Genetic and Morphological Relationship among Korean Hylotelephium Species(Crassulaceae) (한국산 꿩의비름속(돌나물과) 식물의 유전적, 형태적 유연관계)

  • Genetic and morphological studies using morphological and isozymic data from 20 populations of 6 Korea Hylotelephium were conducted to resolve the species relationships and the origin of polyploid species. The results of morphological studies suggest that triploid H. viviparum is closely related to H. verticillatum, and the high frequency alleles of two species were very similar each other in isozyme analyses. Unique fixation of $PGI-1^b$ in two species supports that H. verticillatum should be the diploid ancestor of H. viviparum. However, a unique $ALD-1^c$ allele is present in H. viviparum, and the lack of the common alleles of H. erythrostictum and H. spectabile in this species eliminated them from the candidates of another tetraploid ancestor of H. viviparum. Based on the high morphological and genetic similarities, our results supported that the Korea endemic H. viridescens might be originated from the H. erythrostictum or share a common ancestor with it. Besides, H. ussuriense is closely related with H. erythrostictum and H. viridescens.

Antioxidant Activities and Quality Characteristics of Sulgidduk Added with Portulaca oleracea L. (쇠비름 첨가 설기떡의 품질 특성 및 항산화성)

  • The objective of this study was to develop a healthy Sulgidduk, a kind of rice cake, added with Portulaca oleracea L.. The effects of P. oleracea L. paste (0% 1%, 3%, or 5%) on the quality characteristics of Sulgidduk were evaluated. As the concentration of P. oleracea L. paste increased, pH decreased and acidity increased. The reducing sugar contents (%) increased with the amount of P. oleracea L. paste. The Hunter a and b values of Sulgidduk increased with an increase in P. oleracea L. paste concentration, whereas L value decreased. DPPH radical scavenging activity and SOD like activity of P. oleracea L. Sulgidduk increased with increasing P. oleracea L. paste contents. The sensory results show that overall preference of Sulgidduk with a P. oleracea L paste content of 3% was higher than those of other treatments.

Storage Characteristics and Retrogradation Properties of Sulgidduk Added with Portulaca oleracea L. (쇠비름 첨가 설기떡의 저장성 및 노화 특성)

  • The purpose of this study was to determine the storage characteristics of Sulgidduk, a kind of rice cake, added with Portulaca oleracea L. The effect of P. oleracea L. paste (0, 1, 3, or 5%) on the storage qualities of Sulgidduk was evaluated during storage period at $20{\pm}2^{\circ}C$ for 3 days. As the amount of P. oleracea L. paste increased, loss of water in P. oleracea L. Sulgidduk decreased. Textural properties by texture profile analysis showed that hardness of Sulgidduk added with 5% P. oleracea L. paste was the lowest among treated samples. However, the hardness of all Sulgidduks increased during storage, regardless of the addition amount of P. oleracea L. paste. In accordance with the texture results, differential scanning calorimetry exhibited that the enthalpy of Sulgidduk with 5% P. oleracea L. addition was the lowest, indicating the delaying effect of P. oleracea L. paste on retrogradation of rice cake. From these results, the addition of P. oleracea L. to Sulgidduk extended shelf-life by delaying retrogradation.

A newly naturalized species in Korea: Amaranthus powellii S. Watson (Amaranthaceae) (한국 미기록 귀화식물: 민털비름(비름과))

  • A newly naturalized plant, Amaranthus powellii S. Watson was found Jail-dong, Uijeongbu-si, Gyeonggi-do, Korea. This species is native to Southwestern USA and Northern Mexico, and it is also naturalized in worldwide of temperate regions. This species was distinguished from the related species, A. patulus Bertol. and A. hybridus L., by leave blades broadly elliptic, rhombic or lanceolate, petals unequal, without conspicuous green mid-veins, and utricle gradually narrowed toward the stigma. Korean name was given as 'Min-teol-bi-reum' to A. powellii S. Watson, based on the character, it was hairy less than the three related species, A. patulus Bertol., A. hybridus L. and A. retroflexus L., conspicuously. Descriptions for mophological characters, photographs, illustrations and keys to allied taxa were provided.

