Dipteran Comparison on Carcasses by Decomposition at Different Abandoned Site (사체의 유기장소별 부패진행에 따른 발생 파리류 비교)

  • Because insect visitors was different depending on decomposition stage, dipterans occurred on chicken carcasses depending on postmortem intervals were compared by abandoned sites and decomposition from a serial study on arthropod occurrence from carcasses as forensic indicator. Species occurrence and decomposition of carcasses were different depending on abandoned sites (forest hill, open field, stream, greenhouse and roof), seasons (spring, summer, fall and winter), and situation (burying and non-burying). Development of carcass decomposition was faster at all sites in summer, carcass in greenhouse in winter, and unburied carcasses. Although visiting time of blow flies (Calliphoridae) (Phaenicia sericata, Lucilia illustris, Lucilia sp., Chrysomyia pinguis, and Chrysomyia megacephala) was different depending on abandoned season, their adults were generally collected from carcasses within 2 days. However, there were no visited flies at fresh stage of buried carcasses. The flesh flies (Sarcophagidae) were collected from all sites and seasons, and much faster from unburied carcasses than buried carcasses. Those were collected earlier from carcasses in greenhouse than other sites and occurrence was also shorter. In greenhouse, occurrence time of flesh flies were different depending on season; spring and summer - from fresh to active decay stage, fall - fresh to active decay stage, and winter - advanced decay to remains stage. Calliphora lata, Tricerotopyga calliphoroides, and Aldrichana grahami were dominant species and occurrences were different from other flies. These flies were active mainly from fresh to active stage. Larvae of sarcophagid flies were occurred earlier than those of calliphorid flies on buried carcasses.

생약재의 식품부패 세균에 대한 항균활성

  • 본 연구에서는 식품의 부패에 관여하는 미생물인 Escherichia coli, Pseudomonas aeruginosa, Bacillus subtilis, Staphylococcus aureu, Salmonella typhimurium Lactobacillus plantarum에 항균 활성을 나타내는 생약제의 효능을 살펴보았다. 금산 약초시장에서 구입한 생약재로부터 물 추출물과 70%에탄을 추출물을 제조하여 6종의 세균에 대해 항균활성을 탐색한 결과, 생약재 물 추출물의 경우에 가자 추출물은 E. coli, P. aeruginosa B. subtilis, S. aureus, S. typhimurium, L. plantarum의 6가지 균종에 대해서 모두 높은 항균활성을 나타내었으며, 복분자와 지유의 물추출물에서는 L. plantarum의 균종을 제외한 5가지 균종에 대해 높은 항균 활성을 나타내었다. 또한 소목 과 황련의 추출물에서는 E. coli를 제외한 5가지 균종에서 항균활성을 나타내었으며, 정향 추출물에서는 E. coli균주에 대해서는 약한 항균활성을 나타내었으나, B. subtilis, P. aeruginosa, S. aureus, S. typhimurium의 4종의 균주에 대해서는 우수한 항균활성을 나타내었다. 이 외에 감초 추출물은 P. aeruginosa, 석곡 추출물은 S. aureus, S. typhimurium, 계피 추출물은 S. typhimurium, 산수유 추출물은 B. subtilis, S. aureus, 파고지는 S. aureus균종에 대해서 항균활성을 나타내었다. 70% ethanol 추출물은 대부분의 생약재에서 높은 항균 활성을 나타내었다. 특히 가자, 감초, 강황, 복분자, 석곡, 소목, 오미자, 작약 ,정향, 지유, 파고지, 호장근, 황련 의 13가지 생약재 추출물에서 높은 항균활성을 나타내었다. 가자의 70% ethanol추출물의 경우는 물 추출물의 결과에서 같이 6가지 균종에 대해 모두 높은 항균활성을 나타냈으며, 복분자, 작약, 정향, 지유의 70% ethanol 추출물은 L. plantarum균을 제외한 5가지 균종에 대해 높은 항균 활성을 나타내었다. 소목의 70% ethanol 추출물의 경우에는 E. coli 균을 제외한 5가지 균종에서 높은 항균 활성을 보였으며, 이외에 생약재는 강황, 석곡, 황련, 호장근의 70% ethanol 추출물은 4가지의 균주에 대해서 항균 활성을 나타내었다. 또한 파고지, 오미자의 70% ethanol 추출물은 3가지 균주에 대해서 항균활성을 나타냈으며, 천궁, 육두구, 오수유, 갈근, 계피, 가시오가피의 70% ethanol 추출물은 2가지 균주에 대해 항균 활성을 나타내었다. 항균활성이 우수한 생약재를 농도별로 활성을 조사한 결과, 물 추출물과 10% Ethanol 추출물 모두 낮은 농도에서도 우수한 항균활성을 나타내었다.

