The Effects of Nitrogen Sources on the Expression of Nif Gene in Klebsiella pneumoniae Nif-Lac Fusants (Klebsiella pneumoniae nif-lac 융합변이주의 질소고정 유전자 발현에 미치는 질소원의 효과)

  • The effects of various nitrogen soruces on the expression of nif gene were investigated using nif-lac fusants of Klebsiella pneumoniae. K. pneumoniae UK 2979 was infected with Mudl lysate prepared by heat induction of K. pneumoniae UK 4482. About 80 nif-lac fusants were isolated and designated as LX series. In the prescence of $NH_4^+,\;{\beta}-galactosidase$ activities on nif-lac fusants were greatly repressed. Amino acids, such as serine, glutamine and asparagine, were found to support the growth of K. pneumoniae M5al quite well, and showed a repressive effect on ${\beta}-galactosidase$ activities of nif-lac fusants LX-9 and LX-22 in NFHM. Glutamic acid, histidine and arginine rendered poor growth but high activities of ${\beta}-galactosidase$. Good cell growth and high enzyme activity were observed when complex nitrogen sources, such as casitone, proteose pepone, were employed. ${\beta}-galactosidase$ activities of LX-9 and LX-22 in nitrogen free minimal medium increased sharply within first 4 hours.

Complete genome sequence of Spirosoma aerolatum KACC 17939T, a bacterium related to the DNA repair (DNA 복원에 관련된 박테리아 Spirosoma aerolatum KACC 17939T의 완전한 게놈 서열)

  • A Gram-stain-negative, yellow-pigmented bacterial strain, designated Spirosoma aerolatum KACC $17939^T$, was isolated from a biofilm of car air conditioner collected in Republic of Korea. In this study, we report the complete genome sequence of a bacterium Spirosoma aerolatum KACC $17939^T$ obtained using the PacBio RS II platform. The genome comprised of 7,959,595 bp with the G + C content of 48.3%, the genome included 6,640 genes were predicted, among them, 6,471 genes are protein-coding genes.

전처리공정이 천연유기물질의 움집에 미치는 영향

  • 정수공정은 "원수 -1 침사지 1 전염소 -1 전오존 -1 침전지 1 여과지 1 후오존 1 GAC 4 후염소 1 가정수"로 이루어져 있으며 전처리 공정으로 염 소와 오존을 주입하게 되는데, 여기서 전염소 처리 공정으로 인해 잔류 유리염 소가 수중의 유기물질과 반웅하여 유해성 유기염소 화합물 (i.e.. Trihalomethane, Haloacetonitriles, Chlorophenol 등)을 생성하는 것으로 알려져 있다. 이러한 이유로 고도 정수처리 공정에서는 강한 산화력을 지닌 대체 산화 제로서 오존을 이용하고 있다. 전오존 처리공정은 OOC와 탁도 제거에 있어 응 집제 주입량을 감소시키는 것과 더불어 전오존 효과 (산화, 생분해 증대, 살균 등)를 얻을 수 있는 것으로 알려져 왔다. 그러나 낙동강과 같이 유기물이 많은 원수에 과다한 오존이 주입되면 수중 의 유기물이 저분자화 또는 응집이 어려운 오존 산화물로 변화하여 응집제의 소비가 많아지게 된다. 실제, 오존을 응집 효과에 대해서 pilot plant로 운전한 결과 전오존에 의해 입자성 물질의 제거 효율은 향상된 반면에 유기물 제거는 뚜렷한 효과를 볼 수 없었다고 보고되었다 (류, 1997). 본 연구에서는 물금 지역의 원수와 응집-침전 공정까지 거치는 각각의 처 리수에 대해서 전처리가 응집에 미치는 영향을 천연유기물질 (NOM)의 조성 변화로 파악하였다. 그 방법으로 천연유기물질 (NOM)을 XAD-1, -4 수지를 이 용하여 소수성 물질 (hydrophobic components)과 친수성 물질 (hydrophilic components)로 분리 및 농축하여 용존 유기물이 처리 과정에서 어떻게 변화하 는지를 조사하여 개선된 상수 처리 시스템을 설계하는데 기초 자료를 제공하고 수질 관리 분야에서도 적용하고자 하였다. 상수원수인 물금 지역의 소수성 물질(hydrophobic components)은 75~80%, 친수성 물질(hydrophilic components)은 30~33%정도의 분포를 보였고, 전염소 및 전오존 공정을 거친 처리수에서는 각각 62.2-62.8%, 43.9~49.0% 및 50~ 55%, 40~57% 정도의 분포를 보였다. 그리고 웅집-침전을 거친 처리수에서는 그 분포가 77~82%, 24-48%였다. 전주리 공정을 통하여 소수성 물질(byoghobic components)의 분포가 감 소하는 것을 볼 때 전염소 및 전오존 처리가 용존유기물의 응집에는 오히려 역 효과를 나타내는 것으로 판단된다. 것으로 판단된다.

Stem Rot of English Ivy Caused by Phytophthora cinnamomi (Phytophthora cinnamomi에 의한 아이비줄기썩음병)

  • A stem and root rot disease of English ivy (Hedera helix) was found in the flower market, Jeonju, Chonbuk province, Korea in May 2007. A Phytophthora species was isolated from the diseased part of the plant. Based on the mycological properties and pathogenesis the isolate was identified as Phytophthora cinnamomi. The fungus produce ovoid sporangium which was ovoid to spherical and obpyriform which is noncaducous, non-papillate and averaged $33.8{\times}16{\mu}m$ in dimension. Chlamydospores were abundantly produced on agar media and sized about $19.7{\mu}m$ in diameter. The fungus was heterothallic and A1 mating type. Oospores were measured $26.2{\mu}m$ in size. Optimum temperature for growth of the fungus was 25 to $30^{\circ}C$. Up to our knowledge, this is the first report demonstrating the stem rot on English ivy caused by P. cinnamomi in Korea.

