Post-irradiation Changes in Antioxidant Activity of Spindle Tree Leaves from Different Areas (서식지에 따른 사철나무 잎의 이온화 방사선 조사 후 항산화 활성 변화 차이)

  • As the plants grow in a fixed place, they can be a good indicator which reflects the level of environmental pollution. It is necessary for them to develop a strategy to cope with stress under unfavorable environmental conditions. In this study, spindle trees ($Euonymus$ $japonica$) were collected from a clean area (Kijang) as well as a heavily polluted area (Onsan) to check applicability of irradiation combined with plant bioassay to environmental monitoring. The leaves were irradiated with 0, 50 and 100 Gy of gamma rays, and then evaluated for antioxidative capacity with 1,1-diphenyl-2-picrylhydrazyl (DPPH) assay and superoxide dismutase (SOD) analysis. The result shows that there was no significant changes in SOD and EDA (Electron Donationg Ability) in the samples collected from a polluted area. In the meanwhile, SOD increased in the samples from a clean area until 6 to 10 hours after irradiation, then it decreased gradually until 24 hours after irradiation. In conclusion, the plants in the polluted area have developed higher resistance to oxidative stress induced by ionizing radiation than those in the relatively clean area. Irradiation combined with plant bioassay on enzymatic activities and free radical scavenging capacity has proven to be a possible tool for biomonitoring the environmental pollution.

Solvent Extraction of Trace Mo(VI) in Natural Water Samples by Chelation and Ion-pairing (킬레이트 및 이온쌍 형성을 이용한 자연수 중 극미량 Mo(VI)의 용매추출)

  • The formation of Mo(VI)-alizarin red S chelate ion and its extraction into an organic solvent by ion-pairing were studied for the separative determination of trace Mo(VI) in natural water samples. Natural water 100mL was sampled in 250mL separatory funnel. After Mo(VI)-ARS chelate ion was formed by adding 0.01M alizarin red S solution 0.5mL to the water sample of pH 4.0, 0.2% aliquat-336 chloroform solution 10mL was added and the solution was vigorously shaked for about 30 seconds to from the ion-pair between Mo(VI)-ARS and aliquat-336, completely. The solution was stood for about 90 minutes. And the organic phase was taken for the absorbance measurement of the ion-pair at 520 nm. The content of Mo(VI) in sample was obtained from the standard calibration curve. Several extraction conditions such as pH, adding amounts of alizarin red S and aliquat-336, and shaking and standing times were optimized. This procedure was applied to the analysis of river and tap waters. It could be confirmed from the recoveries of over 99% in samples spiked with a given amount of Mo(VI) that this method was quantitiative in the determination of trace Mo(VI) in a natural water.

Characteristics of Mayonnaise Prepared with Palm Oil (팜유의 사용에 의한 마요네즈의 품질 특성)

  • To know the availability of double fractionated palm olein (DFPL) for vegetable oil in commercial mayonnaise preparation, oxidative stability and stability to cold test of DFPL, soybean oil and blended oils (blended soybean oil with DFPL) were tested. Mayonnaises with these oils were prepared and then emulsion stability at low temperature $(-5^{\circ}C)$ were compared. The oxidative stability of vegetable oil by Rancimat test showed that induction time of DFPL (26.9 hr) was longer when compared with soybean oil (13.4 hr), and became longer with increase of DFPL ratio in the blended oil. Emulsion stability of mayonnaises at low temperature $(-5^{\circ}C)$ was decreased with the increase of DFPL ratio in the blended oil. But, mayonnaise with blended oil of below 20% DFPL was comparable to that with soybean oil only. Among quality characteristics of mayonnaises with soybean oil and blended oil (soybean oil 85% plus DFPL 15%) the latter showed stronger oxidative stability and less flavor reversion during high temperature treatment. This result suggested that the possibility of DFPL to substitute for vegetable oil in the preparation of commercial mayonnaise.