생약재의 식품부패 곰팡이에 대한 항균활성

  • 본 연구에서는 생약재를 사용하여 부패에 관여하는 5종의 곰팡이(Aspergillus niger, Mucor miehei, Penicillium rugullosum, Aspergillus oryzae, Trichoderma reesei)에 대한 항 곰팡이 활성을 탐색하였다. 생약재의 물 추출물과 70% ethanol 추출물을 사용하여 5종의 곰팡이에 대한 항 곰팡이 활성을 측정한 결과, 물 추출물의 경우 갈근 추출물과 황련추출물에서 항 곰팡이 활성이 우수한 것으로 나타났다. 갈근 물 추출물은 A. niger, M. miehei, P. rugullosum의 3가지 균종에 대해서 모두 높은 항 곰팡이 활성을 나타냈으며, 황련 물 추출물은 M. miehei의 균종에 대해서 높은 항 곰팡이 활성을 나타내었다. 70% ethanol 추출물의 항 곰팡이 실험 결과, 물 추출물보다 좋은 항 곰팡이 활성을 나타내었다. 특히, 계피의 70% ethanol 추출물에서는 A. niger, M. miehei, P. rugullosum, A. oryzae, T reesei의 5가지 균종에서 모두 우수한 항 곰팡이 활성을 나타냈으며, 이 외에도 감초의 70% ethanol 추출물에서는 A. niger균주에 대해, 육두구와 호장근 및 황련의 70% ethanol 추출물에서는 M miehei 균주에 대해 우수한 항 곰팡이 활성을 나타냈으며, 또한 초두구의 70% ethanol추출물에서는 T reesei 균주에 대해 좋은 항 곰팡이 활성을 나타내었다. 항 곰팡이 효능이 우수한 생약재를 선별하고 이들 생약재로부터 추출한 다양한 획분을 사용하여 농도별로 항 곰팡이 활성을 조사한 결과, 갈근의 물 추출물의 경우에는 4가지 균종에서 모두 높은 항균활성을 나타냈으며, 낮은 농도에서도 높은 항 곰팡이 활성을 나타내었다. 이 외에도 황련의 물 추출물은 M. miehei 균주에서 우수한 항 곰팡이 활성을 나타내었다. 생약재의 70% Ethanol추출물을 농도별로 제조하여 항 곰팡이 활성을 살펴 본 결과, 70% ethanol추출물에서는 계피와 파고지, 초두구, 황련이 항 곰팡이 활성이 우수하였으며, 특히 계피의 70% ethanol 추출물에서는 5가지 균종에서 모두 우수한 항 곰팡이 활성이 나타났다.

Emission characteristics of odor from salted food materials using Spam (염처리 음식물의 냄새성분 배출특성에 대한 연구: 스팸을 중심으로)

  • In this study, the emission characteristics of volatile and odorant species released from salted meat product (Spam) was investigated as a function of time. Gas samples released from Spam samples were analyzed for volatile organic compounds (VOC) and sulfur compounds (RSC) at five different times for the elapsed (E) days of 0, 1, 3, 6, and 9 (E-0 to E-9) by GC/MS and GC/PFPD system, respectively. Results indicated that reduced sulfur, aldehyde, and ketone groups were the dominant odorants. Especially, hydrogen sulfide was the predominant in concentration and odor activity value (OAV) during the fresh stage. Its concentration was 1465 ${\mu}g/m^3$ (60.0% of the total mass) in E-0 and 455 ${\mu}g/m^3$ (28.0%) in E-1, while its OAV was 19.4 (78.3%: E-0) and 6.02 (41.7%: E-1). On the other hand, the concentration of acetone showed the maximum values in the decaying stage (E-3: 451 (43.2%), E-6: 369 (64.2%), and E-9: 1150 ${\mu}g/m^3$ (70.2%)). Furthermore, the concentration of 2,3-butanedione was also detected considerably from decaying sample (E-3: 17.6 (1.68%), E-6: 16.1 (2.80%), and E-9: 179 ${\mu}g/m^3$ (10.9%)). However, OAV of acetone was insignificant (<0.01%) in the decaying stage, while that of 2,3-butanedione was relatively high in the range of 1.14-11.6 (14.5-76.2% of ${\Sigma}OAV$). It thus confirmed that the major odorant groups generated from Spam samples changed with the progress of decay such as sulfur (fresh stage), aldehyde (intermediate stage), and ketone compounds (decaying stage).

