Effects of Dandelion (Taraxacum coreanum) Extracts on the Mouse Liver with Acute Toxicated by Mercury Chloride (흰민들레(Taraxacum coreanum)추출물이 급성 수은 중독된 생쥐의 간에 미치는 효과)

  • Dandelion has been frequently used as a remedy for women's disease, inflammatory diseases and disorders of the liver and gallbladder. Dandelion extracts water extract, an herbal medication, may have an effect on the activity of hepatic antioxidant enzymes in diabetic rat. This study aims demonstrate the effect of dandelion extracts, one of the natural chelator, on the biochemical and enzyme activity changes in the mouse liver caused by $HgCl_2$. Mice approximately 30 gm in weight were grouped into the control, mercury chloride-treated, and the dandelion extracts-treated after mercury chloride groups. $HgCl_2$ (5 mg/kg) and dandelion extracts (3 g/kg) were delivered orally. Serum AST and ALT were measured, enzyme activity of liver were examined by spectrophotometer and ultrastructural alteration of liver were examined by light and electron microscopy. Dandelion extracts were decreased the increase of serum AST and ALT level induced by mercury. The catalase activity was decreased in the dandelion extracts group. The activity of SOD was dereased, but did not show significant differences. Mercury chloride-treated hepatic cell were irregular nucleus, enlarged and reduced number of mitochodria, enlarged rough endoplasmic reticulum, loss of ribosomes. Cells treated with dandelion extracts were similar to those of the control group. In conclusion, dandelion extracts may protect the mercury-induced toxicity on Liver.

In vivo Radioprotective Effects of Basic Fibroblast Growth Factor in C3H Mice (Basic Fibroblast Growth Factor (bFGF)의 방사선보호작용에 대한 실험적 연구)

  • Purpose : In order to understand in vivo radiation damage modifying of bFGF on jejunal mucosa, bone marrow and the effect of bFGF on the growth of transplanted mouse sarcoma 180 tumor in mice. Materials and Methods : Mice were treated with $6\;{\mu}g$ of bFGF at 24 hours and 4 hours before exposing to 600 cGy, 800 cGy and 1,000 cGy total body irradiation (TBI), and then exposed to 3,000 cGy local radiation therapy on the tumor bearing thigh. Survival and tumor growth curve were plotted in radiation alone group and combined group of bFGF and irradiation (RT). Histologic examination was performed in another experimental group. Experimental groups consisted of normal control, tumor control, RT (radiation therapy) alone, $6\;{\mu}g$ bFGF alone, combined group of $3\;{\mu}g$ bFGF and irradiation (RT), combined group of $6\;{\mu}g$ bFGF and irradiation (RT). Histologic examination was peformed with H-E staining in marrow, jejunal mucosa, lung and sarcoma 180 bearing tumor. Radiation induced apoptosis was determined in each group with the DNA terminal transferase nick-end labeling method ($ApopTag^{\circledR}$ S7100-kit, Intergen Co.) Results : The results were as follows 1) $6\;{\mu}g$ bFGF given before TBI significantly improved the survival of lethally irradiated mice. bFGF would protect against lethal bone marrow syndrome. 2) $6\;{\mu}g$ bFGF treated group showed a significant higher crypt depth and microvilli length than RT alone group (p<0.05). 3) The bone marrow of bFGF treated group showed less hypocellularity than radiation alone group on day 7 and 14 after TBI (p<0.05), and this protective effect was more evident in $6\;{\mu}g$ bFGF treated group than that of $3\;{\mu}g$ bFGF treated group. 4) bFGF protected against early radiation induced apoptosis in intestinal crypt cell but might have had no antiapoptotic effect in bone marrow stem cell and pulmonary endothelial cells. 5) There was no significant differences in tumor growth rate between tumor control and bFGF alone groups (p>0.05). 6) There were no significant differences in histopathologic findings of lung and mouse sarcoma 180 tumor between radiation alone group and bFGF treated group. Conclusions : Our results suggest that bFGF protects small bowel and bone marrow from acute radiation damage without promoting the inoculated tumor growth in C3H mice. Improved recovery of early responding normal tissue and reduced number of radiation induced apoptosis may be possible mechanism of radioprotective effect of bFGF.

Resveratrol Ameliorates NMDA-induced Mitochondrial Injury by Enhanced Expression of Heme Oxygenase-1 in HT-22 Neuronal Cells (NMDA를 처리한 HT-22 신경세포에서 미토콘드리아 손상을 완화하는 레스베라트롤의 보호 효과와 헴 산화효소-1의 역할)