Optimum temperature for growth of the fungus was 25 to $30^{\circ}C$. Up to our knowledge, this is the first report demonstrating the stem rot on English ivy caused by P. nicotianae in Korea.

  • To reduce the redundancy within samples that resulted from uniform sampling method, nonuniform sampling or nonredundant-sample coding methods can be considered. However, it is well known that when conventional nonuniform sampling methods are applied directly to speech signal, the required amount of data is comparable to or mure than that by uniform sampling method like PCM. To overcome this problem, a new nonuniform sampling method is proposed, in which nonuniform sampling is applied to the low-pass filtered speech signal and higher band is compensated by 8 colored Gaussian random noise with various noise levels. By this method, speech signal waveform can be encoded by 1.8 times larger compression ratio than the conventional nonuniform sampling method.

  • A disease causing blights on leaves, stems, flowers, and pods of Vinca (Catharanthus roseus L.) was occurred in Aug. 2003 in Chengdo Peach Experiment Station. A species of Phytophthora was isolated from the diseased tissues. The causal fungus was identified as Phytophthora nicotianae on the basis of mycological characteristics and pathogenicity. Sporangia were ovoid to spherical, noncaducous, prominently papillate and averaged 38.0${\times}$31.0 ${\mu}{\textrm}{m}$ in dimension. Chlamydospores were abundantly produced on agar media and sized about 30.5 fm in diameter, The fungus was heterothallic and Al mating type. Oospores were measured 23.1 ${\mu}{\textrm}{m}$ in size. Optimum temperature for growth of the fungus was 25 to 3$0^{\circ}C$. This is the first report of occurrence of foliage blight of Vinca caused by P. nicotianae in Korea.

  • The off-axis tensile strength of the unidirectional carbon fiber reinforced plastic and the residual strength of impact damaged CFRP were measured and compared with the stress wave factor (SWF) of the specimens. The SWF values were measured to be decreased with the strength reduction in both cases and found to be useful for the nondestructive strength evaluation of unidirectional CFRP. The failure behaviour of the unidirectional CFRP during off-axis tensile testing was also monitored by acoustic emission(AE) method. The AE energy release showed the characteristic feature depending on the off-axis angle and this result was analyzed to be caused by the difference of the expected failure mode depending on the off-axis angle. The failure mode of CFRP was found to be analyzed by investigation of the peak amplitude distribution of AE.

The Isolation of Keratinophilic Fungi from Soils in Chon-ju Area (전주근교의 토양에서 분리된 호각화질성 진균의 오염에 관한 조사)

  • The purpose of this study was to investigate the contamination rates of Keratinophilic fungi in soils In Chonju. From september to November 1986, 98 soil samples were collected at the school and children's play ground, dog barn, pig barn, cow barn, horse barn and fowl barn( total 33 sites ). The samples were collected at the different depths(0~2cm, 30cm, 50cm ) in each of the sites, respectively. Each sample was cultured at 25$^{\circ}C$ according to Modified hair baiting method by using horse mane hair as a bait. The results obtained were summerized as follows ; 1. Eighty one of the 98 soil samples were found to be positive for Keratinophilic fungi. 2. In the examination of keratinophilic fungi in 98 soil samples, the organisms isolated weie as follows; Microsporum gypseum 20, Trichophyton ajelloi 15, Chrysosporium tropicum 40, C. Keratinophilum 24, C. tuberculatum 6 and Chrysosporium spp. like organism 4 strains. 3. The positive rates of keratinophilic fungi in each of the depths were 87.9%(29/33) in surface layer, 90.9%(30/33) in middle layer and 68.8%(22/32) in deep layer. 4. The positive rates of Keratinophilic fungi in each of the sites were 100%(3/3) in horse barn 91.7% (l1/12) in dog barn, 88.9%(16/18) in pig barn, 86.7%(13/15) in cow barn, 76.2%(16/21) in fowl barn and 75.9%(22/29) in school and children's play ground. 5. The isolation rates of M. gypseum, Pathogenic fungi for human and animals, were as follows ; 58.3% in dog barn(surface layers 100%, middle layers 50%, deep layers 25%) and 16.7% in pig barn(surface 33.3%, middle 16.7%, deep 0%). In the cow barn, the isolation rates were 13.3%(surface 40.0%, middle 0%, deep 0%) and 10% in horse barn(surface l00%, middle 100%, depp 100%). In the fowl barn, the isolation rates was 19.1%(surface 28.8%, middle 14.3%, deep 14.3%) and 3.5% in school and children's play ground(surface 0%, middle 0%, deep 11.1%).

Analysis of W-CDMA System with Smart Antenna for Different Bandwidths in Wideband Multipath Channel (광대역 다중경로 채널에서 스마트 안테나를 적용한 W-CDMA 시스템의 대역폭에 따른 성능분석)

  • In this paper, the performance of DS-CDMA system with smart antenna is analyzed for different bandwidths (1.25MHz,5MHz) and different channel environments (rural, urban) in wideband multipath channel. For the analysis of smart antenna system, the vector channel having the spatio-temporal correlation is modeled as a time-variant linear filter in time, and each multipath is assumed as a reflective wave from only one direction (only one cluster) in space. Several multipath is within one chip are distingushed into each one and the strongest signal is selected, DS-CDMA system with smart antenna using wider bandwidth present better performance than that using narrow bandwidth. It is shown that the smart antenna is more effective in urban area when using 2D-RAKE receiver.