Processing of Low Salted and Fermented Shrimp Using Gamma Irradiation Before Optimum Fermentation (새우젓 저염화를 위한 최적 숙성직전의 감마선 조사)

  • Gamma irradiation technology was applied to develop salted and fermented shrimp with lower salt concentration, high sensory quality and storage stability. Shrimp was prepared with 15 and 20% of salt and fermented at $15^{\circ}C$ for 10 weeks. The sample was irradiated at 0, 5.0 and 10.0 kGy right before optimum stage of fermentation. Fermented shrimp with 30% of salt concentration was also prepared as a control. The proximate composition, salinity and Aw were not affected by gamma irradiation. However, pH of irradiated samples was lower than that of non-irradiated samples, probably because irradiation effectively suppressed excessive fermentation by controlling microorganisms. From the results of sensory analysis, it was concluded that fermented shrimp with 15% of salt and irradiated at 10 kGy before optimum fermentation, or 20% of salt and 5 kGy or above were the most effective in terms of sensory quality and storage stability.

Antimicrobial Activity of Salvia miltiorrhiza Bunge Extract and Its Effects on Quality Characteristics in Sulgidduk (단삼 추출물의 향균 활성 및 첨가 설기떡의 품질 특성)

  • In this study, Salvia miltiorrhiza Bunge(Danshen) powder was extracted with ethanol, and its antimicrobial activity was investigated. The ethanol extract of the Danshen had antimicrobial activity against Bacillus subtilis, Escherichia coli, and Staphylococcus aureus. The inhibition zones of the Danshen extract(3 mg/disc) against B. subtilis, E. coli and S. aureus were 13, 12 and 8.5 mm, respectively. To test the food preservation effect of Danshen and determine the optimal ratio of the Danshen extract in a formulation, Sulgidduk samples were prepared with substitutions of 0, 0.25, 0.5, 0.75 and 1% Danshen extract, and then their quality characteristics were investigated over 4 days of storage. According to the results, total cell counts showed a decreasing trend with an increasing amount of added Danshen extract. Moisture contents were not significantly different among the Sulgidduk samples. As the content of Danshen extract increased, the L-values of samples decreased and the a- and b- values increased. For the textural characteristics, the hardness, gumminess, and chewiness of the Sulgidduk samples decreased as the amount of Danshen extract increased; however, they increased with the progression of storage time. In the sensory evaluation, the control group had significantly higher scores for color, flavor and after-taste as compared to the Danshen extract-added groups. With increasing Danshen extract contents, flavor and overall acceptability decreased, while Danshen flavor, bitterness and off-flavor increased. Chewiness was not significantly different among the samples. In conclusion, the results indicate that substituting 0.5% Danshen extract in Sulgidduk is optimal for quality and provides a product with reasonably high overall acceptability.

Antibacterial Activities of Trace Elements in Combination with Food Additives (미량원소 강화 식품소재의 항균효과)

  • Antibacterial activities of the trace elements in combination with the food additives were measured against 6 kinds of food-borne microorganisms such as Escherichia coli, Vibrio parahaemolyticus, Staphylococcus aureus, Bacillus cereus, Bacillus subtilis and Pseudomonas fluorescens. The difference of antibacterial activity was not shown among the kinds of food additives, such as dextrin, gelatin and collagen. Zn and Ge in combination with food additives had strong antibacterial effect. Especially, $1\%$ zinc acetate in combination with $1\%$ gelatin was more effective against P. fluorescens and S. aureus than against Bacillus sp., E. coli and V. parahaemolyticus. Minimum inhibitory concentration of zinc acetate in combination with $1\%$ gelatin appeared to be 0.5 mg/mL on S. aureus and P. fluorescens, and 1.0 mg/mL on E. coli, V. parahaemolyticus, B. cereus and B. subtilis. Minimum bactericidal concentration of zinc acetate in combination with $1\%$ gelatin appeared to be 0.5 mg/mL on P. fluorescens and 1.0 mg/mL on E. coli, V. parahaemolyticus, S. aureus, B. cereus and B. subtilis. The zinc acetate in combination with gelatin showed stronger inhibitory effect in acidic range below pH 6.0, and remained active even after heat treatment at $121^{\circ}C$ for 15 min. In comparison with control, the viable cell counts of fish pastes, which were coated with the solution containing both $1\%$ zinc acetate and $3\%$ gelatin, were decreased by more than 100-fold until the storage of 7 days at $10^{\circ}C$. The results indicate that the combined use of zinc acetate and some food additives could prolong the shelf life of fish pastes by 8 days or more at $10^{\circ}C$.