Rapid Gas Chromatographic Screening of Dairy Products for Volatile and Nonvolatile Organic Acids (기체크로마토그래피법에 의한 유제품내 휘발성 및 비휘발성 유기산의 신속한 스크리닝)

  • A rapid gas chromatographic (GC) profiling method was applied to dairy products (milk and cheeses) for the simultaneous analysis of volatile and nonvolatile organic acids. Cheese samples were first made into aqueous samples by dissolving in water. The aqueous samples were then extracted with organic solvents after the acidification and NaCI saturation. The organic layers (diethyl ether : hexane= 1 : 1) were extracted with $NaHCO_3$ saturated solution with subsequent solid-phase extraction of the aqueous phases using Chromosorb P column/diethyl ether followed by triethylamine treatment. The resulting triethylammonium salts of acids were directly converted into volatile tert.-butyldimethylsilyl derivatives, which were analyzed by dual-capillary column GC, and GC-mass spectrometry. From milk and four cheese samples studied, 31 organic acids including 21 fatty acids and other hydroxy and dioic acids were tentatively identified. The amounts of the fatty acids were different among the kinds of cheese and thus the simplified retention index (RI) spectra of organic acids were useful for the visual pattern recognition of each sample, when the Direct Comparision method between cheese and a blind sample were attempted, it was quickly recognized to be a gouda cheese with the 999 ppt match quality value.

  • This study was conducted to investigate the chemical properties of soils under plastic house cultivation in northern Gyeonggi province(Yangju gun and Paju gun) in 1992. The results are summarized as follows: 1. Distributions of soil texture were in the order of sand loam(65%), silt loam(25%) and loam(5%). 2. Content of soil nutrient varied with difference with soil management practices and years of cultivation and chemical properties of soils. 3. Soil pH, Ca/Mg ratio and Mg/K ratio tended to be low, while other parameters high. 4. The average application rates of poultry dropping, cow dung and piggery waste were 6.0, 4.5, and 2.0ton/10a, respectively.

Frangrance and Chemical Composition of Essential Oil in Cone of Metasequoia (Metasequoia glyptostroboides Hu et Cheng) (메타세콰이아 (Metasequoia glyptostroboides)구과(毬果) 정유의 향취 및 휘발성 화학성분)

  • Essential oil was extracted from cones of metasequoia (Metasequoia glyptostroboides) by steam distillation, fragrance was determined by sensorial analysis and chemical compositions were analyzed by gas chromatography-mass spectrometry with solid-phase microextraction apparatus. Metasequia contained 0.40% of essential oil in the cone. Major impact fragrances of the essential oil were woody, coniferous and herbal, and minor impact fragrances were minty, spicy and oily. There were nine constituents in the essential oil : 8 hydrocarbons and 1 oxide. Constituents were limonene (66.18%), ${\delta}$-3-carene (11.11%), ${\beta}$-caryophyllene (6.66%), ${\beta}$-myrcene (5.92%), ${\beta}$-pinene (4.14%), caryophyllene oxide (2.39%), camphene (2.32%), ${\alpha}$-caryophyllene (0.85%), and tricyclene (0.43%). Herbal and minty frangrances could be due to limonene and ${\delta}$-3-carene, spicy frangrance to caryophyllene, woody and coniferous frangrances to ${\alpha}$-pinene and ${\beta}$-pinene, and oily fragrance to camphene.

Adsorption Column Chromatography for Simultaneous Determination of Multi-pesticide Residues (잔류농약 다성분 동시분석을 위한 흡착 크로마토그래피의 적용)

  • In order to develop the multi-residue purification method for 180 pesticides commonly used in Korea, many analytical methods on individual and multi- pesticide residues in the agricultural commodities and food product were examined. Through the modification of adsorption chromatographic methods used in Europe, the United States and Korea, the Florisil and silica-gel chromatographic systems were developed. Through these purification systems, elution profiles for all pesticides were examined. As the results, 145 pesticides were recovered in the range of 70-120% in Florisil clean-up system. The distribution of pesticides in the elution profile was 12 pesticides in the first fraction, 76 pesticides in the second fraction, 81 pesticides in the third fraction, 60 pesticides in the fourth fraction and 30 pesticides in the last fraction. And, in silica-gel system, 137 pesticides were recovered in the range of 70~120%. The distribution of pesticides in the elution profile was 22 pesticides in the first fraction, 59 pesticides in the second fraction, 102 pesticides in the third fraction, 46 pesticides in the fourth fraction and 8 pesticides in the last fraction.

