Evaluation of Angiotensin -I- Converting Enzyme Inhibitory Activity and Protein Changes of Enzymatic Hydrolysate Extracted from Hanwoo Loin and Round Myosin B (한우 등심과 우둔에서 추출한 Myosin B의 효소적 가수분해물의 단백질 변화와 Angiotensin -I- Converting Enzyme(ACE) 저해효과)

  • This study was performed to determine the protein profiles using sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) and Angiotensin-I-converting enzyme(ACE) inhibitory activity (IC50) as affected by the various meat cuts, digestion times with pepsin. Hydrolysates having the protein concentration of 10 ug/mL had approximately 36∼39% ACE inhibitory activities, regardless of meat cut and digestion time. Protein concentration and ACE inhibitory activity of the diluted hydrolysate increased after 1-hr digestion. In original hydrolysates, ACE inhibitory activities of loin had higher than those of round (P<0.05). In addition, non-heated hydrolysates had higher ACE inhibitory activities than heated counterparts. When myosin B was digested by pepsin more than 1 hr, improved ACE inhibitory activities were observed as compared to the non-digested control.

  • These experiments were conducted to investigate the hydrolysis products on the various oligosaccharides of Bacillus cirulans F-2 $\alpha$-amylase, and the hydrolysis rate on the various raw starches of Bacillus circulans F-2 $\alpha$-amylase, Bacillus amylotiquefaciens $\alpha$-amylase and Rhizopus niveus glucoamylase. The results obtained were as follows : 1. Maltotetraose, maltopentaose, maltohexaose, maltoheptaose and maltooctaose were hydrolyzed, but maltose and maltotriose were not hydrolyzed by Bacillus circulans F-2 $\alpha$-amylase. Among maltotetraose, maltopentaose, maltohexaose, maltoheptaose and maltooctaose, especially maltotetraose was hydrolyzed weakly by Bacillus circulans F-2 $\alpha$-amylase. 2. The Hydrolysis rate of oyster glycogen was slightly lower than soluble starch, amylose and amylopectin. 3. The hydrolysis rate of com starch was higher in shaking incubation than in stationary incubation, but the hydrolysis rate of potato starch was not definite according to kinds of enzyme. 4. On com, rice, arrowroot, high amylose corn, banana, sago, yam and potato starch, Bacillus circulans F-2 $\alpha$-amylase exhibited a remarkably higher hydrolysis rate than Bacillus amyloquefaciems $\alpha$-amylase and Rhizopus niveus glucoamylase.

  • Bovine whey protein are rich in cysteine, which is the rate limiting amino acid for synthesis of antioxidant glutathione(GSH). Some strains of Lactobacillus caseihas been reported to contain high level of GSH in cell extracts. The objective ofthis study was to determine whether enzymatically hydrolyzed whey protein isolate(WPI) and cell extract of Lb. casei HY2782 could increase intracellular GSH concentrations and protect against oxidant induced cell death in human prostate epithelial cell line (designated as RWPE1, and PC3MMM2 cells). Treatment of RWPE1 cellsandPC3MMM2 cells with hydrolyzed WPI (500g/ml) significantly increased GSH by28.2% and38.4% respectively. Compared with control cells receiving no hydrolyzed WPI(P<0.05). hydrolyzed WPI and Lb casei HY2782 cell extracts significantly protected RWPE1 and PC3MMM2 cellsfrom oxidant induced cell death compared with controls receiving no WPI. DNA damage associated with oxidant treatment was demonstrated by single cell gel (SCG) electrophoresis.

Lignocellulose Biodegradation and Interaction between Cellulose and Lignin under Sulfate Reducing Conditions (황산염 환원 조건에서 리그노셀룰로오스의 분해 및 리그닌과 셀룰로오스의 상호작용)

  • In this study, the biodegradation test on lignocellulose under sulfate reducing conditions was carried out. In particular, the interaction between cellulose and lignin was investigated with various g-cellulose/g-lignin (C/L) ratios: 42.15, 4.59, 2.51, 1.14 and 0.7. It was shown that the rate of cellulose degradation decreased in proportion to the lignin content. Assuming first order degradation kinetics, the consequences of competitive inhibition were graphically shown for different C/L ratios. The relation between cellulose reduction rate and C/L ratio was expressed by logarithm function with a determination coefficient of 0.97. Lignocellulose reduction rate was also described as a logarithm function of C/L ratio showing a inhibition effect by lignin. In the mean time, the rate of lignin decomposition was higher at C/L ratio of 2.51 and 1.14 compared with C/L ratios of 4.59 and 0.7, indicating that excessive extra carbon source is not appropriate for lignin biodegradation.

The Effect of Environmental Factors on the Hydrolysis Characteristics of Lipase (환경인자가 리파제의 가수분해 특성에 미치는 영향)