Effect of Ethanol Extract from Portulaca oleracea L. on the Lipid Metabolism in Rats fed Hypercholesterol Diet (고콜레스테롤혈증 유발식이에서의 쇠비름 에탄올 추출물이 흰쥐의 지질대사에 미치는 영향)

  • 쇠비름(Portulaca oleracea L.)은 쇠비름과(Portulacaceae)에 속하는 일년생 초본으로 우리나라 각지에 5~9월에 걸쳐 자생하고 있다. 한국보다는 중국에서 식용으로 애용되고 있으며, 해열, 이뇨, 산혈 등의 효능이 있어 임질, 요도염, 유종, 대하증, 임파선염 등에 약용으로 사용하고 있다. 쇠비름의 성분으로는 vitamin $B_1$ 및 C, saponin, tannin, flavonoid, dopamine, noradrenalin 등이 보고되었다. 쇠비름에 관한 연구로는 항산화효과, 항균효과 및 해독효과가 발표되었으나, 고지혈증에 관한 연구는 미흡한 실정이다. 따라서 본 연구에서는 고콜레스테를 유발식이에서 쇠비름 에탄을 추출물이 혈청 중 지질에 미치는 영향에 대해 검토하고자 하였다. 실험동물은 6주령의 Sprague Dawley 흰쥐 수컷을 각 실험군당 10마리씩, AIN-93M을 기본으로 하여 대조군(고지방식이), 쇠비름 에탄올 추출물 투여군의 3군(고지방식이+25mg/kg, 75mg/kg, 125mg/kg)으로 나누어 4주간 실험 식이를 급여하고 사육기간 중 사료 섭취량, 체중증가율, 식이효율을 측정하였다. 실험 종료 후, 혈액을 채취하여 혈액에서의 total lipid, total cholesterol, triglyceride, HDL-cholesterol은 효소법을 이용한 kit(아산제약)을 사용하여 측정하였고, 심혈관계 질환의 위험도에 이용되는 동맥경화지수는(atherogenic index, AI) 계산식을 이용하여 산출하였다. 실험결과, 최종체중, 체중증가율, 식이효율 및 장기무게는 실험군 간의 차이를 나타내지 않았다. 그러나 고지방식이에 쇠비름 에탄올 추출물을 125mg/kg의 농도로 병합 투여했을 때 혈중의 total lipid, total cholesterol 및 triglyceride는 대조군에 비해 유의적으로 감소하였으며, 고지방식이에 의해 증가된 동맥경화지수도 대조군에 비하여 유의적으로 저하되었다. 그리고 고지방식이로 증가된 ALT 및 AST 활성이 쇠비름 에탄올 추출물 투여로 유의적으로 감소하였다. 따라서 쇠비름 에탄올 추출물은 혈청 중 지질 개선 효과와 간 기능 개선효과가 있는 것으로 사료되나, 어떤 성분이 콜레스테롤 저하 효과를 나타내는지 보다 자세한 연구가 요구되어 진다.

Two new naturalized species from Korea, Amaranthus hybridus and Crepis tectorum (한국 미기록 귀화식물: 긴털비름(Amaranthus hybridus)과 나도민들레 (Crepis tectorum))

  • Two new naturalized species, Amaranthus hybridus and Crepis tectorum, are reported here. Descriptions and illustrations of the species and comparison of morphology with related species are provided. Amaranthus hybridus was first found at Nanjido, Seoul and is widely distributed in the central and southern part of Korean peninsula. New Korean name is given as "Ginteolbireum". It is well distinguished from the related species, A. retroflexus and A. patulus, by its late flowering time (September to October), sparse hair on the leaves, similar length between tepal and fruit, and long inflorescence without division. Crepis tectorum was first found at the entrance of Weoljeongsa Temple in Pyongchang-gun, Gangwon-do and spreads rapidly to other areas. New Korean name is given as "Nadomindulle".