Distribution of Heavy Metal in the Cell Components of Heavy Metal-Tolerant Microorganisms (중금속내성균의 세포내 중금속 분포)

  • Heavy metal-tolerant microorganisms, such as Pseudomonas putida, P. aeruginosa, P. chlororaphis and P. stutzeri which possessed the ability to accumulate cadmium, lead, zinc and copper, respectively, were isolated from industrial wastewaters and mine wastewaters polluted with various heavy metals. The distribution of heavy metal in the cell components, and amino acid compositions, was investigated. The distribution of heavy metal in the cell fractions of each heavy metal-tolerant microorganism grown for 20 hours in the basal medium containing 100mg/l of each heavy metal was investigated. In the case of cadmium-tolerant P. putida, lead-tolerant P. aeruginosa and copper-tolerant P. stutzeri, approximately $50{\sim}60%,\;30{\sim}40%$ and $10{\sim}17%$ of each heavy metal absorbed were distributed to cell wall, cell membrane and cytoplasm fractions, respectively. In the case of zinc-tolerant P. chlororaphis, approximately 32%, 55% and 13% of zinc were distributed to cell wall, cell membrane and cytoplasm fractions, respectively. These results indicated that the cell wall was a major adsorbing fraction of cadmium, lead and copper, and the cell membrane was that of zinc. Total amino acid content per gram of the cell grown in the culture media with heavy metal was higher than that of the cell grown in the culture media without heavy metal, and the content of acidic amino acids, such as aspartic acid(Asp.+Asn.) and glutamic acid(Glu.+Gln.) was higher than that of basic amino acids, such as histidine, lysine and arginine.

Petrological study on the Miocene Dangsari volcanic rocks, eastern part of Ulsan city, southeastern Korea (울산 동부 마이오세 당사리화산암류에 대한 암석학적 연구)

  • The Miocene volcanic rocks in the Dangsari area, eastern part of Ulsan city, are mainly composed of andesite lava flows and pyroclastic rocks. The andesite lavas are identified as two-pyroxone andesite, comprising phenocrysts of augite ($Wo_{43.2}$ $En_{41.0}$ $Fs_{15.8}$ ) and hyperthene ($Wo_{2.7}$ $65.8_{En}$ $_{Fs}$ 31.5). The andesitic pyroclastic rocks are largely composed of pyroclastic breccias with alternating tuff-breccia and lapilli tuff, which showing planar layering, and minor amount of andesitic tuff with thin deposits of interlayered tuffaceous shale. According to the petrochemical data, andesitic rocks belong to medium-K calc-alkaline andesite. The position of bulk composition on the AFM diagram and the presence of normative quartz and hypersthene indicate that the volcanic rocks are calc-alkaline. The trace element composition and REE patterns of andesite, which are characterized by a high LILE/HFSE ratio and enrichment in LREE, suggest that they are typical of continental margin arc calc-alkalic volcanic rocks produced in the subduction environment. On the discrimination diagram, the Dangsari volcanic rocks fall into the fields of subduction related continental margin arc volcanic province. The primary magic melts may be derived from about 15% partial melting of mantle wedge in the upper mantle under destructive plate margin. And the melt evolved to calc-alkaline andesite magma by fractional crystallization and the magma was a little contaminated with crustal materials.

Petrology of host granites and enclaves from the Bohyeonsan area, Euiseong Basin (의성분지 보현산 일대 화강암류와 포획암에 대한 암석학적 연구)

  • Mafic microgranular enclaves (MME) occur in the granites from the Bohyunsan area. The host granites are generally of granodioritic and granitic compositions. The MME can be divided into magic mineral clusters, quartz diorite and diorite according to their occurrence. Halter variation diagrams show linear trends between the MME and the host granites. Though the rim compositions of plagioclase in the host granites and the MME are similar the core compositions of plagioclase in some host granites show abnormally high An content. The Mg/(Mg+Fe) ratio of hornblende in the host granites gradually increase from the core to the rim. The chemical composition of minerals in the host granites had been affected by more marc magma composition. The modelling of major elements of the MME and hybrid host granites also indicate that they result from simple mingling/mixing between a dioritic magma and the host granite magma. The MME are thus interpreted to be globules of a more mafic magma which intruded the granite magma. Partial equilibration has been achieved between the MME and the host granites after they were commingled with each other.