  • The effects of environmental and compositon factors, such as reaction time, metal ions, pH, agitation speed, the weight ratio of water to oil, and the weight of enzyme, on the hydrolysis of oils by Lipase-OF were investigated. In case of oils with low melting point, the optimum temperature of hydrolysis were the enzyme activity was maximum was 37$^{\circ}C$. However, when the melting temperature was higher than 4$0^{\circ}C$, the optimum temperature was around the fusion temperature. The activity of Lipase-OF decreased very rapidly with increase of temperature in the range of higher than 45$^{\circ}C$ and the activity perished above $65^{\circ}C$. The effect of agitation speed was investigated from 150 to 650 rpm. The hydrolysis of oils increased as the agitation speed increased up to 350 rpm, but it did not increase any more above 350 rpm. The weight ratio of water to oil was changed from 1 : 9 to 9 : 1 for the investigation of the effect on the hydrolusis. The weight ratio for maximum hydrolysis was 1 : 1. $Ca^{2+}\;and\;Mg^{2+}$ among various metal ions had some effect on the stimulation of hydrolysis. The optimum concentration of the ions was about 100ppm at which the hydrolysis increased, compared with that of distilled water, by 2 to 3%. The Optimum pH of Lipase-OF was 7. The hydrolysis decreased as the pH decreased as the pH decreased and also decreased as the pH increased. The content of enzyme affected the hydrolysis of oil. The hydrolysis increased with the content of Lipase-OF in the range of less than 0.013 wt% of substrate. However, the increase of hydrolysis with the content of Lipase-OF ceased above 0.013 wt%. The experiments investigating the effect of environmental and composition factors on the hydrolysis of oils showed that the optimum temperature was 37$^{\circ}C$, the pH 7, the concentration of $Ca^{2+}\;or\;Mg^{2+}$ 100 ppm, the agitation speed 350 rpm, the weight ratio of water to oil 1 : 1, and the content of Lipase-OF 0.013 wt% of substrate.

  • The purpose of this study is screening of microfloras involved in hydrolysis of seaweed tenella, seaweed fusiforme and green laver. This is a part of studies on the hydrolysis of seaweed using microorganisms. First, about two hundred microflora samples were obtained from mountain, rice field, dry field, sea, seaside and fish market in the vicinity of Yeosu. Thirty-three microflora samples were screened from the destruction of tissue in sea tangle and sea mustard. It was sufficient that results of the naked eye observation were obtained at eight microflora samples as a feces of bull, a decayed pine tree, a soil of dry field, the mud of the banks in a rice field, the water of a ditch in a rice field, the weed of the banks in a rice field, the water in a rice field and leaved in the air. Above all, extraction rate and contents of reducing sugar in extracts of seaweeds added a decayed pine tree(sample No. 8) and the water of a ditch in a rice field(sample No. 27) were showed high value. And the value of chemical analysis of the sample is much better in comparison with control. Accordingly the hydrolysis of seaweed using microorganisms in the inside of these microflora samples can be possible.

  • Experiments were conducted to determine effects of whole and processed corns on in situ disappearance rates of nutrients in the rumen and in vitro degradability of dry matter by rumen microorganisms. Whole corn(WC) was processed into four different types; ground corn(GC), cracked corn(CC), flaked corn(FC), and soaked corn(SC). In the in situ experiment, the rate of ruminal DM disappearance after 48 hour incubation was highest in GC(76.1%) and lowest in WC and SC(12% ; P<0.01). The rate of ruminal CP disappearance after 48 hour incubation was highest in GC and CC(48 and 38%, respectively; P<0.01). The rate of ruminal OM disappearance after 48 hour incubation was highest in GC(76.14%) and lowest in WC and SC(11.82 and 12.26%, respectively; P<0.01). In the in vitro experiment, the two-stage incubation technique was used to measure digestibilities of whole and processed corns. The digestibility of DM was higher in GC, CC and FC(86.95, 85.84 and 82.29%, respectively) than in WC(15.36%; P<0.01).

전해질 첨가 전기/UV 공정을 이용한 염료의 제거

  • UV공정은 NaCl 첨가량 증가에 따라 RhB 분해율이 감소하였고, 전기분해 공정에서는 증가하는 경향을 보였으나 전기/UV 공정에서 NaCl 농도가 0.25 g/L까지 RhB 분해속도가 증가하였으며 최적 NaCl 농도는 0.25 g/L로 판단되었다. UV와 전기분해 단일 공저의 COD 제거 합보다 전기/UV 공정에서의 COD 제거율이 높아 복합 공정에 대한 시너지 효과가 나타나는 것으로 사료되었다.

  • Biofiltration of volatile organic compounds (VOCs) was performed for 80 days in a biofilter packed with peat. The empty bed residence time was 3.2 min. for a gas mixture of isoprene, dimethyl sulfide, chloroform. benzene, trichlorethylene, toluene, m0xylene, o-xylene and styrene. After 34 days of acclimatization the removal efficiency for a 83 g/㎥ gas input was 93% at $25^{circ}C$ and 73% at $45^{circ}C$, respectively. The maximum cell density at $25^{circ}C$ was 1.12$\times$10(sup)8 cells/g. Removal efficiencies of m-xylene and toluene (91%) were better than that of benzene (86%). The first quarter of the packed column removed 60% of the incoming VOCs.

  • Composting facilities are operated with air and moisture control. Composting effects on two operating factors was analysed changing aeration rate with and without water addition to maintain the optimun moisture level. Though the composting facilties are provided with appropriate surroundings for compositing, operating temperature is set for decomposition rate. Accordingly control of decomposition phases was analysed by modeling the process of high and low decomposition phases with various operating temperature. A composting model of "The Library of Compost Engineering Software" developed by Roger T. Haug Inc. in U.S.A. was applied in modeling. As result of this study, operation with optimum moisture has more sensitive temperature to aeration fluctuation and lead to higher reaction rate with lower aeration than operation with poor moisture. Decomposition rate in composting facilities depend on slow decomposition phase because high rate decomposing substances already have been decomposed before entire process is not completed. In order to enhance decomposition rate of organics, effective decomposition in slow decomposition phase needs to be focused.
