning of apple wine were investigated during wine brewing. The decrease of total phenol was remarkably inhibited by the addition of sodium metabisulfite. In the meantime, auto-oxidation of catechol in a model system increased proportionally as the reaction pH and temperature increased. Catechol oxidation, however, was not detected at $4^{\circ}C$ below pH 5.0. Polyacrylamidegel electrophoretic patterns showed that the apple (Jonathan) indicated 4 bands with polyphenol oxidase activity, designated a, b, c and d whose Rm were 0.21, 0.30, 0.41 and 0.51, respectively. Among these, 2 bands, a and c remained until 5th day fermentation and only c band after 6th day fermentation. After pasteurization of apple wine at $60^{\circ}C$ for 30min, c band also remained.

Lipoxygenase and Off-flavor Development in Some Frozen Foods (일부냉동식품에서의 Lipoxygenase와 이취발생관계)

  • Several tests were conducted to study lipoxygenase activity and off-flavor developement in frozen sweet corn. Fresh corn contained about 60% of total lipoxygenase activity in the germ section. When non-blanched frozen sweet corn was stored at $-10^{\circ}F$, it developed off-flavor and most significant changes in the flavor profile of off-flavored sweet corn was $4{\sim}5$ times higher hexanal peaks. The high hexanal peaks observed in the sterilized sweet corn with added lipoxygenase, alone and in combination with other enzymes, suggested the fact that high hexanal peaks in off-flavored sweet corn could be due to an oxidative reaction of lionleic acid (and other unsaturated fatty acids) catalyzed by lipoxygenase. Based on lipoxygenase activity and linoleic acid content in sweet corn, this reaction occur most heavily in the germ section of sweet corn. There was a significant relationship between flavor score of frozen stored corn-on-the-cob and hexanal peak in the germ section of corn-on-the-cob. This result indicated that hexanal peak could be used as an objective index of off-flavor development in frozen sweet corn.