  • N-methyl-D-aspartate (NMDA) receptors have received considerable attention regarding their involvement in glutamate-induced neuronal excitotoxicity. Resveratrol has been shown to exhibit neuroprotective effects against this kind of overactivation, but the underlying cellular mechanisms are not yet clearly understood. In this study, HT-22 neuronal cells were treated with NMDA in Mg2+-free buffer and subsequently used as an experimental model of glutamate excitotoxicity to elucidate the mechanisms of resveratrol-induced neuroprotection. We found that NMDA treatment causes a drop in MTT reduction ability, disrupts inside-negative transmembrane potential of mitochondria, depletes cellular ATP levels, and stimulates intracellular ROS production. Double fluorescence imaging studies demonstrated an increased formation of mitochondrial permeability transition (MPT) pores accompanied by apoptotic cell death, while cobalt protoporphyrin and bilirubin showed protective effects against NMDA-induced mitochondrial injury. On the other hand, zinc protoporphyrin IX significantly attenuated the protective effects of resveratrol which was itself shown to enhance heme oxygenase-1 (HO-1) mRNA and protein expression levels. In cells transfected with HO-1 small interfering RNA, resveratrol failed to suppress the NMDA-induced effects on MTT reduction ability and MPT pore formation. The present study suggests that resveratrol may prevent mitochondrial injury in NMDA- treated HT-22 cells and that enhanced expression of HO-1 is involved in the underlying cellular mechanism.

Effects of The pilose antler of Cervus Korean TEMMINCK var. mantchuricus Swinhoe(DAS), herbal acupuncture solution on suppression of collagenolysis and bone resorption in mouse calvarial osteoblasts (녹용약침액이 mouse의 두개골 골아세포에서 collagen용해와 골재흡수에 미치는 효과)

  • 본 실험에서는 녹용 약침액의 항 골재흡수 속성을 조사하였다. PTH, $1,25(OH)_2D_3$와 IL-1을 각각 골재흡수 인자로 사용하여 생쥐의 두개골에서 osteoblast 세포를 격리, 배양, 그리고 자극시켰을 때 collagenolysis의 증가를 보였다. 두 가지를 동시에 사용한 결과, IL-1은 골재흡수성을 촉진시키고 재 흡수력을 생산하였다. In vitro에서의 세포독성 결과는 $1-200{\mu}g/ml$의 녹용 약침액 농도 분포에서 무세포독성을 보였다. 또한 녹용 약침액은 생쥐의 두개골 골아세포 내에서 PTH (2 unit/ml), IL-$1{\alpha}$ (1 ng/ml), $1,25(OH)_2D_3$ (10 ng/ml), IL-$1{\alpha}$ 및 IL-$1{\beta}$로 인해 유발된 collagenolysis에 대해서 대항하는 보호활동성을 나타내었다. 녹용약침액은 IL-$1{\alpha}$ 와 IL-$1{\beta}$로 인해 유발된 collagenolysis에 대항하는 보호활동성을 지녔다. DAS는 IL-$1{\alpha}$와 IL-$1{\beta}$로 인해 촉진된 골재 흡수력을 억제하는 효과를 보였다. 이와 같은 결과는 녹용약침액이 골다공증과 연관된 질환에 대해서 매우 안정적인 임상적 사용이 가능한 것을 관찰할 수 있으므로 추후 이와 관련한 지속적인 연구가 필요할 것으로 사료되었다.

  • 수제는 하천에서의 흐름 방향과 유속을 제어하여 하안 또는 제방을 유수에 의한 침식작용으로부터 보호하기 위해 호안 또는 하안 전면부에 설치하는 구조물이다. 수제를 설치하는 목적은 하안 및 제방의 보호, 유로의 제어, 그리고 수환경의 개선 등을 들 수 있다. 국내에는 해방 이후 수제의 시공 사례가 거의 없었으나, 최근 들어 자연형 하천파 하천생태계 복원에 대한 관심이 점증하면서 수제에 대한 관심이 커지고 있다. 그러나 현재 국내에는 수제설계에 대한 실증적인 지침이 미비하고 국외의 기준을 검증 없이 소개하는 수준이어서 현장의 설계단계에서 수제공의 채택을 망설이게 하는 원인이 되고 있다. 이 연구에서는 국내${\cdot}$외 수제의 현황과 기술 동향을 파악하여 수제의 기하학적 특성인 수제길이, 수제길이, 그리고 설치각도의 3가지 설계인자를 조합하여 실험을 실시하였다. LSPIV 기법을 적용하여 측정한 유속자료를 분석한 결과 수제역 내 평균유속이 유입부 유속에 비해 $40\%$ 이하로 감소하는 것을 관찰할 수 있었다. 유속성분을 통하여 수제역 내 와도 및 사분면 검토를 실시한 결과 수제역 내에서는 비교적 안정적인 수치를 보이고 있어 호안침식방지에 유리한 것으로 보인다. 수제의 길이가 하폭의 $15\%$ 이하인 경우 상향 또는 하향수제가, $20\%$ 이상인 경우 직각수제가 유리한 것으로 나타났다.

Gastroprotective Effect of Korean Rice-Wine (Yakju) (한국 전통약주의 위보호 효과)

  • Gastroprotective effects of Korean rice-wine (Yakju) in two types of acute experimental gastric ulcer induced in rats and in mice were evaluated. Korean rice-wine were administered to 24-hr fasted rats 30 min before administration of 60% EtOH in 150 mM HCl or absolute ethanol. Korean rice-wine prevented formation of gastric ulcers induced by 60% EtOH in 150 mM HCl at oral doses of 250-1,000mg/kg and reduced gastric ulcers induced by absolute ethanol at oral doses of 62.5-1,000mg/kg, and inhibitory effect against 30% alcohol treatment for 7 days (twice/day). These results suggest Korean rice-wine have inhibitory effects on gastric lesion and ulceration.