Effects of Zizyphus jujuba var. boeunesis Extracts on the Growth of Intestinal Microflora and Its Antioxidant Activities (대추 추출물이 장내 미생물의 생육에 미치는 영향 및 항산화 활성)

  • This study was carried out to investigate the effects of jujube extracts on intestinal microflora, along with their antioxidant activities, according to extraction method. The antimicrobial activities of the extracts were measured using the agar diffusion method with a jujube extract concentration of 50 mg/mL. Neither the first nor second jujube extracts were inhibitory against the tested intestinal bacteria. However, water extracts of jujube significantly enhanced the growth of lactic acid bacteria, especially Bifidobacterium bifidum and Bifidobacterium adolescentis. Total phenol compounds and flavonoid compounds were higher in the 1st than in the 2nd water extracts. The EDA values of both water and ethanol extracts increased in proportion to the extract concentration. The 1st water extract showed the highest value among all the others, which was 85.60% at the concentration of 0.05 mg/mL. Furthermore, the 1st water extract showed stronger antioxidant activity than the other samples with an activity of 679.91 mg AA eq/g. These results support the potential use of jujube water extracts as a functional food component and a valuable resource for the development of nutraceutical foods, to increase the growth of Bifidobacterium spp. in the human intestine.

Antimicrobial Activity of Paeonia japonica Extract and Its Quality Characteristic Effects in Sulgidduk (백작약 추출물의 항균효과 및 설기떡의 품질특성에 미치는 영향)

  • In this study, Paeonia japonica powder was extracted with ethanol, and its antimicrobial activity was investigated. The ethanol extract of the P. japonica had antimicrobial activity against Bacillus subtilis, Escherichia coli and Staphylococcus aureus. As the content of the P. japonica extract increased, the L-values of the samples decreased and the a- and b- values increased. In regards to the textural characteristics, the hardness, gumminess, and chewiness of the Sulgidduk samples decreased as the amount of P. japonica extract increased; however, they increased with the progression of storage time. Adhesiveness, springiness and cohesiveness were not significantly different at the different P. japonica extract concentrations and decreased with storage time. In the sensory evaluation, the control group had significantly higher scores for color, flavor and after taste as compared to the P. japonica extract added groups. When the P. japonica extract content was increased, the flavor and overall acceptability decreased, while Bakjakyak flavor, bitterness and off-flavor increased. Softness was not significantly different among the samples. In conclusion, the results indicate that substituting $0.25{\sim}1%$ P. japonica extract in Sulgidduk is optimal for quality and provides a product with reasonably high overall acceptability.

A New Purple Sweetpotato Cultivar for Table Use 'Yeonjami' (식용 자색고구마 신품종 '연자미')

  • 'Yeonjami' is a new sweetpotato variety developed for table use by Bioenergy Crop Research Center, National Institute of Crop Science (NICS), RDA in 2008. This variety was selected from the cross between 'Ayamurasakki' and 'Poly Cross' in 2000, and seedling and line selections were practiced from 2001 to 2003. Preliminary and advanced yield trials were carried out from 2004 to 2005. The regional yield trials were conducted at six locations from 2006 to 2008. 'Yeonjami' has cordate leaf, green vine and petiole, long elliptic storage root, purple skin and light purple flesh color of storage root. The average yield of storage root was 25.9 ton/ha in the regional yield trials, which was 15% higher than that of 'Sinjami' variety. Number of storage roots over 50 gram per plant was 2.9, and the average weight of one storage root was 153 gram. This variety was partly-resistant to Fusarium wilt and nematode. In addition, steamed 'Yeonjami' has higher polyphenol contents as 139.6 mg/100 g and higher biological activities as, which may improve the bioactivity in human.
