Isolation and Characteristics of Microorganisms Producing Extracellular Enzymes from Jeju Traditional Fermented Soybean Paste (Doenjang) (제주전통된장으로부터 세포외효소 분비능이 우수한 미생물의 분리 및 특성)

  • Bacteria strains with high activities of extracellular enzymes (protease, fibrinolytic enzyme, amylase, cellulase, and lipase) were isolated from Jeju traditional fermented soybean paste (Doenjang), and characterized by 16S rRNA gene sequence analysis and physiological properties. Protease activities were higher in JR14, JR19, JR25, JR32, JR38, JR47, and JR64 than Bacillus subtilis KCCM 12027 (standard strain). Amylase activities were shown in JR6, JR25, JR38, JR56 and JR81, while not in KCCM12027. Cellulase activities were higher in JR6, JR14, JR48, and JR65 than those of other isolated strains and KCCM 12027 whereas lipase activities were the higher in JR-14 and JR-48. Thrombolytic activity in JR19 with high hemolysis activity were 192% compared with that of plasmin as a positive control. Zymogram analysis indicated that the thrombolytic active strains had 4~5 bands in the molecular weight range of 25~75 kDa. Gene sequence analysis of rRNA revealed that the isolated stains had 99% homology with Bacillus species, and the thrombolytic active stain JR19 was B. stratosphericus $41KF2a^T$.

  • We isolated microorganisms presenting high enzymatic activities for amylase, cellulase, protease, lipase or fibrinolysis from Korean traditional soybean sauce and paste. Then, the physiological properties and 16S rRNA sequences of isolated microorganisms were analyzed. All of the isolated 13 strains possessing high extra cellular enzyme activities have higher amylase and cellulase activities than Bacillus subtilis KACC 10114. All the selected strains have protease activities except for D2-14. Except D8-8 and K4-1, other strains have lipase activity. D2-7, D8-8 and K4-1 strains have higher fibrinolytic activities than others, while D8-2 strain has no activity. Most of the selected strains showed antibacterial activity even in gram positive and gram negative bacteria and yeast. Gene sequence analysis of 16S rRNA from isolated strains revealed that all the selected strains were member of Bacillus species.

  • The aim of the research is to provide the boundary conditions for segregation of normal strength grade and high fluidity concrete mixture (so called mid-fluidity concrete) with rheology parameters. Since the normal strength grade concrete mixture has a relatively high water-to-cement ratio and no SCMs, it is easy to be segregated when superplasticizer is added. Hence, to achieve the mid-fluidity concrete of normal strength grade and high fluidity, preventing segregation of the mixture is inevitable. In this research, using two superplasticizers with different solid concentrations, the flow behaviors and rheological behaviors were assessed by increasing fluidity until the segregation happened. According to the experiment in this research, an unusual behavior in rheology parameters was observed when the concrete mixture started to be segregated. From this results and report, it is expected to contribute on the definition of segregation with rheological test methods.

  • 제올라이트는 다양한 유기질 분리의 촉매제 및 광학, 화학 센서, 기체 분리 등의 고기능 소재로서 크게 주목받고 있으며, 그 중 MFI type(ZSM-5, Silicalite-1) 제올라이트는 주로 석유화학공정에 주로 이용되고 있고, 분리막으로서 이산화탄소의 분리/회수 및 물/유기 혼합물의 분리 등에 대한 연구가 활발히 진행되고 있다. 본 연구에서는 분리막 제조에 유리할 것으로 판단되어지는 적합한 크기와 형상을 갖춘 나노크기의 제올라이트 분말을 수열합성법과 마이크로웨이브 합성법을 이용하여 합성하였으며, 위의 조건으로 다공질 알루미나 지지 체 위에서 알루미나/제올라이트 분리막 제조를 하는데 성공하였다. 또 한 다양한 조건(시간, 온도, 조성)에 따른 막의 두께변화와 균열발생정도를 관찰한 결과 합성시간, 건조온도에 따라서 같은 조건의 분리막 사이에서도 현격한 차이가 나타남을 알 수 있었다. 얻어진 MFI type제올라이트 분말과 분리막은 XRD, SEM, BET, TGA, FT-IR등의 분석수단을 이용하여 물성평가를 실시하였다.