  • 본 연구는 2005. 7. 1 시행 공포된 전파법 시행령 개정안 내용 중 전력선통신설비의 주파수 대역이 9kHz${\sim}$450kHz에서 그 상한선 범위가 30MHz까지 확대 시행됨과 관련하여 전력선통신설비가 단파대 무선통신에 혼신을 야기할 수 있음에 따라 그 영향여부를 평가하는 방법을 제시하고 향후 전력선통신설비를 운용함에 있어서 단파대 무선통신에 영향을 회피하기 위한 대책방안을 제안하는데 있다. 전력선통신의 운용주파수 확대와 관련하여 정보통신부 전파연구소에서 전력선통신이 단파대 무선통신의 혼신여부에 대하여 측정 관찰해 왔으며 그 간섭정도를 판단하기 위해 노력해 왔다. 본 연구에서는 전파수신기, 신호발생기 및 SINAD(Signal to Noise and Distortion) Meter를 사용한 측정방법을 제시하고 있으며 이는 무선(RF : Radio Frequency) 환경에 적합한 측정방법으로 기존의 EMC(electromagnetic compatibility) 환경에 의한 한계를 극복할 수 있다. 또한, 본 연구에서는 전력선통신설비가 단파대 해상이동통신에 영향을 최소화하기 위하여 보호구역을 설정 또는 해당 주파수에 대하여 운용금지하는 방안을 제시함으로써 무선통신환경을 보호하는데 그 목적이 있다.

  • 전 세계적으로 9.11로 대표되는 자살테러, 비행기테러, 화학가스 테러 등에 의한 인명피해가 늘고 있다. 1986년 김포공항 폭발사고, 1995년 음진리교 지하철 사린가스 사건, 2001년 9.11 사건, 2003년 필리핀 공항 폭탄테러 사건 등의 큰 테러 외에도 작은 사건은 무수히 많다. 이러한 테러 사태를 미연에 방지하기 위한 지능형 영상 보안 감시 시스템이 사회적으로 연구되어왔고 도입되어 왔다. 기존의 지능형 영상 보안 시스템은 모션 디렉션 기반으로 금지구역의 침입자로 판단되는 사람을 찾는 기능 중심으로 발전해왔다. 또한 보안요원의 관찰 모니터를 통해 화면을 계속 주시하는 한계 시간의 존재와 모니터 증가에 따른 한계 시간은 더 급감하게 되는 것으로 판단되었다. 이에 본 연구를 통해 유기물 탐지의 자동화를 추구하고, 보안 요원의 감시 업무를 도와 국가 주요 시설물의 유기물 자동 탐지를 통해 테러로부터 안전하게 보호하고자 한다.

The Effect of Career Group Counseling Program using Cinema on Self Identity and Career Decision-making Self-efficacy of Adolescent on Probation (영화를 활용한 진로 집단상담 프로그램이 보호관찰 청소년의 자아정체감과 진로결정 자기효능감에 미치는 효과)

  • Purpose: This study was to examine the effect of career group counseling program using cinema for the improving self identity and career decision-making self-efficacy of adolescents on probation. Methods: The subject were 36 (Experimental. 20, Control. 16) adolescents on probation who were referred to N mental health center in G city South Korea, but 2 of the experimental group did not complete the program. The program consisted of 8 sessions and was conducted from November, 2008 to February, 2009. The study variables were 'self identity' and 'career decision-making self-efficacy' surveyed by self-reported questionnaires. Results: Self identity (t=-2.797, p=.005), and career decision-making self-efficacy (t=-3.354, p=.001) changed more significantly in the experimental group than in control group after career group counseling program using cinema. Conclusion: Based on these results, career group counseling program using cinema for adolescents on probation was especially effective in increasing self identity and career decision-making self-efficacy.

Nursing Students' Experiences of Observing the Use of Physical Restraints: A Qualitative Study (간호대학생의 신체보호대 사용 관찰 경험: 질적 연구)

  • Purpose: The purpose of this study was to understand the experiences of final-year undergraduate nursing students in observing the use of physical restraints on patients in the course of clinical practice. Methods: Three focus group interviews were conducted with 12 Korean nursing students who could provide sufficient information about their observation of physical restraints. The collected data was analyzed through conventional content analysis. Results: The data were classified into four theme clusters, and nine themes. The four theme clusters included 'recognized as an unavoidable means,' 'experienced problems with the use of physical restraints,' 'realized the importance of the nurse's role and efforts,' and 'aspire to learn about correct use of physical restraints.' While nursing students recognized the necessities and problems of using physical restraints in clinical practice, and the importance of nurses' role and effort, the results found that education related to the use of physical restraints should be more systematic within the nursing curriculum. Conclusion: This study highlights the necessity of educating nursing students to ensure they acquire accurate knowledge and awareness regarding the use of physical restraints, and suggests the inclusion of systematic guidelines through simulation or extracurricular activities.
