본연구는 조경소재로 이용가능성이 크지만 현재 자생지가 파괴되어 복원이 요구되고 있는 야생자란의 대량번식을 위해 무균배양시 배지 내 담함량의 변화와 펩톤, 트립톤 등의 첨가가 종자발아와 계대배양 후 유식물의 생육에 미치는 영향을 알아보고자 실시하였다. 배지 내 펩톤과 트립톤의 첨가는 발아에 영향을 주지는 않았지만, 당의 함량은 그 농도가 10g/L까지 증가함에 따라 발아율을 높였다. 또한 발아 후 유식물의 생육시 당의 첨가는 뿌리의 생육을 두드러지게 향상시켰으며, 생체중도 거의 2~3배정도 많았다. 하이포넥스 배지(대조구)에서는 높은 발아율을 보였지만 유식물의 생육은 트립톤 첨가배지(2g/L)에서 많았는데 엽수, 뿌리수, 엽장 근장, 생체중 등이 모두 다른 처리구의 2~3배에 이르는 초기생육을 보였다. 계대배양 이후의 생육상은 펩톤 첨가배지에서 가장 많은 생육량을 보였는데 특히 엽장과 엽폭 그리고 근장이 다른 처리구보다 월등히 높은 경향을 나타냈다. 생체중도 한 개체당 0.18g으로 가장 높게 나타나 펩톤의 첨가가 계대배양 이후의 생육을 크게 촉지시키는 것으로 나타났다. 결론적으로 하이포넥스 배지에 트립톤 2g/L를 첨가하였을 때 발아율과 유식물의 생육이 다른 배지에 대해 매우 양호한 것으로 나타나 자생 자란의 종자발아용 배지로 가장 적당한 것으로 사료된다. 그리고 이후 계대배양시에는 펩톤의 첨가 (3g/L)가 유식물의 생육을 가장 촉진시키는 것으로 나타났다.
This study was conducted to examine the growth characteristics of seedlings of several major fruit vegetables and compare them between plants grown with the conventional plug seedling system (Plug) and those grown with the cylindrical paper pot system (CPP). There were no significant differences in shoot growth characteristics, such as plant height, leaf area, dry weight, and fresh weight, between tomato, cucumber, and watermelon plants grown with the CPP and Plug systems. The total root lengths of tomato and cucumber plants grown with Plug were longer than those grown with CPP at the beginning of seedling growth, and the total root lengths of watermelon grown with Plug were longer than those grown with CPP throughout the whole seedling period. The length of root that separated five steps according to the diameter of the root of tomato did not differ between CPP and Plug, but those of cucumber and watermelon were 20-251% longer with Plug than with CPP for all kinds of roots. The fresh weight of Plug-grown tomato roots was about 30% heavier than that of CPP-grown tomato during the seedling period, but the difference decreased as growth progressed. Finally, there was no difference between plants before and after transplanting. The fresh weights of the Plug-grown plants were about 20-30% heavier than those of CPP-grown ones in terms of the whole seedling. After transplanting to the Wagner pot, the shoot growth of Plug-grown watermelon plants in terms of dry weight was low until 7 days had passed. However, this tendency was reversed from 8 days after transplantation onward. In conclusion, there were no significance differences in the growth of shoots between plants grown with the CPP and Plug systems. However, root development was better with Plug than with CPP.
Increase in nodulation and nitrogen fixation was achieved partly through the isolation of supernodulating soybean mutant plants. This experiment was conducted to compare nodulation, nitrogen fixation, and early growth characters of wild type 'Bragg' with those of its supernodulating soybean mutant, 'nts 382' and 'nts 246'. At 31 days after planting, nodule dry weight of nts mutants was 2.5 to 3.7 times greater than that of Bragg. Higher nodulation of nts mutants showed the reduced top growth, indicating that photosynthates might be translocated and used for nodule growth attached to the root system. Total acetylene reduction activity was higher in nts mutants than Bragg, whereas specific acetylene reduction activity of nts mutant was the half of that of Bragg. Mixture of nts mutants and Bragg did not affect nodulation characters each other, suggesting that factors affecting supernodulating characters exist inside rather than outside the root system.
