  • The effects of forest fires on the activity of microbial communities in topsoil and subsoil were investigated. Samples were collected from Korean forest soils comprising mainly igneous and sedimentary rocks. Analysis of beta-glucosidase, found higher microbial activity in sedimentary rocks than in igneous rocks. Enzyme activity was not observed immediately after fire, but was restored over time. The enzyme activity of subsoil was inhibited by 33~46% compared with that in the topsoil, regardless of soil damage. The effect of fire on the availability of microbial substrate was investigated using EcoPlate. The percentages of average well color development values of damaged and normal topsoil were 52.7~56.8% and 62.3~83.6%, respectively. Forest fires appear to affect the diversity and substrate availability of the subsoil microbial community by accelerating the decomposition of soil organic matter. The Shanon index, representing microbial biodiversity, was high in the topsoil of all samples; it was higher for soil microorganisms in sedimentary rocks than in igneous rocks, and higher in topsoil than in subsoil.

  • This study was conducted to predict the distribution of rare·endemic plants (Sophora koreensis Nakai) in the border forests where wildfire damage occurred and to quantify the damage. For this purpose, we tried to derive more accurate results through forest area damage (NBR) according to the Burn severity of wildfires, damage by tree species type (Vegetation map), and MaxEnt model. For Burn severity analysis, satellite imagery (Landsat-8) was used to analyze Burn severity (ΔNBR2016-2015) and to derive the extent of damage. To prepare the Vegetation map, the land cover map prepared by the Ministry of Environment, the Vegetation map prepared by the Korea Forest Service, and the vegetation survey conducted by itself were conducted to prepare the clinical map before and after the forest fire. Lastly, for MaxEnt model analysis, the AUC value was derived by using the habitat coordinates of Sophora koreensis Nakai based on the related literature and self-report data. As a result of combining the Maxent model analysis data with the Burn severity data, it was confirmed that 45.9% of the 44,760 m2 of habitat (predicted) area of Sophora koreensis Nakai in the wildfire damaged area or 20,552 m2, was damaged.

  • This study was carried out to evaluate the weight loss rates, carbon and nitrogen dynamics of wood stakes following soil amendment treatments (CLB: compound fertilizer + lime + biochar; LB: lime + biochar) in a post-fire restoration area, Ulsan Metropolitan city, southern Korea. Soil amendments in the fire-disturbed area were applied to two-times (Mar. and Jun. 2015, 2016) during the study period. Wood stakes on Mar. 2015 were buried at a top 15cm of mineral soil in two soil amendment and control treatments of Liriodendron tulipifera, Prunus yedoensis, Quercus acutissima, Pinus thunbergii plantations and an unplanted area in the post-fire restoration area. Wood stakes were collected at Oct. 2015, Mar. 2016 and Oct. 2016 to measure weight loss rates, organic carbon and nitrogen concentrations. Weight loss rates of wood stakes were not significantly affected by soil amendment treatments. However, remaining carbon of wood stakes were lowest in the control treatment (43.7%), followed by the CLB (71.3%) and the LB (71.6%) treatments. Remaining nitrogen of wood stakes was less in the control treatment (29.7%) compared with the LB treatment (52.6%). The results indicate that carbon and nitrogen mineralization of wood stakes in post-fire restoration area were delayed by soil amendment treatments.

  • Forest soils were analyzed on their biological and physicochemical properties for the ecological restoration of burnt forest soil using organic wastes and proper microorganisms. Three kinds of soil samples were collected from undamaged soil(US), naturally restoring soil(NS) and artificially restoring soil(AS). All soil samples were sandy soil and acidic soil, ranged pH 5.34~5.78. Moisture content was higher in the soil of NS region. And the others were similar. Total organic matter and soluble sugar were higher at the surface, generally. Heterotrophic soil microbes were abundant at the surface soil of NS and subsoil of AS. Dehydrogenase, cellulase and phosphatase activities were higher at the NS soil. Especially, Dehydrogenase activity as primary index of soil microbial process showed high correlationship with moisture content(r=0.90, P < 0.05).

  • Changes of biochemical and genetic diversity of microbial communities in the soil damaged by a forest fire were analyzed. Soil samples were collected from Gangnung area where a forest fire was broken out in 2000. Two soil samples were from the burnt area, one from the naturally restoring soil (NS) and the other from the artificially restoring soil (AS). A normal, unaffected soil sample (US) was also included as a control. For the biochemical diversity, each sample was directly applied to the BIOLOG system, and the cluster analysis through a statistic process (SPSS) were performed. Genetic diversity was analyzed through DGGE using 16S-rDNA amplified from soil DNA. Among the samples tested, top soils of US and NS, and sub soil of NS revealed more than 70% of the similarity value in biochemical diversity. In case of genetic diversity, however, the similarity value was found to be in the range of 53% to 68% in all samples. This result indicates that the biochemical diversity is not always correlated with the genetic diversity in the analysis of microbial communities.

  • We have isolated two bacterial strains, FM-02 and AL-02, which produced EPS from forest soil for the restoration of forest fire by promoting soil aggregation. FM-02 was found to be Gram negative rod and belong to Beta Proteobacterium sp. through 16s-rDNA sequence analysis, and AL-02 was Gram positive rod and showed 81% of similarity to Zoogloea sp. through the analysis of 16s-rDNA sequence. FM-02 and AL-02 produced about 1.8g and 8.3g of EPS, respectively, per 1L of culture as dry weight. Flocculation activity (FA) was also measured in two strains. FM-02 showed 2.31 FA against active carbon, and AL-02 showed 6.21 FA against kaolin clay. From these results, we expect that AL-02 strain will be applied as a good biological material for the reduction of forest soil erosion by wild and rain after fire through promoting coagulation of soil particles.
