A Study for the Adaptation of Simulation of Uniaxial Compressive Strength Test for Concrete in 3-Dimensional Particle Bonded Model (3차원 입자 결합 모델에서 콘크리트의 일축압축실험 모사 적용성 연구)

  • In an uniaxial compressive test of a concrete standard specimen (150$\times$300 mm) the crack initiation and extension with the stress increase are the major reason of the failure, which is similar to the breakage of the particle bonding in the simulation by using particle bonded model, especially particle flow code in 3 dimensions (PFC3D) developed by Itasca Consulting Group Inc. That is the main motive to study the possibility of an uniaxial compressive strength test simulation. It is important to investigate the relationship between the micro-parameters and the macro-properties because the 3-dimensional particle bonded model uses the spherical particles to analyze the physical phenomena. Contact bonded model used herein has eight micro-parameters and there are five macro-properties; Young's modulus, Poisson's ratio, uniaxial compressive strength and the crack initiation stress and the ratio concerning the crack propagation with the stress. To simulate the compressive test we made quantitative relationships between the micro-parameters and the macro-properties by using the fractional factorial design and various sensitivity analyses including regression analysis, which result in the good agreement with the previous studies. Also, the stress-stain curve and the crack distribution over the specimen given by PFC3D showed the mechanical behavior of the concrete standard specimen under the uniaxial compression. It is concluded that the particle bonded model can be a good tool for the analyzing the mechanical behavior of concrete under the uniaxial compressive load.

The Effects of Calcium and Phenothiazine Derivatives on the Thermotropic Phase Transition of Acidic Phospholipid Bilayers (산성 인지질 이중층의 열적 상전이에 미치는 칼슘과 페노치아진 유도체의 영향)

  • The effects of phenothiazine derivatives and calcium on the thermotropic phase transition of bilayers in dipalmitoyl phosphatidylcholine (DPPC) and dipalmitoyl phosphatidic acid (DPPA) liposomes were investigated with differential scanning calorimeter (DSC). Bilayers underwent an abrupt organizational changes at a characteristic temperature when heated. Such temperature-dependent transition was particularly striking and sharp in the bilayers prepared from pure phospholipids. The ability of phenothiazine derivatives to modify the phase transition of phospholipids liposomes was measured by a broadening of the phase transition profile, that transition began to appear at lower temperature than which occurs in untreated liposomes. Calcium ion caused a large upward shift in the transition temperature of DPPC:DPPA (34:66mol%) liposomes. When the liposomes were first incubated with calcium ion followed by phenothiazine derivatives, disappearance of the broad curve centering at $73^{\circ}C$ indicated displacement of calcium ion by phenothiazine derivatives at the anionic site. It is supposed that calcium ion and phenothiazine derivatives might compete with each other on the head group of acidic phospholipid.

Fabrication and Characterization of Porous PLLA Scaffolds with Gentamicin Sulfate Release System (겐타마이신 설페이트를 서방화한 다공성 PLLA 지지체의 제조와 물성평가)

  • PLLA scaffold loaded with gentamicin sulfate (GS) was prepared by emulsion freeze-drying method for the prevention of infection and the improvement of wettability. i.e., the cell- and tissue-compatibility. GS-loaded PLLA scaffolds were characterized by scanning electron microscopy (SEM), mercury porosimetry and blue dye intrusion, and the GS release pattern was analyzed by high performance liquid chromatography (HPLC). GS-loaded PLLA scaffolds with porosity above 50%, medium pore size ranging from 30 to 57 ${\mu}{\textrm}{m}$ (with larger pore diameters greater than 150 ${\mu}{\textrm}{m}$), and specific pore area in the range of 35 to 75($m^2$ /g )were manufactured by varying processing parameter as GS concentration. It was observed that GS-loaded PLLA scaffolds were highly porous with good interconnections between pores for allowing cell adhesion and growth. These scaffolds may be applicable for scaffold as structures that facilitate either tissue regeneration or repair during reconstructive operations.