Separation of Carbon Dioxide Using Pelletized Zeolite Adsorbent with Amine Impregration (아민 함침 성형 제올라이트 흡착제를 이용한 이산화탄소 분리)

  • In order to separate of carbon dioxide in the combustion exhaust gas, monoethanolamine (MEA) and piperazine (PZ) impregnated zeolite 13X adsorbents were used. A series of adsorbents were synthesized by impregnating 30, 50 and 70 wt% of MEA and PZ respectively on zeolite 13X pellet. XRD, FT-IR and BET were used to characterize the properties of impregnated pelletized zeolite 13X absorbents. In order to investigate the separation characteristics of carbon dioxide, zeolite pellet, MEA impregnated zeolite and PZ impregnated zeolite pellet were investigated at 25, 50 and $75^{\circ}C$. Amine impregnated pelletized zeolite absorbent has shown that adsorption decreases with increasing temperature. Finally, the carbon dioxide adsorption capacity when emission temperature of the combustion exhaust gas, PZ impregnated zeolite pellet was 1.8 times than zeolite pellet as well as 20 times than MEA impregnated zeolite pellet.

The Reason for the Loss of Photosynthetic Activity in Isolated Spinach Chloroplasts during Photosynthesis (분리된 시금치 엽록체에서 광합성중 광합성능이 소실되는 이유)

  • $ CO_2$ fixation of isolated intact spinach chloroplasts under saturating light began to decrease after 20 min, and stopped completely after 1 h. To identify the lesion sites for the die off, reconstituted chloroplast system was used with chloroplasts collected at several phases of time course. $ CO_2$ fixation was inhibited in the reconstituted chloroplasts made of thylakoids and stroma in the later phases, but showed a higher degree of inhibition by the participation of thylakoids than that of stroma in the later phases. Measurement of photophosphrylation and NADP reduction revealed that a severed thylakoidal damage was occurred at the later phases. This results indicate that the lesion sites for the die off are in the thylakoid.the thylakoid.

  • five strains of Streptomyces spp. with high Productivity of glucose isomerase (15-30 units/ml) were obtained among 280 microbial strains isolated from 150 soil samples. These strains produced glucose isomerase with xylose as an inducer. These 5 strains were also identified to be different strains of Streptomyces spp.:streptomyces sp. K-14, K-53, K-71, K-77 and K-733. It was found that Streptomyces sp. K-14 produced the highest enzyme activity. The spore chains of these strains were rectiflexible and spore surface was smooth except Steptomyces sp. K-77 and K-733, with spiny surface.

퇴비에서 분리한 우모(牛毛) 분해균 Bacillus pumilis RS7에 의한 우모분해산물이 식물성장에 미치는 영향

  • 전 세계적으로 방대하게 배출되는 도축 부산물인 우모는 높은 영양적 가치를 가짐에도 불구하고 현재 사용하는 물리화학적 처리법의 여러 단점으로 효율적으로 이용되지 못하고 있는 실정이다. 그 결과 2차 오염을 발생시키지 않는 경제적 방법인 생물학적 처리에 대한 연구의 필요성이 커지고 있다. 본 연구에서는 그러한 방법 중 대표적인 방법으로 미생물이 생산하는 keratinase를 이용한 생분해에 대해 연구를 하기 위해 경남 일대 퇴비화 볏짚에서 keratinolytic protease 생성능이 우수한 균주인 RS7을 분리하였고 생화학적 동정법과 16S rDNA를 이용한 동정결과 B. pumilus로 동정되었다. 본 실험에서 분리된 B. pumilus는 기존에 활발히 연구되어 있지 않는 균주로써 native feather 분해도가 기존에 알려진 Bacillus 속들이 3일 정도에서 분해 완료되는 것과는 달리 36시간 ${\sim}$48시간 내에 깃대까지 완전히 분해하였다. 또한 분리 균주의 경제적인 분해능을 토대로 분해산물이 식물 생장에 주는 영향과 아미노산 함유량을 검토한 결과 기존의 화학적 분해에 의한 분해산물보다 아미노산 함유량이 4배가량 많았으며 식물 생장에 있어서도 생장율과 개화시점으로 미루어볼 때 비료로써의 역할을 수행할 수 있었다.

Isolation of the Microbes Having Cyanobacteria Lytic Activity from Blooming Reservoirs (수화발생 저수지로부터 남조류 분해능을 가지는 미생물의 분리)

  • We have from water samples of Kwalim, Dochang, and Mulwang reservoirs in Kyonggi-Do, where cyanobacteria blooming occurred. Isolated microbes which have lytic activity for cyanobacteria. Water samples were smeared on the Anabaena cylindrica lawn and incubated in light chamber at $28^\circ{C}$, under 3000 lux for 13 days. A fungus having cyanobacterial lytic activity was isolated from the samples of Dochang reservoir. The isolate was identified as Cryptococcus laurentii by Vitek system. From the culture of the isolate, four major extracellular protein bands (29, 35.2, 40.9, 51.1 kDa) have been detected and the 29 kDa protein band was more thickly appeared in the culture with cyanobacteria.