This study has been conducted to assess and analyze the damage of Codonopsis lanceolata by differential inoculation levels of Meloidogyne hapla in pot and plot. As the density of M. hapla increased, early growth of C. lanceolata was inhibited 60% in vine and 54% in root. However, growth of C. lanceolata was not remarkably different by density of M. hapla in the plot test. This result may be attribute to low density of M. hapla in the plot.
Superoxide dismutase (SOD) activities were investigated from the portions of shoots and roots in accordance with developmental stage and in response to environmental stress and antioxidants using Rhodiola-seedlings. The rates of SOD activities were revealed highly at the portion of roots and its tip of seedlings in the latter stages rather than the initial stages. SOD activities of seedlings in the initial stages treated with sodium chloride and cadmium as environmental stressors showed the decrease by 15 and 30% with respect to the control, respectively. However, in spite of stressor-treatments, the activities in the roots were increased according to the growth period showing a maximum rate of up to 45%. Also, SOD activities of the seedling treated with ascorbic acid as a antioxidant were increased by 46% of control value, but this was similar to the rate revealed in the presence of stressors. These results suggest that SOD activities in Rhodiola-seedlings may be related with the important defence-system against injurious environments.
The experiment was investigated the effects of lower root zone temperature on shoot and root growth of 'Akihime' strawberries in aeroponics in cold season. Root growth was decreased with decrease of root zone temperature, especially in lateral roots. Elongation of main roots was highest in $18^{\circ}C$ of root zone temperature. Number of lateral roots and length of lateral roots were lowest in $8^{\circ}C$ of root zone temperature. Lower root zone temperature resulted significantly in decrease of fresh weight of root and shoot and leaf area of strawberry. But there were no significant statistical differences in shoot fresh weight and leaf area in $8^{\circ}C$ and $13^{\circ}C$ of root zone temperature. Leaf length, leaf width and number of leaves of strawberry were decreased in lower root zone temperature. The results of this experiment will be utilized in the winter season cultivation for strawberry in hydroponics.
This experiment investigated the effect of various moisture levels on the top and root growth of barley and its yield. Resistant varieties to excess moisture injury shortened plant height, but tillered more, having creeping plant type compared with ordinary one, and greater portion of root of this varieties occured at the deeper soil depths with longer root length. However, susceptible one to excess moisture injury showed an inversed trend when compared with the untreated one. Oversaturated treatment compared with the untreated increased a little bit soil temperature and Eh of soil. Increased root activity and increased yield were obtained with resistant varieties compared with the untreated. Lower root activity and less yield than those of the untreated were observed in both extreme treatments. Yield reduction in both the extreme treatments was mainly due to reduction of tiller number, grain number and 1, 000-grain weight. Highest yield reduction was marked at the stage of internode elongation in the oversaturated and at the booting stage in the overdried treatment.
This study was conducted to elucidate the effects of green manure crop on growth characteristics and saponin contents of Platycodin grandiflorum Ridix roots for organic farming. Rye, hairy vetch and rye+hairy vetch were cultivated as green manure crops with several different methods before transplanting P. grandiflorum Radix. In root width and number of fine-roots, when the highest scores (23.9 mm, 25.7) was recorded in rye pre-cultibated plot in $2^{nd}$ year cultivation. Fresh weight of the Platycodin grandiflorum Ridix root was no significant difference among the different green manure crop treatments, but fresh weights of the Platycodin grandiflorum Ridix root of all green manure crop treatments were higher than that of control. The total content of saponin was highest detected in Platycodin grandiflorum Ridix roots of the hairy vetch plot (1,106 mg/100 g) and rye plot (1,693 mg/100 g) in $1^{st}$ and $2^{nd}$ year cultivation, respectively. Therefore, we confirmed that precultivation of the green manure crops showed significant differences in the growth characteristics and saponin contents of the Platycodin grandiflorum Ridix root, and growth and saponin contents in the Platycodin grandiflorum Ridix root of rye precultivated plot was much higher in $2^{nd}$ year cultivation than $1^{st}$ year cultivation.
