Effect of pH, Temperature, and added Sucrose on the Production of Vitamin $B_{12}$ and Riboflavin by Bacillus megaterium and Enterobacter aerogenes (온도, pH 및 첨가된 Sucrose가 Bacillus megaterium 과 Enterobacter aerogenes 에 의한 비타민 $B_{12}$ 와 Riboflavin 생산에 미치는 영향)

  • Optimal pH temperature and sucrose content for the production of vitamin B$_{12}$ and riboflavin by Bacillus megaterium and Enterobacter aerogenes was studied by microbiological analysis. Optimal pH for the production of B$_{12}$ was 6.0 by B. megaterium while the pH for E. aerogenes was 5.0. However, upon the addition of sucrose the optimal pH for B. megaterium shifted to 7.5 but E. aerogenes remained at pH 5.0. In the absence of sucrose, pH did not influence the yields of riboflavin produced by either bacterium. Addition of sucrose stimulated synthesis of riboflavin by both bacteria. Temperature had little effect on the production of vitamins by either bacterium.

Changes in β-Carotene, Vitamin E, and Folate Compositions and Retention Rates of Pepper and Paprika by Color and Cooking Method (고추와 파프리카의 색과 조리방법의 차이에 따른 베타카로틴, 비타민 E, 엽산의 함량과 잔존율)

  • ${\beta}-Carotene$, vitamin E, and folate contents according to color and species of Capsicum annuum were investigated. In addition, four C. annuum species were cooked by using different methods (boiling, pan-cooking, pan-frying, deep-frying, steaming, roasting, and microwaving), and retention rates of ${\beta}-Carotene$, vitamin E, and folate affected by color and species were evaluated. Weight loss was observed in all paprika and pepper species using all cooking methods. ${\beta}-Carotene$ and folate contents of C. annuum samples were significantly different by color, species, and cooking method (P<0.05), whereas vitamin E contents were not significantly affected by species. ${\beta}-Carotene$, vitamin E, and folate contents (mg/100 g) of raw C. annuum showed ranges of 95.4 (green pepper)~2,441.1 (red pepper), 0.7 (green paprika)~4.2 (red paprika), and 6.2 (green pepper)~148.7 (red pepper). As a result, the highest ${\beta}-Carotene$, vitamin E, and folate contents were detected in deep fried red pepper, roasted red pepper, and boiled red pepper, respectively, among the cooked samples. Retention rates of C. annuum varied by cooking method, resulting in ranges of 77.3% (boiled red pepper)~356.4% (roasted green pepper) for ${\beta}-Carotene$, 2.0% (microwaved red pepper)~789.3% (deep-fried green paprika) for vitamin E, and 20.9% (microwaved red pepper)~445.1% (steamed green pepper) for folate. Over 75% retention rates were observed for ${\beta}-Carotene$, vitamin E, and folate in all samples except for red pepper. However, in red pepper, vitamin E and folate retentions were remarkably lowered by deep frying and micro-waving, resulting in rates of 2.0~32.1%. This study provides information on effective cooking methods for vitamin retention in C. annuum depending on color and species.

Effects of Vitamin E enhanced transgenic soybean cultivation on insect diversity (비타민 E 강화콩 재배가 곤충다양성에 미치는 영향)

  • This study was carried out to develop of environmental risk assessments and the biosafety guide for Vitamin E enhanced transgenic soybean at LMO (Living Modified Organism) isolation field. In LMO quarantine area of National Institute of Agricultural Sciences, insect species diversities and population densities on vitamin E enhanced transgenic soybean and non-GM soybeans (Willams 82 and Seoritae) were investigated. A total of 17,717 individuals of 77 species from 8 orders were collected in LMO isolation field. In three type soybeans field, total of 5,250 individuals in Vitamin E enhanced transgenic soybean, 5,510 individuals in Willams 82, and 6,957 individuals in Seoritae were collected, respectively. There was no difference between the population densities of insect pests, natural enemies and other insects on Vitamin E enhanced transgenic soybean and Willams 82, while natural enemies density on Seoritae was higher than on Vitamin E enhanced transgenic soybean, but insect pests density on Vitamin E enhanced transgenic soybean was higher. These results provided the insects diversity for risk assessment survey of Vitamin E enhanced transgenic soybean and suggested that the guideline could be useful to detect LMO crops.

Detection and Measurement of Retinoic Acid in Human Liver Samples (간 조직내의 Retinoic Acid 검출 및 측정 방법)

  • Retinoic acid. the active metabolite of vitamin A. was detected in the human liver for the first time using a new method. A rapid and sensitive technique has been developed using gradient-elution. reverse-phase high performance liquid chromatography. This assay. with simultaneous multiwavelength detection at 294nm, 325nm and 450nm after saponifcation of liver samples. allows us seperation and quantitation of vitamin E, retinoic acid, total retinoids and various carotenoids in one small sample. The proportion of retinoic acid to total retinoids in human liver appears to be quite $consistent(2.4\pm0.2%$ ). With low vitamin A storage in liver, detection at another wavelenth 354nm would increase the sensitivity for retinoic acid of small quantity This method of analysis could be used for other tissues like red blood cells, plasma or serum, also. Hepatic retinoic acid level with total retinoids and carotenoids would serve a better indicator of functional vitamin A nutriture especially for those with disease requiring needle biopsy of liver.