Inhibition of Tobacco Mosaic Virus Infection by the Crude Sap Extracted from Amaranthaceae Plants (비름과식물즙액에 의한 담배 모자이크 바이러스의 감염억제효과)

  • Crude sap, which was extracted from six Amaranthaceae plants, inhibited local lesion formation on Nicotiana glutinosa by tobacco mosaic virus(TMV) infection. Especially the remark. able inhibitory effect to TMV infection was shown on leaves of N. glutinosa precoated with the sap from Amaranthus mangostanus. The inhibitory activity of the sap from A. mangostanus was stable to storage in vitro for I day and to dilution 1/4 of the sap with distilled water. However, its activity was lost when the sap was heated at $70^{\circ}C\;to\;100^{\circ}C$ for 10 minutes. When the leaves of N. glutinosa precoated with the sap were sprayed with water, the inhibitory effect to TMV infection was maintained for 2 days. The A. mangostanus sap readjusted pH 3, pH 5, or pH 9 with 1 N HCl or 1 N NaOH did not decline the inhibitory action but the sap absorbed with $5\%\;to\;15\%$ charcoal completely lost their action. The protein components purified from A. mangostanus sap revealed three major bands by $5\%\;to\;15\%$ polyacrylamide gel electrophoresis and the top component of which showed the inhibitory action to TMV infection.

  • 1) Optimum temperature was $15^{\circ}C$ for C. bursa-pastoris, $20^{\circ}C$ for C. album, $30^{\circ}C$ for P. oleracea, A. lividus, E. crus-galli, D. sanguinalis, and 4 showed wide range of germination temperature. 2) Emergence of C. bursa-pastoris, and C. album was best at $14.8^{\circ}C$ of soil temperature, E. crus-galli at $23^{\circ}C$, E. indica, A. lividis and P. oleracea at $27.1^{\circ}C$ and A. retroflexus and D. sanguinalis at $31.1^{\circ}C$. 3) A. retroflexus and P. oleracea started to germinate at 30% water absorption stage and A. lividus, C. album, S. viridis, and D. sanguinalis at 40% and E. indica at 70%. 4) Germination of weed species was decreased as PEG 6000 induced osmotic potential lowered. C. album, P. oleracea, D. sanguinalis, A. lividus, and Solanum nigrum were germinated at -5.0 bar osmotic potential and C. album and P. oleracea were germinated at -7.0 bar.

The Effect of Fermented Extracts of Portulaca oleracea against Campylobacter jejuni (쇠비름 추출물 발효액이 Campylobacterjejuni의 증식에 미치는 영향)

  • One of the main microorganisms causing diarrheal diseases is Campylobacter jejuni. Purslane or Portulaca oleracea is an edible plant containing polyphenols that has been widely used as a folk remedy for treatment of diarrhea for a long time. This study was performed to investigate the antimicrobial activity of fermented P. oleracea extracts made with probiotics and plant-origin lactic acid bacteria(PLAB) isolated from P. oleracea against C. jejuni. Lactobacillus rhamnosus, L. acidophilus, L. bulgaricus, L. delbrueckii, L. plantarum, Leuconostoc mesenteroides and Bifidobacterium longum were applied to P. oleracea to make a fermentation broth of purslane. Leuconostoc mesenteroides and the lactic acid bacteria isolated from P. oleracea grew best in the fermentation broth of P. oleracea extracts when the broth was combined with 2% yeast extract, 1% peptone, and 0.05 to 1% potassium phosphate. The number of viable cells in the fermentation broth containing purslane extracts after 48 hours increased to $1{\times}10^{12}\;CFU/m{\ell}$ and remained at $1.3{\times}10^{10}\;CFU/m{\ell}$ after refrigeration for 2 weeks. The pH and acidity of purslane-fermented broth after 48 hours of fermentation was 3.7 and 3.14, respectively, which show that the fermentation broth was within the range of the general standards of fermented dairy products. The antimicrobial activity of the fermented P. oleracea extracts was determined using the liquid culture method. The 10 $mg/m{\ell}$ concentration of the fermented P. oleracea extract made with Leuconostoc mesenteroides and the lactic acid bacteria isolated from purslane showed the strongest antimicrobial activity against C. jejuni. The fermentation broth of purslane with the probiotics retarded the growth of C. jejuni for 48 hours at $42^{\circ}C$.