Characterization and Partial Purification of Storage Protein-i Receptor in the Fat Body of Hyphantria cunea (미국흰불나방 지방체에서 저장단백질-1의 수용체의 특성과 부분정제)

  • In vitro tissue culture of fat body of Hyphantria cunea in the medium containing [35S]-methionine reveaied that storage protein-i (SP-1) is taken up into fat body of prepupae and 1-day-old pupae. Using Western blotting and ligand binding method, we were able to identify the protein band of the SP-1 receptor protein. For the partial purification, the membrane proteins of fat body cells were solubilixed with 1% Triton X-1OO and applied to anion exchange chromatography. The results revealed the molecular weight of the receptor protein to be about 80 kl)a in SDSPAGE, and the P1 was estimated to be about 6.1. The mobility of the receptor protein in 8D8-PAGE was highly dependent on both temperature during electrophoresls and the condition of samples whether they were in reducing or nonreducing.

  • 서울대학교 병원에서는 약2년간의 이분야에 대한 기초 및 임상연구 결과를 토대로 하여 D.S.R.System을 개발하고 있다. 현재 개발하고 있는 Systme은 다음과 같다. 1) X선 발생장치 및 Control부분 2) 산란효과 제거를 위한 fore-slit 및 after-slit과 구동장치 3) X선을 광신호로 변환하기 위한 rare earth screen과 광신호를 전기적인 신호로 변환시키기 위한 photo-diode array detector의 결합체, 4) data 수집부분, 증폭장치와 A/D변환기, 고속 data 저장용 기억장치 부분 5) data 처리와 display를 위한 Bit-slice processor 5) Picture Archiving and Communication Systems (PACS) 이러한 부분 이외에 SNR을 향상시키고 좁은 부위의 region of interest를 보다 정확하게 관찰하기 위해서 511p/mm의 고밀도 선형검출기를 병행으로 사용하고 있다. 이 선형검출기를 이용하여 일부의 영상을 관찰하여 그 결과를 Wiener filter 등의 방법의 매개변수 선정에 이용하여 전체적인 영상의 질을 향상시키고자 하는 것이다.

  • Finite element vibration analysis of the trial 5x5 partial fuel assembly in the still water was performed using the substructure method. ANSYS software was used as a finite element modeling and modal analysis tool. The calculated natural frequencies of the partial fuel assembly were more consistent with the experimental results for the identical test model compared to the much larger solid model. This modeling technique can be utilized for the fuel assembly dynamic behavior analysis under normal operation, seismic and loss-of-coolant-accident analysis.

Physiology characteristics of genus Hypocrea isolated at oyster mushrooms of the shelf cultivation (균상재배 느타리버섯에서 분리한 Hypocrea속 균의 생리적 특성)

  • Hypocrea disease is the most severe disease of oyster mushroom cultivation in Korea. Physiological and ecological studies were performed on the pathogens (Hypocrea spp.) to obtain basic information for developing the integrated disease management system. Fourteen isolates of Hypocrea were collected from oyster mushroom house in five areas. Pathogenic fungi causing disease of oyster mushroom were identified as Hypocrea sp. based on morphological characteristics and pathogenicity. Two isolates (H-1, H-12) showed the fastest growth at $15^{\circ}C$ but four isolates (H-8, H-9, H-13, H-14) showed slower growth than those of other isolates at $20^{\circ}C$ and $25^{\circ}C$. Stroma with ascocarps and ascospore were produced on PDA under fluorescent light. The five isolates produced stroma with ascocarps and ascospores. Formation of fruiting body of strains H-14 of Hypocrea were the best out of all the strains on the potato dextrose agar (PDA). Also, fruiting bodies and ascospores were completely produced under fluorescent light. The growth of the isolates was correlated with total carbon content. The stroma of the isolates was formed mainly in histidine and asparagine treatment and especially in histidine-70 and asparagine-100 treatment. In the test of pathogenicity, after and before spawning showed very fast incidence of disease.
